Integrin binding peptides facilitate growth and interconnected vascular-like network formation of rat primary cortical vascular endothelial cells in vitro

Neovascularization and angiogenesis in the brain are important physiological processes for normal brain development and repair/regeneration following insults. Integrins are cell surface adhesion receptors mediating important function of cells such as survival, growth and development during tissue organization, differentiation and organogenesis. In this study, we used an integrin-binding array platform to identify the important types of integrins and their binding peptides that facilitate adhesion, growth, development, and vascular-like network formation of rat primary brain microvascular endothelial cells. Brain microvascular endothelial cells were isolated from rat brain on post-natal day 7. Cells were cultured in a custom-designed integrin array system containing short synthetic peptides binding to 16 types of integrins commonly expressed on cells in vertebrates. After 7 days of culture, the brain microvascular endothelial cells were processed for immunostaining with markers for endothelial cells including von Willibrand factor and platelet endothelial cell adhesion molecule. 5-Bromo-2′-dexoyuridine was added to the culture at 48 hours prior to fixation to assess cell proliferation. Among 16 integrins tested, we found that α5β1, αvβ5 and αvβ8 greatly promoted proliferation of endothelial cells in culture. To investigate the effect of integrin-binding peptides in promoting neovascularization and angiogenesis, the binding peptides to the above three types of integrins were immobilized to our custom-designed hydrogel in three-dimensional(3 D) culture of brain microvascular endothelial cells with the addition of vascular endothelial growth factor. 利用静电纺丝技术制备了有序和无序聚己内酯(PCL)纤维支架。通过扫描电镜(SEM)、X射线衍射(XRD)和万能试验机对支架结构与形态、结晶性能及力学性能进行了表征;将体外培养的细胞接种至支架表面,用激光共聚焦显微镜(LSCM)和SEM观察了细胞在支架表面的生长情况。研究结果表明,电纺丝得到的纤维直径均一,有序化纤维支架中86.14%的纤维沿着确定方向有序排列;有序纤维拉伸强度为(27.63±0.75)MPa,比无序纤维的拉伸强度增大了232%。弹性模量基本不变,断裂伸长率明显降低;有序纤维对细胞的定向生长具有良好的引导作用。

Vulnerable atherosclerotic plaques are responsible for most cardiovascular diseases(CVDs).Folate receptor(FR)positive activated macrophages were thought to be a prominent component in the development of vulnerable *** objective of this study is to develop folate conjugated two-dimensional(2D)Pd@Au nanomaterials(Pd@Au-PEG-FA)for targeted multimodal imaging of the FRs in advanced atherosclerotic *** and imaging studies(single photon emission computed tomography(SPECT),computed tomography(CT)and photoacoustic(PA)imaging)were performed to confirm the prolonged blood half-life and enrichment of radioactivity in atherosclerotic *** signals were detected in vivo with SPECT,CT and PA imaging in heavy atherosclerotic plaques,which were significantly higher than those of the normal aortas after injection of Pd@*** studies with preinjection of excess FA could effectively reduce the targeting ability of Pd@Au-PEG-FA in atherosclerotic plaques,further demonstrating the specific binding of Pd@Au-PEG-FA for plaque *** characterization revealed that the signal of probe was in accordance with the high-risk *** summary,the Pd@Au-PEG-FA has favorable pharmacokinetic properties and provides a valuable approach for detecting high-risk plaques in the presence of FRs in atherosclerotic plaques.

Anti-corrosion inspection engineers are responsible for 14 degassing stations, which complete anti-corrosion examinations of desalination and dehydration containers under maintenance, detecting potential corrosion, drawing up the diagram illustrations and inspection reports of Rumaila's oil field and gas system, guiding the completion of equipment sandblasting and anti-corrosion inspections, and ensuring the fulfillment of other relevant work reports. Anti-corrosion inspection engineers can be called the "health doctors" of oil and gas systems.

选用目前最为先进与常用的4点弯曲疲劳试验,对常用的包括70#基质沥青和常用的改性沥青在内的10种沥青混合料进行了15℃及1 000×10-6应变控制下的疲劳试验,并进行二次疲劳试验,兼顾了改性沥青的自愈合效果,完成了相同沥青用量下和相同体积设计目标下的各沥青混合料的疲劳性能对比,以及针对相同高温性能和疲劳性能的双向分级,以提供实际工程时混合料设计之前的参考,最后还依据所有试验数据绘制了相应的疲劳性能和高温性能分区散点图以供直观参考.

为研究动态热环境下人体心理反应预测，基于模糊集理论及人与周围热环境瞬时热量传递的合理分析，提出一种动态热环境中人体热感觉模糊综合评判的方法。在本质上，人体外表面同周围环境之间换热热流通量的变化决定了人体在热环境中热感觉值的变化规律。该方法选取换热热流通量变化值作为特征变量，因而降低了求解问题的复杂度。数值模拟算法采用分层递阶模块式结构、基于知识的推理系统和模糊信息并行处理，具有结构简单、易修改扩充、计算速度快和占用存储空间少等优点。将模拟结果同微气候环境室的实验数据相比较，验证了算法的有效性。
