Effects of Heterologously Overexpressing PIP5K-Family Genes in Arabidopsis on Inflorescence Development

Castor is one of the top 10 oil crops in the world and has extremely valuable uses.Castor inflorescences directly affect yield,so the study of inflorescence development is very important in increasing castor yield.Our previous studies have shown that the PIP5K gene family(PIP5Ks)is associated with inflorescence development.In this study,to determine the function of each PIP5K gene in castor,a female Lm-type castor line,aLmAB2,was used to determine the relative expression levels of the PIP5Ks in castor inflorescences.Six PIP5K genes were heterologously overexpressed in Arabidopsis thaliana,the relative expression of each gene and the effect on plants was determined in A.thaliana,and the relationships among the PIP5Ks in castor were inferred.The expression levels of the PIP5Ks in the female Lm-type castor line aLmAB2 were analyzed.The relative expression levels of the PIP5K9 and PIP5K11 genes were high(p<0.05)in isofemale inflorescences,and those of PIP5K1,PIP5K2,PIP5K6,and PIP5K8 were high(p<0.05)in female inflorescences but low(p<0.05)in bisexual inflorescences.The PIP5Ks were heterologously overexpressed in A.thaliana,and T3-generation plants with stable genetic resistance,i.e.,AT-PIP5K^(+)plants(AT-PIP5K1^(+),AT-PIP5K2^(+),AT-PIP5K6^(+),AT-PIP5K8^(+),AT-PIP5K9^(+),and ATPIP5K11^(+) plants),were obtained.Biological tests of the AT-PIP5K+plants showed that the growth of the main stem was significantly delayed in AT-PIP5K+plants compared with Columbia wild-type(WT)A.thaliana plants;the PIP5K1 and PIP5K2 genes promoted lateral stem growth and flower and silique development;and the PIP5K6,PIP5K8,PIP5K9 and PIP5K11 genes inhibited lateral stem growth and flower and silique development.The correlations among PIP5Ks in castor suggest that there may be a synergistic relationship among PIP5K1,PIP5K2,and PIP5K6 in castor inflorescences,and PIP5K8,PIP5K9,and PIP5K11 are complementary to the other three genes.

Probiotic-derived exopolysaccharide protects against Staphylococcus aureus bloodstream infection in mice

Staphylococcus aureus (S. aureus) infection is a global public health problem， searching and developing green alternatives for antibiotics are urgently required. In this study， the exopolysaccharides (EPS) produced by Lactobacillus helveticus WXD191 were extracted and purified. Structure analysis suggested that the EPS contained Ara， Man， Gal， GalN， Glc， GlcN， and GlcA， and possessed a molecular of 84.2 kDa. Methylation combined with NMR spectroscopy analysis revealed that the backbone of EPS was →3)-β-D-Galp-(1→3)-β-D-GlcpNAc-(1→4)-β-D-GlcpA-(1→3- Man-1→2-Man-1→2，6-Man-1→2，6-Man-1→. Congo red analysis and CD spectrum indicated the existence of α-helices. Crystalline characteristics， scanning electron microscopy， and thermogravimetric analysis revealed that EPS formed thermally stable amorphous with a small amount of microcrystalline structure and a rough and porous surface. Meanwhile， The S. aureus bloodstream infection model was used to evaluate the protection efficiency for systemic infection induced by S. aureus and found that the EPS could enhance survival as well as reduce bacterial burden and proinflammatory chemokines. Collectively， these results suggest that EPS isolated from Lactobacillus helveticus was a competitive candidate for defense against S. aureus infection.

An ultrasensitive and selective sandwich-type fluorescent immunosensor based on fluorescence resonance energy transfer(FRET) between CdSe/ZnS quantum dots(QDs) and Au nanorods(AuNRs) was developed for the rapid detection of porcine reproductive and respiratory syndrome virus(PRRSV).Modified CdSe/ZnS QDs and AuNRs were bioconjugated with primary antibodies M and GP5,respectively.When the target antigen was present,a well-known sandwich-type form CdSe/ZnS QDs-M antibody-antigen-GP5 antibody-AuNRs was produced,and thus reducing the distance between the QDs and NRs.As a result,FRET occurred and the fluorescence intensity of CdSe/ZnS QDs decreased,and this decrease was used to determine the antigen concentration.Ultrasensitive detection of PRRSV at low concentrations in swine serum was achieved with a limit of detection of 0.55 TCID50/mL and a linear detection range of 10~1-3.5 × 10~4 TCID50/mL.Therefore,the fluorescent immunosensor is selective and highly sensitive;specifically,it can detect PRRSV at low concentrations in swine serum over a large concentration range.This proposed detection method can facilitate the development of high-performance fluorescent immunosensors for the detection of PRRSV and other antigens.

