Functional analysis of the novel mitochondrial tRNA^(Trp)and tRNA^(Ser(AGY))variants associated with type 2 diabetes mellitus

BACKGROUND Mutations in mitochondrial tRNA(mt-tRNA)genes that result in mitochondrial dysfunction play important roles in type 2 diabetes mellitus(T2DM).We previously reported a large Chinese pedigree with maternally inherited T2DM that harbors novel mt-tRNA^(Trp)A5514G and tRNA^(Ser(AGY))C12237T variants,however,the effects of these mt-tRNA variants on T2DM progression are largely *** To assess the potential pathogenicity of T2DM-associated m.A5514G and m.C12237T variants at genetic,molecular,and biochemical *** Cytoplasmic hybrid(cybrid)cells carrying both m.A5514G and m.C12237T variants,and healthy control cells without these mitochondrial DNA(mtDNA)variants were generated using trans-mitochondrial *** features,including mt-tRNA steady-state level,levels of adenosine triphosphate(ATP),mitochondrial membrane potential(MMP),reactive oxygen species(ROS),mtDNA copy number,nicotinamide adenine dinucleotide(NAD+)/NADH ratio,enzymatic activities of respiratory chain complexes(RCCs),8-hydroxy-deoxyguanine(8-OhdG),malondialdehyde(MDA),and superoxide dismutase(SOD)were examined in cell lines with and without these mt-tRNA *** Compared with control cells,the m.A5514G variant caused an approximately 35%reduction in the steady-state level of mt-tRNA^(Trp)(P<0.0001);however,the m.C12237T variant did not affect the mt-tRNA^(Ser(AGY))steady-state level(P=0.5849).Biochemical analysis revealed that cells with both m.A5514G and m.C12237T variants exhibited more severe mitochondrial dysfunctions and elevated oxidative stress than control cells:ATP,MMP,NAD+/NADH ratio,enzyme activities of RCCs and SOD levels were markedly decreased in mutant cells(P<0.05 for all measures).By contrast,the levels of ROS,8-OhdG and MDA were significantly increased(P<0.05 for all measures),but mtDNA copy number was not affected by m.A5514G and m.C12237T variants(P=0.5942).CONCLUSION The m.A5514G variant impaired mt-tRNA^(Trp)metabolism,which subsequent

Carbon dots-derived three-dimensional-ordered macroporous host with high penetrability and catalytic activity for LSBs under lean electrolyte

The practical energy density of lithium-sulfur batteries(LSBs)is seriously limited by the high electrolyte-to-sulfur ratios(E/S).The E/S can be reduced by employing hosts with easy-to-infiltrate structure that ensures the uniform distribution of a lean electrolyte and high catalytic activity that can suppress"shuttle effect"via accelerating the slow conversion of soluble poly sulfides to insoluble *** the easy-to-infiltrate structures,the three-dimensional-ordered macroporous(3DOM)structure is easier to scale preparation and more suitable for the existing industrial ***,it is difficult to obtain the 3DOM with(1)high penetrability due to the high viscosity of frequently-used organic polymer precursors and(2)high catalytic activity due to the low confinement effect,meaning that uniformly dispersed small-sized catalytic materials are difficult to load on ***,using carbon dots(CDs)with both organic and inorganic properties as a precursor and aggregation limiting agent,a 3DOM host with high penetrability and homodispersed Ni particles of small sizes(Ni-CDs-3DOM)was *** this host:(1)CDs precursor with suitable viscosity can improve the penetrability.(2)CDs can effectively inhibit the agglomeration of Ni particles.(3)Uniformly dispersed small-size Ni particles offer high-efficiency catalytic activity toward sulfur ***,the Ni-CDs-3DOM/sulfur cathode exhibits high sulfur utilization and stable cycling performance even under high sulfur loading(5.5 mg·cm^(-2))and low E/S(6.5μl·mg^(-1)).This work indicates the usefulness of CDs in constructing hosts for LSBs with high energy density.

The room temperature strong coupling between the photonic modes of micro/nanocavities and quantum emitters(QEs) can bring about promising advantages for fundamental and applied *** the electric fields(EFs) by using plasmonic modes and reducing their losses by applying dielectric nanocavities are widely employed approaches to achieve room temperature strong ***,ideal photonic modes with both large EFs and low loss have been ***,we propose the abnormal anapole mode (AAM),showing both a strong EF enhancement of~70-fold (comparable to plasmonic modes) and a low loss of 34 meV,which is much smaller than previous records of isolated all-dielectric *** realizing strong coupling,we further show that by replacing the normal anapole mode with the AAM,the lasing threshold of the AAM-coupled QEs can be reduced by one order of magnitude,implying a vital step toward on-chip integration of nanophotonic devices.

Two polyoxovanadoborates with over 100 nuclei assembled from a {V_(14)Co_(12)B_(72)} cluster

Combining theoretical prediction and experimental exploration, the first sandwich-type polyoxovanadoborate(POVB) with an odd number of nuclei, [V_(11)B_(26)O_(73)(OH)_(10)]^(8-)(1), was discovered under hydrothermal conditions. Density-functional theory(DFT)gives a rationalization for the existence of this structure and the structural feature is consistent with the {V_NB_(2N+X)}(X = 2, 4, 6, 8)general formula. Excitingly, when Co and 1,10-phenanthroline(phen) are introduced into the synthesis of vanadoborates system,a 100-nuclei [Co_6(phen)_(10)(H_(2)O)_8][V_(14)Co_(12)B_(72)O_(144)(OH)_(20)]·28H_(2)O(2) and a 102-nuclei [Co_6(phen)_(14)(H_(2)O)_(4)][V_(14)Co_(12)B_(72)O_(144)(OH)_(20)]·23H_(2)O(3) with the same core structure of [V_(14)Co_(12)B_(72)]^(12-)are obtained. The vase-shaped [V_(14)Co_(12)B_(72)] polyanion is a multilayered sandwich-type structure containing seven distinct structural layers and is the largest known POVB. Furthermore, 2demonstrates exceptional thermal and chemical stability and serves as an efficient and stable catalyst for the photoreduction of carbon dioxide.

