Recombinant Mouse Canstatin Inhibits Chicken Embryo Chorioallantoic Membrane Angiogenesis and Endothelial Cell Proliferation

Human canstatin, a 24 kD fragment of the alpha2 chain of type IV collagen, has been proved to be one of the most effective inhibitors of angiogenesis and tumor growth. To investigate in vivo antiangiogenesis activity and in vitro effects on endothelial cell proliferation of recombinant mouse canstatin, the cDNA of mouse canstatin was introduced into an expression vector pQE40 to construct a prokaryotic expression vector pQE-mCan. The recombinant mouse canstatin efficiently expressed in E. coli M15 after IPTG induction was monitored by SDS-PAGE and by Western blotting with an anti-hexahistidine tag antibody. The expressed mouse canstatin, mainly as inclusion bodies, accounted for approximately 35% of the total bacterial proteins. The inclusion bodies were washed, lysed and purified by the nickel affinity chromatography to a purity of approximately 93%. The refolded mouse canstatin was tested on the chicken embryo chorioallantoic membranes (CAM), and a large number of newly formed blood vessels were significantly regressed. In addition, recombinant mouse canstatin potently inhibited endothelial cell proliferation with no inhibition on non-endothelial cells. Taken together, these findings demonstrate that the recombinant mouse canstatin effectively inhibited angiogenesis of the chicken embryo in a dose-dependent manner and specially suppressed in vitro the proliferation of human umbilical vein endothelial cells.

Separation of salidroside from the fermentation broth of engineered Escherichia coli using macroporous adsorbent resins

Salidroside(8-O-β-D-glucoside of tyrosol),a plant-derived natural product,is used for treatment of hypoxia,fatigue and aging *** availability of salidroside is restricted since it is extracted from3-5 years old Rhodiola roots,which grow very slowly in the cold region of northern hemisphere of *** laboratory has constructed an engineered Escherichia coli and established a fermentation process to produce salidroside from *** this article,nine macroporous resins from polarity to non-polarity,including NKA-9,S-8,AB-8,SP825,D101,LSA-8,lx-12,lx-18 and lx-68 resins,were tested to separate salidroside from fermentation *** static and dynamic experiments,the weakly polar SP825 resin had a better separation efficiency among nine *** adsorption kinetic and isotherm of salidroside on the SP825 resin were determined,and the pseudo-second-order kinetic model and Langmuir model could be fitted *** effects of the pH on adsorption and ethanol concentration on desorption were investigated,and an optimal separation process was *** adsorption for salidroside in the SP825 resin column was conducted with loading 150 ml at pH 7,and desorpted by washing 50 ml of80%ethanol *** the best process conditions,the purity and yield of salidroside in the final product were 91.6%and 74.0%,*** results showed that the macroporous SP825 resin would be feasible and effective to prepare salidroside and has promising application in the downstream process of microbial fermentation.

Cu(In,Ga)Se2 absorbers prepared by electrodeposition for low-cost thin-film solar cells

Reducing the manufacturing cost of solar cells is necessary to their industrial production. Electrodepositing is an effective, non-vacuum method which is very suitable for cutting the manufacturing cost of thin films as well as developing its large-scale industrial production. In this study, about 1-μm-thick Cu（In,Ga）Se2（CIGS） precursors were electrodeposited on Mo/glass substrates in aqueous solution utilizing a three-electrode potentiostatic *** was used as complexing agent, and all parameters of electrodeposition were precisely *** that, the electrodeposited precursors were selenized in a Se atmosphere with different heating ramp rates（60 and 600℃·min^（-1））. High-quality CIGS films were obtained, and their characteristics were investigated by X-ray fluorescence, scanning electron microscopy, energydispersive spectroscopy, X-ray diffraction, Raman spectra and near-infrared-visible（NIR-Vis） spectra. The results reveal that there are many differences between the properties of the films under different heating rates. Finally,CIGS solar cells were fabricated using a fast and a slow heating rate. The maximum efficiencies achieved for the films selenized at 60 and 600℃-min^（-1） are 3.15% and 0.71%, respectively.

