Diversified synthetic strategies are extremely important for the structural diversity of uranium-containing polyoxometalates(U-POMs)and their functional expansion.Herein,two sandwich-type U-POMs were reported,which are Na_(5.6)K_(6.4)[(UO_(2))(H_(2)O)(TeW_(9)O_(33))]_(2)·21H_(2)O(UTeW_(9))and Na_(8.9)K_(2.7)H_(1.19)[K_(0.79)(UO_(2))_(2.21)(PW_(9)O_(34))_(2)]·12H_(2)O(UPW_9)using in-situ strategy and lacunary precursor strategy,respectively.UTeW_9 shows a typical Keggin-typeopen or half-sandwich configuration.

BACKGROUND Human-derived gastric cancer organoids(GCOs)are widely used in gastric cancer research;however,the culture success rate is generally low.AIM To explore the potential influencing factors,and the literature on successful culture rates of GCOs was reviewed using meta-analysis.METHODS PubMed,Web of Science,and EMBASE were searched for studies.Two trained researchers selected the studies and extracted data.STATA 17.0 software was used for meta-analysis of the incidence of each outcome event.The adjusted Methodological Index for Non-Randomized Studies scale was used to assess the quality of the included studies.Funnel plots and Egger’s test were used to detect publication bias.Subgroup analyses were conducted for sex,tissue source,histo-logical classification,and the pathological tumor-node-metastasis(pTNM)cancer staging system.RESULTS Eight studies with a pooled success rate of 66.6%were included.GCOs derived from women and men had success rates of 67%and 46.7%,respectively.GCOs from surgery or biopsy/endoscopic submucosal dissection showed success rates of 70.9%and 53.7%,respectively.GCOs of poorly-differentiated,moderately-differentiated and signet-ring cell cancer showed success rates of 64.6%,31%,and 32.7%,respectively.GCOs with pTNM stages I-II and III-IV showed success rates of 38.3%and 65.2%,respectively.Y-27632 and non-Y-27632 use showed success rates of 58.2%and 70%,respectively.GCOs generated with collagenase were more successful than those constructed with Liberase TH and TrypLE(72.1%vs 71%,respectively).EDTA digestion showed a 50%lower success rate than other methods(P=0.04).CONCLUSION GCO establishment rate is low and varies by sex,tissue source,histological type,and pTNM stage.Omitting Y-27632,and using Liberase TH,TrypLE,or collagenase yields greater success than EDTA.

BACKGROUND Three-dimensional organoid culture systems have been established as a robust tool for elucidating mechanisms and performing drug efficacy testing.The use of gastric organoid models holds significant promise for advancing personalized medicine research.However,a comprehensive bibliometric review of this burgeoning field has not yet been published.AIM To analyze and understand the development,impact,and direction of gastric organoid research using bibliometric methods using data from the Web of Science Core Collection(WoSCC)database.METHODS This analysis encompassed literature pertaining to gastric organoids published between 2010 and 2023,as indexed in the WoSCC.CiteSpace and VOSviewer were used to depict network maps illustrating collaborations among authors,institutions and keywords related to gastric organoid.Citation,co-citation,and burst analysis methodologies were applied to assess the impact and progress of research.RESULTS A total of 656 relevant studies were evaluated.The majority of research was published in gastroenterology-focused journals.Globally,Yana Zavros,Hans Clevers,James M Wells,Sina Bartfeld,and Chen Zheng were the 5 most productive authors,while Hans Clevers,Huch Meritxell,Johan H van Es,Marc Van de Wetering,and Sato Toshiro were the foremost influential scientists in this area.Institutions from the University Medical Center Utrecht,Netherlands Institute for Developmental Biology(Utrecht),and University of Cincinnati(Cincinnati,OH,United States)made the most significant contributions.Currently,gastric organoids are used mainly in studies investigating gastric cancer(GC),Helicobacter pylori-infective gastritis,with a focus on the mechanisms of GC,and drug screening tests.CONCLUSION Key focus areas of research using gastric organoids include unraveling disease mechanisms and enhancing drug screening techniques.Major contributions from renowned academic institutions highlight this field’s dynamic growth.