Synthesis of uniform two-dimensional MoS_(2) films via thermal evaporation

Two-dimensional(2D)molybdenum disulfide(MoS_(2))holds great potential for various applications such as electronic devices,catalysis,lubrication,anti-corrosion and so *** evaporation is a versatile thin film deposition technique,however,the conventional thermal evaporation techniques face challenges in producing uniform thin films of MoS_(2) due to its high melting temperature of 1375℃.As a result,only thick and rough MoS_(2) films can be obtained using these *** address this issue,we have designed a vacuum thermal evaporation system specifically for large-scale preparation of MoS_(2) thin *** using K2MoS4 as the precursor,we achieved reliable deposition of uniform polycrystalline MoS_(2) thin films with a size of 50 mm×50 mm and controllable thickness ranging from 0.8 to 2.4 *** approach also allows for patterned deposition of MoS_(2) using shadow masks and sequential deposition of MoS_(2) and tungsten disulfide(WS_(2)),similar to conventional thermal evaporation ***,we have demonstrated the potential applications of the obtained MoS_(2) thin films in field effect transistors(FETs),memristors and electrocatalysts for hydrogen evolution reaction(HER).

Radial growth in Qinghai spruce is most sensitive to severe drought in the Qilian Mountains of Northwest China

Global warming and frequent extreme drought events lead to tree death and extensive forest decline,but the underlying mechanism is not *** drought years,cambial development is more sensitive to climate change,but in different phenological stages,the response rela-tionship is ***,the dynamic relationship between tree radial growth and climatic/environmental fac-tors needs to be *** thus continuously monitored radial growth of Qinghai spruce(Picea crassifolia Kom.)and environmental factors from January 2021 to November 2022 using point dendrometers and portable meteorological weather stations in the central area of the Qilian *** relationship and stability between the radial growth of Qinghai spruce and environmental factors were compared for different levels of drought in 2021 and *** year 2022 had higher temperatures and less precipitation and was drier than *** with 2021,the growing period in 2022 for Qinghai spruce was 10 days shorter,maximum growth rate(Grmax)was 4.5μm·d^(-1) slower,and the initiation of growth was 6 days *** of Qinghai spruce was always restricted by drought,and the stem radial increment(SRI)was more sensitive to precipitation and air relative *** changes in cumulative radial growth were divided into four phenological stages according to the time of growth onset,cessation,and maximum growth rate(Grmax)of Qinghai *** responses of SRI to climate change were stronger in Stage 3 and Stage 4 of 2021 and stronger in Stage 1(initiation growth stage)and Stage 3 of *** results provide important information on the growth of the trees in response to drought and for specific managing forests as the climate warms.

p-d Orbital Hybridization Engineered Single-Atom Catalyst for Electrocatalytic Ammonia Synthesis

The rational design of metal single-atom catalysts（SACs） for electrochemical nitrogen reduction reaction（NRR） is challenging. Two-dimensional metal–organic frameworks（2DMOFs） is a unique class of promising SACs. Up to now, the roles of individual metals, coordination atoms, and their synergy effect on the electroanalytic performance remain unclear. Therefore, in this work, a series of 2DMOFs with different metals and coordinating atoms are systematically investigated as electrocatalysts for ammonia synthesis using density functional theory calculations. For a specific metal, a proper metalintermediate atoms p-d orbital hybridization interaction strength is found to be a key indicator for their NRR catalytic activities. The hybridization interaction strength can be quantitatively described with the p-/d-band center energy difference（Δd-p）, which is found to be a sufficient descriptor for both the p-d hybridization strength and the NRR performance. The maximum free energy change（ΔGmax） and Δd-p have a volcanic relationship with OsC4（Se）4located at the apex of the volcanic curve, showing the best NRR performance. The asymmetrical coordination environment could regulate the band structure subtly in terms of band overlap and *** work may shed new light on the application of orbital engineering in electrocatalytic NRR activity and especially promotes the rational design for SACs.

Estimation of surface geometry on combustion characteristics of AP/HTPB propellant under rapid depressurization

The 2D sandwich model serves as a potent tool in exploring the influence of surface geometry on the combustion attributes of Ammonium perchlorate/Hydroxyl-terminated polybutadiene(AP/HTPB)propellant under rapid pressure *** thickness of the sandwich propellant is derived from slicing the 3D random particle packing,an approach that enables a more effective examination of the micro-flame *** analysis of the predicted burning characteristics has been performed with experimental *** findings demonstrate a reasonable agreement,thereby validating the precision and soundness of the *** on the typical rapid depressurization environment of solid rocket motor(initial combustion pressure is 3 MPa and the maximum depressurization rate is 1000 MPa/s).A-type(a flatter surface),B-type(AP recesses from the combustion surface),and C-type(AP protrudes from the combustion surface)propellant combustion processes are numerically *** comparison of the evolution of gas-phase flame between 0.1 and 1 ms,it is discerned that the flame strength and form created by the three sandwich models differ significantly at the beginning stage of depressurization,with the flame structures gradually becoming harmonized over *** are drawn by comparison extinction times:the surface geometry plays a pivotal role in the combustion process,with AP protrusion favoring combustion the most.