Objective Hepatic stellate cells(HSCs)play a crucial role in liver ***-stage liver fibrosis is reversible and intimately associated with the state of ***-like factor 4(KLF4)plays a pivotal role in a wide array of physiological and pathological *** study aimed to investigate the effect of KLF4 on the proliferation,apoptosis and phenotype of quiescent HSCs Methods We designed a KLF4 lentiviral vector and a KLF4 siRNA lentiviral vector,to upregulate and silence KLF4 expression in human HSC lx-2 cells via *** proliferation was assessed using the CCK-8 *** cytometry was used to detect the cell cycle distribution and apoptosis *** blotting was used to determine the levels of some quiescence and activation markers of HSCs Results Overexpression of KLF4 significantly increased the levels of E-cadherin and ZO-1,which are quiescent HSC markers,while significantly decreased the levels of N-cadherin and a-SMA,known activated HSC *** contrast,cell proliferation and apoptosis rates were elevated in lx-2 cells in which KLF4 expression was silenced Conclusion KLF4 inhibits the proliferation and activation of human lx-2 *** might be a key regulatory protein in the maintenance of HSC quiescence and may serve as a target for the inhibition of hepatic fibrosis.

Experimental investigation on indoor daylight environment of building with Cadmium Telluride photovoltaic window

Photovoltaic(PV)windows have received more and more attention in recent years since their active energy-saving *** the surface covered with solar cell modules,the indoor daylight environment of PV windows is obviously different with clear glass ***,despite many scholars have studied the indoor daylight environment of PV windows,there few investigations study it from the perspective of human subjective visual *** this paper,the indoor daylight environment and human visual comfort of building with Cadmium Telluride Photovoltaic(CdTe-PV)window were ***,the parameters of indoor daylight environment and subjective questionnaires in rooms with CdTe-PV window and clear glass window were analyzed *** the basis of this,combined with indoor working surface illuminance and results of subjective questionnaires,the daylight illuminance threshold of human visual comfort was investigated by the method of Mean Bias Degree(MBD).Finally,an evaluation model for indoor daylight environment of buildings with CdTe-PV window was developed by Fuzzy Comprehensive Evaluation *** results showed that the working surface illuminance of CdTe-PV window was lower than that of clear glass room,the CCT of different windows room had a minor gap and the CdTe-PV window room was closer to the recommended range that was *** for CRI,both the CdTe-PV window room and the clear glass room could meet the visual comfort requirements of office ***,it was found that the requirement of human visual comfort was met when indoor working surface illuminance varies between 500-2200lx in the room with CdTe-PV *** last,according to the comprehensive evaluation model proposed in this paper,it was found that the indoor daylight environment of buildings with CdTe-PV window was excellent in the present experiment.

Artificial evolution of OsEPSPS through an improved dual cytosine and adenine base editor generated a novel allele conferring rice glyphosate tolerance

Exploiting novel endogenous glyphosate-tolerant alleles is highly desirable and has promising potential for weed control in rice breeding. Here,through fusions of different effective cytosine and adenine deaminases with nCas9-NG, we engineered an effective surrogate two-component composite base editing system, STCBE-2, with improved C-to-T and A-to-G base editing efficiency and expanded the editing window. Furthermore,we targeted a rice endogenous OsEPSPS gene for artificial evolution through STCBE-2-mediated near-saturated mutagenesis. After hygromycin and glyphosate selection, we identified a novel OsEPSPS allele with an Asp-213-Asn(D213N)mutation(OsEPSPS-D213N) in the predicted glyphosate-binding domain, which conferred rice plants reliable glyphosate tolerance and had not been reported or applied in rice breeding. Collectively, we developed a novel dual base editor which will be valuable for artificial evolution of important genes in crops. And the novel glyphosate-tolerant rice germplasm generated in this study will benefit weeds management in rice paddy fields.

