Background:Toll-like receptor 5(TLR5)-mediated pathways play critical roles in regulating the hepatic immune response and show hepatoprotective effects in mouse models of hepatic diseases.However,the role of TLR5 in experimental models of liver regeneration has not been reported.This study aimed to investigate the role of TLR5 in partial hepatectomy(PHx)-induced liver regeneration.Methods:We performed 2/3 PHx in wild-type(WT)mice,TLR5 knockout mice,or TLR5 agonist CBLB502 treated mice,as a model of liver regeneration.Bacterial flagellin content was measured with ELISA,and hepatic TLR5 expression was determined with quantitative PCR analyses and flow cytometry.To study the effects of TLR5 on hepatocyte proliferation,we analyzed bromodeoxyuridine(BrdU)incorporation and proliferating cell nuclear antigen(PCNA)expression with immunohistochemistry(IHC)staining.The effects of TLR5 during the priming phase of liver regeneration were examined with quantitative PCR analyses of immediate early gene mRNA levels,and with Western blotting analysis of hepatic NF-κB and STAT3 activation.Cytokine and growth factor production after PHx were detected with real-time PCR and cytometric bead array(CBA)assays.Oil Red O staining and hepatic lipid concentrations were analyzed to examine the effect of TLR5 on hepatic lipid accumulation after PHx.Results:The bacterial flagellin content in the serum and liver increased,and the hepatic TLR5 expression was significantly up-regulated in WT mice after PHx.TLR5-deficient mice exhibited diminished numbers of BrdU-and PCNA-positive cells,suppressed immediate early gene expression,and decreased cytokine and growth factor production.Moreover,PHx-induced hepatic NF-κB and STAT3 activation was inhibited in Tlr5–/–mice,as compared with WT mice.Consistently,the administration of CBLB502 significantly promoted PHx-mediated hepatocyte proliferation,which was correlated with enhanced production of proinflammatory cytokines and the recruitment of macrophages and neutrophils in the liver.Furthermore,Tlr5–/–

Bismuth(Bi),as an alloy-based anode material,has attracted much atte ntion in the developme nt of sodiumion hybrid capacitors(SIHCs)due to its high theoretical capacity.However,the volume expansion of the Bi-based anode during the sodiation/desodiation process results in limited rate capability.In the present work,a porous Bi-based composite was constructed by a one-step hydrothermal method,and Bi was encapsulated in ligninderived nitrogen-doped porous carbon(Bi@LNPC)after carbonization.The obtained Bi nanoparticles could effectively adapt to the strain and shorten the diffusion distance of Na^(+).In addition,porous carbon skeleton provides a rigid conductive network for electronic transportation.Therefore,the assembled sodium-ion half-cell with Bi@LNPC anode shows ultra-high-rate capability.When the current density was enhanced from 0.1 to 50 A·g^(-1),the specific capacity decreased slightly from 351.5 to 342.8 mAh·g^(-1).Even at an extremely high current density of 200 A·g^(-1),it retains 81.3%capacity retention when compared to a current density of 1 A·g^(-1).The SIHCs assembled by Bi@LNPC show a high energy density of 63 Wh·kg^(-1).This work provides an effective method for developing high-rate Bi anode materials for sodium-ion hybrid capacitors(SIHCs)and sodium-ion batteries(SIBs).

BACKGROUND Gastric cancer(GC)is one of the most aggressive malignancies with limited therapeutic options and a poor prognosis.Resveratrol,a non-flavonoid poly-phenolic compound found in a variety of Chinese medicinal materials,has shown excellent anti-GC effect.However,its exact mechanisms of action in GC have not been clarified.AIM To identify the effects of resveratrol on GC progression and explore the related molecular mechanisms.METHODS Action targets of resveratrol and GC-related targets were screened from public databases.The overlapping targets between the two were confirmed using a Venn diagram,and a“Resveratrol-Target-GC”network was constructed using Cyto-scape software version 3.9.1.The protein-protein interaction(PPI)network was constructed using STRING database and core targets were identified by PPI network analysis.The Database for Annotation,Visualization and Integrated A total of 378 resveratrol action targets and 2154 GC disease targets were obtained from public databases,and 181 intersection targets between the two were screened by Venn diagram.The top 20 core targets were identified by PPI network analysis of the overlapping targets.GO function analysis mainly involved protein binding,identical protein binding,cytoplasm,nucleus,negative regulation of apoptotic process and response to xenobiotic stimulus.KEGG enrichment analysis suggested that the involved signaling pathways mainly included PI3K-AKT signaling pathway,MAPK signaling pathway,IL-17 signaling pathway,TNF signaling pathway,ErbB signaling pathway,etc.FBJ murine osteosarcoma viral oncogene homolog(FOS)and matrix metallopeptidase 9(MMP9)were selected by differential expression analysis,and they were closely associated with immune infiltration.Molecular docking results showed that resveratrol docked well with these two targets.Resveratrol treatment arrested the cell cycle at the S phase,induced apoptosis,and weakened viability,migration and invasion in a dose-dependent manner.Furthermore,resveratrol could exhibit anti-GC effect by regula

