Human microorganisms,including bacteria,fungi,and viruses,play key roles in several physiological and pathological processes.Some studies discovered that tumour tissues once considered sterile actually host a variety of microorganisms,which have been confirmed to be closely related to oncogenesis.The concept of intratumoural microbiota was subsequently proposed.Microbiota could colonise tumour tissues through mucosal destruction,adjacent tissue migration,and hematogenic invasion and affect the biological behaviour of tumours as an important part of the tumour microenvironment.Mechanistic studies have demonstrated that intratumoural microbiota potentially promote the initiation and progression of tumours by inducing genomic instability and mutations,affecting epigenetic modifications,promoting inflammation response,avoiding immune destruction,regulating metabolism,and activating invasion and metastasis.Since more comprehensive and profound insights about intratumoral microbiota are continuously emerging,new methods for the early diagnosis and prognostic assessment of cancer patients have been under examination.In addition,interventions based on intratumoural microbiota show great potential to open a new chapter in antitumour therapy,especially immunotherapy,although there are some inevitable challenges.Here,we aim to provide an extensive review of the concept,development history,potential sources,heterogeneity,and carcinogenic mechanisms of intratumoural microorganisms,explore the potential role of microorganisms in tumour prognosis,and discuss current antitumour treatment regimens that target intratumoural microorganisms and the research prospects and limitations in this field.

Influenza A virus(IAV)binds sialic acid receptors on the cell surface to enter the host cells,which is the key step in initiating infection,transmission and pathogenesis.Understanding the factors that contribute to the highly efficient entry of IAV into human cells will help elucidate the mechanism of viral entry and pathogenicity,and provide new targets for intervention.In the present study,we reported a novel membrane protein,C1QTNF5,which binds to the hemagglutinin protein of IAV and promotes IAV infection in vitro and in vivo.We found that the HA1 region of IAV hemagglutinin is critical for the interaction with C1QTNF5 protein,and C1QTNF5 interacts with hemagglutinin mainly through its N-terminus(1–103 aa).In addition,we further demonstrated that overexpression of C1QTNF5 promotes IAV entry,while blocking the interaction between C1QTNF5 and IAV hemagglutinin greatly inhibits viral entry.However,C1QTNF5 does not function as a receptor to mediate IAV infection in sialic acid-deficient CHO-Lec2 cells,but promotes IAV to attach to these cells,suggesting that C1QTNF5 is an important attachment factor for IAV.This work reveals C1QTNF5 as a novel IAV attachment factor and provides a new perspective for antiviral strategies.

Deep learning-based subseasonal to seasonal precipitation prediction in southwest China:Algorithm comparison and sensitivity to input features

The prediction of precipitation at subseasonal to seasonal(S2S)timescales remains an enormous challenge because of the gap between weather and climate predictions.This study compares three deep learning algorithms,namely,the long short-term memory recurrent(LSTM),gated recurrent unit(GRU),and recurrent neural network(RNN),and selects the optimal algorithm to establish an S2S precipitation prediction model.The models were evaluated in four subregions of the Sichuan Province:the Plateau,Valley,eastern Basin,and western Basin.The results showed that the RNN model had better performance than the LSTM and GRU models.This could be because the RNN model had an advantage over the LSTM model in the transformation of climate indices with positive and negative variations.In the validation of test datasets,the RNN model successfully predicted the precipitation trend in most years during the wet season(May-October).The RNN model had a lower prediction bias(within±10%),higher sign accuracy of the precipitation trend(~88.95%),and greater accuracy of the maximum precipitation month(>0.85).For the prediction of different lead times,the RNN model was able to provide a stable trend prediction for summer precipitation,and the time correlation coefficient score was higher than that of the National Climate Center of China.Furthermore,this study proposed a method to measure the sensitivity of the RNN model to different input features,which may provide unprecedented insights into the nonlinear relationship and complicated feedback process among climate systems.The results of the sensitivity distribution are as follows.First,the Niño 4 and Niño 3.4 indices were equally important for the prediction of wet season precipitation.Second,the sensitivity of the snow cover on the Tibetan Plateau was higher than that in the Northern Hemisphere.Third,an opposite sensitivity appeared in two different patterns of the Indian Ocean and sea ice concentrations in the Arctic and the Barents Sea.

