Neuropeptide Y receptor Y8b(npy8br)regulates feeding and digestion in Japanese medaka(Oryzias latipes)larvae:evidence from gene knockout

Neuropeptide Y receptor Y8(NPY8R)is a fish-specific receptor with two subtypes,NPY8AR and *** in expression levels during physiological processes or in vivo regulation after ventricular injection suggest that NPY8BR plays an important role in feeding regulation;this has been found in only a few fish,at *** order to better understand the physiological function of npy8br,especially in digestion,we used clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated protein 9(Cas9)technology to generate npy8br-/-japanese medaka(Oryzias latipes).We found that the deletion of npy8br in medaka larvae affected their feeding and digestion ability,ultimately affecting their ***,npy8br deficiency in medaka larvae resulted in decreased feed intake and decreased expression levels of orexigenic genes(npy and agrp).npy8br-/-medaka larvae fed for 10 d(10th day of feeding)still had incompletely digested brine shrimp(Artemia nauplii)in the digestive tract 8 h after feeding,the messenger RNA(mRNA)expression levels of digestion-related genes(amy,lpl,ctra,and ctrb)were significantly decreased,and the activity of amylase,trypsin,and lipase also significantly *** deletion of npy8br in medaka larvae inhibited the growth and significantly decreased the expression of growth-related genes(gh and igf1).Hematoxylin and eosin(H&E)sections of intestinal tissue showed that npy8br-/-medaka larvae had damaged intestine,thinned intestinal wall,and shortened intestinal *** far,this is the first npy8br gene knockout model established in fish and the first demonstration that npy8br plays an important role in digestion.

Effect of ChREBP on carbohydrate feeding regulation in mandarin fish Siniperca chuatsi

Carnivorous fish have poor tolerance to carbohydrate in feed and low utilization rate of ***,the balance of carbohydrate and lipids in the nutrient metabolism of carnivorous fish,the ef fective conversion and utilization of carbohydrate and lipids,and the feedback regulation of feeding are the key links for the e fficient utilization of carnivorous fish *** response element binding protein(ChREBP)is a new transcription factor found in recent years in the glucose signaling pathway,and can also participate in feeding *** performed in-vivo and in-vitro experiments to reveal the role of ChREBP in the glucose metabolism and feeding in mandarin *** mRNA expression of ChREBP and appetite regulatory factors were measured after intraperitoneal injection of glucose in mandarin fish Siniperca chuatsi and cotransfection with glucose and glucose+siRNA in the hypothalamic cells in mandarin *** results reveal that at hour 2 and 4 post intraperitoneal injection with 1 mg/g BW glucose,the blood glucose level of the mandarin fish increased significantly,but food intake decreased significantly,and it also displayed a significantly increased ChREBP mRNA expression levels in *** hour 4 post injection,hypothalamic ChREBP mRNA level was significantly increased,whereas the mRNA expression levels of appetite genes neuropeptide Y(npy)and agouti-related peptide(AgRP)were decreased *** the glucose concentration was 40 mmol/L,the expression level of ChREBP mRNA in mandarin fish hypothalamic cells was significantly up-regulated,but the expression level of appetite gene npy mRNA was *** siRNA and glucose were co-transfected into mandarin fish brain cells,the expression level of chrebp mRNA was significantly decreased,and the appetite gene npy mRNA was significantly *** results indicated that glucose regulated food intake through the modulation of appetite gene npy by ChREBP.

Effects of dietary tert-butylhydroquinone on domoic acid metabolism and transcription of detoxification-related liver genes in red sea bream Pagrus major

Domoic acid (DA) is a neuroexcitatory amino acid that is produced by Pseudo-nitzschia during harmful algal blooms (HAB). Accumulation of DA can be transferred through food chain and cause neuronal damage in marine animal and in human. Like other algal toxins, DA was suggested to increase the oxidative stress and increase the detoxification-related gene expression in fish. The widely used food antioxidant, tert-butylhydroquinone (tBHQ), was known to induce a wide range of antioxidative potentials such as elevation of the glutathione levels and glutathione S-transferases (GSTs), via the activation of antioxidant response elements (AREs). In this study, the influences of dietary tBHQ on domoic acid (DA) metabolism and detoxification-related gene transcription were investigated both in vivo and in vitro. Oral administration of tBHQ resulted in significant decreases of DA accumulation of liver tissues in which red sea bream were fed with a single dose of 10 mg DA and 100 mg tBHQ per kg body weight per fish. Real-time PCR further revealed that the mRNA levels of AHR/ARNT/CYP1A1/GSTA1/GSTR were up-regulated in the above liver tissues at 72 h post tBHQ treatment. In consistence, tBHQ exposure also resulted in increased mRNA transcription of GSTA1, GSTA2 and GSTR in cultured red sea bream hepatocytes. Collectively, our findings in this research suggested that the dietary intake of tBHQ accelerated DA metabolism in fish, through mechanisms involving altered transcription of detoxification- related liver genes.

