Metabolic reprogramming: a new option for the treatment of spinal cord injury

Spinal cord injuries impose a notably economic burden on society,mainly because of the severe after-effects they cause.Despite the ongoing development of various therapies for spinal cord injuries,their effectiveness remains unsatisfactory.However,a deeper understanding of metabolism has opened up a new therapeutic opportunity in the form of metabolic reprogramming.In this review,we explore the metabolic changes that occur during spinal cord injuries,their consequences,and the therapeutic tools available for metabolic reprogramming.Normal spinal cord metabolism is characterized by independent cellular metabolism and intercellular metabolic coupling.However,spinal cord injury results in metabolic disorders that include disturbances in glucose metabolism,lipid metabolism,and mitochondrial dysfunction.These metabolic disturbances lead to corresponding pathological changes,including the failure of axonal regeneration,the accumulation of scarring,and the activation of microglia.To rescue spinal cord injury at the metabolic level,potential metabolic reprogramming approaches have emerged,including replenishing metabolic substrates,reconstituting metabolic couplings,and targeting mitochondrial therapies to alter cell fate.The available evidence suggests that metabolic reprogramming holds great promise as a next-generation approach for the treatment of spinal cord injury.To further advance the metabolic treatment of the spinal cord injury,future efforts should focus on a deeper understanding of neurometabolism,the development of more advanced metabolomics technologies,and the design of highly effective metabolic interventions.

Experimental Study of Temperature Control Based on Composite Phase Change Materials during Charging and Discharging of Battery

This study is to utilize the heat-absorbing and releasing capabilities of phase change materials(PCM)to regulate the surface temperature fluctuations of batteries during charging and discharging.The goal is to keep the battery within the optimal operating temperature range.The impact of PCM thickness and phase change temperature on battery temperature is investigated by encircling a cylindrical battery with a PCM ring.To improve the thermal conductivity of PCM,expanded graphite(EG) is added to make a composite phase change material(CPCM),and the effects of various EG mass ratios on battery surface temperature and CPCM utilization level are investigated.The findings indicate that increasing PCM thickness effectively extends temperature control time,but its impact is limited.The difference in phase change temperature of PCM controls the battery temperature in different temperature ranges.Lower phase change temperatures are unsuitable for controlling battery temperature in high temperature environments.The addition of EG enhances the thermal conductivity of PCM,leading to further control of battery temperature.The results show that the addition of 6%(mass ratio) EG to CPCM extends the effective temperature control time by 11 min and improves by 28% compared to a single PCM.The CPCM utilization is also more satisfactory and achieved a balance between heat storage and thermal conductivity in a battery thermal management system(BTMS) based on PCM.

A comparative study of data-driven battery capacity estimation based on partial charging curves

With its generality and practicality, the combination of partial charging curves and machine learning(ML) for battery capacity estimation has attracted widespread attention. However, a clear classification,fair comparison, and performance rationalization of these methods are lacking, due to the scattered existing studies. To address these issues, we develop 20 capacity estimation methods from three perspectives:charging sequence construction, input forms, and ML models. 22,582 charging curves are generated from 44 cells with different battery chemistry and operating conditions to validate the performance. Through comprehensive and unbiased comparison, the long short-term memory(LSTM) based neural network exhibits the best accuracy and robustness. Across all 6503 tested samples, the mean absolute percentage error(MAPE) for capacity estimation using LSTM is 0.61%, with a maximum error of only 3.94%. Even with the addition of 3 m V voltage noise or the extension of sampling intervals to 60 s, the average MAPE remains below 2%. Furthermore, the charging sequences are provided with physical explanations related to battery degradation to enhance confidence in their application. Recommendations for using other competitive methods are also presented. This work provides valuable insights and guidance for estimating battery capacity based on partial charging curves.

