Comparative transcriptomic analysis reveals the molecular changes of acute pancreatitis in experimental models

BACKGROUND Acute pancreatitis(AP)encompasses a spectrum of pancreatic inflammatory conditions,ranging from mild inflammation to severe pancreatic necrosis and multisystem organ *** the challenges associated with obtaining human pancreatic samples,research on AP predominantly relies on animal *** this study,we aimed to elucidate the fundamental molecular mechanisms underlying AP using various AP *** To investigate the shared molecular changes underlying the development of AP across varying severity *** AP was induced in animal models through treatment with caerulein alone or in combination with lipopolysaccharide(LPS).Additionally,using Ptf1αto drive the specific expression of the hM3 promoter in pancreatic acinar cells transgenic C57BL/6J-hM3/Ptf1α(cre)mice were administered Clozapine N-oxide to induce ***,we conducted RNA sequencing of pancreatic tissues and validated the expression of significantly different genes using the Gene Expression Omnibus(GEO)*** Caerulein-induced AP showed severe inflammation and edema,which were exacerbated when combined with LPS and accompanied by partial pancreatic tissue *** with the control group,RNA sequencing analysis revealed 880 significantly differentially expressed genes in the caerulein model and 885 in the caerulein combined with the LPS *** Encyclopedia of Genes and Genomes enrichment analysis and Gene Set Enrichment Analysis indicated substantial enrichment of the TLR and NOD-like receptor signaling pathway,TLR signaling pathway,and NF-κB signaling pathway,alongside elevated levels of apoptosis-related pathways,such as apoptosis,P53 pathway,and phagosome *** significantly elevated genes in the TLR and NOD-like receptor signaling pathways,as well as in the apoptosis pathway,were validated through quantitative real-time PCR experiments in animal *** from the GEO database revealed that only MYD88 concurred in both mouse panc

为评估水体生态健康,量化和确定供水输入的浮游藻类群落对受水水体浮游藻类生长的影响程度具有重要意义。该研究在不同季节的调水期测定了供、受水水体的理化因子,同时利用藻类生长潜力试验比较和分析了不同比例的供、受水水体混合处理后浮游藻类细胞密度和群落结构变化。结果显示,调水期间受水水库的氮磷营养盐浓度没有显著变化(P>0.05)。不同季节的藻类生长潜力试验显示,夏、冬季处理组的浮游藻类细胞密度峰值时间比对照组提前了4 d,且密度峰值随着混合比例的提高而增大;但夏季培养末期(20 d)处理组的浮游藻类密度均低于对照组,同时夏、秋和冬季处理组的实际藻类密度峰值均低于理论估算值,分别低10%、17%~24%和31%。处理组的群落结构变化表现为夏季受水水库的绝对优势种转变为输入的优势种,秋、冬季绝对优势种的相对丰度随调水比例的提高而增加,明显受到供水优势种生长的影响。供、受水水体的营养盐浓度差异是影响藻类生长的主要因素,持续调水显著影响受水水体浮游藻类的群落结构,但在一定程度上降低了发生藻类水华的风险。

Simple bone cysts in children treated with intracystic fibrin sealant injection

Simple bone cyst （SBC）, also known as a unicameral or solitary cyst, is a benign fluid-filled cavity found primarily at the proximal ends of long bones in children. Treatment is warranted if the pain or a pathologic fracture occurs or if a child is at risk of pathologic fractures. The spectrum of treatments proposed for SBC management ranges from simple Kirschner wire drill-hole, resection or curettage with or without bone grafting to using a cannulated screw for continuous decompression, steroid injection and the latest experience of the use of percutaneous autologous marrow grafting. However, clinical practice of these treatments is limited because of their unsatisfactory effect and possible complications.

Role of gut microbiota on intestinal barrier function in acute pancreatitis

Acute pancreatitis(AP)is a common gastrointestinal ***15%-20%of patients develop severe *** inflammatory response syndrome and multiple organ dysfunction syndrome may be caused by the massive release of inflammatory cytokines in the early stage of severe AP,followed by intestinal dysfunction and pancreatic necrosis in the later stage.A study showed that 59%of AP patients had associated intestinal barrier injury,with increased intestinal mucosal permeability,leading to intestinal bacterial translocation,pancreatic tissue necrosis and infection,and the occurrence of multiple organ dysfunction ***,the real effect of the gut microbiota and its metabolites on intestinal barrier function in AP remains *** review summarizes the alterations in the intestinal flora and its metabolites during AP development and progression to unveil the mechanism of gut failure in AP.

