Ribosomal RNAs(rRNAs) provide the structural framework of ribosomes and play critical roles in protein *** ribosome biogenesis,rRNAs acquire various modifications that can influence the structure and catalytic activity of ***,rRNA modifications in plants have yet to be fully ***,we proposed a method to purify rRNAs by a successive isolation with different strategies,including poly A-based m RNA depletion and agarose gel electrophoresis-based purification,with which highly pure rRNAs could be *** addition,we developed a liquid chromatography-electrospray ionization-tandem mass spectrometry(LC-ESI-MS/MS) method to systematically profile and characterize modifications from the isolated highly pure plant 18S rRNA and 25S ***-ESI-MS/MS analysis showed that 10 and 12 kinds of modifications were present in plant 18S rRNA and 25S rRNA,***,among these identified modifications,2 kinds of modifications of N^(2),N^(2)-dimethylguanosine(m^(2,2)G)and N^(6),N^(6)-dimethyladenosine(m^(6,6)A) in 18S rRNA,and 4 kinds of modifications of m^(2,2)G,m^(6,6)A,N7-methylguanosine(m^(7)G) and 3-methyluridin(m^(3)U) in 25S rRNA,were first reported to be present in ***,exposure of Arabidopsis thaliana to cadmium(Cd) led to significant changes of modifications in both 18S rRNA and 25S rRNA of plants,indicating that rRNA modifications play important roles in response to environmental *** discovery of new modifications in plant rRNAs improves the spectra of plant rRNA modifications and may promote the investigation of the functional roles of plant ribosomes in regulating gene expression.

Active demethylation of 5-methylcytosine(5mC)can be realized through ten-eleven translocation(TET)dioxygenase-mediated oxidation of 5mC to 5-hydroxymethylcytosine(5hmC),5-formylcytosine(5fC),and 5-carboxylcytosine(5caC),followed by thymine DNA glycosylase(TDG)-initiated base excision repair(BER).The TDG-BER pathwaymay lead to the generation of DNA strand breaks,potentially compromising genome ***,direct decarboxylation of TET-produced 5caC is highly attractive because this mechanism allows for conversion of 5mC to cytosine without the formation of DNA strand ***,cleavage of the C–C bond in 5caC in human cells remains an open *** examined this reaction in cell extract and live cells using 5caC-carrying hairpin DNA *** incubation with whole-cell protein extract or transfection into human cells,we monitored the transformation of 5caC to cytosine through direct decarboxylation or BER using liquid chromatography–tandem mass spectrometry(LCMS/MS)analyses at both the mononucleotide and oligodeoxynucleotide *** results clearly showed the direct conversion of 5caC to cytosine in human cells,providing evidence to support a novel pathway for active DNA demethylation.

RNA molecules harbor diverse chemical modifications that play important regulatory roles in a variety of biological processes. Up to date, more than 150 modifications have been identified in various RNA species.[1] Most of these modifications occurring in DNA and RNA are the methylation of nucleic acids. It has been demonstrated that many of these methylation modifications are reversible and undergo dynamic demethylation and the hydroxylation of the methylated nucleosides may be a general pathway for the demethylation of DNA and RNA methylation. However, so far few other hydroxylmethylation modification has yet been reported existence in mammals. In the current study, we developed a neutral enzymatic digestion method for the mild digestion of nucleic acids followed by liquid chromatography-electrospray ionization-tandem mass spectrometry（LC-ESI-MS/MS） analysis. With the established method, we reported the existence of a new hydroxylmethylated nucleosides, N2-hydroxymethylguanosine（hm2 G）, in mammalian RNA. Our study suggests that RNA hydroxylmethylation may be a new prevalent group of modifications existing in RNA, which expands the diversity of nucleic acid modifications and should exert regulatory functions in living organisms.

