A novel strategy of tannery sludge disposal–converting into biochar and reusing for Cr(Ⅵ)removal from tannery wastewater

Tannery sludge with high chromium content has been identified as hazardous solid waste due to its potential toxic effects.The safety disposal and valorization of the tannery sludge remains a challenge.In this study,the chromium stabilization mechanism was systematically investigated during chromium-rich tannery sludge was converted to biochar and the removal performance of the sludge biochar(SBC)for Cr(Ⅵ)from tannery wastewater was also investigated.The results showed that increase in pyrolysis temperature was conductive to the stabilization of Cr and significant reduction of the proportion of Cr(Ⅵ)in SBC.It was confirmed that the stabilization of chromium mainly was attributed to the embedding of chromium in the C matrix and the transformation of the chromium-containing substances from the amorphous Cr(OH)_(3)to the crystalline state,such as(FeMg)Cr_(2)O_(5).The biochar presented high adsorption capacity of Cr(Ⅵ)at low pH and the maximal theoretical adsorption capacity of SBC produced at 800°C can reach 352 mg Cr(Ⅵ)/g,the process of which can be well expressed by Langmuir adsorption isotherm and pseudo second order model.The electrostatic effect and reduction reaction were dominantly responsible for the Cr(Ⅵ)adsorption by SBC800.Overall,this study provided a novel strategy for the harmless disposal and resource utilization for the solid waste containing chromium in leather industry.

Biomimetic nanoparticles drive the mechanism understanding of shear-wave elasticity stiffness in triple negative breast cancers to predict clinical treatment

In clinical practice,we noticed that triple negative breast cancer(TNBC)patients had higher shear-wave elasticity(SWE)stiffness than non-TNBC patients and a higherα-SMA expression was found in TNBC tissues than the non-TNBC tissues.Moreover,SWE stiffness also shows a clear correlation to neoadjuvant response efficiency.To elaborate this phenomenon,TNBC cell membrane-modified polylactide acid-glycolic acid(PLGA)nanoparticle was fabricated to specifically deliver artesunate to regulate SWE stiffness through inhibiting CAFs functional status.As tested in MDA-MB-231 and E0771 orthotopic tumor models,CAFs functional status inhibited by 231M-ARS@PLGA nanoparticles(231M-AP NPs)had reduced the SWE stiffness as well as attenuated hypoxia of tumor as tumor soil loosening agent which amplified the antitumor effects of paclitaxel and PD1 inhibitor.Single-cell sequencing indicated that the two main CAFs(extracellular matrix and wound healing CAFs)that produces extracellular matrix could influence the tumor SWE stiffness as well as the antitumor effect of drugs.Further,biomimetic nanoparticles inhibited CAFs function could attenuate tumor hypoxia by increasing proportion of inflammatory blood vessels and oxygen transport capacity.Therefore,our finding is fundamental for understanding the role of CAFs on affecting SWE stiffness and drugs antitumor effects,which can be further implied in the potential clinical theranostic predicting in neoadjuvant therapy efficacy through non-invasive analyzing of SWE imaging.

Common and distinct regulation of human and mouse brown and beige adipose tissues： a promising therapeutic target for obesity

Obesity, which underlies various metabolic and cardio- vascular diseases, is a growing public health challenge for which established therapies are inadequate. Given the current obesity epidemic, there is a pressing need for more novel therapeutic strategies that will help adult individuals to manage their weight. One promising therapeutic intervention for reducing obesity is to enhance energy expenditure. Investigations into human brown fat and the recently discovered beige/brite fat have galvanized intense research efforts during the past decade because of their pivotal roles in energy dissi- pation. In this review, we summarize the evolution of human brown adipose tissue （hBAT） research and dis- cuss new in vivo methodologies for evaluating energy expenditure in patients. We highlight the differences between human and mouse BAT by integrating and comparing their cellular morphology, function, and gene expression profiles. Although great advances in hBAT biology have been achieved in the past decade, more cellular models are needed to acquire a better under- standing of adipose-specific processes and molecular mechanisms. Thus, this review also describes the development of a human brown fat cell line, which could provide promising mechanistic insights into hBAT function, signal transduction, and development. Finally, we focus on the therapeutic potential and current limi- tations of hBAT as an anti-glycemic, anti-lipidemic, and weight loss-inducing ＇metabolic panacea＇.