OBJECTIVE:To investigate the impact of Yemazhui(Herba Eupatorii lindleyani,HEL)against lipopolysaccharide(LPS)-induced acute lung injury(Ali)and explore its underlying mechanism in ***:The chemical constituents of HEL were analyzed by ultra-high performance liquid chromatographyquadrupole time-of-flight mass spectrometry ***,HEL was found to suppress LPS-induced Ali in ***-week-old male Sprague-Dawley rats were randomly divided into 6 groups:control,LPS,Dexamethasone(Dex),HEL low dose 6 g/kg(HEL-L),HEL medium dose 18 g/kg(HEL-M)and HEL high dose 54 g/kg(HEL-H)*** model rats were intratracheally injected with 3 mg/kg LPS to establish an Ali *** counts,lung wet/dry weight ratio,as well as myeloperoxidase(MPO)activity were determined followed by the detection with hematoxylin and eosin staining,enzyme linked immunosorbent assay,quantitative real time polymerase chain reaction,western blotting,immunohistochemistry,and ***,to explore the effect of HEL on Ali-mediated intestinal flora,we performed 16s rRNA sequencing analysis of intestinal ***:HEL attenuated LPS-induced inflammation in lung tissue and intestinal flora *** study indicated that HEL suppressed the lung coefficient and wet/dry weight ratio of LPS-induced Ali in rats,inhibited leukocytes exudation and MPO activity,and improved the pathological injury of lung *** addition,HEL reduced the expression of tumor necrosis factoralpha,interleukin-1beta(IL-1β)and interleukin-6(IL-6)in bronchoalveolar lavage fluid and serum,and inhibited nuclear displacement of nuclear factor kappa-B p65(NF-κBp65).And 18 g/kg HEL also reduced the expression levels of toll-like receptor 4(TLR4),myeloid differentiation factor 88,NF-κBp65,phosphorylated inhibitor kappa B alpha(phospho-IκBα),nod-like receptor family pyrin domain-containing 3 protein(NLRP3),IL-1β,and interleukin-18(IL-18)in lung tissue,and regulated intestinal flora disturban

Incidence of congenital heart disease in Beijing, China

Background The incidence of congenital heart disease has been studied in developed countries for many years, but rarely in the mainland of China. Fetal echocardiographic screening for congenital heart disease was first performed in Beijing in the early 2000s, but the impact was not clear. The current study was undertaken to determine the incidence of congenital heart disease in Beijing, China and to estimate the impact of fetal echocardiography on the incidence of liveborn congenital heart disease.Methods The study involved all infants with congenital heart disease among the 84 062 total births in Beijing during the period of January 1 and December 31, 2007. An echocardiographic examination was performed on every baby suspected to have congenital heart disease, prenatally or/and postnatally.Results A total of 686 infants were shown to have congenital heart disease among 84 062 total births. The overall incidence was 8.2/1000 total births. Mothers of 128 of 151 babies diagnosed prenatally were chosen to terminate the pregnancy. Two of the 151 infants died in utero. A specific lesion was identified for each infant and the frequencies of lesions were determined for each class of infants （total births, stillbirths and live births）. The incidence of congenital heart disease in stillbirths and live births was 168.8/1000 and 6.7/1000, respectively. The difference between the incidence of total birth and the incidence of live birth was statistically significant （P 〈0.001）. Conclusions The incidence of liveborn congenital heart disease in Beijing is within the range reported in developed countries. Fetal echocardiography reduce significantly the incidence of liveborn congenital heart disease.

P-xylene(p-C_(8)H_(10))is extremely harmful and dangerous to human health due to high toxicity and strong *** sensitive material to effectively detect p-xylene is of *** this paper,perovskite single crystal(C_(4)H_(9)NH_(3))_(2)PbI_(4) has been successfully synthesized via solution *** obtained product was analyzed by single crystal X-ray *** the space group Pbca,orthorhombic(C_(4)H_(9)NH_(3))_(2)PbI_(4) layered perovskite structure consists of an extended two-dimensional network of corner-sharing PbI_(6) *** layer perovskite sheets of distorted PbI_(6) octahedron alternated with protonated n-butylammonium cation bilayers,which offers many advantages and provides the possibility of forming a gas sensor device based on the change of *** functional theory(DFT)simulations regarding the adsorption energy revealed that this organicinorganic hybrid perovskite compound has excellent selectivity toward p-xylene compared with other gases including C_(2)H_(5)OH,C_(6)H_(6),CH_(2)Cl_(2),HCHO,CH_(3)COCH_(3) and C_(7)H_(8).The calculation of electron density,density of states and electron density difference showed the sensing mechanism of p-C_(8)H_(10) is mainly derived from physical adsorption-desorption in view of electron transfer.

Gully morphology is an important part of loess landform *** with gully development,the variation of its cross section is the most significant,and it can intuitively reflect the characteristics of the lateral widening of the gully ***,in-depth research of the variation of the cross-sectional morphology of the gully is helpful to understanding the development process of the loess *** on the DEMs(Digital Elevation Model)of nine periods of an indoor simulated loess small watershed,this paper studies the evolution model of a complete branch ditch in the watershed using the digital terrain analysis theory and *** show that with the development of the gully,the average gradient of the gully slope continuously decreases,and the slope morphology is mostly a concave slope along the slope *** degree of downward concave first increases and then gradually tends to be *** gully erosion mode is gradually transformed from downward cutting erosion to lateral *** more mature the gully development,the lower the depth of gully bottom cutting is compared with the width of gully ***,the surface cutting depth tends to be stable and the slope is ***,the transformation law of the slope morphology of the gully cross section with the development of the gully is studied,and the prediction model of the transformation of the slope morphology of the gully cross section is established by using the Markov *** Markov model can better reflect the dynamic change of the slope morphology of the gully cross section,which is of great significance to revealing the external performance and internal mechanism of the gully morphology.
