Ferroptosis inhibitors reduce celastrol toxicity and preserve its insulin sensitizing effects in insulin resistant HepG2 cells

Objective:Research has shown that celastrol can effectively treat a variety of diseases,yet when passing a certain dosage threshold,celastrol becomes toxic,causing complications such as liver and kidney damage and erythrocytopenia,among others.With this dichotomy in mind,it is extremely important to find ways to preserve celastrol’s efficacy while reducing or preventing its toxicity.Methods:In this study,insulin-resistant Hep G2(IR-Hep G2)cells were prepared using palmitic acid and used for in vitro experiments.IR-Hep G2 cells were treated with celastrol alone or in combination with Nacetylcysteine(NAC)or ferrostatin-1(Fer-1)for 12,24 or 48 h,at a range of doses.Cell counting kit-8assay,Western blotting,quantitative reverse transcription-polymerase chain reaction,glucose consumption assessment,and flow cytometry were performed to measure celastrol’s cytotoxicity and whether the cell death was linked to ferroptosis.Results:Celastrol treatment increased lipid oxidation and decreased expression of anti-ferroptosis proteins in IR-Hep G2 cells.Celastrol downregulated glutathione peroxidase 4(GPX4)m RNA.Molecular docking models predicted that solute carrier family 7 member 11(SLC7A11)and GPX4 were covalently bound by celastrol.Importantly,we found for the first time that the application of ferroptosis inhibitors(especially NAC)was able to reduce celastrol’s toxicity while preserving its ability to improve insulin sensitivity in IR-Hep G2 cells.Conclusion:One potential mechanism of celastrol’s cytotoxicity is the induction of ferroptosis,which can be alleviated by treatment with ferroptosis inhibitors.These findings provide a new strategy to block celastrol’s toxicity while preserving its therapeutic effects.

Adaptive fault-tolerant control for non-minimum phase hypersonic vehicles based on adaptive dynamic programming

In this paper,a novel adaptive Fault-Tolerant Control(FTC)strategy is proposed for non-minimum phase Hypersonic Vehicles(HSVs)that are affected by actuator faults and parameter uncertainties.The strategy is based on the output redefinition method and Adaptive Dynamic Programming(ADP).The intelligent FTC scheme consists of two main parts:a basic fault-tolerant and stable controller and an ADP-based supplementary controller.In the basic FTC part,an output redefinition approach is designed to make zero-dynamics stable with respect to the new output.Then,Ideal Internal Dynamic(IID)is obtained using an optimal bounded inversion approach,and a tracking controller is designed for the new output to realize output tracking of the nonminimum phase HSV system.For the ADP-based compensation control part,an ActionDependent Heuristic Dynamic Programming(ADHDP)adopting an actor-critic learning structure is utilized to further optimize the tracking performance of the HSV control system.Finally,simulation results are provided to verify the effectiveness and efficiency of the proposed FTC algorithm.

Energy-saving and environmentally friendly photocatalysis has emerged as a popular research area in response to issues with energy scarcity and environmental degradation.Due to the unique layer-like structure,BiOX（Cl,Br,I） is frequently used in photocatalysis.However,inherent flaws in BiOX,such as an inappropriate band gap and low carrier separation efficiency,restrict its capacity for photocatalysis.Owing to the tunable grouping layer,alloying engineering is employed to optimize the intrinsic properties of BiOX and alloyed BiOX becomes a promising photocatalytic material.This review describes the structure of BiOX,where tunable halogen layers provide favorable conditions for the implementation of alloying engineering to improve intrinsic properties.The article compares the effects and mechanisms of alloying engineering on the optimization of the energy band structure and carrier behavior of BiOX,and lists various modification methods used to improve the optimization of the intrinsic properties by alloying engineering,including defect engineering,morphology control as well as the synergy between alloying and other modification methods（bismuth-rich strategies,cation doping,construction of heterojunctions and plasma resonance effects）.Subsequently,applications of alloyed BiOX in energy and environmental fields are summarized,including contaminant degradation,antibacterial,CO2reduction,nitrogen fixation and organic synthesis.Finally,we summarize the current challenges and future directions of alloyed BiOX.It is expected that this work will provide guidance and assistance for an in-depth study and understanding of the mechanisms of alloying engineering to optimize intrinsic properties and design alloyed BiOX with higher photocatalytic activity.

