Distinct roles of H3K27me3 and H3K36me3 in vernalization response, maintenance, and resetting in winter wheat

Winter plants rely on vernalization, a crucial process for adapting to cold conditions and ensuring successful reproduction. However,understanding the role of histone modifications in guiding the vernalization process in winter wheat remains limited. In this study, we investigated the transcriptome and chromatin dynamics in the shoot apex throughout the life cycle of winter wheat in the field. Two core histone modifications, H3K27me3 and H3K36me3, exhibited opposite patterns on the key vernalization gene VERNALIZATION1(VRN1),correlating with its induction during cold exposure. Moreover, the H3K36me3 level remained high at VRN1 after cold exposure, which may maintain its active state. Mutations in FERTILIZATION-INDEPENDENT ENDOSPERM(TaFIE) and SET DOMAIN GROUP 8/EARLY FLOWERING IN SHORT DAYS(TaSDG8/TaEFS), components of the writer complex for H3K27me3 and H3K36me3, respectively, affected flowering time. Intriguingly, VRN1 lost its high expression after the cold exposure memory in the absence of H3K36me3. During embryo development,VRN1 was silenced with the removal of active histone modifications in both winter and spring wheat, with selective restoration of H3K27me3 in winter wheat. The mutant of Tafie-cr-87, a component of H3K27me3 “writer” complex, did not influence the silence of VRN1during embryo development, but rather attenuated the cold exposure requirement of winter wheat. Integrating gene expression with H3K27me3 and H3K36me3 patterns identified potential regulators of flowering. This study unveils distinct roles of H3K27me3 and H3K36me3 in controlling vernalization response, maintenance, and resetting in winter wheat.

The spread of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)has resulted in significant casualties and put immense strain on public health systems worldwide,leading to economic recession and social unrest.In response,various prevention and control strategies have been implemented globally,including vaccine and drug development and the promotion of preventive measures.Implementing these strategies has effectively curbed the transmission of the virus,reduced infection rates,and gradually restored normal social and economic activities.However,the mutations of SARS-CoV-2 have led to inevitable infections and reinfections,and the number of deaths continues to rise.Therefore,there is still a need to improve existing prevention and control strategies,mainly focusing on developing novel vaccines and drugs,expediting medical authorization processes,and keeping epidemic surveillance.These measures are crucial to combat the Coronavirus disease(COVID-19)pandemic and achieve sustained,long-term prevention,management,and disease control.Here,we summarized the characteristics of existing COVID-19 vaccines and drugs and suggested potential future directions for their development.Furthermore,we discussed the COVID-19-related policies implemented over the past years and presented some strategies for the future.

Phenology of different types of vegetation and their response to climate change in the Qilian Mountains,China

The Qilian Mountains(QM)possess a delicate vegetation ecosystem,amplifying the evident response of vegetation phenology to climate change.The relationship between changes in vegetation growth and climate remains complex.To this end,we used MODIS NDVI data to extract the phenological parameters of the vegetation including meadow(MDW),grassland(GSD),and alpine vegetation(ALV))in the QM from 2002 to 2021.Then,we employed path analysis to reveal the direct and indirect impacts of seasonal climate change on vegetation phenology.Additionally,we decomposed the vegetation phenology in a time series using the trigonometric seasonality,Box-Cox transformation,ARMA errors,and Trend Seasonal components model(TBATS).The findings showed a distinct pattern in the vegetation phenology of the QM,characterized by a progressive shift towards an earlier start of the growing season(SOS),a delayed end of the growing season(EOS),and an extended length of the growing season(LOS).The growth cycle of MDW,GSD,and ALV in the QM species is clearly defined.The SOS for MDW and GSD occurred earlier,mainly between late April and August,while the SOS for ALVs occurred between mid-May and mid-August,a one-month delay compared to the other vegetation.The EOS in MDW and GSD were concentrated between late August and April and early September and early January,respectively.Vegetation phenology exhibits distinct responses to seasonal temperature and precipitation patterns.The advancement and delay of SOS were mainly influenced by the direct effect of spring temperatures and precipitation,which affected 19.59%and 22.17%of the study area,respectively.The advancement and delay of EOS were mainly influenced by the direct effect of fall temperatures and precipitation,which affected 30.18%and 21.17%of the area,respectively.On the contrary,the direct effects of temperature and precipitation in summer and winter on vegetation phenology seem less noticeable and were mainly influenced by indirect effects.The indirect effect of winter precipitation is the main fac

Rapid report of source parameters of 2023 M6.2 Jishishan,Gansu earthquake sequence

