Dehydroabietic acid chemosensitizes drug-resistant acute lymphoblastic leukemia cells by downregulating survivin expression

Objective:To explore the mechanism of drug resistance in acute lymphoblastic leukemia and the anti-tumor effect of combination therapy of dehydroabietic acid and vincristine against acute lymphoblastic leukemia ***:Acute lymphoblastic leukemia cells REH and CCRFCEM were employed to detect the anti-tumor effect of vincristine and doxorubicin on proliferation and apoptosis using EdU assay,human active caspase-3 Quantikine ELISA kit,and flow ***-resistant REH cells(REH-R),survivin knockdown and overexpressing REH cells were established to verify the role of survivin in drug ***,in vitro and in vivo assays were performed to determine the effect of dehydroabietic acid on the cytotoxicity of ***:Vincristine and doxorubicin markedly suppressed proliferation and induced apoptosis of REH and CCRF-CEM *** expression was upregulated in REH-R cells compared with REH *** of survivin expression obviously restored the sensitivity of REH-R cells to *** phosphorylation was also increased in REH-R cells compared to REH *** addition,LY294002,a PI3k/Akt pathway blocker,inhibited survivin expression and enhanced cytotoxicity of vincristine to REH-R *** acid effectively reduced survivin expression in REH-R cells,thereby enhancing the therapeutic effect of vincristine on drug-resistant *** overexpression markedly reduced the effect of dehydroabietic acid on enhancing the anti-proliferation and inducing apoptosis effect of ***,the combination of dehydroabietic acid with vincristine significantly extended the survival rate in a mouse xenograft model of acute lymphoblastic leukemia,compared with vincristine treatment ***:Dehydroabietic acid may be used as a potential candidate for the treatment of acute lymphoblastic leukemia in combination with vincristine.

Combined Transplantation of Neural Stem Cells and Olfactory Ensheathing Cells Improves the Motor Function of Rats with Intracerebral Hemorrhage

Objective To investigate the effects of combined transplantation of neural stem cells （NSC） and olfactory ensheathing cells （OEC） on the motor function of rats with intracerebral hemorrhage. Methods In three days after a rat model of caudate nucleus hemorrhage was established, NSCs and OEC, NSC, OEC （from embryos of Wistar rats） or normal saline were injected into bematomas of rats in combined transplantation group, NSC group, OEC group, and control group, respectively. Damage of neural function was scored before and in 3, 7, 14, 30 days after operation. Tissue after transplantation was observed by immunocytochemistry staining. Results The scores for the NSC, OEC and co-transplantation groups were significantly lower in 14 and 30 days after operation than in 3 days after operation （P〈0.05）. The scores for the NSC and OEC groups were significantly lower than those for the control group only in 30 days after operation （P〈0.05）, while the difference for the NSC-OEC group was significant in 14 days after operation （P〈0.05）. Immunocytochemistry staining revealed that the transplanted OEC and NSC could survive, migrate and differentiate into neurons, astrocytes, and oligodendrocytes. The number of neural precursor cells was greater in the NSC and combined transplantation groups than in the control group. The number of neurons differentiated from NSC was significantly greater in the co-transplantation group than in the NSC group. Conclusion Co-transplantation of NSC and OEC can promote the repair of injured tissue and improve the motor fimction of rats with intracerebral hemorrhage.

ATM Activation is Key in Vasculogenic Mimicry Formation by Glioma Stem-like Cells

Objective Vasculogenic mimicry(VM)is a novel vasculogenic process integral to glioma stem cells(GSCs)in glioblastoma(GBM).However,the relationship between VM and ataxia-telangiectasia mutated(ATM)serine/threonine kinase activation,which confers chemoradiotherapy resistance,remains *** We investigated VM formation and phosphorylated ATM(pATM)levels by CD31/GFAPperiodic acid-Schiff dual staining and immunohistochemical staining in 145 GBM *** stem-like cells(GSLCs)derived from the formatted spheres of U87 and U251 cell lines and their pATM level and VM formation ability were examined using western blot and three-dimensional *** the examination of the function of pATM in VM formation by GSLCs,ATM knockdown by shRNAs and deactivated via ATM phosphorylation inhibitor KU55933 were *** VM and high pATM expression occurred in 38.5% and 41.8% of tumors,respectively,and were significantly associated with reduced progression-free and overall *** with VM-positive GBMs exhibited higher pATM levels(r_(s)=0.425,P=0.01).The multivariate analysis established VM as an independent negative prognostic factor(P=0.002).Furthermore,GSLCs expressed high levels of pATM and formed vascular-like networks in *** inactivation or knockdown hindered VM-like network formation concomitant with the downregulation of pVEGFR-2,VE-cadherin,and laminin *** VM may predict a poor GBM prognosis and is associated with pATM *** propose that pATM promotes VM through extracellular matrix modulation and VE-Cadherin/pVEGFR-2 activation,thereby highlighting ATM activation as a potential target for enhancing anti-angiogenesis therapies for GBM.