Unsupervised Multi-Expert Learning Model for Underwater Image Enhancement

Underwater image enhancement aims to restore a clean appearance and thus improves the quality of underwater degraded images.Current methods feed the whole image directly into the model for enhancement.However,they ignored that the R,G and B channels of underwater degraded images present varied degrees of degradation,due to the selective absorption for the light.To address this issue,we propose an unsupervised multi-expert learning model by considering the enhancement of each color channel.Specifically,an unsupervised architecture based on generative adversarial network is employed to alleviate the need for paired underwater images.Based on this,we design a generator,including a multi-expert encoder,a feature fusion module and a feature fusion-guided decoder,to generate the clear underwater image.Accordingly,a multi-expert discriminator is proposed to verify the authenticity of the R,G and B channels,respectively.In addition,content perceptual loss and edge loss are introduced into the loss function to further improve the content and details of the enhanced images.Extensive experiments on public datasets demonstrate that our method achieves more pleasing results in vision quality.Various metrics(PSNR,SSIM,UIQM and UCIQE) evaluated on our enhanced images have been improved obviously.

The overdiagnosis of prostate cancer(PCa)caused by nonspecific elevation serum prostate-specific antigen(PSA)and the overtreatment of indolent PCa have become a global problem that needs to be solved urgently.We aimed to construct a prediction model and provide a risk stratification system to reduce unnecessary biopsies.In this retrospective study,clinical data of 1807 patients from three Chinese hospitals were used.The final model was built using stepwise logistic regression analysis.The apparent performance of the model was assessed by receiver operating characteristic curves,calibration plots,and decision curve analysis.Finally,a risk stratification system of clinically significant prostate cancer(csPCa)was created,and diagnosis-free survival analyses were performed.Following multivariable screening and evaluation of the diagnostic performances,a final diagnostic model comprised of the PSA density and Prostate Imaging-Reporting and Data System(PI-RADS)score was established.Model validation in the development cohort and two external cohorts showed excellent discrimination and calibration.Finally,we created a risk stratification system using risk thresholds of 0.05 and 0.60 as the cut-off values.The follow-up results indicated that the diagnosis-free survival rate for csPCa at 12 months and 24 months postoperatively was 99.7%and 99.4%,respectively,for patients with a risk threshold below O.05 after the initial negative prostate biopsy,which was significantly better than patients with higher risk.Our diagnostic model and risk stratification system can achieve a personalized risk calculation of csPCa.It provides a standardized tool for Chinese patients and physicians when considering thenecessity of prostatebiopsy.

Effect of Al_(2)O_(3)/SiO_(2) mass ratio and CaO content on viscosity and structure of slag for pyrometallurgical processing of spent automotive catalysts

The effect of Al_(2)O_(3)/SiO_(2) mass ratio and CaO content on the viscosity and structure of the CaO-Al_(2)O_(3)-SiO_(2)-6MgO-1.5ZrO_(2)-1.5CeO_(2) slag was investigated.The results show that with the increase in Al_(2)O_(3)/SiO_(2) mass ratio,the viscous flow units within the slag gradually change from Si-O-Si to Al-O-Al and Al-O-Si.Furthermore,the substitution of Al_(2)O_(3) for SiO_(2) leads to the transformation of Si-O bonds towards weaker Al-O bonds,which weaken the bond strength of the aluminosilicate networks,thus leading to a decrease in the viscosity of slag.The increase in CaO content effectively promotes the depolymerization of the aluminosilicate networks,resulting in a significant decrease in the viscosity of the slag.The slag with Al_(2)O_(3)/SiO_(2) of 0.7-1.5 and CaO of 30 wt.%shows promise as the reference slag system for the Fe-collection smelting of spent automotive catalysts due to its good comprehensive performance.

