Cancer of unknown primary(CUP)is a rare disease characterized by metastases in which the primary tumor is of unknown origin.The cerebrospinal fluid(CSF)tumor microenvironment of CUP is still unknown.A Chinese male was diagnosed with leptomeningeal metastases from CUP(CUP-LM)based on the following medical examination results:partial leptomeningeal enhancement by brain magnetic resonance imaging,few malignant cells of diverse morphology in CSF,and no abnormalities or lymphadenopathy by systemic examination.

Metasurface-based imaging has attracted considerable attention owing to its compactness,multifunctionality,and subwavelength coding capability.With the integration of computational imaging techniques,researchers have actively explored the extended capabilities of metasurfaces,enabling a wide range of imaging methods.We present an overview of the recent progress in metasurface-based imaging techniques,focusing on the perspective of computational imaging.Specifically,we categorize and review existing metasurface-based imaging into three main groups,including(i)conventional metasurface design employing canonical methods,(ii)computation introduced independently in either the imaging process or postprocessing,and(iii)an end-to-end computation-optimized imaging system based upon metasurfaces.We highlight the advantages and challenges associated with each computational metasurface-based imaging technique and discuss the potential and future prospects of the computational boosted metaimager.

The regeneration of Tetracentron sinense Oliv.is poor in the understory and in open areas due to the charac-teristics of natural regeneration of the species on forest edges and in gaps.It is unclear whether different light intensities in various habitats affect eco-physiological characteristics of saplings and their natural regeneration.In this study,the light intensity in T.sinense habitats was simulated by artificial shading(L1:100%NS(natural sunlight)in the open;L2:50%NS in a forest gap or edge;L3:10%NS in understory)to investigate differences in morphology,leaf structure,physiology,and photosynthesis of 2-year-old sap-lings,and to analyze the mechanism of light intensity on sapling establishment.Significant differences were observed in morphology(including leaf area,and specific leaf area)under different light intensities.Compared to L1 and L3,chloroplast structure in L2 was intact.With increasing time,superoxide dismutase(SOD)and catalase(CAT)activities in L2 became gradually higher than under the other light intensities,while malondialdehyde(MDA)content was opposite.Shading decreased osmoregulation substance contents of leaves but increased chlorophyll.The results suggest that light intensities significantly affect the eco-physiological characteristics of T.sinense saplings and they would respond most favorably at intermediate levels of light by optimizing eco-physiological characteristics.Therefore,50%natural sunlight should be created to promote saplings establishment and population recovery of T.sinense during in situ conservation,including sowing mature seeds in forest edges or gaps and providing appropriate shade protection for seedlings and saplings in the open.

Geothermal energy utilization in China had been promoted actively during the 13^(th)Five-Year Plan period(2016-2020),with resultant achievements showing unique Chinese characteristics.Buildings(measured by areas)heated/cooled by intermediate-to-deep geothermal energy increased more rapidly than those by shallow geothermal energy.Geothermal power generation was restricted by the distribution of the resource and the sufficiency of local power supply.With the proposition of“dual carbon goal”by the central government of China and the coming era of great development of renewable energy,geothermal energy will be provided more development opportunities in room heating/cooling and power generation in the country ever than before.However,tackling technology bottlenecks and introduction of appropriate policy support are still urgently needed for nurturing a healthy environment for geothermal industry.Analysis suggests that a near-to medium-term development of geothermal energy shall be focused on a comprehensive and efficient utilization both through heating/cooling and power generation.Room heating/cooling by intermediate-to-deep and shallow geothermal energy shall be promoted nationwide according to local resource endowment and economic conditions.Geothermal power generation in Yunnan and Tibet shall be pushed forward in an orderly way.For medium-and long-term development,focus shall be put on power generation by using low to medium temperature geothermal energy and hot dry rocks.The"hot spring+"model shall be adapted in steps on a regular basis.To achieve these,measures including attaching importance to tackling key technologies,promoting standards for geothermal energy development and utilization,strengthening geothermal energy exploration and evaluation,and seeking for necessary policy support shall be taken.

Background:Since RNA sequencing has shown that induced pluripotent stem cells(iPSCs)share a common antigen profile with tumor cells,cancer vaccines that focus on iPSCs have made promising progress in recent years.Previously,we showed that iPSCs derived from leukemic cells of patients with primary T cell acute lymphoblastic leukemia(T-ALL)have a gene expression profile similar to that of T-ALL cell lines.Methods:Mice with T-ALL were treated with dendritic and T(DC-T)cells loaded with intact and complete antigens from T-ALL-derived iPSCs(T-ALL-iPSCs).We evaluated the safety and antitumor efficiency of autologous tumor-derived iPSC antigens by flow cytometry,cytokine release assay,acute toxicity experiments,long-term toxicity experiments,and other methods.Results:Our results indicate that complete tumor antigens from T-ALL-iPSCs could inhibit the growth of inoculated tumors in immunocompromised mice without causing acute and long-term toxicity.Conclusion:T-ALL-iPSC-based treatment is safe and can be used as a potential strategy for leukemia immunotherapy.