Pyrolysis Empirical Modeling of Polyester Glass Fiber Reinforced Plastics Using Sestak-Berggren Model Method

Polyester Glass Fiber Reinforced Plastic(Polyester GFRP),a thermosetting plastic comprised of glass fiber and polyester polymer compounds,is extensively utilized in high-speed *** its pyrolysis mechanism is crucial as it significantly influences the combustion characteristics and fire safety ***,kinetic research on polyester GFRP primarily focuses on employing the Coats-Redfern method to derive a theoretical kinetic ***,the pyrolysis process of polyester GFRP is complex and the aforementioned theoretical model fails to accurately describe the pyrolysis ***,this study seeks to utilize the Sestak and Berggren(SB) model as a methodological approach to reveal the complex reaction mechanism during the pyrolysis *** on thermogravimetric analysis,the entire pyrolysis process of polyester GFRP is divided into two primary ***,model-free methods are employed to ascertain the activation energy and pre-exponential *** results show that the fitted empirical models of the two main pyrolysis stages are f(α)=(1-α)^(1.47)[-ln(1-α)]^(1.50) and f(α)=(1-α)^(1.77)[-ln(1-α)]^(1.72),*** predicted results are in good agreement with experimental data under different heating rates,which indicates that the empirical model can sufficiently describe the pyrolysis process of polyester GFRP.

Tailoring sulfur-doped and nitrogen-defective carbon nitride nanosheets for efficient photo-controlled radical polymerizations

Despite the challenges that remain,the synergistic adjustment of various microstructures and photochemical parameters of graphitic carbon nitride(g-C_(3)N_(4))in photocatalytic reactions holds promises for improving catalytic efficiency and reducing energy ***,sulfur-doped and nitrogen-defective g-C_(3)N_(4)(n-SC_(3)N_(x))nanosheets were designed and elaborately *** resultant n-SC_(3)N_(x)possessed a precisely defined 2D layer structure with extensive porosity and incremental specific surface *** photoinduced electron transfer-reversible addition-fragmentation chain transfer(PET-RAFT)polymerization of vinyl monomers with low dispersity,excellent temporal control and high chain-end fidelity was achieved under mild blue light irradiation in a nondegassed *** to their ultrathin nanostructures with nitrogen defects and sulfur dopants,n-SC_(3)N_(x)was capable of catalyzing RAFT polymerization in aqueous solutions at significantly accelerated rates,which were nearly 8 times faster compared to bulk g-C_(3)N_(4).The ease of separation and efficient reusability in subsequent polymerizations was enabled by the heterogeneous nature of n-SC_(3)N_(x).The appeal of this approach was illustrated by the fact that utilizing a reusable and metal-free photocatalyst in aqueous environments allowed for the synthesis of polymers with molecular weight up to 300 kg mol^(-1) and a dispersity of 1.32.

Numerical Analysis of Surcharge Effect on Stability and Interaction Mechanism of Slope-Pile-Footing System

To investigate the stability and interaction mechanism of the slope-pile-footing system under surcharge effects,the finite difference method(FDM)was adopted to analyze the response laws of the stability of the reinforced slope,evolution of the critical slip surface,stress characteristic of retaining structures,deformation and failure modes of the slope foundation and building footing under surcharge parameters,including the surcharge intensity,the surcharge position,and the surcharge *** results show that surcharge parameters significantly affect the stability and the deformation characteristics of the slope-pile-footing *** speaking,with the increasing surcharge intensity and the decreasing surcharge position and width,the deformation and failure mode of the system will gradually evolve in a direction that is harmful to its *** interaction mechanism of the slope-pile-footing system is further clarified as the load transfer of the building footing,the generation of the additional stress in the slope foundation,and the adjustment of pile bending moment due to the stress ***,the safety of anti-slide piles will determine the stability of the slope foundation and building *** findings are expected to provide guidance for the comprehensive development and utilization of filled slopes after reinforcement.