裂缝预测是非常规储层预测的重要内容。裂缝密度不仅反映了裂缝的发育程度,也是影响裂缝孔隙度和渗透率的重要参数。文中提出了一种应用纵波反射振幅分方位傅里叶系数正、余弦分量加权的裂缝密度反演方法。基于线性滑动裂缝等效介质理论,分别推导了含油和含气条件下HTI介质纵波反射振幅傅里叶系数与裂缝密度的关系,并利用二阶傅里叶系数预测裂缝对称轴方位角;研究了地震数据含噪声时二阶傅里叶系数正、余弦分量信噪比之比随裂缝对称轴方位角的变化规律,并提出应用二阶傅里叶系数正、余弦分量加权的裂缝密度反演方法。模型试算结果表明,所提的裂缝密度反演方法具有较强的抗噪性和稳定性;将其应用于实际地震数据,裂缝密度预测结果与测井信息吻合,验证了方法的有效性。

Objective: Few studies have explored the suitability of endometrial thickness (EMT) as a predictor of ectopic pregnancy (EP) in frozen embryo transfer (FET) cycles. Our study aimed to investigate the association between EMT and EP and explore the cutoff value of EMT to predict EP.Methods: This retrospective cohort study included women aged 20-45 years undergoing FET between March 2015 and September 2021. The primary outcomes and measured variables were EP and EMT, respectively. The threshold effect of EMT on EP was calculated based on a constructed smooth curve fitting. The patients were divided into two groups based on the threshold for comparison. A subgroup analysis was conducted to examine whether the relationship between EMT and EP remained consistent across the different subgroups. Additionally, a generalized linear model was used to analyze the interactions between the treatment factors and EMT in EP.Results: In total, 27,549 FET cycles were included, with 27,086 intrauterine and 463 ectopic pregnancies. The risk of EP in the EMT ≤8.7 mm group was 3.36 times higher than that in the EMT >8.7 mm group (odds ratio [OR]: 3.36, 95% confidence interval [CI]: 2.57-4.05). Subgroup analysis further indicated that a thin EMT was a risk factor for EP, independent of other risks. We also noted that diminished ovarian reserve, tubal factor infertility, transfer of two embryos, or transfer of cleavage-stage embryos may further enhance the risk of EP in patients with thin endometrium.Conclusions: Thin endometrium was associated with an increased risk of EP. Patients with EMT less than 8.7 mm need close medical observation and management after embryo transfer. In addition, diminished ovarian reserve, tubal factor infertility, transfer of two embryos, or transfer of cleavage-stage embryos may significantly enhance the effect of a thin endometrium on the risk of EP.

Combinatorial metabolic engineering of Saccharomyces cerevisiae for improved production of 7-dehydrocholesterol

7-Dehydrocholesterol(7-DHC),a key pharmaceutical intermediate in the production of vitamin D3,has a wide range of applications.To explore fermentative synthesis of 7-DHC,a 7-DHC-producing Saccharomyces cerevisiae strain was constructed by blocking the competitive pathway,eliminating rate-limiting steps,altering global reg-ulation,and pathway compartmentalization.After blocking the competitive pathway by disrupting ERG5 and ERG6 and introducing DHCR24 from Gallus gallus,S.cerevisiae produced 139.72 mg/L(17.04 mg/g dry cell weight,hereafter abbreviated as DCW)7-DHC.Subsequent alteration of global regulation by deleting ROX1 and overexpressing UPC2-1 increased 7-DHC production to 217.68 mg/L(37.56 mg/g DCW).To remove the accu-mulated squalene,the post-squalene pathway was strengthened by co-overexpression of PGAL1-driven ERG11 and PGAL10-driven ERG1,which improved 7-DHC titer and yield to 281.73 mg/L and 46.78 mg/g DCW,respectively,and reduced squalene content by 90.12%.We surmised that the sterol precursors in the plasma membrane and peroxisomes may not be accessible to the pathway enzymes,thus we re-localized DHCR24p and Erg2p-GGGGS-Erg3p to the plasma membrane and peroxisomes,boosting 7-DHC production to 357.53 mg/L(63.12 mg/g DCW).Iron supplementation further increased 7-DHC production to 370.68 mg/L in shake flasks and 1.56 g/L in fed-batch fermentation.This study demonstrates the power of global regulation and subcellular relocalization of key enzymes to improve 7-DHC synthesis in yeast.