Postoperative accurate pain assessment of children and artificial intelligence: A medical hypothesis and planned study

Although the pediatric perioperative pain management has been improved in recent years,the valid and reliable pain assessment tool in perioperative period of children remains a challenging task.Pediatric perioperative pain management is intractable not only because children cannot express their emotions accurately and objectively due to their inability to describe physiological characteristics of feeling which are different from those of adults,but also because there is a lack of effective and specific assessment tool for children.In addition,exposure to repeated painful stimuli early in life is known to have short and long-term adverse sequelae.The short-term sequelae can induce a series of neurological,endocrine,cardiovascular system stress related to psychological trauma,while long-term sequelae may alter brain maturation process,which can lead to impair neurodevelopmental,behavioral,and cognitive function.Children’s facial expressions largely reflect the degree of pain,which has led to the developing of a number of pain scoring tools that will help improve the quality of pain mana-gement in children if they are continually studied in depth.The artificial inte-lligence(AI)technology represented by machine learning has reached an unprecedented level in image processing of deep facial models through deep convolutional neural networks,which can effectively identify and systematically analyze various subtle features of children’s facial expressions.Based on the construction of a large database of images of facial expressions in children with perioperative pain,this study proposes to develop and apply automatic facial pain expression recognition software using AI technology.The study aims to improve the postoperative pain management for pediatric population and the short-term and long-term quality of life for pediatric patients after operational event.

Nursing effect of narrative nursing intervention on postoperative patients with severe lung cancer

BACKGROUND Lung cancer is a common disease with high mortality,and psychological support is very important in the diagnosis and treatment of postoperative patients with cancer pain.AIM To explore the application effect of the narrative nursing method in postoperative lung cancer patients in the intensive care unit.METHODS A total of 120 patients diagnosed with lung cancer and experiencing cancer-related pain were randomly allocated into two groups:an observation group and a control group,each consisting of 60 cases.The control group was given routine analgesic and psychological care,while the research group applied the five-step narrative nursing method based on routine care,comparing the visual analogue scale scores,sleep status,anxiety and depression status,and quality of life of the two groups of patients before and after the intervention.RESULTS The pain scores,anxiety scores,and depression scores of the study group were lower than those of the control group after the intervention using the narrative nursing method,and the difference was statistically significant(Pbined with cancerous pain can help patients to correctly recognize their disease,adjust their mentality,establish confidence,alleviate patients'subjective pain feelings,and improve their adverse emotions.

In recent years,lithium-sulfur batteries（LSBs）are considered as one of the most promising new generation energies with the advantages of high theoretical speeific capacity of sulfur(1675 mAh·g-1),abundant sulfur resources,and environmental friendliness storage technologies,and they are receiving wide attention from the industry.However,the problems of the shuttle effect and lithium dendrite growth in LSBs have limited their practical application,so there is a need to find ways to solve these problems.It is an excellent choice to use novel materials to modify battery materials.Among those novel materials,the metal-organic framework（MOF） has the properties of regular pores and controllable structure.When applied as a positive electrode and diaphragm,it can restrain the shuttle effect and lithium dendrite growth,especially since it shows excellent performance in diaphragm modification.Therefore,various design strategies and synthesis methods of MOF-modified separators are reviewed in this paper,and the applications of MOF in LSBs separators in different forms are introduced,including the composite of MOF and carbon-based materials,the compounding of MOF and polymer,self-carbonization to form MOF-derived materials.At the same time,different characterization techniques are systematically reviewed to obtain the physical and chemical properties of MOF particles and the working mechanism of MOF-modified diaphragm,which provides a basis for further research in this field.Finally,some future research trends and directions are put forward to fully tap the future commercial potential of MOF-modified diaphragm in LSBs.

1 Project overview The“Beishan No.1”,the world’s first-ever hard rock Tunnel Boring Machine(TBM)tailored for high-gradient spiral tunnels,constitutes a pivotal element within the Beishan Underground Research Laboratory(URL)initiative in China.Beishan URL,the first URL for geological disposal of high level radioactive waste(HLW)in China,is a national key construction project listed in the“13th Five-Year Plan”.In 2019,subsequent to receiving approval from the China Atomic Energy Authority,the Beijing Research Institute of Uranium Geology(BRIUG),serving as the project’s owner,initiated its construction.This underground facility,categorized as a“third generation”URL for HLW disposal,i.e.,area-specific URL,was located in Beishan,Jiuquan City,Gansu Province,China,following more than three decades of rigorous research on site selection.