Studying the variable energy band structure for energy storage materials in charge/discharge process

So far,a clear understanding about the relationship of variable energy band structure with the corresponding charge-discharge process of energy storage materials is still lacking.Here,using optical spectroscopy(red-green-blue(RGB)value,reflectivity,transmittance,UV-vis,XPS,UPS)to studyα-Co(OH)_(2) electrode working in KOH electrolyte as the research object,we provide direct experimental evidence that:(1)The intercalation of OH-ions will reduce the valence/conduction band(VB and CB)and band gap energy(Eg)values;(2)The deintercalation of OH-ions corresponds with the reversion of VB,CB and E_(g) to the initial values;(3)The color of Co(OH)_(2) electrode also exhibit regular variations in RGB value during the charge-discharge process.

Reconstruction of osteochondral(OC)defects represents an immense challenge due to the need for synchronous regeneration of special stratified tissues.The revolutionary innovation of bioprinting provides a robust method for precise fabrication of tissue-engineered OCs with hierarchical structure;however,their spatial living cues for simultaneous fulfilment of osteogenesis and chondrogenesis to reconstruct the cartilage-bone interface of OC are underappreciated.Here,inspired by natural OC bilayer features,anisotropic bicellular living hydrogels(ABLHs)simultaneously embedding articular cartilage progenitor cells(ACPCs)and bone mesenchymal stem cells(BMSCs)in stratified layers were precisely fabricated via two-channel extrusion bioprinting.The optimum formulation of the 7%GelMA/3%AlgMA hydrogel bioink was demonstrated,with excellent printability at room temperature and maintained high cell viability.Moreover,the chondrogenic ability of ACPCs and the osteogenic ability of BMSCs were demonstrated in vitro,confirming the inherent differential spatial regulation of ABLHs.

Photoreceptor cell degeneration leads to blindness, for which there is currently no effective treatment. Our previous studies have shown that Lycium barbarum(L. barbarum) polysaccharide(LBP) protects degenerated photoreceptors in rd1, a transgenic mouse model of retinitis pigmentosa. L. barbarum glycopeptide(Lb GP) is an immunoreactive glycoprotein extracted from LBP. In this study, we investigated the potential protective effect of Lb GP on a chemically induced photoreceptor-degenerative mouse model. Wild-type mice received the following: oral administration of Lb GP as a protective pre-treatment on days 1–7;intraperitoneal administration of 40 mg/kg N-methylN-nitrosourea to induce photoreceptor injury on day 7;and continuation of orally administered Lb GP on days 8–14. Treatment with Lb GP increased photoreceptor survival and improved the structure of photoreceptors, retinal photoresponse, and visual behaviors of mice with photoreceptor degeneration. Lb GP was also found to partially inhibit the activation of microglia in N-methyl-N-nitrosourea-injured retinas and significantly decreased the expression of two pro-inflammatory cytokines. In conclusion, Lb GP effectively slowed the rate of photoreceptor degeneration in N-methyl-N-nitrosourea-injured mice, possibly through an anti-inflammatory mechanism, and has potential as a candidate drug for the clinical treatment of photoreceptor degeneration.

Yuzaoea gen. nov., a new biraphid diatom(Bacillariophyceae)genus and its phylogenetic significance

The flexed frustules in pennate diatoms are usually associated with monoraphid diatoms. Interestingly, we found a biraphid diatom species with flexed frustules in an offshore intertidal beach environment on Weizhou Island,Beihai City, Guangxi Zhuang Autonomous Region, China. Therefore, based on morphological characteristics, we described a new genus of diatoms Yuzaoea sinensis gen. et sp. nov. CH Li, HH Liu, YH Gao & CP Chen. The frustule of this genus is characterized by heterogeneous frustule with one concave valve and one convex valve,complete raphe on both valves, straight and moderately eccentric raphe, uniseriate striae and girdle bands with a single row of areolae. The most identifying feature of this genus was the flexed frustule, which is rare in biraphid diatoms and common in monoraphid diatoms. We compared the morphometric characteristics of genus Yuzaoea with genus Rhoikoneis and several genera within the family Rhoicospheniaceae, including Rhoicosphenia,Campylopyxis, and Cuneolus. Phylogenetic analyses based on SSU rRNA and rbc L showed that the genus Yuzaoea was the sister group to the clade of Rhoicosphenia with a high support value(bootstrap values = 100%), and the clade “Yuzaoea+Rhoicosphenia” was sister to the clade of monoraphid diatoms, in which the genera Achnanthidium, Planothidium and some Cocconeis with high support values(bootstrap = 100%).Morphologically, the genus Yuzaoea shares many morphological features with monoraphid diatoms like genera Achnanthidium and Planothidium and the members within the Rhoicospheniaceae. Therefore, based on a combined morphological studies and phylogenetic results we suggested that this branch may represented the evolution of one kind monoraphid diatoms, from biraphid diatoms(e.g. genus Yuzaoea), to incompleted biraphid diatoms(e.g. genera Rhoicosphenia, Campylopyxis), to monoraphid diatoms(e.g. genera Achnanthidium and Planothidium).

