Tomato(Solanum lycopersicum) stress resistance and fruit total soluble solid(TSS) content have changed dramatically during selective breeding, and transcriptome variation has played a critical role in this rewiring. However, the single tomato reference genome impedes characterization of whole-transcriptome variation during domestication and breeding at the population level. Here, we constructed a pan-transcriptome of orange-stage tomato fruit, and investigated global expression presence/absence variation(e PAV) and differentially expressed genes(DEGs) based on RNA sequencing(RNA-seq) data from 399 tomato accessions. A total of 7 181 genes absent from the reference genome were identified, 6 122 of which were e PAV genes during tomato domestication and breeding including resistance genes such as late blight resistance gene PIM_DN29746_c0_g3_i1 and peroxidase P7-like gene PIM_DN30274_c0_g2_i1. In addition, 3 629 genes were significantly differentially expressed during tomato selection, among which 19 genes were associated with the reduced fruit TSS content of modern tomato cultivars, including LIN5, TIV1, and seven novel sugar transporter genes. Our results indicate that natural and artificial selection greatly shaped the tomato transcriptome, thereby altering the fruit TSS content and resistance to abiotic and biotic stresses.

Bond strength of different adhesives to normal and caries-affected dentins

Background Currently, several systems of dentin substrate-reacting adhesives are available for use in the restorative treatment against caries. However, the bond effectiveness and property of different adhesive systems to caries-affected dentin are not fully understood. The objective of this study was to evaluate the bond strength of different adhesives to both normal dentin （ND） and caries-affected dentin （CAD） and to analyze the dentin/adhesive interracial characteristics. Methods Twenty eight extracted human molars with coronal medium carious lesions were randomly assigned to four groups according to adhesives used. ND and CAD were bonded with etch-and-rinse adhesive AdperTM Single Bond 2 （SB2） or self-etching adhesives Clearfil SE Bond （CSE）, Clearfil S3 Bond （CS3）, iBond GI （IB）. Rectangular sticks of resin-dentin bonded interfaces 0.9 mm2 were obtained. The specimens were subjected to microtensile bond strength （pTBS） testing at a crosshead speed of 1 mm/min. Mean pTBS was statistically analyzed with analysis of variance （ANOVA） and Student-Newman-Keuls tests. Interfacial morphologies were analyzed by Scanning Electron Microscopy （SEM）. Results Etch-and-rinse adhesive AdperTM Single Bond 2 yielded high bond strength when applied to both normal and caries-affected dentin. The two-step self-etching adhesive Clearfil SE Bond generated the highest bond strength to ND among all adhesives tested but a significantly reduced strength when applied to CAD. For the one-step self-etching adhesives, Clearfil S3 Bond and iBond GI, the bond strength was relatively low regardless of the dentin type. SEM inteffacial analysis revealed that hybrid layers were thicker with poorer resin tag formation and less resin-filled lateral branches in the CAD than in the ND for all the adhesives tested. Conclusion The etch-and-rinse adhesive performed more effectively to caries-affected dentin than the self-etching adhesives.

全氟烷基酸(PFAAs)是一类在水环境中广泛存在的有机污染物,具有潜在的生物累积性和多种生物毒性.为探究长链PFAAs与其短链替代品在浮游动物大型溞体内的竞争富集作用,通过比较典型长链PFAAs存在和不存在两个暴露条件下短链PFAAs在大型溞体内的生物富集,考察PFAAs在大型溞体内的吸收速率常数(ku)、排出速率常数(ke)和稳态生物浓缩系数(BCFss)等指标的变化规律,探讨典型长链PFAAs与其短链替代品的竞争富集作用机制.结果表明,PFAAs在大型溞体内的ku和BCFss值分别与全氟碳原子数、膜−水分配系数和蛋白质结合系数显著正相关,且长链PFAAs对短链PFAAs的生物富集具有明显抑制作用,导致短链PFAAs的BCFss值降低56%~80%.长链与短链PFAAs在蛋白质结合点位的竞争作用可能是短链PFAAs生物富集受到抑制的机制之一.

Circulatory responses to nasotracheal intubation： comparison of GlideScope videolaryngoscope and Macintosh direct laryngoscope

