Four multi-catheterized lactating goats were used in a 4×4 Latin square experiment to investigate the responses of amino acid metabolism in portal-drained viscera(PDV),liver,and mammary glands to short-term varying supplies of methionine(Met).During the last 45 h in each experimental period,goats were fasted for 12 h and then abomasally infused with an amino acid(AA)mixture plus glucose for 33 h.Treatments consisted of graded removal of Met from an infused AA mixture to achieve Met content in the infusate of 100%(complete),60%,30%,or 0%that in casein.Graded Met removal decreased the pro-duction of milk,milk protein,lactose,and fat linearly whilst also decreasing arterial Met concentration linearly(Plinearly(P0.1)was maintained as Met supply declined.This was achieved through increased mammary affinity(Plinearly decreased mammary uptake-to-milk output ratios of Met(Plinearly(0.05linearly decreased circulating concentration of prolactin and linearly increased insulin concentration(Plinear increase in mammary blood flow.The observed decreases in milk protein production as Met supply decreased appear to be a result of regulatory events which may have been driven by decreased circulating prolactin,rather than as a result of decreased mammary Met uptake.

The mechanistic target of rapamycin complex 1(mTORC1)integrates various types of signal inputs,such as energy,growth factors,and amino acids to regulate cell growth and proliferation mainly through the 2 direct downstream targets,eukaryotic translation initiation factor 4 E-binding protein 1(4 EBP1)and ribosomal protein S6 kinase 1(S6 K1).Most of the signal arms upstream of mTORCl including energy status,stress signals,and growth factors converge on the tuberous sclerosis complex(TSC)-Ras homologue enriched in brain(Rheb)axis.Amino acids,however,are distinct from other signals and modulate mTORCl using a unique pathway.In recent years,the transmission mechanism of amino acid signals upstream of mTORCl has been gradually elucidated,and some sensors or signal transmission pathways for individual amino acids have also been discovered.With the help of these findings,we propose a general picture of recent advances,which demonstrates that various amino acids from lysosomes,cytoplasm,and Golgi are sensed by their respective sensors.These signals converge on mTORCl and form a huge and complicated signal network with multiple synergies,antagonisms,and feedback mechanisms.

网络虚拟化作为云计算的关键技术,虽然提高了资源的灵活性和效率,但也带来了性能优化的挑战。文章首先分析网络虚拟化的原理及其在云环境中的应用,其次提出针对性的优化策略,最后进行实验分析。实验结果表明,优化后的资源利用率、响应时间、网络延迟、吞吐量、数据传输速率均优于优化前,且高负载下网络能够稳定运行,无性能下降。

Relationship between FHIT Expression and HPV Status in Vulvar Squamous Cell Carcinomas

OBJECTIVE To investigate the relationship between fragilehistidine triad (FHIT) depletion and human papillomavirus (HPV)status in the vulvar squamous cell carcinomas (VSCC).METHODS Immunohistochemical method was used to detectthe expression of FHIT protein in 42 cases of VSCC and 10 casesof normal vulvar tissues. PCR was used to detect HPV infectionstatus. We analyzed the relationship of the expression of FHITprotein between the 2 groups: HPV positive and HPV negative, aswell as the clinically pathological characteristics.RESULTS The expression of FHIT was positive in all normal vul-var tissues, and 71.4% was depletion in VSCC (P < 0.01). Abnormalexpression of FHIT was significantly correlated with pathologicalgrade (P < 0.05). There was a significant difference between FHITdepletion and HPV infection based on the statistial analysis (P <0.05).CONCLUSION The FHIT depletion is related to occurrenceand development of VSCC, and the abnormal expression of FHITsignificantly correlates with HPV infection.

The goal of the current study is to prepare polyclonal antibody against bovine intestinal peptide transporter I (bPepTI) in order to develop assay for immunological assessment of protein levels. Antigenicity of the entire bPepTI was analyzed with DNAStar, and a fragment with high antigenicity (bPepTI ORF 1369 - 1695) was selected, cloned in pGEX-6p-1 vector, resulting in a recombinant plasmid GST-BP, which verified by double enzyme digestion and sequenced, the recombinant plasmid was introduced to BL21. Exogenous expression was induced by IPTG and validated by Western blot analysis. The recombinant protein was isolated, purified and used for production of antiserum in mice. The specificity of antiserum was evaluated with immunobloting and titer was estimated with ELISA. Results indicate that the antibody against bPepTI was produced. The optimal GST-BP antigen embedding concentration was 0.5 μg/ml. The optimal dilution was 1:400. An indirect ELISA assay indicates the effective dilution was 1:102400.