Chimeric antigen receptor(CAR)T-cell therapy has demonstrated clinical response in treating both hematologic malignancies and solid tumors.Although instances of rapid tumor remissions have been observed in animal models and clinical trials,tumor relapses occur with multiple therapeutic resistance mechanisms.Furthermore,while the mechanisms underlying the long-term therapeutic resistance are well-known,short-term adaptation remains less understood.However,more views shed light on short-term adaptation and hold that it provides an opportunity window for long-term resistance.In this study,we explore a previously unreported mechanism in which tumor cells employ trogocytosis to acquire CAR molecules from CAR-T cells,a reversal of previously documented processes.This mechanism results in the depletion of CAR molecules and subsequent CAR-T cell dysfunction,also leading to short-term antigen loss and antigen masking.Such type of intercellular communication is independent of CAR downstream signaling,CAR-T cell condition,target antigen,and tumor cell type.However,it is mainly dependent on antigen density and CAR sensitivity,and is associated with tumor cell cholesterol metabolism.Partial mitigation of this trogocytosis-induced CAR molecule transfer can be achieved by adaptively administering CAR-T cells with antigen density-individualized CAR sensitivities.Together,our study reveals a dynamic process of CAR molecule transfer and refining the framework of clinical CAR-T therapy for solid tumors.

Minimizing interfacial energy losses in inverted perovskite solar cells by a dipolar stereochemical 2D perovskite interface

Inverted perovskite solar cells(PSCs) have attracted broad research and industrial interest owing to their suppressed hysteresis,cost-effectiveness,and easy-fabrication.However,the issue of non-radiative recombination losses at the n-type interface between the perovskite and fullerene has impeded further improvement of photovoltaic performance.Here,we modify the n-type interface of FAPbI_(3) perovskite films by constructing a stereochemical two-dimensional(2D) perovskite interlayer,in which the organic cations comprise both pyridine and ammonium groups.The pyridine N donor can create stable bonding with the surface-uncoordinated Pb on the perovskite,thereby passivating the shallow-level defects and enhancing the air stability of the film.Furthermore,the pyridine N donor also offers a positive polar interface to decrease the surface work function of the perovskite film,enabling n-type modification.Ultimately,we employ a p-i-n photovoltaic(PV) device with the positive dipole interlayer at perovskite/fullerene contact and achieve remarkable photoelectric conversion efficiency(PCE) of 22.0%.

A Robust Superhydrophobic Smart Coating with Reversible Thermochromic and Photochromic Property

Both thermochromic and photochromic coating have attracted many attentions due to their widely applications,but the low stability is a big obstacle.Inspired by the lotus leaf,to endow the chromic coating with superhydrophobicity is a possible solution.In this research,a dual response coating was prepared by adding photochromic and thermochromic particles simultaneously.The prepared sample demonstrated at least four-state color switching,which can be successfully used in tactile imaging,multi-color fabric,erasable record,and security labels.The superhydrophobicity was achieved by introducing vinyl-terminated polydimethylsiloxane,which not only offers low surface energy but also can cross-link with the particles to increase the adhesion.Thus,the prepared sample maintained superhydrophobicity after various kinds of destruction(such as sandpaper abrasion,corrosive liquid attack,ultrasonic treatment,UV irradiation,and high-speed drops/turbulent jets impact).Even though the superhydrophobicity can be destroyed by plasma etching,it can be recovered after 12 h at room temperature.
