Previous studies have suggested that the incidence of post-traumatic stress disorder in earthquake rescue workers is relatively high. Risk factors for this disorder include demographic characteristics, earthquake-related high-risk factors, risk factors in the rescue process, personality, social support and coping style. This study examined the current status of a unit of 1 040 rescue workers who participated in earthquake relief for the Wenchuan earthquake that occurred on May 12th, 2008. Post-traumatic stress disorder was diagnosed primarily using the Clinician-Administered Post-traumatic Stress Disorder Scale during structured interviews. Univariate and multivariate sta-tistical analyses were used to examine major risk factors that contributed to the incidence of post-traumatic stress disorder. Results revealed that the incidence of this disorder in the rescue group was 5.96%. The impact factors in univariate analysis included death of family members, contact with corpses or witnessing of the deceased or seriously injured, near-death experience, severe injury or mental trauma in the rescue process and working at the epicenter of the earthquake. Correlation analysis suggested that post-traumatic stress disorder was positively correlated with psychotic and neurotic personalities, negative coping and low social support. Impact factors in mul-tivariate logistic regression analysis included near-death experience, severe injury or mental trauma, working in the epicenter of the rescue, neurotic personality, negative coping and low social support, among which low social support had the largest odds ratio of 20.42. Findings showed that the oc-currence of post-traumatic stress disorder was the result of the interaction of multiple factors.

Protein ubiquitination is essential for diverse cellular functions including spermatogenesis.The tripartite motif(TRIM)family proteins,most of which have E3 ubiquitin ligase activity,are highly conserved in mammals.They are involved in important cellular processes such as embryonic development,immunity,and fertility.Our previous studies indicated that Trim69,a testis-specific expressed TRIM family gene,potentially participates in the spermatogenesis by mediating testicular cells apoptosis.In this study,we investigated the biological functions of Trim69 in male mice by established Trim69 knockout mice with CRISPR/Cas9 genomic editing technology.Here,we reported that the male Trim69 knockout mice had normal fertility.The adult knockout mice have shown that the appearance of testes,testis/body weight ratios,testicular histomorphology,and the number and quality of sperm were consistent with wild-type mice.These results indicated that the E3 ubiquitin ligase protein Trim69 was not essential for male mouse fertility,and it might be compensated by other TRIM family members such as Trim58 in Trim69-deficiency testis.This study would help to elucidate the functions of tripartite motif protein family and the regulation of spermatogenesis.

Internal ribosome entry sites(IRESs)are functional RNA elements that can directly recruit ribosomes to an internal position of the mRNA in a cap-independent manner to initiate translation.Recently,IRES elements have attracted much attention for their critical roles in various processes including translation initiation of a new type of RNA,circular RNA(circRNA),with no5’cap to support classical cap-dependent translation.Thus,an integrative data resource of IRES elements with experimental evidence will be useful for further studies.In this study,we present IRESbase,a comprehensive database of IRESs,by curating the experimentally validated functional minimal IRES elements from literature and annotating their host linear and circular RNAs.The current version of IRESbase contains 1328 IRESs,including 774 eukaryotic IRESs and 554 viral IRESs from 11 eukaryotic organisms and 198 viruses,respectively.As IRESbase collects only IRES of minimal length with functional evidence,the median length of IRESs in IRESbase is 174 nucleotides.By mapping IRESs to human circRNAs and long non-coding RNAs(lncRNAs),2191 circRNAs and 168 lncRNAs were found to contain at least one entire or partial IRES sequence.IRESbase is available at http://***/cgi-bin/iresbase/index.php.

The increase in occurrence and severity of cyanobacteria blooms is causing increasing concern;moreover,human and animal health is affected by the toxic effects of Microcystin-LR released into the water.In this paper,a floating photocatalyst for the photocatalytic inactivation of the harmful algae Microcystis aeruginosa(M.aeruginosa)was prepared using a simple sol-gel method,i.e.,coating g-C_(3)N_(4) coupled with Bi-doped TiO_(2) on Al_(2)O_(3)-modified expanded perlite(CBTA for short).The impact of different molar ratios of Bi/Ti on CBTA was considered.The results indicated that Bi doping in TiO_(2) inhibited photogenerated electron-hole pair recombination.With 6 h of visible light illumination,75.9%of M.aeruginosa(initial concentration=2.7106 cells/L)and 83.7%of Microcystin-LR(initial concentration=100μg/L)could be removed with the addition of 2 g/L CBTA1%(i.e.,Bi/Ti molar ratio=1%).The key reactive oxygen species(ROSs)in the photocatalytic inactivation process are h+andOH.The induction of the Bi^(4+)/Bi^(3+)species by the incorporation of Bi could narrow the bandgap of TiO_(2),trap electrons,and enhance the stability of CBTA-1%in the solutions with coexisting environmental substances.

Generation of isogenic single and multiplex gene knockout mice by base editing-induced STOP

Although CRISPR/Cas9 has been widely used to generate knockout mice, two major limitations remain:the founders usually carry a mixture of genotypes, and mosaicism harboring multiple genotypes.Therefore, it takes a long time to get homozygous mutants. Recently developed base editing(BE) system,which introduces C-to-T conversion without double strand DNA cleavage, has been used to introduce artificial stop codons(i-STOP) to prematurely terminate translation, providing a cleaner strategy for genome engineering. Using this strategy, we generated CD160 KO and VISTA/CD160 double KO mice by microinjection of a single sg RNA targeting CD160 and a mixture of sg RNAs targeting VISTA and CD160,respectively. The BE system induced STOP efficiently in mouse embryos and consequently in founder mice without detectable off-target. Most interestingly, the majority of the mutants harbor same genetic modifications, indicating we generated isogenic single and multiplex gene mutant mice by BE-induced STOP. We also obtained homozygous mutant mouse in F1 mice, demonstrating the accelerated strategy in generating animal models.