In less than a century,Asia’s largest apex predator,the tiger(Panthera tigris),has been relegated to isolated populations surviving in only a small fraction of its historical *** Medog region,located in Xizang Zizhiqu(Tibet),China,is an important stronghold for this ecologically important apex *** enhance our knowledge of the status of tigers in the Medog region,we carried out a systematic camera trapping survey combined with socioecological data collection between April 2020 and May *** surveyed a 1769 km2 area across the Medog region,with a total valid sampling effort of 43163 camera days from 322 camera *** also collected socioecological data from 27 forest *** documented tigers at eight camera trapping stations at two villages separated by the Jinzhu Tsangpo River,confirming the presence of at least one adult male individual within the Medog *** new records were observed over 80 km northeast of the first record in ***,residents reported tiger sightings in 2020 and likely tiger pugmarks in *** spatially and temporally separate records of tigers in the Medog region provide valuable data for informing species-oriented conservation management and highlight the importance of the region for tiger movement and population ***,although encouraging,our survey also detected human activities that may threaten tigers and their potential *** the future survival of tigers in the Medog region entails anti-poaching patrols and long-term camera trapping *** addition,it is also important to identify and establish corridors that connect the Medog region with the adjacent Zangnan region,which also harbors an important population of tigers.

Manifold regularization(MR)provides a powerful framework for semi-supervised classification using both the labeled and unlabeled *** constrains that similar instances over the manifold graph should share similar classification out-puts according to the manifold *** is easily noted that MR is built on the pairwise smoothness over the manifold graph,i.e.,the smoothness constraint is implemented over all instance pairs and actually considers each instance pair as a single ***,the smoothness can be pointwise in nature,that is,the smoothness shall inherently occur“everywhere"to relate the behavior of each point or instance to that of its close *** in this paper,we attempt to de-velop a pointwise MR(PW_MR for short)for semi-supervised learning through constraining on individual local *** this way,the pointwise nature of smoothness is preserved,and moreover,by considering individual instances rather than instance pairs,the importance or contribution of individual instances can be *** importance can be described by the confidence for correct prediction,or the local density,for *** provides a different way for implementing manifold smoothness Finally,empirical results show the competitiveness of PW_MR compared to pairwise MR.

The interaction of bromothymol blue（BB） with human serum albumin（HSA） was studied by electrochemical techniques and a sensitive method for proteins assay was developed. When BB interacted with HSA, the voltammetric peak current value of BB decreased linearly with the concentration of HSA in a range of 1.0--40.0 mg/L, and the peak potential shifted negatively. Based on the results, a sensitive assay method for proteins, such as HSA, bovine serum albumin（BSA）, and egg albumin etc. was established. This method was further applied to determining the HSA in healthy human blood samples, and the results are not significantly different from those obtained by the classic Coomassie Brilliant Blue G-250 spectrophotometic method. The detecting conditions of this method were optimized and the interaction mechanism was discussed. The results show that the electrochemical parameters（formal potential E^0, standard rate constant of the electrode reaction ks, parameter of kinetic nα） of BB have no obvious changes before and after the interaction, which indicate that BB can interact with HSA, forming an electrochemical non-active complex. The equilibrium constant（βs） and the binding ratio（m） for this complex were calculated. The m is 4 and βs is 1.41 × 10^19. This method is fast, simple, highly sensitive, and has good selectivity, which can be used in clinical measurements.

As one of the top four commercially important species in China,yellow croaker(Larimichthys polyactis)with two geographic subpopulations,has undergone profound changes during the last several *** is widely comprehended that understanding its population dynamics is critically important for sustainable management of this valuable fishery in *** only two existing population dynamics models assessed the population of yellow croaker using short time-series data,without considering geographical *** this study,Bayesian models with and without hierarchical subpopulation structure were developed to explore the spatial heterogeneity of the population dynamics of yellow croaker from 1968 to *** hypotheses were constructed to test potential temporal patterns in yellow croaker’s population *** variations in population dynamics characteristics among space and time were found through this *** population growth rate was revealed to increase since the late 1980s,and the catchability increased more than twice from 1981 to *** East China Sea’s subpopulation witnesses faster growth,but suffers from higher fishing pressure than that in the Bohai Sea and Yellow *** global population and two subpopulations all have high risks of overfishing and being overfished according to the MSY-based reference points in recent *** conservative management strategies with subpopulation considerations are imperative for the fishery management of yellow croaker in *** methodology developed in this study could also be applied to the stock assessment and fishery management of other species,especially for those species with large spatial heterogeneity data.