光强和光谱对鳜摄食、生长及相关基因表达的影响

为了探讨光强和光谱对鳜(Sinioerca chautsi)行为、摄食和生长性能的影响,对不同光强和光谱条件下养殖8周的鳜的摄食、生长进行观测，并检测相关基因的表达情况。对鳜摄食的影响研究发现,光强在10、25和50lx下时鳜的摄食比例较高,显著高于5、300和500 lx组(P0.05)。另外,10 lx光强下生长相关基因igf1在肌肉中的表达量显著高于其他组,同时促食欲基因npy和agrp显著上调(P0.05), 500 lx条件下pomc表达显著高于10和200 lx组(P<0.05)。绿光组的增重率和特定生长率显著升高(P<0.05),观察到生长性能的改善。在红光条件下观察到了相反的结果,增重率和特定生长率显著低于其他组(P<0.05),肝体比显著大于其他各组(P<0.05)。此外,红光和绿光组igf1在肝脏中的表达量显著高于蓝光组(P<0.05)。促食欲基因npy表达在绿光组中显著升高(P<0.05),agrp在红光、绿光和蓝光组中的表达量均显著低于白光组(P<0.05)。综上所述,鳜更加偏好低光强和短波长的光,并且在这种光照条件下摄食行为更加活跃且具有较好的生长性能。

Seasonal variation of gut Cyanophyta contents and liver GST expression of mud carp(Cirrhina molitorella) and Nile tilapia(Oreochromis niloticus) in the tropical Xiangang Reservoir(Huizhou,China)

Liver glutathione S-transferase(GST) plays a major role in the detoxification of microcystins(MCs) via conjugation to glutathione(GSH).We evaluated the relationship between seasonal variation in fish gut contents and the expression of GST isoforms in mud carp(Cirrhina molitorella) and Nile tilapia(Oreochromis niloticus).We quantified the abundance and diversity of plankton in the water column and foregut of mud carp and Nile tilapia in the tropical Xiangang Reservoir between October 2007 and July *** mRNA expression of 7 liver GST isoforms was determined by real-time *** gut contents of both species were dependent on the amount and type of phytoplankton and zooplankton in the *** expression of liver GST genes in Nile tilapia and mud carp was positively and negatively correlated,respectively,with the abundance of toxic cyanobacteria in the *** expression of liver GST mRNA was correlated to the abundance of toxic cyanobacteria in the gut contents of both species,suggesting that mRNA expression of GST isoforms could be used as a biomarker in Nile tilapia and mud carp to monitor cyanobacteria blooms in reservoirs.

Gluconeogenesis during starvation and refeeding phase is affected by previous dietary carbohydrates levels and a glucose stimuli during early life in Siberian sturgeon(Acipenser baerii)

Gluconeogenesis responses was assessed during a short starvation period and subsequent refeeding in Siberian sturgeon(Acipenser baerii) previously fed different dietary carbohydrates levels and experienced to a glucose stimuli during early life. The sturgeon larvae were previously fed either a high glucose diet(G) or a low glucose diet(F) from the first feeding to yolk absorption(8 to 12 d post-hatching [dph]). Each group of fish was sub-divided into 2 treatments at 13 dph and was fed either a high-carbohydrate diet(H) or a low carbohydrate diet(L) until 20 wk. In the current study, the fish in 4 groups(GL, FL, GH and FH) were experienced to starvation for 21 d following by re-feeding of their corresponding diets for 21 *** were sampled at postprandial 6 and 24 h before starvation(P6 h and P24 h), starvation 7,14 and 21 d(S7, S14 and S21) and 1, 7,14 and 21 d during refeeding(R1, R7, R14 and R21). Plasma samples during refeeding were taken at P6 h at each time point. Glycaemia levels, liver and muscle glycogen contents,activities and mRNA levels of hepatic gluconeogenic enzymes were examined. We found that both dietary carbohydrate levels and early glucose stimuli significantly affected the metabolic responses to starvation and refeeding in Siberian sturgeon(P < 0.05). During prolonged starvation, Siberian sturgeon firstly mobilized the liver glycogen and then improved gluconeogenesis when the dietary carbohydrates were abundant, whereas preserved the liver glycogen stores at a stable level and more effectively promoted gluconeogenesis when the dietary carbohydrates are absent to maintain glucose *** refeeding, as most teleostean, Siberian sturgeon failed controlling the activities and mRNA levels of phosphoenolpyruvate carboxykinase cytosolic forms(PEPCK-C), fructose-1,6-bisphosphatase(FBPase),but particularly controlled phosphoenolpyruvate carboxykinase mitochondrial forms(PEPCK-M) activities and mRNA expression of glucose-6-phosphatase(G6 Pase,