In the spread of SARS-C oV-2,there have been multiple waves of replacement between strains,each of which having a distinct set of mutations.The first wave is a group of four mutations(C241 T,C3037 T,C14408 T and A23403 G [this being the amino acid change D614 G];all designated 0 to 1 below).This DG(D614 G) group,fixed at the start of the pandemic,is the foundation of all subsequent waves of strains.Curiously,the DG group is absent in early Asian samples but present(and likely common) in Europe from the beginning.European data show that the high fitness of DG1111 requires the synergistic effect of all four mutations.However,the European strains would have had no time to evolve the four DG mutations(0 to1),had they come directly from the early Asian DG0000 strain.Very likely,the European DG1111 strain had acquired the highly adaptive DG mutations in pre-pandemic Europe and had been spreading in parallel with the Asian strains.Two recent reports further support this twin-beginning interpretation.There was a period of two-way spread between Asia and Europe but,by May 2020,the European strains had supplanted the Asian strains globally.This large-scale replacement of one set of mutations for another has since been replayed many times as COVID-19 progresses.

As survival rates improve and detection technologies advance,the occurrence of multiple primary cancers(MPCs)has been increasing.Approximately 16%of cancer survivors develop a subsequent malignancy,with lung cancer often developing after esophageal cancer due to potential“field cancerization”effects.Despite this observation,the genetic heterogeneity underlying MPCs remains understudied.However,the recent emergence of genetic testing has expanded the scope of investigations into MPCs to investigate signatures underlying cancer predisposition.This report reveals 3 unprecedented TP53 fusion mutations in a Chinese patient afflicted by MPCs,namely,AP1M2–TP53(A1;T11)fusion,TP53–ILF3(T10;I13)fusion,and SLC44A2–TP53(S5;T11)fusion.This patient exhibited an extended period of survival after diagnosis of extensive-stage small cell lung cancer,which occurred 6 years after the diagnosis of esophageal squamous cell cancer.This unique reportmay provide supplementary data that enhance our understanding of the genetic landscape ofMPCs.

The recently discovered type IX secretion system(T9SS)is limited to the Bacteroidetes phylum.Cytophaga hutchin-sonii,a member of the Bacteroidetes phylum widely spread in soil,has complete orthologs of T9SS components and many T9SS substrates.C.hutchinsonii can efficiently degrade crystalline cellulose using a novel strategy,in which bacterial cells must be in direct contact with cellulose.It can rapidly glide over surfaces via unclear mech-anisms.Studies have shown that T9SS plays an important role in cellulose degradation,gliding motility,and ion assimilation in C.hutchinsonii.As reported recently,T9SS substrates are N-or O-glycosylated at their C-terminal domains(CTDs),with N-glycosylation being related to the translocation and outer membrane anchoring of these proteins.These findings have deepened our understanding of T9SS in C.hutchinsonii.In this review,we focused on the research progress on diverse substrates and functions of T9SS in C.hutchinsonii and the glycosylation of its substrates.A model of T9SS functions and the glycosylation of its substrates was proposed.

Ulcerative colitis(UC)is a chronic and recurrent inflammatory bowel disease(IBD)that has become a major gastroenterologic problem during recent decades.Numerous complicating factors are involved in UC development such as oxidative stress,inflammation,and microbiota disorder.These factors exacerbate damage to the intestinal mucosal barrier.Spirulina platensis is a commercial alga with various biological activity that is widely used as a functional ingredient in food and beverage products.However,there have been few studies on the treatment of UC using S.platensis aqueous extracts(SP),and the underlying mechanism of action of SP against UC has not yet been elucidated.Herein,we aimed to investigate the modulatory effect of SP on microbiota disorders in UC mice and clarify the underlying mechanisms by which SP alleviates damage to the intestinal mucosal barrier.Dextran sulfate sodium(DSS)was used to establish a normal human colonic epithelial cell(NCM460)injury model and UC animal model.The mitochondrial membrane potential assay 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)and staining with Annexin V-fluorescein isothiocyanate(FITC)/propidium iodide(PI)and Hoechst 33258 were carried out to determine the effects of SP on the NCM460 cell injury model.Moreover,hematoxylin and eosin(H&E)staining,transmission electron microscopy(TEM),enzyme-linked immunosorbent assay(ELISA),quantitative real-time polymerase chain reaction(qPCR),western blot,and 16S ribosomal DNA(rDNA)sequencing were used to explore the effects and underlying mechanisms of action of SP on UC in C57BL/6 mice.In vitro studies showed that SP alleviated DSS-induced NCM460 cell injury.SP also significantly reduced the excessive generation of intracellular reactive oxygen species(ROS)and prevented mitochondrial membrane potential reduction after DSS challenge.In vivo studies indicated that SP administration could alleviate the severity of DSS-induced colonic mucosal damage compared with the control group.Inhibition of inflammation and oxidati
