Oxygen tension modulates cell function in an in vitro three-dimensional glioblastoma tumor model

Hypoxia is a typical feature of the tumor microenvironment,one of the most critical factors affecting cell behavior and tumor ***,the lack of tumor models able to precisely emulate natural brain tumor tissue has impeded the study of the effects of hypoxia on the progression and growth of tumor *** study reports a three-dimensional(3D)brain tumor model obtained by encapsulating U87MG(U87)cells in a hydrogel containing type I *** also documents the effect of various oxygen concentrations(1%,7%,and 21%)in the culture environment on U87 cell morphology,proliferation,viability,cell cycle,apoptosis rate,and ***,it compares two-dimensional(2D)and 3D *** comparison purposes,cells cultured in flat culture dishes were used as the control(2D model).Cells cultured in the 3D model proliferated more slowly but had a higher apoptosis rate and proportion of cells in the resting phase(G0 phase)/gap I phase(G1 phase)than those cultured in the 2D ***,the two models yielded significantly different cell ***,hypoxia(e.g.,1%O2)affected cell morphology,slowed cell growth,reduced cell viability,and increased the apoptosis rate in the 3D *** results indicate that the constructed 3D model is effective for investigating the effects of biological and chemical factors on cell morphology and function,and can be more representative of the tumor microenvironment than 2D culture *** developed 3D glioblastoma tumor model is equally applicable to other studies in pharmacology and pathology.

A novel LAAS pseudo-range error over-bound method based on improved pseudo-range error distribution model

The high level of safety demand of civil aviation requests local area augmentation system （LAAS） extremely high navigation integrity performance. A new LAAS pseudo-range error overbound method is proposed in this paper to improve the integrity of LAAS. Firstly, a more practical pseudo-range error distribution model is established. Then, by calculating the relationship between the statistical uncertainty of the model parameter and the integrity risk, a new method is proposed to calculate the pseudo-range error over-bound model. This method can effectively reduce the inflation factor and the resulting conservativeness of the over-bound model. Comparative experiments show that the method proposed in this paper performs better and satisfies the requirements of real applications.

Alzheimer’s disease is the most frequent form of dementia characterized by the deposition of amyloid-beta plaques and neurofibrillary tangles consisting of hyperphosphorylated *** amyloid-beta plaques has been a primary direction for developing Alzheimer’s disease treatments in the last ***,existing drugs targeting amyloid-beta plaques have not fully yielded the expected results in the clinic,necessitating the exploration of alternative therapeutic *** evidence unravels that astrocyte morphology and function alter in the brain of Alzheimer’s disease patients,with dysregulated astrocytic purinergic receptors,particularly the P2Y1 receptor,all of which constitute the pathophysiology of Alzheimer’s *** receptors are not only crucial for maintaining normal astrocyte function but are also highly implicated in neuroinflammation in Alzheimer’s *** review delves into recent insights into the association between P2Y1 receptor and Alzheimer’s disease to underscore the potential neuroprotective role of P2Y1 receptor in Alzheimer’s disease by mitigating neuroinflammation,thus offering promising avenues for developing drugs for Alzheimer’s disease and potentially contributing to the development of more effective treatments.

Electroless plating of Ni-Zn(Fe)-P alloy on carbon steel sheets

The autocatalytic deposition of Ni-Zn(Fe)-P alloys were carried out on carbon steel sheets from bath containing nickel sulfate, zinc sulfate, sodium hypophosphite, sodium citrate and boric acid. The effects of pH and mole ratio of NiSO4/ZnSO4 on the deposition rate and the composition of deposit were studied. It is found that the presence of zinc sulfate in the bath has an inhibitory effect on the alloy deposition. As a consequence, the mole fraction of zinc in the deposits never reaches high value, which is less than 18.0%. The structure and surface morphology of Ni-Zn(Fe)-P coatings were charactered by XRD and SEM. The alloys plated at all conditions consist of amorphous phase coexisting with a crystalline cubic Ni phase. The surface morphology of coating is dependent on the deposition parameters.

Experimental and Computational Analysis of the Unstable Flow Structure in a Centrifugal Compressor with a Vaneless Diffuse

The unstable flow phenomena in compressors, such as stall and surge, are closely related to the e ciency and the operating region. It is indispensable to capture the unstable flow structure in compressors and understand the mechanism of flow instability at low flow rates. Cooperated with the manufacturer, an industrial centrifugal compressor with a vaneless di user is tested by its performance test rig and our multi-phase dynamic measurement system. Many dynamic pressure transducers are circumferentially mounted on the casing surface at seven radial locations, spanning the impeller region and the di user inlet region. The pressure fields from the design condition to surge are measured in details. Based on the multi-phase dynamic signals, the original location of stall occurring can be determined. Meanwhile, the information of the unstable flow structure is obtained, such as the circumferential mode and the propagating speed of stall cells. To get more details of the vortex structure, an unsteady simulation of this tested compressor is carried out. The computational result is well matched with the experimental result and further illustrates how the unstable flow structure in the impeller region gradually a ects the stability of the total machine at low flow rates. The dynamic mode decomposition(DMD) method is applied to get the specific flow pattern corresponding to the stall frequency. Both experimental and computational analysis show that the flow structure at a particular radial location in the impeller region has a great impact on the stall and surge. Some di erences between the computational and experimental result are also discussed. Through these two main analytical methods, an insight into the unstable flow structure in an industrial compressor is gained. The result also plays a crucial role in the guidance of the compressor stabilization techniques.