Bromine and oxygen redox species mediated highly selective electro-epoxidation of styrene

Olefin epoxidation is an essential transformation and arouses great interest among the scientific community for the key role of epoxide in the chemical industry.Traditional methods suffer from harmful oxidants and require harsh operating conditions and complex separation processes.Electro-epoxidation through bromine redox reactions is an eco-friendly strategy.However,the lack of efficient electrodes always limits the selectivity of epoxidation and results in massive energy consumption.Herein,we found that GF-CoS_(2)/CoS heterostructures anchored on graphite felt(GF-CoS_(2)/CoS)can efficiently facilitate the generation of bromine redox species on the anode.The synergistic cooperation between the bromine redox species and oxygen redox species(generated on a graphite rod cathode)can establish a benign electro-epoxidation system for olefin substrates in an ambient environment.Under the optimized reaction conditions,the epoxidation of styrene can achieve a yield of 97% at 30 mA cm^(-2).Importantly,the applied voltage(4-5 V)is roughly half of the Pt-based electrode system(7.8-9.3 V),hence,saving half of the energy.We demonstrated that our proposed strategy could also be applied to other olefins.Because of the simple,sustainable,and economic advantages of the epoxidation process,we believe this strategy is highly desirable for technical applications.

Constitutive activation of RAS-RAF-MEK-ERK signaling pathway(MAPK pathway)frequently occurs in many cancers harboring RAS or RAF oncogenic mutations.Because of the paradoxical activation induced by a single use of BRAF or MEK inhibitors,dual-target RAF and MEK treatment is thought to be a promising strategy.In this work,we evaluated erianin is a novel inhibitor of CRAF and MEK1/2 kinases,thus suppressing constitutive activation of the MAPK signaling pathway induced by BRAF V600E or RAS mutations.KinaseProfiler enzyme profiling,surface plasmon resonance(SPR),isothermal titration calorimetry(ITC),cellular thermal shift assay,computational docking,and molecular dynamics simulations were utilized to screen and identify erianin binding to CRAF and MEK1/2.Kinase assay,luminescent ADP detection assay,and enzyme kinetics assay were investigated to identify the efficiency of erianin in CRAF and MEK1/2 kinase activity.Notably,erianin suppressed BRAF V600E or RAS mutant melanoma and colorectal cancer cell by inhibiting MEK1/2 and CRAF but not BRAF kinase activity.Moreover,erianin attenuated melanoma and colorectal cancer in vivo.Overall,we provide a promising leading compound for BRAF V600E or RAS mutant melanoma and colorectal cancer through dual targeting of CRAF and MEK1/2.

Experiments on particle cluster behaviors in a fast fluidized bed

A three-dimensional(3D) fast fluidized bed with the riser of 3.0 m in height and 0.1 m in inner diameter was established to experimentally study the cluster behaviors of Geldart B particles. Five kinds of quartz sand particles(dp= 0.100, 0.139, 0.177, 0.250 and 0.375 mm and ρp= 2480 kg·m^(-3)) were respectively investigated, with the total mass of the bed material kept as 10 kg. The superficial gas velocity in the riser ranges from 2.486 to 5.594 m·s^(-1) and the solid mass flux alters from 30 to 70 kg·((m^(-2)·s))^(-1). Cluster characteristics and evolutionary processes in the different positions of the riser were captured by the cluster visualization systems and analyzed by the self-developed binary image processing. The results found four typical cluster structures in the riser,i.e., the macro stripe-shaped cluster, saddle-shaped cluster, U-shaped cluster and the micro cluster. The increasing superficial gas velocity and particle sizes result in the increasing average cluster size and the decreasing cluster time fraction, while the solid mass flux in the riser have the reverse influences on the cluster size and time fraction. Additionally, clusters in the upper region of the riser often have the larger size and time fraction than that in the lower region. All these effects of operating conditions on clusters become less obvious when particle size is less than 0.100 mm.