River width and depth as key factors of diurnal activity energy expenditure allocation for wintering Spot-billed Ducks in the Xin'an River Basin

Rivers are important habitats for wintering waterbirds.However,they are easily influenced by natural and human activities.An important approach for waterbirds to adapt to habitats is adjusting the activity time and energy expenditure allocation of diurnal behavior.The compensatory foraging hypothesis predicts that increased energy expenditure leads to longer foraging time,which in turn increases food intake and helps maintain a constant energy balance.However,it is unclear whether human-disturbed habitats result in increased energy expenditure related to safety or foraging.In this study,the scan sample method was used to observe the diurnal behavior of the wintering Spot-billed Duck(Anas poecilorhyncha) in two rivers in the Xin’an River Basin from October 2021 to March 2022.The allocation of time and energy expenditure for activity in both normal and disturbed environments was calculated.The results showed that foraging accounted for the highest percentage of time and energy expenditure.Additionally,foraging decreased in the disturbed environment than that in the normal environment.Resting behavior showed the opposite trend,while other behaviors were similar in both environments.The total diurnal energy expenditure of ducks in the disturbed environment was greater than that in the normal environment,with decreased foraging and resting time percentage and increased behaviors related to immediate safety(swimming and alert) and comfort.These results oppose the compensatory foraging hypothesis in favor of increased security.The optimal diurnal energy expenditure model included river width and water depth,which had a positive relationship;an increase in either of these two factors resulted in an increase in energy expenditure.This study provides a better understanding of energy allocation strategies underlying the superficial time allocation of wintering waterbirds according to environmental conditions.Exploring these changes can help understand the maximum fitness of wintering waterbirds in response to nature and hum

The effects of exercise interventions on brain-derived neurotrophic factor levels in children and adolescents:a meta-analysis

Brain-derived neurotrophic factor is a crucial neurotrophic factor that plays a significant role in brain health. Although the vast majority of meta-analyses have confirmed that exercise interventions can increase brain-derived neurotrophic factor levels in children and adolescents, the effects of specific types of exercise on brain-derived neurotrophic factor levels are still controversial. To address this issue, we used meta-analytic methods to quantitatively evaluate, analyze, and integrate relevant studies. Our goals were to formulate general conclusions regarding the use of exercise interventions, explore the physiological mechanisms by which exercise improves brain health and cognitive ability in children and adolescents, and provide a reliable foundation for follow-up research. We used the Pub Med, Web of Science, Science Direct, Springer, Wiley Online Library, Weipu, Wanfang, and China National Knowledge Infrastructure databases to search for randomized controlled trials examining the influences of exercise interventions on brain-derived neurotrophic factor levels in children and adolescents. The extracted data were analyzed using Review Manager 5.3. According to the inclusion criteria, we assessed randomized controlled trials in which the samples were mainly children and adolescents, and the outcome indicators were measured before and after the intervention. We excluded animal experiments, studies that lacked a control group, and those that did not report quantitative results. The mean difference(MD;before versus after intervention) was used to evaluate the effect of exercise on brain-derived neurotrophic factor levels in children and adolescents. Overall, 531 participants(60 children and 471 adolescents, 10.9–16.1 years) were included from 13 randomized controlled trials. Heterogeneity was evaluated using the Q statistic and I^(2) test provided by Review Manager software. Development of a new cerebral ischemia reperfusion model of Mongolian gerbils and standardized evaluation system