Cardiovascular computed tomography angiography(CTA)is a widely used imaging modality in the diagnosis of cardiovascular disease.Advancements in CT imaging technology have further advanced its applications from high diagnostic value to minimising radiation exposure to patients.In addition to the standard application of assessing vascular lumen changes,CTA-derived applications including 3D printed personalised models,3D visualisations such as virtual endoscopy,virtual reality,augmented reality and mixed reality,as well as CT-derived hemodynamic flow analysis and fractional flow reserve(FFRCT)greatly enhance the diagnostic performance of CTA in cardiovascular disease.The widespread application of artificial intelligence in medicine also significantly contributes to the clinical value of CTA in cardiovascular disease.Clinical value of CTA has extended from the initial diagnosis to identification of vulnerable lesions,and prediction of disease extent,hence improving patient care and management.In this review article,as an active researcher in cardiovascular imaging for more than 20 years,I will provide an overview of cardiovascular CTA in cardiovascular disease.It is expected that this review will provide readers with an update of CTA applications,from the initial lumen assessment to recent developments utilising latest novel imaging and visualisation technologies.It will serve as a useful resource for researchers and clinicians to judiciously use the cardiovascular CT in clinical practice.

Highly clinical and genetic heterogeneity of neurodevelopmental disorders presents a major challenge in clinical genetics and medicine.Panoramic variation analysis is imperative to analyze the disease phenotypes resulting from multilocus genomic variation.Here,a Pakistani family with parental consanguinity was presented,characterized with severe intellectual disability(ID),spastic paraplegia,and deafness.Homozygosity mapping,integrated single nucleotide polymorphism(SNP)array,whole-exome sequencing,and whole-genome sequencing were performed,and homozygous variants in TMEM141(c.270G>A,p.Trp90^(*)),DDHD2(c.411+767_c.1249-327del),and LHFPL5(c.250delC,p.Leu84^(*))were identified.A Tmem141^(p.Trp90^(*)/p.Trp90^(*))mouse model was generated.Behavioral studies showed impairments in learning ability and motor coordination.Brain slice electrophysiology and Golgi staining demonstrated deficient synaptic plasticity in hippocampal neurons and abnormal dendritic branching in cerebellar Purkinje cells.Transmission electron microscopy showed abnormal mitochondrial morphology.Furthermore,studies on a human in vitro neuronal model(SH-SY5Y cells)with stable shRNA-mediated knockdown of TMEM141 showed deleterious effect on bioenergetic function,possibly explaining the pathogenesis of replicated phenotypes in the cross-species mouse model.Conclusively,panoramic variation analysis revealed that multilocus genomic variations of TMEM141,DDHD2,and LHFPL5 together caused variable phenotypes in patient.Notably,the biallelic loss-of-function variants of TMEM141 were responsible for syndromic ID.

In this paper,we consider the initial-boundary value problem(IBVP)for the micropolar Naviers-Stokes equations(MNSE)and analyze a first order fully discrete mixed finite element scheme.We first establish some regularity results for the solution of MNSE,which seem to be not available in the literature.Next,we study a semi-implicit time-discrete scheme for the MNSE and prove L2-H1 error estimates for the time discrete solution.Furthermore,certain regularity results for the time discrete solution are establishes rigorously.Based on these regularity results,we prove the unconditional L2-H1 error estimates for the finite element solution of MNSE.Finally,some numerical examples are carried out to demonstrate both accuracy and efficiency of the fully discrete finite element scheme.