NQO1 Mediates Lenvatinib Resistance by Regulating ROS-induced Apoptosis in Hepatocellular Carcinoma

Objective Hepatocellular carcinoma(HCC)is the third leading cause of cancer-associated death *** a first-line drug for advanced HCC treatment,lenvatinib faces a significant hurdle due to the development of both intrinsic and acquired resistance among patients,and the underlying mechanism remains largely *** present study aims to identify the pivotal gene responsible for lenvatinib resistance in HCC,explore the potential molecular mechanism,and propose combinatorial therapeutic targets for HCC *** Cell viability and colony formation assays were conducted to evaluate the sensitivity of cells to lenvatinib and ***-Seq was used to determine the differences in transcriptome between parental cells and lenvatinib-resistant(LR)*** upregulated genes were analyzed by GO and KEGG ***,qPCR and Western blotting were employed to determine the relative gene expression ***,the intracellular reactive oxygen species(ROS)and apoptosis were detected by flow *** PLC-LR and Hep3B-LR were *** was a total of 116 significantly upregulated genes common to both LR cell *** GO and KEGG analyses indicated that these genes were involved in oxidoreductase and dehydrogenase activities,and reactive oxygen species ***,NAD(P)H:quinone oxidoreductase 1(NQO1)was highly expressed in LR cells,and was involved in the lenvatinib *** high expression of NQO1 decreased the production of ROS induced by lenvatinib,and subsequently suppressed the *** combination of lenvatinib and NQO1 inhibitor,dicoumarol,reversed the resistance of LR *** The high NQO1 expression in HCC cells impedes the lenvatinib-induced apoptosis by regulating the ROS levels,thereby promoting lenvatinib resistance in HCC cells.

Efficacy of hepatic resection vs transarterial chemoembolization for solitary huge hepatocellular carcinoma

AIM: To compare the efficacy of hepatic resection(HR) and transarterial chemoembolization(TACE) for patients with solitary huge(≥ 10 cm) hepatocellular carcinoma(HCC).METHODS: Records were retrospectively analyzed of 247 patients with solitary huge HCC, comprising 180 treated by HR and 67 by TACE. Long-term overall survival(OS) was compared between the two groups using the Kaplan-Meier method, and independent predictors of survival were identified by multivariate analysis. These analyses were performed using all patients in both groups and/or 61 pairs of propensity score-matched patients from the two groups. RESULTS: OS at 5 years was significantly higher in the HR group than the TACE group, across all patients(P = 0.002) and across propensity score-matchedpairs(36.4% vs 18.2%, P = 0.039). The two groups showed similar postoperative mortality and morbidity. Multivariate analysis identified alpha-fetoprotein ≥ 400 ng/m L, presence of vascular invasion and TACE treatment as independent predictors of poor ***: Our findings suggest that HR can be safe and more effective than TACE for patients with solitary huge HCC.

Spatiotemporal distribution of regional landslides and erosion rates in Southeastern Tibet

The difficulty in estimating ages of regional landslides hampers to assess frequency of landslides and hence to quantitatively assess regional landslide *** this study,we used radiocarbon dating of organic sediment on boulder rock varnish to estimate landslide ages in Langxian(lx)arid region in southeastern *** of rock varnish with organic sediment were collected on site for radiocarbon dating,leading to landslide ages from 1880±30 to 18,430±30 yr *** measure surface roughness characteristics of 109 remotely-mapped large bedrock landslide deposits,we estimated average standard deviation of slope(SDS)over an area of~640 km^(2) by calculating the slope gradient of each raster cell and using a rectangular moving window method in Arc Map from a 5 m-resolution Digital Elevation Model generated from helicopterobtained *** estimated landslide ages(t)with average surface roughness of mapped landslide deposits(R)quantified by SDS,we fit an exponential landslide deposits surface roughness-age function(t=1.47×10^(6)×e^(-1).46R,r2=0.63)that was used to estimate regional landslide ages in *** conclude that three periods with clusters of regional landslides in lx were revealed by different surface roughness of landslide deposits combing roughness-age function,with the values of 5563-7455 yr B.P.,1724-4151 yr B.P.,and 960-1287 yr B.P..Furthermore,we used our estimates of landslide ages to quantify landslide erosion rates of three corresponding hillslopes in lx ranging from 0.50 to 2.42 mm *** rock varnish radiocarbon dating provides us a feasible option for timing regional landslides of arid regions,the epistemic uncertainty in the dating method should arouse our attention,which could be reduced by increasing the number of samples.

D-serine reduces memory impairment and neuronal damage induced by chronic lead exposure