Background The therapeutic effect of Bifidobacterium and Lactobacillus on necrotizing enterocolitis(NEC)in very-low-birth-weight preterm infants was controversial,and we aimed to explore the exact impact of the two probiotics.Methods The PubMed,EMBASE,Web of Science and Cochrane Library were systematically searched for studies published from January 1,2010 to February 28,2019.Results were combined with fixed-effect model or random-effect model with specific conditions.Sensitivity analysis was conducted by the trim-and-fill method,and the Begger's and Egger's test were used to measure publication bias.Results The meta-analysis included 16 original articles with 4632 very-low-birth-weight preterm infants.With respect to the intervention of Bifidobacterium,we estimated non-significant decrease in the morbidity of NEC with a risk ratio(RR)of 0.75[95%confidence internal(CI)0.56-1.01,P=0.06].Regarding the effect of Lactobacillus,there was no evidence of significant lower risk in the incidence of NEC(RR=0.67,95%CI 0.39-1.17,P=0.16).The use of mixture of probiotics(Bifidobacterium and Lactobacillus)reduced the risk of NEC in the probiotics group(RR=0.45,95%CI 0.25-0.80,P=0.007).Conclusion The mixture of Bifidobacterium and Lactobacillus could prevent the morbidity of NEC in very-low-birth-weight preterm infants.But Bifidobacterium or Lactobacillus alone did not show this effect.

高角度裂缝的存在导致地下介质呈现方位各向异性,具有水平对称轴的横向各向同性介质中反射系数随方位角的变化可以近似表示为傅里叶级数的形式.傅里叶系数的大小取决于背景介质和裂缝参数,傅里叶系数的相位角取决于裂缝对称轴方向.常规傅里叶级数分析利用逐个时间采样点的方位地震数据求和计算傅里叶系数,计算结果易受噪声影响.为了提高傅里叶系数估计的抗噪性和稳定性,将常规傅里叶级数分析与AVAz(振幅随方位角变化)反演方法相结合,提出一种基于整形正则化的AVAz二阶傅里叶系数反演方法.常规整形正则化约束中的整形算子形态固定,提出一种基于方位各向异性强度的自适应整形算子,利用基于常规傅里叶级数分析的二阶傅里叶系数初步估计结果计算阻尼因子,结合一阶差分矩阵构建自适应整形算子,达到在反演中自适应地调整整形算子形态的目的.理论测试表明,基于自适应整形正则化的AVAz反演方法具有较强的抗噪性,能够有效提高二阶傅里叶系数反演的稳定性.实际叠前地震数据应用结果显示裂缝预测结果与测井资料相吻合,证明了基于自适应整形正则化AVAz反演进行裂缝预测的有效性.

单味中药的开发是目前中药领域技术创新的一个重要方向,研究其知识产权综合性保护体系,对单味药材的全链条创新企业的知识产权保护具有重要意义。基于此,该文以九芝堂药业构建的裸花紫珠知识产权综合性保护体系作为模型进行实证研究,从专利、行政保护、商标、外观设计、非物质文化遗产等适用于中药知识产权的保护形式进行分析,对九芝堂药业构建的综合性保护体系值得借鉴之处和不足之处进行探讨,并提出如下建议:(1)重视原料药材种植/炮制方法的专利申请;(2)重视地理标志、道地药材、植物新品种的保护;(3)积极推进产品、技术升级;(4)产品迭代时期申请数据保护;(5)注重专利、行政保护的布局时机;(6)注重提高商誉;(7)提高知识产权保护意识,推进产学研深度融合。希望从事单味药材开发的创新企业能够从裸花紫珠实例中借鉴经验,并且优化策略,为相关产品更好地进行知识产权保护。