The M6.2 earthquake in Jishishan,Gansu Province,on December 18,2023,caused extraordinary earthquake disasters.It was located in the northern part of the north−south seismic zone,which is a key area for earthquake monitoring in China.The newly built dense strong motion stations in this area provide unprecedented conditions for high-precision earthquake relocation,especially the earthquake focal depth.This paper uses the newly built strong motion and traditional broadband seismic networks to relocate the source locations of the M3.0 and above aftershocks and to invert their focal mechanisms.The horizontal error of earthquake location is estimated to be 0.5−1 km,and the vertical error is 1−2 km.The focal depth range of aftershocks is 9.6−14.6 km,distributed in a 12-km-long strip with SSE direction.Aftershocks in the south are more concentrated horizontally and vertically,while aftershocks in the north are more scattered.The focal mechanisms of the main shock and aftershocks are relatively consistent,and the P-axis orientation is consistent with the regional strain direction.There is a seismic blank area of M3.0 and above,about 3−5 km between the main shock and aftershocks.It is suggested that the energy released by the main shock rupture is concentrated in this area.Based on the earthquake location and focal mechanism of the main shock,it is inferred that the Northern Lajishan fault zone is the seismogenic structure of the main shock,and the main shock did not occur on the main fault,but on a secondary fault.The initial rupture depth and centroid depth of the main shock were 12.8 and 14.0 km,respectively.The source rupture depth may not be the main reason for the severe earthquake disaster.

A CMIP6-based assessment of regional climate change in the Chinese Tianshan Mountains

Climate warming profoundly affects hydrological changes,agricultural production,and human society.Arid and semi-arid areas of China are currently displaying a marked trend of warming and wetting.The Chinese Tianshan Mountains(CTM)have a high climate sensitivity,rendering the region particularly vulnerable to the effects of climate warming.In this study,we used monthly average temperature and monthly precipitation data from the CN05.1 gridded dataset(1961-2014)and 24 global climate models(GCMs)of the Coupled Model Intercomparison Project Phase 6(CMIP6)to assess the applicability of the CMIP6 GCMs in the CTM at the regional scale.Based on this,we conducted a systematic review of the interannual trends,dry-wet transitions(based on the standardized precipitation index(SPI)),and spatial distribution patterns of climate change in the CTM during 1961-2014.We further projected future temperature and precipitation changes over three terms(near-term(2021-2040),mid-term(2041-2060),and long-term(2081-2100))relative to the historical period(1961-2014)under four shared socio-economic pathway(SSP)scenarios(i.e.,SSP1-2.6,SSP2-4.5,SSP3-7.0,and SSP5-8.5).It was found that the CTM had experienced significant warming and wetting from 1961 to 2014,and will also experience warming in the future(2021-2100).Substantial warming in 1997 was captured by both the CN05.1 derived from interpolating meteorological station data and the multi-model ensemble(MME)from the CMIP6 GCMs.The MME simulation results indicated an apparent wetting in 2008,which occurred later than the wetting observed from the CN05.1 in 1989.The GCMs generally underestimated spring temperature and overestimated both winter temperature and spring precipitation in the CTM.Warming and wetting are more rapid in the northern part of the CTM.By the end of the 21st century,all the four SSP scenarios project warmer and wetter conditions in the CTM with multiple dry-wet transitions.However,the rise in precipitation fails to counterbalance the drought induced by escalating temperatu

Ever since the concept of"plant cell totipotency"was first proposed in the early twentieth century,plant regeneration has been a major focus of study.Regeneration-mediated organogenesis and genetic transformation are important topics in both basic research and modern agriculture.Recent studies in the model plant Arabidopsis thaliana and other species have expanded our understanding of the molecular regulation of plant regeneration.The hierarchy of transcriptional regulation driven by phytohormone signaling during regeneration is associated with changes in chromatin dynamics and DNA methylation.Here,we summarize how various aspects of epigenetic regulation,including histone modifications and variants,chromatin accessibility dynamics,DNA methylation,and microRNAs,modulate plant regeneration.As the mechanisms of epigenetic regulation are conserved in many plants,research in this field has potential applications in boosting crop breeding,especially if coupled with emerging single-cell omics technologies.

