The development and maturation of the CRISPR/Cas genome editing system provides a valuable tool for plant functional genomics and genetic *** available genome-editing tools have a limited number of targets,restricting their application in genetic *** this study,we developed a novel CRISPR/Cas9 plant ultra-multiplex genome editing system consisting of two template vectors,eight donor vectors,four destination vectors,and one primer-design software *** combining the advantages of Golden Gate cloning to assemble multiple repetitive fragments and Gateway recombination to assemble large fragments and by changing the structure of the amplicons used to assemble sg RNA expression cassettes,the plant ultra-multiplex genome editing system can assemble a single binary vector targeting more than 40 genomic loci.A rice knockout vector containing 49 sg RNA expression cassettes was assembled and a high co-editing efficiency was *** plant ultra-multiplex genome editing system advances synthetic biology and plant genetic engineering.

Tailoring the alloy composition,which induces the hard secondary phase to increase hardness and strength to improve the wear features,is a feasible approach for developing wear-resistant metal ***,a group of(AlCoCrFeNi)_(100–x)Sc_(x)(x=0–2.0,at%)high-entropy alloys(HEAs)are designed and the phase compositions and wear behaviors are ***-doped HEA series contain the primary body-centered cubic(BCC)and eutectic phases,in which the eutectic phase is composed of the alternately grown BCC and Laves *** addition promotes the diffusion of Ni atoms from BCC phase to form the Sc-rich Laves phase at the grain *** hardness increases due to solid solution strengthening and second phase *** the second phase strengthening plays a more significant role relative to solid solution *** phase and the oxides caused by wear heating prevent the direct contact between friction pair and HEAs,thus inducing a decreased wear rate from 6.82×10^(−5) to 3.47×10^(−5)m^(3)·N^(−1)·m^(−1).Moreover,the wear mechanism changes from adhesive wear,abrasive wear and oxidative wear to abrasive wear and oxidative wear.

Initially defined high entropy alloys(HEAs)usually exhibit a single-phase solid-solution ***,two and/or more types of phases in HE As possibly induce the desired microstructure features,which contribute to improving the wear properties of HE ***,we prepare a series of(AlCoCrFeNi)_(100-x)Hf_(x)(x=0,2,4and 6;at%) HEAs and concern their phase compositions,micro structures and wear *** leads to the formation of(Ni,Co)_(2)Hf-type Laves phase and tailors the microstructure from a body-centered cubic(BCC) singlephase structure to a hypoeutectic *** increased hardness from~HV 512.3 to~HV 734.1 is due to solid-solution strengthening,grain refinement strengthening and precipitated phase *** a few oxides(Al_(2)O_(3)+Cr_(2)O_(3)) caused by the wear heating contribute to an 85.5% decrease in wear rate of the HEA system from6.71×10^(-5) to 0.97×10^(-5) m^(3)·N^(-1)·m^(-1).In addition,Hf addition changes the wear mechanism from abrasive wear,mild oxidative wear and adhesive wear to oxidative wear and adhesive wear.

Retaining the self-released chalcogenate at reconstructed cobalt sites by self-transformed carbonate regulation for boosted oxygen evolution

The in-situ generated oxyanions at electrochemically reconstructed catalysts from metal-based nonoxide compounds have been proven to significantly accelerate oxygen evolution reaction（OER） ***,it remains a challenge to retain these self-released oxyanions at reconstructed catalysts,hindering its utilization as a tool to develop efficient OER ***,we demonstrate a versatile selftransformed carbonate regulation strategy to efficiently retain the self-released chalcogenate at Co oxyhydroxides reconstructed from carbon-incorporated Co selenides under OER *** selftransformed CO32-can induce electron accumulation and narrow d bond at Co sites to facilitate the Co3d-O 2p orbital hybridization between Co sites and SeOx2-for enhanced SeOx2-retention,which can accelerate the rate-limiting step for*OOH formation during *** to CoOOH-SeOx2-with limited SeOx2-residues,CoOOH-CO32-/SeOx2-with elevated SeOx2-retention by CO32-regulation exhibited a 5.6-fold increase in current density and a remarkable lower Tafel slope towards *** strategy paves a rational avenue to design efficient catalysts for electrooxidation reactions through finely regulating self-released oxyanions at reconstructed structures.