Global research on cognitive behavioural therapy for schizophrenia from 2000 to 2019:a bibliometric analysis via CiteSpace

Background Cognitive behavioural therapy(CBT)is considered to be an effective treatment for schizophrenia.In recent years,researchers have published clinical research results on CBT for schizophrenia.However,there is currently a lack of bibliometric analysis on CBT for schizophrenia.Aims To understand the current situation and research trends of CBT for schizophrenia,and to provide valuable information for researchers in this field.Methods Literature was retrieved from the Web of Science Core Collection database.CiteSpace was used to visually analyse the current status of research on CBT for schizophrenia,as well as trends and points of general interest on this topic.Results A total of 1151 papers were retrieved.The annual number of publications has increased 417.65%from 2000 to 2019.Schizophrenia Research was the most active journal.McGorry enjoys considerable influence.The University of London is deemed to be the most active research institution.England and the USA retain the highest scientific productivity in this field.China and Norway might make more contributions to this field in the future.The most important research topics are randomised controlled trials of CBT for schizophrenia,meta-analysis,scale selection and research on the improvement of quality of life.Results from the following keywords‘psychoeducation’,‘insomnia’,‘individual’,‘mental illness’,‘major depressive disorder’,‘efficacy’,‘internalised stigma’and‘remediation’indicated new research frontiers in this field.Conclusions The field of CBT for schizophrenia is progressing and has great potential.The level and quality of research in this field is high.The proposal of research hotspots and frontiers serves as a direction for researchers in this field.Researchers around the world should strengthen their cooperation and communication to promote further development of this field.

Cellular senescence provides a protective barrier against tumorigenesis in precancerous or normal tissues upon distinct stressors.However,the detailed mechanisms by which tumor cells evade premature senescence to malignant progression remain largely elusive.Here we reported that RBM4 adversely impacted cellular senescence to favor glutamine-dependent survival of esophageal squamous cell carcinoma(ESCC)cells by dictating the activity of LKB1,a critical governor of cancer metabolism.The level of RBM4 was specifically elevated in ESCC compared to normal tissues,and RBM4 overexpression promoted the malignant phenotype.RBM4 contributed to overcome H-RAS-or doxorubicin-induced senescence,while its depletion caused P27-dependent senescence and proliferation arrest by activating LKB1-AMPK-mTOR cascade.Mechanistically,RBM4 competitively bound LKB1 to disrupt the LKB1/STRAD/MO25 heterotrimeric complex,subsequently recruiting the E3 ligase TRIM26 to LKB1,promoting LKB1 ubiquitination and degradation in nucleus.Therefore,such molecular process leads to bypassing senescence and sustaining cell proliferation through the activation of glutamine metabolism.Clinically,the ESCC patients with high RBM4 and low LKB1 have significantly worse overall survival than those with low RBM4 and high LKB1.The RBM4 high/LKB1 low expression confers increased sensitivity of ESCC cells to glutaminase inhibitor CB-839,providing a novel insight into mechanisms underlying the glutamine-dependency to improve the efficacy of glutamine inhibitors in ESCC therapeutics.

Supramolecular conducting microfibers from amphiphilic tetrathiafulvalene-based organogelator

An amphiphilic tetrathiafulvalene molecule was designed and readily synthesized. The am-TTF can gelate a variety of organic solvents in view of multiple intermolecular interactions, especially in polar solvent.SEM observation provided clear evidence for the self-assembled micro/nanofibers morphologies in gelation state. Moreover, XRD measurements indicated the formation of highly-ordered columnar structures. The FT-IR spectra revealed that the formation of mixed-valence states with the absorption over 1700 cm^(-1), showing the semiconductive behaviors with the conductivity of 10^(-4)S/cm. The am-TTF based conducting fibers could be promising candidates for organic electronics.

Mediating effect of interpersonal relations on negative emotions and dysmenorrhea in female adolescents

Background Good interpersonal relationships can improve the negative mood and, to a certain extent, may help relieve dysmenorrhea symptoms. However, there has been no study examining the role of interpersonal relationships in dysmenorrhea and how they may interact with negative emotions. Aims To investigate the connection between negative affect and dysmenorrhea, and the role of interpersonal relationships plays in this relationship. Methods The Cox Menstrual Symptom Scale (CMSS), short-form of Depression Anxiety Stress Scale, Interpersonal Comprehensive Diagnostic Scale and UCLA (University of California at Los Angeles) Loneliness Scale were applied to 855 adolescent nursing students to collect information on severity and frequency of dysmenorrheal symptoms, negative emotions (including depression, anxiety and stress), interpersonal problems and subjective experience of loneliness among them, and the relationship among these variables was examined. Results (1) Both the severity and frequency of dysmenorrhea were positively correlated with negative emotions, loneliness experience and interpersonal problems, while negatively correlated with age at menarche.(2) The negative emotions, romantic relationship and menarche age, but not loneliness and interpersonal problems, significantly contributed to both the severity and frequency of dysmenorrhea.(3) Mediation analysis revealed that the effects of loneliness and interpersonal problems on dysmenorrhea were totally mediated by negative emotions. Conclusions This investigation indicates that the relationship between interpersonal relationships and dysmenorrhea is mediated by negative emotions.

山丘区小流域洪峰模拟结果与降雨时空分布密切相关。以河南省荥阳市王宗店流域“7·20”山洪灾害为例,构建了15种时空分布的10 min尺度降雨情景,综合采用中国山洪水文模型和方差分析理论评估降雨时空情景变化对洪峰模拟的影响。结果表明:中国山洪水文模型可以较好地模拟“7·20”暴雨洪水过程。不同时间情景下10 min降雨量均匀分布模拟得到的洪峰流量较小、峰现时间较晚;非均匀分布时最大10 min降雨量发生时间越晚,模拟得到的洪峰流量越大、峰现时间越晚。空间情景变化主要影响洪峰流量模拟,采用泰森多边形法模拟得到的洪峰流量最大。时间情景变化是洪峰流量和峰现时间模拟结果不确定性的最主要来源,空间情景变化和时空情景耦合变化仅影响洪峰流量模拟。