Eco-physiological characteristics of Tetracentron sinense Oliv.saplings in response to different light intensities

The regeneration of Tetracentron sinense Oliv.is poor in the understory and in open areas due to the charac-teristics of natural regeneration of the species on forest edges and in gaps.It is unclear whether different light intensities in various habitats affect eco-physiological characteristics of saplings and their natural regeneration.In this study,the light intensity in T.sinense habitats was simulated by artificial shading(L1:100%NS(natural sunlight)in the open;L2:50%NS in a forest gap or edge;L3:10%NS in understory)to investigate differences in morphology,leaf structure,physiology,and photosynthesis of 2-year-old sap-lings,and to analyze the mechanism of light intensity on sapling establishment.Significant differences were observed in morphology(including leaf area,and specific leaf area)under different light intensities.Compared to L1 and L3,chloroplast structure in L2 was intact.With increasing time,superoxide dismutase(SOD)and catalase(CAT)activities in L2 became gradually higher than under the other light intensities,while malondialdehyde(MDA)content was opposite.Shading decreased osmoregulation substance contents of leaves but increased chlorophyll.The results suggest that light intensities significantly affect the eco-physiological characteristics of T.sinense saplings and they would respond most favorably at intermediate levels of light by optimizing eco-physiological characteristics.Therefore,50%natural sunlight should be created to promote saplings establishment and population recovery of T.sinense during in situ conservation,including sowing mature seeds in forest edges or gaps and providing appropriate shade protection for seedlings and saplings in the open.

Neuromorphic circuits based on memristors: endowing robots with a human-like brain

Robots are widely used,providing significant convenience in daily life and production.With the rapid development of artificial intelligence and neuromorphic computing in recent years,the realization of more intelligent robots through a pro-found intersection of neuroscience and robotics has received much attention.Neuromorphic circuits based on memristors used to construct hardware neural networks have proved to be a promising solution of shattering traditional control limita-tions in the field of robot control,showcasing characteristics that enhance robot intelligence,speed,and energy efficiency.Start-ing with introducing the working mechanism of memristors and peripheral circuit design,this review gives a comprehensive analysis on the biomimetic information processing and biomimetic driving operations achieved through the utilization of neuro-morphic circuits in brain-like control.Four hardware neural network approaches,including digital-analog hybrid circuit design,novel device structure design,multi-regulation mechanism,and crossbar array,are summarized,which can well simulate the motor decision-making mechanism,multi-information integration and parallel control of brain at the hardware level.It will be definitely conductive to promote the application of memristor-based neuromorphic circuits in areas such as intelligent robotics,artificial intelligence,and neural computing.Finally,a conclusion and future prospects are discussed.

NiCo/Al_(2)O_(3)nanocatalysts for the synthesis of 5-amino-1-pentanol and 1,5-pentanediol from biomass-derived 2-hydroxytetrahydropyran

Al_(2)O_(3)-supported monometallic Ni,Co,and bimetallic Ni-Co nanocatalysts originated from layered double hydroxide precursors were synthesized by co-precipitation method,and used for the synthesis of useful 5-amino-1-pentanol(5-AP)and 1,5-pentanediol(1,5-PD)by reductive amination(RA)or direct hydrogenation of biofurfuralderived 2-hydroxytetrahydropyran(2-HTHP),respectively.In both reactions,the yield of the target products decreased monotonously with the increasing amounts of Co in the NiCo/Al_(2)O_(3)catalysts,owing probably to the replacement of highly reactive Ni by Co component with inferior hydrogenation activity at the low reaction temperature of 60℃.However,the incorporation of Co could improve the reducibility of the NiCo/Al_(2)O_(3)bimetallic catalysts and promote the reaction stability of the catalysts,especially for Ni_(2)Co1/Al_(2)O_(3),in both reactions with over 180 h time-on-stream.Characterization of the catalysts before and after the reaction showed that the incorporating Co could inhibit the sintering of metal particles and hinder the surface oxidation of the more reactive Ni0species,thanks to the formation of Ni-Co alloy in the bimetallic catalysts.DFT-based modeling of the reaction mechanisms is also performed,supporting the reaction pathway proposed previously and also the much higher activity of Ni in the RA of 2-HTHP as compared with Co.

Evaluation of the safety and efficiency of cytotoxic T cell therapy sensitized by tumor antigens original from T-ALL-iPSC in vivo

Background:Since RNA sequencing has shown that induced pluripotent stem cells(iPSCs)share a common antigen profile with tumor cells,cancer vaccines that focus on iPSCs have made promising progress in recent years.Previously,we showed that iPSCs derived from leukemic cells of patients with primary T cell acute lymphoblastic leukemia(T-ALL)have a gene expression profile similar to that of T-ALL cell lines.Methods:Mice with T-ALL were treated with dendritic and T(DC-T)cells loaded with intact and complete antigens from T-ALL-derived iPSCs(T-ALL-iPSCs).We evaluated the safety and antitumor efficiency of autologous tumor-derived iPSC antigens by flow cytometry,cytokine release assay,acute toxicity experiments,long-term toxicity experiments,and other methods.Results:Our results indicate that complete tumor antigens from T-ALL-iPSCs could inhibit the growth of inoculated tumors in immunocompromised mice without causing acute and long-term toxicity.Conclusion:T-ALL-iPSC-based treatment is safe and can be used as a potential strategy for leukemia immunotherapy.