Background:The Mongolian gerbil is an excellent laboratory animal for preparing the cerebral ischemia model due to its inherent deficiency in the circle of Willis.However,the low incidence and unpredictability of symptoms are caused by numerous complex variant types of the circle.Additionally,the lack of an evaluation system for the cer-ebral ischemia/reperfusion(I/R)model of gerbils has shackled the application of this model.Methods:We created a symptom-oriented principle and detailed neurobehavioral scoring criteria.At different time points of reperfusion,we analyzed the alteration in locomotion by rotarod test and grip force score,infarct volume by triphenyltetrazo-lium chloride(TTC)staining,neuron loss using Nissl staining,and histological charac-teristics using hematoxylin-eosin(H&E)straining.Results:With a successful model rate of 56%,32 of the 57 gerbils operated by our method harbored typical features of cerebral I/R injury,and the mortality rate in the male gerbils was significantly higher than that in the female gerbils.The suc-cessfully prepared I/R gerbils demonstrated a significant reduction in motility and grip strength at 1 day after reperfusion;formed obvious infarction;exhibited typi-cal pathological features,such as tissue edema,neuronal atrophy and death,and vacuolated structures;and were partially recovered with the extension of reperfu-sion time.Conclusion:This study developed a new method for the unilateral common carotid artery ligation I/R model of gerbil and established a standardized evaluation system for this model,which could provide a new cerebral I/R model of gerbils with more practical applications.

提出了一种用于测量材料介电常数的微波传感器,其特点是成本低、测量精度高。在传统的互补裂环结构上进行优化设计,实现了更大范围的测量区域;并加载了金属通孔结构,将电场限制在测量区域内,达到了更好的测量效果。结果显示传感器的测量灵敏度为2.89%。传感器在空载时谐振频率为2GHz;当样品加载到测量区域时,测量区域的电场将发生变化,从而导致频移。选取了介电常数值为1~10的样品进行了模拟加载试验,得到了介电常数与谐振频率的拟合经验关系式,同时采用5种常见的高频板材对拟合公式进行了验证。结果表明:传感器具有很高的测量精度,整体测量误差为0.8%,证实了利用该传感器测量材料介电常数的可行性。

目的基于核转录因子红系2相关因子2(Nrf2)/血红素加氧酶1(HO-1)/依赖还原型辅酶/醌氧化还原酶1(NQO1)信号通路探究芍药苷对妊娠期糖尿病(GDM)大鼠糖代谢、炎症、氧化应激的改善作用及潜在机制。方法将以高脂高糖饲料喂养的雌性大鼠和以普通饲料喂养的雄性大鼠合笼,收集妊娠期大鼠,单次腹腔注射链脲佐菌素复制GDM模型。将造模成功的大鼠随机分为模型组,盐酸二甲双胍组(灌胃200 mg/kg盐酸二甲双胍),芍药苷低、高剂量组(分别灌胃45、90 mg/kg芍药苷),芍药苷+ML385组(灌胃90 mg/kg芍药苷并腹腔注射30 mg/kg Nrf2抑制剂ML385),每组12只;另取以普通饲料喂养的妊娠期大鼠12只,作为对照组。各组大鼠给予相应药物/生理盐水,每天1次,连续干预2周。检测其糖代谢指标[空腹血糖(FBG)、空腹胰岛素(FINS)、胰岛素抵抗指数(HOMA-IR)]、血清炎症因子[白细胞介素6(IL-6)、肿瘤坏死因子α(TNF-α)]和肾组织氧化应激指标[超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)]水平,观察肾组织病理变化,并检测肾组织中Nrf2、HO-1、NQO1蛋白的表达情况。结果与对照组相比,模型组大鼠肾组织病变明显,可见肾小球萎缩、肾小管上皮细胞水肿变性和大量炎症细胞浸润;FBG、FINS水平,HOMA-IR,血清IL-6、TNF-α水平,以及肾组织中MDA水平均显著升高(P<0.05);肾组织中SOD、GSH-Px水平和Nrf2、HO-1、NQO1蛋白的表达水平均显著降低(P<0.05)。与模型组相比,芍药苷低、高剂量组大鼠肾组织病变有所减轻,上述各定量指标均显著改善,且芍药苷高剂量组的改善效果更优(P<0.05);ML385可显著逆转芍药苷对上述指标的改善作用(P<0.05)。结论芍药苷可改善GDM大鼠的糖代谢异常、炎症和肾组织氧化应激损伤,上述作用可能与激活Nrf2/HO-1/NOQ1信号通路有关。