Predicting Resting-state Functional Connectivity With Efficient Structural Connectivity

The complex relationship between structural connectivity(SC) and functional connectivity(FC) of human brain networks is still a critical problem in neuroscience. In order to investigate the role of SC in shaping resting-state FC, numerous models have been proposed. Here, we use a simple dynamic model based on the susceptible-infected-susceptible(SIS) model along the shortest paths to predict FC from SC. Unlike the previous dynamic model based on SIS theory, we focus on the shortest paths as the principal routes to transmit signals rather than the empirical structural brain network. We first simplify the structurally connected network into an efficient propagation network according to the shortest paths and then combine SIS infection theory with the efficient network to simulate the dynamic process of human brain activity. Finally, we perform an extensive comparison study between the dynamic models embedded in the efficient network, the dynamic model embedded in the structurally connected network and dynamic mean field(DMF) model predicting FC from SC. Extensive experiments on two different resolution datasets indicate that i) the dynamic model simulated on the shortest paths can predict FC among both structurally connected and unconnected node pairs; ii) though there are fewer links in the efficient propagation network, the predictive power of FC derived from the efficient propagation network is better than the dynamic model simulated on a structural brain network; iii) in comparison with the DMF model,the dynamic model embedded in the shortest paths is found to perform better to predict FC.

Analysis of Allele Specific Expression——A Survey

Allele specific expression is essential for cellular programming and development and the diversity of cellular phenotypes. Traditional analysis methods utilize RNA and depend on single nucleotide polymorphisms,thus to suffer from limited amount of materials for analysis. The rapid development of next-generation sequencing technologies provides more comprehensive and powerful approaches to analyze the genomic, epigenetic, and transcriptomic data, and further to detect and measure allele specific expressions. It will potentially enhance the understanding of the allele specific expressions, their complexities, and the effect on biological processes. In this paper, we extensively review the state-of-art enabling technologies and tools to analyze, detect, and measure allele specific expressions, compare their features, and point out the future trend of the methods.

Assessing the corrosion protection property of coatings loaded with corrosion inhibitors using the real-time atmospheric corrosion monitoring technique

The atmospheric corrosion monitoring(ACM)technique has been widely employed to track the real-time corrosion behavior of metal ***,limited studies have applied ACM to the corrosion protection properties of organic *** study compared a bare epoxy coating with one containing zinc phosphate corrosion inhibitors,both applied on ACM sensors,to observe their corrosion protection properties over *** with artificial damage via scratches were exposed to immersion and alternating dry and wet environments,which allowed for monitoring galvanic corrosion currents in *** the corrosion tests,the ACM currents of the zinc phosphate/epoxy coating were considerably lower than those of the blank epoxy *** trend in ACM current variations closely matched the results obtained from regular electrochemical tests and surface *** alignment highlights the potential of the ACM technique in evaluating the corrosion protection capabilities of organic *** with the blank epoxy coating,the zinc phosphate/epoxy coating showed much-decreased ACM current values that confirmed the effective inhibition of zinc phosphate against steel corrosion beneath the damaged coating.

Postnatal development of rat retina:a continuous observation and comparison between the organotypic retinal explant model and in vivo development

The organotypic retinal explant culture has been established for more than a decade and offers a range of unique advantages compared with in vivo experiments and cell ***,the lack of systematic and continuous comparison between in vivo retinal development and the organotypic retinal explant culture makes this model controversial in postnatal retinal development ***,we aimed to verify the feasibility of using this model for postnatal retinal development studies by comparing it with the in vivo *** this study,we showed that postnatal retinal explants undergo normal development,and exhibit a consistent structure and timeline with retinas in ***,we used SOX2 and PAX6 immunostaining to identify retinal progenitor *** then examined cell proliferation and migration by immunostaining with Ki-67 and doublecortin,***-67-and doublecortin-positive cells decreased in both in vivo and explants during postnatal retinogenesis,and exhibited a high degree of similarity in abundance and distribution between ***,we used Ceh-10 homeodomain-containing homolog,glutamate-ammonia ligase(glutamine synthetase),neuronal nuclei,and ionized calcium-binding adapter molecule 1 immunostaining to examine the emergence of bipolar cells,Müller glia,mature neurons,and microglia,*** timing and spatial patterns of the emergence of these cell types were remarkably consistent between in vivo and explant *** study showed that the organotypic retinal explant culture model had a high degree of consistency with the progression of in vivo early postnatal retina *** findings confirm the accuracy and credibility of this model and support its use for long-term,systematic,and continuous observation.