微塑料可以在水中吸附重金属并作为载体共同迁移,且微塑料会在水中受光照而老化,影响其吸附能力和吸附机制.本研究通过XRD、FTIR、SEM/EDS等分析手段对聚丁二酸丁二醇酯基可降解微塑料(PBS-MPs)进行了表征,探究了光老化过程对PBS-MPs吸附水中Pb(Ⅱ)的影响及其机制.结果表明,光老化能增加PBS-MPs表面亲水性含氧基团及活性吸附位点数量,对Pb(Ⅱ)的最大吸附容量从64.13μg·g^(-1)提升至362.06μg·g^(-1);Pb(Ⅱ)在P-PBS上的吸附符合Elovich动力学模型,吸附机制包括表面络合、静电作用和孔填充/扩散作用.光老化会强化可降解微塑料对水中Pb(Ⅱ)的吸附,从而促进Pb(Ⅱ)的迁移,二者共存可对水环境带来更为严重的生态风险.

CircAST:Full-length Assembly and Quantification of Alternatively Spliced Isoforms in Circular RNAs

Circular RNAs(circ RNAs),covalently closed continuous RNA loops,are generated from cognate linear RNAs through back splicing events,and alternative splicing events may generate different circ RNA isoforms at the same locus.However,the challenges of reconstruction and quantification of alternatively spliced full-length circ RNAs remain unresolved.On the basis of the internal structural characteristics of circ RNAs,we developed Circ AST,a tool to assemble alternatively spliced circ RNA transcripts and estimate their expression by using multiple splice graphs.Simulation studies showed that Circ AST correctly assembled the full sequences of circ RNAs with a sensitivity of 85.63%–94.32%and a precision of 81.96%–87.55%.By assigning reads to specific isoforms,Circ AST quantified the expression of circ RNA isoforms with correlation coefficients of 0.85–0.99 between theoretical and estimated values.We evaluated Circ AST on an in-house mouse testis RNA-seq dataset with RNase R treatment for enriching circ RNAs and identified 380 circ RNAs with full-length sequences different from those of their corresponding cognate linear RNAs.RT-PCR and Sanger sequencing analyses validated 32 out of 37 randomly selected isoforms,thus further indicating the good performance of Circ AST,especially for isoforms with low abundance.We also applied Circ AST to published experimental data and observed substantial diversity in circular transcripts across samples,thus suggesting that circ RNA expression is highly regulated.Circ AST can be accessed freely at https://***/xiaofengsong/CircAST.

露天煤矿排土场土层重构对于生态重建具有重要意义，为研究接种深色有隔内生真菌(dark septate endophytes, DSE)对不同重构土层模式下玉米根系水分的利用效应，采用土柱模拟培养试验，设置4种类型土层处理，每种土层类型下设置接菌及对照处理，共8组处理。结果表明：掺黄土20%处理下玉米根长密度最大，分别为掺黄土0%、10%和40%的3.2、2.4、2.8倍，水分胁迫后根系具有向下生长、吸取深层水分的能力；基于δ18O值的MixSIAR模型水源分析，掺黄土0%处理下玉米主要利用0～25 cm处的水分，水分利用效率达到80%；掺黄土10%处理下玉米主要利用15～35 cm处的水分，水分利用效率达到64%；而掺黄土20%处理下玉米对0～25 cm处的水分利用效率仅为36%，对25～45 cm处水分利用率达到64%，说明掺黄土20%处理下玉米主要利用土壤深层水分。接种DSE提高了植物吸收更深层水的能力，掺黄土20%处理下水分利用深度向下增加了5 cm，掺黄土20%基质中接菌处理在干旱胁迫后植物根系提水量达到最大，生长期总提水量较不接菌处理提升了45%；在不接菌条件下掺黄土20%植物根系提水量是掺黄土0%的1.45倍，而在接菌条件下掺黄土20%植物根系提水量可达到掺黄土0%的1.72倍。综上认为，接种DSE及土层重构均对提升植物提水能力具有显著作用。此外，本研究结果对露天矿区排土场土层重构过程中土壤改良及植物的水分利用效率提供实验参考依据。

Atomic Co/Ni dual sites with N/P-coordination as bifunctional oxygen electrocatalyst for rechargeable zinc-air batteries

Metal-nitrogen-carbon(M-N-C)single-atom catalysts exhibit desirable electrochemical catalytic properties.However,the replacement of N atoms by heteroatoms(B,P,S,etc.)has been regarded as a useful method for regulating the coordination environment.The structure engineered M-N-C sites via doping heteroatoms play an important role to the adsorption and activation of the oxygen intermediate.Herein,we develop an efficient strategy to construct dual atomic site catalysts via the formation of a Co_(1)-PN and Ni1-PN planar configuration.The developed Co_(1)-PNC/Ni1-PNC catalyst exhibits excellent bifunctional electrocatalytic performance in alkaline solution.Both experimental and theoretical results demonstrated that the N/P coordinated Co/Ni sites moderately reduced the binding interaction of oxygen intermediates.The Co_(1)-PNC/Ni1-PNC endows a rechargeable Zn-air battery with excellent power density and cycling stability as an air-cathode,which is superior to that of the benchmark Pt/C+IrO_(2).This work paves an avenue for design of dual single-atomic sites and regulation of the atomic configuration on carbon-based materials to achieve high-performance electrocatalysts.