OBJECTIVE:To elucidate the mechanism of the nourishing Yin and purging fire Chinese herbal mixture(NYPF)in delaying light-induced premature puberty in rats.METHODS:Twenty-one days old female SpragueDawley rats were randomly assigned to normal group(N),long light exposure group(L),NYPF and normal saline group(NS).Rats in the L,NYPF and NS groups were exposed to 16 h:350 lux light/8 h:dark,while rats in the N group were exposed to 12 h:50 lux light/12 h:dark.NYPF and normal saline was administered to the rats in the NYPF group or NS group,respectively,from day 21.Five rats in every group were sacrificed at 9 p.***.on day 28(P28),on the day when rat's vulva opened in the L group(L-VO),on the day when the first estrous interphase occurred in rats of L group(L-E1),and on the day when the second estrous interphase occurred in rats of L group(L-E2),respectively.RESULITS:On day 34,all rats in the L group,80%of rats in the NS group,40%of rats in the N group,and 20%of rats in the NYPF group showed complete opening of the vulva.At P28,m RNA level of hypothalamic kisspeptin(Kiss-1)in the L group was significantly higher than that in the N group(P<0.05).The rats in the L and NS groups had significantly lower hypothalamic argininephenylalanine-amide(RFamide)-related peptide 3(RFRP-3)m RNA levels than those in the N group(P<0.05),whereas RFRP-3 m RNA level was significantly higher in the NYPF group than that in the L group(P<0.05).At L-VO,the ovarian index of the L and NS groups was significantly higher than that of the N group(P<0.05)and estradiol(E2)level of the NYPF group was significantly lower than that of the N and NS groups(P<0.05);hypothalamic Kiss-1 m RNA level in the L and NS groups was significantly higher than that in the N and NYPF groups(P<0.05),whereas hypothalamic RFRP-3 m RNA level in the L,NYPF,and NS groups was significantly lower than that in the N group(P<0.05).At L-E1,E2 level of the L and NS groups was significantly higher than that of the N group(P<0.01),whereas it was significantly lower in the NYPF group

TX-80 low-transformation-temperature(LTT)welding wire was used to replace the traditional ER 307Si welding wire to realize the connection of 22SiMn2TiB armor steel in manual overlay welding.The previously existing issues,such as welding cracks,large welding deformation,and severe welding residual stress,were solved to ensure good strength and ductility requirements.In particular,with the same welding conditions,TX-80 LTT wire eliminates welding cracks.It reduces the welding deformation no matter the base pretreatment of pre-setting angle or no pre-setting angle.By comparison,it was found that the microstructure at the TX-80 weld is mainly composed of martensite and a small amount of retained austenite.In contrast,the microstructure of the ER 307Si weld consists of a large amount of austenite and a small amount of skeleton-like ferrite.The variation trend of residual stress and microhardness from the weld to the base were investigated and compared with the mechanical properties of base materials.The TX-80 and the ER 307Si tensile samples elongation is 6.76%and 6.01%,while the ultimate tensile strengths are 877 and 667 MPa,respectively.The average impact toughness at room temperature of the ER 307Si weld is 143.9 J/cm^(2),much higher than that of the TX-80 weld,which is only 36.7 J/cm^(2).The relationship between impact and tensile properties with microstructure species and distribution was established.In addition,the fracture surface of the tensile and the impact samples was observed and analyzed.Deeper dimples,fewer pores,larger radiation zone,and shear lips of TX-80 samples indicate better tensile ductility and worse impact toughness than those of ER 307Si weld.

The Lamb shift of a quantum emitter in close proximity to a plasmonic nanostructure can be three or more orders of magnitude larger than that in the free space and is ultra-sensitive to the emitter position and polarization.We demonstrate that this large Lamb shift can be sensitively observed from the scattering or absorption spectrum dip shift of the coupled system when the plasmonic nanoparticle or tip scans the emitter.Using these observations,we propose a scanning optical scattering imaging method based on the plasmonic-enhanced Lamb shift with achieves sub-nanometer resolution.Our method is based on the scattering or absorption spectrum of the plasmon-emitter coupling system,which is free of the fluorescence quenching problem and easier to implement in a plasmon-emitter coupling system.In addition,our scheme works even if the quantum emitter is slightly below the dielectric surface,which can bring about broader applications,such as detecting atoms and molecules or quantum dots above or under a surface.