Huge volume changes of Si during lithiation/delithiation lead to regeneration of solid-electrolyte interphase(SEI)and consume electrolyte.In this article,γ-glycidoxypropyl trimethoxysilane(GOPS)was incorporated in Si/PEDOT:PSS electrodes to construct a flexible and conductive artificial SEI,effectively suppressing the consumption of electrolyte.The optimized electrode can maintain 1000 mAh g^−1 for nearly 800 cycles under limited electrolyte compared with 40 cycles of the electrodes without GOPS.Also,the optimized electrode exhibits excellent rate capability.The use of GOPS greatly improves the interface compatibility between Si and PEDOT:PSS.XPS Ar+etching depth analysis proved that the addition of GOPS is conducive to forming a more stable SEI.A full battery assembled with NCM 523 cathode delivers a high energy density of 520 Wh kg^−1,offering good stability.

BYSL contributes to tumor growth by cooperating with the mTORC2 complex in gliomas

Objective:BYSL,which encodes the Bystin protein in humans,is upregulated in reactive astrocytes following brain damage and/or inflammation.We aimed to determine the role and mechanism of BYSL in glioma cell growth and survival.Methods:BYSL expression in glioma tissues was measured by quantitative real-time PCR,Western blot,and immunohistochemistry.In vitro assays were performed to assess the role of BYSL in cell proliferation and apoptosis.Protein interactions and co-localization were determined by co-immunoprecipitation and double immunofluorescence.The expression and activity of the AKT/m TOR signaling molecules were determined by Western blot analysis,and the role of BYSL in glioma growth was confirmed in an orthotopic xenograft model.Results:The BYSL m RNA and protein levels were elevated in glioma tissues.Silencing BYSL inhibited glioma cell proliferation,impeded cell cycle progression,and induced apoptosis,whereas overexpressing BYSL protein led to the opposite effects.We identified a complex consisting of BYSL,RIOK2,and m TOR,and observed co-localization and positive correlations between BYSL and RIOK2 in glioma cells and tissues.Overexpressing BYSL or RIOK2 increased the expression and activity of AKT/m TOR signaling molecules,whereas downregulation of BYSL or RIOK2 decreased the activity of AKT/m TOR signaling molecules.Silencing BYSL or RIOK2 decreased the growth of the tumors and prolonged the lifespan of the animals in an orthotopic xenograft model.Conclusions:High expression of BYSL in gliomas promoted tumor cell growth and survival both in vitro and in vivo.These effects could be attributed to the association of BYSL with RIOK2 and m TOR,and the subsequent activation of AKT signaling.

Biomimetic giant membrane vesicles,with size and lipid compositions comparable to cells,have been recognized as an attractive experimental alternative to living systems.Due to the similarity of their membrane structure to that of body cells,cell-derived giant plasma membrane vesicles have been used as a membrane model for studying lipid/protein behavior of plasma membranes.However,further application of biomimetic giant membrane vesicles has been hampered by the side-efects of chemical vesiculants and the utilization of osmotic bufer.We herein develop a facile strategy to derive giant membrane vesicles(GMVs)from mammalian cells in biofriendly medium with high yields.Tese GMVs preserve membrane properties and adaptability for surface modifcation and encapsulation of exogenous molecules,which would facilitate their potential biological applications.Moreover,by loading GMVs with therapeutic drugs,GMVs could be employed for drug transport to tumor cells,which represents another step forward in the biomedical application of giant membrane vesicles.Tis study highlights biocompatible GMVs with biomimicking membrane surface properties and adaptability as an ideal platform for drug delivery strategies with potential clinical applications.