Effects of dietary protein levels on the long-term growth response and fitting growth models of gibel carp(Carassius auratus gibelio)

A 41-wk growth trial was conducted to evaluate the effects of dietary protein levels on the long-term growth response and fitting growth models of gibel carp(Carassius auratus gibelio) with an initial body weight of 1.85 ± 0.17 g. The dietary protein levels were designed at 320(P32), 360(P36). 400(P40).and 440 g/kg(P44), respectively. The growth curves of the gibel carp for each group were fitted and analyzed with four nonlinear regression models(Gompertz. logistic. von Bertalanffy and Richards). The final body weights(mean ± SD) of the fish were 226 ± 6.231 ± 7.242 ± 2, and 236 ± 2 g for P32, P36, P40,and P44. respectively. Feed conversion ratio of P40 and P44 groups was significantly lower than that of P32 and P36 groups(P 0.05). The growth response of the gibel carp for each group was the best fitted by Richards model with the lowest Chi^2, residual sum of squares and residual variance, then Gompertz and von Bertalanffy growth models, but the logistic model did not fit the data well justified by Chi^2 values. The optimal protein level(400 g/kg) prolonged the stage of fast growth and predicted the highest asymptotic weight, which was close to the harvest size in practice.

为研究中枢神经对营养物的直接感知及外周组织的间接感知,以翘嘴鳜(Siniperca chuatsi)为试验对象,通过脑室注射和饲料缺乏方式探索组氨酸或缬氨酸对翘嘴鳜摄食的影响。脑室注射试验中,对照组注射磷酸盐缓冲液(PBS),试验组注射组氨酸或缬氨酸或同时注射一般性调控阻遏蛋白激酶2(general control non-derepressible-2,GCN2)的抑制剂GCN2iB,检测注射后1 h和4 h翘嘴鳜的摄食量变化。饲料缺乏试验中,对照组投喂氨基酸完全饲料,试验组投喂组氨酸或缬氨酸缺乏饲料,检测翘嘴鳜的摄食量、GCN2信号通路相关因子及食欲基因的变化。结果显示,与对照组相比,注射组氨酸或缬氨酸均显著降低了翘嘴鳜的摄食量;同时注射GCN2iB和组氨酸可使翘嘴鳜的摄食量恢复至正常水平。与对照组相比,投喂缺乏组氨酸或缬氨酸的饲料,翘嘴鳜的摄食量显著减少,且GCN2信号通路下游激活转录因子4(activating transcriptional factor 4,atf4)mRNA表达水平显著降低,下丘脑中食欲因子神经肽Y基因(neuropeptide Y,npy)的mRNA表达水平显著降低,但刺鼠相关蛋白(agouti-related protein,agrp)、促黑素细胞皮质素原(pro-opiomelanocortin,pomc)、可卡因-苯丙胺转录调节肽(amphetamine-regulated transcript,cart)基因的mRNA表达水平无显著变化。综上所述,组氨酸和缬氨酸通过脑室注射和饲料缺乏的方式均可抑制翘嘴鳜摄食,且翘嘴鳜摄食行为的变化与GCN2信号通路有关,饲料缺乏组氨酸或缬氨酸可能是通过下调促食欲因子npy基因的表达起到抑食作用。

翘嘴鳜pck1及t1r1基因DNA甲基化在其转食人工饲料中的作用

为探究翘嘴鳜(Sinipercachuatsi)磷酸烯醇式丙酮酸羧基酶(phosphoenolpyruvatecarboxykinase,PEPCK)及鲜味受体(taste receptor type 1 member 1, T1R1)基因DNA甲基化在其转食人工饲料中的作用,实验检测了易驯食(可转食人工饲料)及不易驯食翘嘴鳜(只接受活鱼)的脑、肝脏和尾鳍中pck1和t1r1基因的表达及启动子区域CpG岛的DNA甲基化水平。结果显示, pck1基因主要在肝脏中表达(P0.05);在易驯食组脑t1r1表达量显著低于不易驯食组(P0.05)。研究表明,pck1基因启动子区域的DNA甲基化可能通过调控pck1基因的表达,促进翘嘴鳜转食人工饲料;而t1r1基因的甲基化可能不是调控其表达以促进翘嘴鳜转食的主要方式。