Desiccation and osmotic resistance mechanism of Cronobacter spp. isolated from powdered infant formula

Cronobacter spp. has strong resistance to desiccation and high permeability in Enterobacteriaceae， and powdered infant formula (PIF) is one of the main contamination routes. In recent years， the contamination of Cronobacter spp. in PIF incidents occur from time to time， causing infant serious diseases or death. In this investigation， matrix-assisted laser desorption/ionization time of flight mass spectrometry was used to identify the phenotypes of 35 Cronobacter strains isolated from PIF and its processing environment. Subsequently， the isolates were evaluated for drying and osmotic pressure tolerance. The results showed that the deactivation rate of the strains ranged from 9.01% to 77.57%， and the highest osmotic pressure condition the strains could tolerate was 6% NaCl. In addition， there was a positive correlation between biofilm formation ability and desiccation resistance. Combined with transcriptomics， Cronobacter spp. can activate biofilm synthesis， produce more trehalose， accumulate betaine and electrolytes to stabilize intracellular structure under the two treatment conditions. 31 and 43 genes were found related to desiccation and permeability resistance， respectively. And some genes (cysM， thuF， ycjO， et al) were found to be associated with two tolerances for the first time.

Most triaxial-vectorial magnetic field measurements with spin-exchange relaxation free(SERF)atomic magnetometer(AM)are based on the quasi-steady-state solution of the Bloch ***,the responding speed of these methods is greatly limited because the frequency of the modulation signal should be slow enough to ensure the validity of the quasi-steady-state *** this work,a new model to describe the response of the three-axis sensitive SERF AM with high modulation frequency is presented and *** response of alkali-atomic spin to high-frequency modulation field is further investigated by solving the Bloch equation in a modulation-frequency-dependence *** solution is well verified by our experiments and can offer a reference for selection of modulation *** result shows a potential to achieve a SERF AM operating in a geomagnetic field without heavy aluminum shielding when the modulation frequencies are selected properly.

Multiple myeloma survival in New South Wales, Australia, by treatment era to 2020

Objective: Australia has relatively high multiple myeloma(MM) incidence and mortality rates. Advancements in MM treatment over recent decades have driven improvements in MM survival in high-income countries;however, reporting in Australia is limited. We investigated temporal trends in population-wide MM survival across 3 periods of treatment advancements in New South Wales(NSW), ***: Individuals with an MM diagnosis in the NSW Cancer Registry between 1985 and 2015 with vital follow-up to 2020, were categorized into 3 previously defined treatment eras according to their diagnosis date(1985±1995, chemotherapy only;1996±2007, autologous stem cell transplantation;and 2008±2015, novel agents including proteasome inhibitors and immunomodulatory drugs). Both relative survival and cause-specific survival according to Fine and Gray's competing risks cumulative incidence function were calculated by treatment era and age at ***: Overall, 11,591 individuals were included in the study, with a median age of 70 years at diagnosis. Five-year relative survival improved over the 36-year(1985±2020) study period(31.0% in 1985±1995;41.9% in 1996±2007;and 56.1% in 2008±2015). For individuals diagnosed before 70 years of age, the 5-year relative survival nearly doubled, from 36.5% in 1985±1995 to 68.5% in 2008±2015. Improvements for those > 70 years of age were less pronounced between 1985±1995 and 1996±2007;however, significant improvements were observed for those diagnosed in 2008±2015. Similar overall and age-specific patterns were observed for causespecific survival. After adjustment for gender and age at diagnosis, treatment era was strongly associated with both relative and cause-specific survival(P < 0.0001).Conclusions: Survival of individuals with MM is improving in Australia with treatment advances. However, older age groups continue to experience poor survival outcomes with only modest improvements over time. Given the increasing prevalence of MM in

Glycine Attenuates Myocardial Fibrosis in Myocardial Infarction in Rats Partly through Modulating Signal Transducer and Activator of Transcription 3/Nuclear Factor-κB/Transforming Growth Factor-β axis

Objective: Inflammation and fibrosis are strongly associated with each other. Glycine is present in various traditional Chinese medicines and exhibits anti-inflammatory activity. However, the effects of glycine on myocardial fibrosis(MF) in rats with myocardial infarction(MI) have not been reported. The purpose of this study is to investigate the effects of glycine therapy on MF and comprehend its underlying mechanisms. Materials and Methods: Left anterior descending artery ligation-induced MI in Sprague Dawley rats was leveraged to assess the therapeutic effects of Glycine. Rats received either normal saline or glycine(0.5 mg/g bodyweight) for 7 days. Results: Glycine upregulated cardiac ejection fraction and fractional shortening to improve cardiac function, as evaluated by echocardiography. Histological and immunohistochemical analyses demonstrated that glycine could decrease inflammatory cell infiltration and alleviate collagen deposition. Western blotting revealed that nuclear factor-κB(NF-κB)-mediated inflammatory signaling was also downregulated by glycine treatment. The expression of signal transducer and activator of transcription 3(STAT3), tumor necrosis factor-α, and transforming growth factor-β(TGF-β) was decreased significantly in the glycine-treated group compared to the model group. Thus, glycine plays a protective role against myocardial ischemia and subsequent MF. Conclusion: The protective effects of glycine were achieved partly through STAT3/NF-κB/TGF-β signaling pathway.