脂质代谢是动物机体能量转化代谢的重要生理过程,与畜禽生长发育、脂肪沉积等密切相关。白藜芦醇是一种广泛存在于自然界中的芪类多酚化合物,可通过腺苷酸活化蛋白激酶和过氧化物酶体增殖物激活受体等多条途径调节畜禽脂质代谢。目前的研究发现,白藜芦醇对畜禽不同类型脂肪组织的脂质代谢存在差异影响。本文综述了白藜芦醇调控脂质代谢的机制途径及在畜禽生产中的应用研究进展,以期为白藜芦醇的精准化开发与应用提供参考。

Lactobacillus adheres to intestinal epithelial cells and yeast fungus cells with the aid of adhesion factors expressed on its cell surface. To identify adhesion factors nature on the surface of Lactobacillus, an adhesion experiment was carried out by pre-treating the Lactobacillus supernatant with different concentrations of bovine serum albumin, trypsin and 100°C for 10min. Additionally, intestinal epithelial cells were treated with sodium iodate, trypsin and sugar inhibition tests to characterize the receptors in Lactobacillus that interact with intestinal epithelial cells. It was demonstrated that Lactobacillus adhesion ability was decreased (P Lactobacillus cells was identified as a D-mannose glycoprotein. This observation was confirmed after treating intestinal epithelial cells with sodium iodate and trypsin, and sugar inhibition tests. Wild type Lactobacillus can agglutinate yeast fungus cells but after being exposured to mannose, agglutination to yeast fungus cells is lost or reduced. Results from this study we also got that inactivated and live bacteria that similarly adhere to intestinal epithelial cells.

Adhesive ability was tested in seven Lactobacillus solated from the chicken digestive tract after cultivation with CaCo-2 cells (intestinal epithelial cells), MDCK (dog kidney) cells and CEF (chicken embryo fiber) cells. We noted the following important observations regarding the adhesive ability between different Lactobacillus strains and three cell types: the adhesive interaction between the SDnB7, SDnE1 and SDnA3 lactobacillus strains and CaCo-2cells was greater compared to controls, the adhesive effect between SDnB1 and CEF cells was also greater than controls and Lactobacillus showed only minimal adherence to MDCK cells. Incubation time also affected Lactobacillus adherence to CaCo-2cells: adhesive ability was optimal at 37°C when incubated for 2 days and this was confirmed by a local increase in the concentration of Lactobacillus around CaCo-2 cells when incubated for 24 h as opposed to 3 h. Adherence ability in lactobacillus was also tested at various concentrations (108, 107, 106, 105 and 104 ). The number of Lactobacillus that adhered around the cells was significantly increased in the treatment with 108 bacterial cells. Transmission electron microscopy revealed that the cellularity of the junction between CaCo-2cells and Lactobacillus was not compromised. Transmission electron microscopy also revealed that the thalli fabric structure remained intact.

For the experiment, 8 newborn male Holstein calves were selected that had the same feeding environment, and were of similar ages. They were randomly divided into 2 groups, with 4 in each group. The treatments consisted of feeding active probiotics (Group P) and a normal fed control group (Group C). The growth performance and blood indices were measured;rumen fluid samples were collected after weaning, and 16SrDNA sequencing and LC-MS metabolome detection were performed. Compared with the control group, relative abundances of Deltaproteobacteria, Desulfovibrionales, Bacteroidales_ BS11_gut_group, Desulfovibrionaceae, Bacteroidales_S24-7_group, Acetobacteraceae, Ruminococcaceae_NK4A214_group, Asaia, [Ruminococcus] gauvreauii_group, Desulfovibrio, Kingella, Selenomonas, Lachnoclostridium in group P were significantly different (P of 2-methylbenzoic acid and myo-inositol were significantly increased (P 0.05). These results showed that compared with normally fed calves, the growth performance and blood indices of probiotic-fed calves were changed, but the differences were not significant.
