Blockade of CD300A enhances the ability of human NK cells to lyse hematologic malignancies

Objective: The human cluster of differentiation(CD)300A, a type-i transmembrane protein with immunoreceptor tyrosine-based inhibitory motifs, was investigated as a potential immune checkpoint for human natural killer(NK) cells targeting hematologic malignancies(HMs).Methods: We implemented a stimulation system involving the CD300A ligand, phosphatidylserine(PS), exposed to the outer surface of malignant cells. Additionally, we utilized CD300A overexpression, a CD300A blocking system, and a xenotransplantation model to evaluate the impact of CD300A on NK cell efficacy against HMs in in vitro and in vivo settings. Furthermore, we explored the association between CD300A and HM progression in ***: Our findings indicated that PS hampers the function of NK cells. increased CD300A expression inhibited HM lysis by NK cells. CD300A overexpression shortened the survival of HM-xenografted mice by impairing transplanted NK cells. Blocking PS–CD300A signals with antibodies significantly amplified the expression of lysis function-related proteins and effector cytokines in NK cells, thereby augmenting the ability to lyse HMs. Clinically, heightened CD300A expression correlated with shorter survival and an “exhausted” phenotype of intratumoral NK cells in patients with HMs or solid ***: These results propose CD300A as a potential target for invigorating NK cell-based treatments against HMs.

Study on the effect and mechanism of Yanghe decoction Huacai(阳和汤化裁) on tissue repair of Yin syndrome after anal fistula surgery

OBJECTiVE: To observe the clinical efficacy and safety of Yanghe decoction Huacai(阳和汤化裁) for the repair of Yin syndrome wounds with slow-healing after anal fistula surgery. METHODS: A total of 120 patients with slow-healing negative wounds with after low-grade anal fistula surgery who met the inclusion criteria were divided into a treatment group and a control group based on a random number table method, with 60 patients in the treatment group and 60 patients in the control group. The treatment group was given Yanghe decoction Huacai in combination with routine treatment; the control group was only given routine treatment, in which the wound surface was disinfected with iodine, and then covered with sterile gauze. The course of treatment in both groups was 10 d. After treatment, the wound secretion score, wound granulation tissue score, the expression levels of basic fibroblast growth factor(bFGF), transforming growth factor β1(TGF-β1), and epidermal growth factor(EGF) in the wound, wound healing time and clinical efficacy were compared. RESULTS: There was no significant difference in age or gender between the two groups(P > 0.05). On the 10th and 15th days after the surgery, the wound secretion scores were higher in the treatment group than in the control group(P ing the two groups at the 10th and 15th day after surgery, the granulation tissue growth scores in the treatment group were better than the in control group(P ion, the expression levels of bFGF, TGF-β1 and EGF factors in the treatment group were stronger than those in the control group. A highly sensitive electrochemical sensing platform based on Zn-CuGaO@CMK-3 signal amplification for simultaneous detection of the sunset yellow and tartrazine in foods

in this work， a highly sensitive electrochemical sensor based on Zn-doped copper gallium oxide@ordered mesoporous carbon (Zn-CuGaO2@CMK-3) for signal amplification was successfully developed for the simultaneous detection of sunset yellow （SY） and tartrazine （TZ） in foods. Compared with CuGaO2@CMK-3， Zn-CuGaO2@CMK-3 offered enhanced conductivity and catalytic properties owing to the improved carrier density， which was beneficial to the electrooxidation of SY and TZ. Under the optimal testing conditions， the constructed Zn-CuGaO2@CMK-3/GCE sensor offered a wide linear concentration range （0.25 to 100.00 μmol/L） for the detection of both SY and TZ. The limits of detection for SY and TZ were 0.044 μmol/L and 0.059 μmol/L， respectively. Recovery experiments were performed in milk， white vinegar and biscuit samples， yielding satisfactory recoveries （82.70%-114.80%）. Furthermore， the sensor was successfully applied to the determination of the SY and TZ residues in two kinds of carbonated drinks， and the results were nearly consistent with those detected by the high performance liquid chromatography （HPLC） method （P＞0.05）.

Facile synthesis of magnetic covalent organic framework nanocomposites for the enrichment and quantification of trace organophosphorus pesticides in fruit juice

Organophosphorus pesticides (OPPs) in foods pose a serious threat to human health， motivating the development of novel analytical methods for their rapid detection and quantification. A magnetic covalent organic framework (M-COF) adsorbent for the magnetic solid-phase extraction (MSPE) of OPPs from foods was reported. M-COF was synthesized by the Schiff base condensation reaction of 1，3，5-tris (4-aminophenyl)benzene and 4，4-biphenyldicarboxaldehyde on the surface of amino-functionalized magnetic nanoparticles. Density–functional theory (DFT) calculations showed that adsorption of OPPs onto the surface of M-COF involved hydrophobic effects， Van der Waals interactions， π–π interactions， halogen-N bonding， and hydrogen bonding. Combined with gas chromatography–mass spectrometry (GC–MS) technology， the MSPE method features low limits of detection for OPPs (0.002–0.015 μg/L)， good reproducibility (1.45%–6.14%)， wide linear detection range (0.01–1 μg/L， R≥ 0.9935)， and satisfactory recoveries (87.3%–110.4%). The method was successfully applied for the trace analysis of OPPs in spiked fruit juices.