Background The GlideScope videolaryngoscope （GSVL） has been shown to have no special advantage over the Macintosh direct laryngoscope （MDL） in attenuating the circulatory responses to orotracheal intubation, but no study has compared the circulatory responses to nasotracheal intubation （NTI） using the two devices. This prospective randomized clinical study was designed to determine whether there was a clinically relevant difference between the circulatory responses to NTI with the GSVL and the MDL. Methods Seventy-six adult patients were randomly allocated equally to the GSVL group and the MDL group. After induction of anesthesia, NTI was performed. Non-invasive blood pressure （BP） and heart rate （HR） were recorded before induction （baseline values） and immediately before intubation （post-induction values）, at intubation and every minute for a further five minutes. During the observation, times required to reach the maximum values of systolic BP （SBP） and HR, times required for recovery of SBP and HR to postinduction values and incidence of SBP and HR percent changes 〉 30% of baseline values were also noted. The product of HR and systolic BP, i.e. rate pressure product （RPP）, and the areas under SBP and HR vs. time curves （AUGsBP and AUGHR） were calculated. Results The NTI with the GSVL resulted in significant increases in BP, HR and RPP compared to postinduction values, but these circulatory changes did not exceed baseline values. BPs at all measuring points, AUGSBP, maximum values of BP and incidence of SBP percent increase 〉 30% of baseline value during the observation did not differ significantly between groups. However, HR and RPP at intubation and their maximum values, AUGHR and incidence of HR percent increase 〉 30% of baseline value were significantly higher in the MDL group than in the GSVL group. 采用99Tcm-rh-AnnexinⅤ作为凋亡显像示踪剂,观察小鼠放疗后早期肿瘤组织内Survivin、Caspase-3蛋白表达。结果显示,放疗组肿瘤组织内99Tcm-rh-AnnexinⅤ分布、TUNEL检测阳性细胞数及Caspase-3蛋白表达均明显多于对照组,Survivin蛋白表达A组明显高于B组,差异均存在显著性(P均1)。相关性研究表明,肿瘤组织的放射性摄取与TUNEL阳性细胞数呈明显正相关(r=0.942,P=0.000);肿瘤组织内Survivin蛋白表达与Caspase-3蛋白表达呈明显负相关(r=-0.836,P=0.000)。肿瘤组织内99Tcm-Annex-inⅤ分布与Caspase-3蛋白表达呈明显正相关(r=0.948,P=0.000),与Survivin蛋白表达呈明显负相关(r=-0.819,P1)。以上结果提示,肿瘤组织内99Tcm-rh-AnnexinⅤ的分布可以反映放疗后早期肿瘤组织细胞凋亡的状况以及凋亡调控蛋白Survivin、Caspase-3表达水平的变化。

长江流域不同植被对2022年极端气象干旱事件的响应

基于植被对气象干旱事件的响应特征构建了植被敏感性及脆弱性的评估方法:基于长序列降水及潜在蒸散发数据和日诱导叶绿素荧光指数构建日尺度标准化指数,以2022年长江特大气象干旱事件为例,计算相关系数(CC)、累积响应时间(RT)及响应敏感度(RS)评估植被对干旱响应的敏感性;利用历时响应比(DRR)和烈度响应比(IRR)评估植被对干旱事件响应的脆弱性.结果表明:对于2022年长江流域特大气象干旱事件,(1)上游常绿针叶林及草甸敏感性低(中位CC=0.36,0.44;中位RS=-1.72,-1.58);中下游耕地敏感性最高(中位CC=0.68,中位RS=-1.37),稀树及多树草原敏感性高于林地(中位CC:林地=0.41~0.49,多树草原=0.64,稀树草原=0.61;中位RS:林地=-1.61~-1.57,多树草原=-1.44,稀树草原=-1.40).(2)上游常绿针叶林脆弱性高于其他林地(中位DRR:针叶林=1.04,其他林地=0.85~0.97;中位IRR:针叶林=0.94,其他林地:IRR=0.76~0.88);草甸脆弱性高于中下游多树及稀树草原(中位DRR:草甸=1.28,多树草原=0.74,稀树草原=0.88;中位IRR:草甸=1.24,多树草原=0.74,稀树草原=0.80);耕地脆弱性低(中位DRR=0.77IRR=0.72).(3)空间分布上,上游金沙江流域是高脆弱性热点区域;中下游的四川盆地及汉江流域耕地聚集,是高敏感性热点区域.基于研究结论,当极端干旱事件发生时,可根据不同植被的响应特点开展植被监测和恢复措施,从而保护植被不发生退化和永久性损害.

Spatiotemporal residual noise in terrestrial earth observation products,often caused by unfavorable atmospheric conditions,impedes their broad *** users prefer to use gap-filled remote sensing products with time series reconstruction(TSR)*** currently available implementations of TSR to large-volume datasets is time-consuming and challenging for non-professional users with limited computation or storage *** study introduces a new open-source software package entitled‘HANTS-GEE’that implements a well-known and robust TSR algorithm,*** ANalysis of Time Series(HANTS),on the Google Earth Engine(GEE)platform for scalable reconstruction of terrestrial earth observation *** tasks can be conducted on user-defined spatiotemporal extents when raw datasets are available on *** to site-based and regional-based case evaluation,the new tool can effectively eliminate cloud contamination in the time series of earth observation *** with traditional PC-based HANTS implementation,the HANTS-GEE provides quite consistent reconstruction results for most terrestrial vegetated *** HANTS-GEE can provide scalable reconstruction services with accelerated processing speed and reduced internet data transmission volume,promoting algorithm usage by much broader user *** our knowledge,the software package is thefirst tool to support full-stack TSR processing for popular open-access satellite sensors on cloud platforms.
