Mechanomedicine:From Mechanogenomics to Mechano-Immunoengineering

Mechanobiology is a rapidly growing field that has expanded significantly over the past 40 years. There have been great examples of mechanobiology research on the cardiovascular, musculoskeletal, and respiratory systems. The field has evolved from the system level to the organ and tissue levels, and down to the cellular and molecular levels, benefiting from scientific progress and technological developments in molecular and cell biology, biomaterials, imaging techniques, and computational tools. Additionally, the interdisciplinary integration of mechanobiology with cell engineering, tissue engineering, and drug delivery has led to promising mechanomedicine applications in both therapeutics and ***, I will discuss two examples at the molecular and cellular levels:(1) mechanical regulation of epigenomics for cell reprogramming, and (2) engineering the mechanical properties of artificial antigen-presenting cells (APCs) for immune cell modulation and cancer *** memory of its identity is determined by the epigenetic state of the cells. However, how cells control the epigenetic state and thus cell fate is not well understood, and how mechanical factors such as surface topography, cell morphology,mechanical properties of the extracellular matrix (ECM), and the mechanical deformation of cells regulate the epigenetic state is not clear. In an early study, we showed that biophysical cues, in the form of parallel microgrooves on the surface of cell-adhesive substrates, can replace the effects of small-molecule epigenetic modifiers and significantly improve reprogramming efficiency from fibroblasts to induced pluripotent stem cells. The mechanism relies on the mechanomodulation of the cells’ epigenetic state through the activity of histone deacetylase and H3 methyltransferase. To enhance the DC electrical performance of cross-linked polyethylene(XLPE),the graftable antioxidant methacrylic acid 2-hydroxy-3-(4-anilinoanilino)propyl ester(GA),which contains carbonyl and amino groups,and the crosslinking coagent trimethylolpropane trimethacrylate(TMPTMA),which contains carbonyl groups,are individually or co-grafted onto *** slightly higher deep trap density introduced by higher grafting concentration of TMPTMA results in more significant suppression effect of conductance current and enhanced breakdown strength at a lower temperature,while the suppression effect for the conductance current at 90°C becomes weakened due to limited trap ***,the deeper energy level introduced by GA suppresses the conductance current and improves the electrical strength of XLPE at 90°C more *** co-grafting,the conductance current of XLPE in a wide range temperature can be significantly reduced,especially for the temperature dependence of conductance current,which is beneficial to suppress the field strength *** results of thermally stimulated current and molecular simulation show that the polar groups of two monomers introduce deep charge traps in *** co-grafting system ensures the rationality of the crosslinking reaction kinetics and does not affect the cable manufacturing.

Layered rare-earth hydroxides(LREHs) draw wide research interest because of their peculiar crystal structure,rich interlayer chemistry and abundant functionality of the RE element,but are limited to the two categories of RE_(2)(OH)_(5)A·nH_(2)O(A:typical of Cl^(-)or NO_(3)^(-)) and RE_(2)(OH)_(4)SO_(4)·nH_(2)*** the other hand,rare-earth oxysulfates(RE_(2)O_(2)SO_(4)) have attracted attention due to their properties of large-capacity oxygen storage,low-temperature magnetism and luminescence,but their preparation procedure mostly involves toxic SO_(x) gases and/or complicated *** this work,RE_(2)(OH)_(2)CO_(3)SO_(4)·nH_(2)O as a new family of LREHs(RE=Gd-Lu lanthanides and Y) were produced via hydrothermal reaction,from which phase-pure RE_(2)O_(2)SO_(4) was derived via subsequent annealing at 800℃ in air without the involvement of SO_(x),The compounds were thoroughly characterized to reveal the intrinsic influence of lanthanide contraction(RE^(3+) radius) on crystal structure,thermal behavior(dehydroxylation/decarbonation/desulfurization),vibrational property and crystallite *** analyzing the photoluminescence of Eu^(3+) and Sm^(3+)in the Gd_(2)O_(2)SO_(4) typical host it is found that the 617 nm(Eu^(3+),λ_(ex)=275 nm) and 608 nm(Sm^(3+),λ_(ex)=407 nm) main emissions can retain as high as ^(7)9.6% and 85.5%of their room-temperature intensities at 423 K,with activation energies of ~0.19 and 0.21 eV for thermal quenching,*** also indicates that both the phosphors have the potential for optical temperature sensing via the fluorescence intensity ratio(FIR) technology,whose maximum relative sensitivity reaches -2.70%/K for Eu^(3+)and 1.73%/K for Sm^(3+) at 298 K.

The advent of tyrosine kinase inhibitors(TKI)targeting BCR-ABL has drastically changed the treatment approach of chronic myeloid leukemia(CML),greatly prolonged the life of CML patients,and improved their ***,TKI resistance is still a major problem with CML patients,reducing the efficacy of treatment and their quality of *** resistance is mainly divided into BCR-ABL-dependent and BCR-ABL-independent ***,the main clinical strategy addressing TKI resistance is to switch to newly developed ***,data have shown that these new drugs may cause serious adverse reactions and intolerance and cannot address all resistance ***,finding new therapeutic targets to overcome TKI resistance is crucial and the ubiquitin-proteasome system(UPS)has emerged as a *** UPS mediates the degradation of most proteins in organisms and controls a wide range of physiological *** recent years,the study of UPS in hematological malignant tumors has resulted in effective treatments,such as bortezomib in the treatment of multiple myeloma and mantle cell *** CML,the components of UPS cooperate or antagonize the efficacy of TKI by directly or indirectly affecting the ubiquitination of BCR-ABL,interfering with CML-related signaling pathways,and negatively or positively affecting leukemia stem *** of these molecules may help overcome TKI resistance and treat *** this review,the mechanism of TKI resistance is briefly described,the components of UPS are introduced,existing studies on UPS participating in TKI resistance are listed,and UPS as the therapeutic target and strategies are discussed.

Influence of Recycled Concrete Fine Powder on Durability of Cement Mortar

In this paper,the durability of cement mortar prepared with a recycled-concrete fine powder(RFP)was examined;including the analysis of a variety of aspects,such as the carbonization,sulfate attack and chloride ion erosion *** results indicate that the influence of RFP on these three aspects is *** carbonization depth after 30 days and the chloride diffusion coefficient of mortar containing 10%RFP decreased by 13.3%and 28.19%.With a further increase in the RFP content,interconnected pores formed between the RFP particles,leading to an acceleration of the penetration rate of CO_(2)and Cl^(−).When the RFP content was less than 50%,the corrosion resistance coefficient of the compressive strength of the mortar was 0.84-1.05 after 90 days of sulfate *** the expansion and cracking of the mortar was effectively alleviated due to decrease of the gypsum *** electron microscope(SEM)analysis has confirmed that 10%RFP contributes to the formation of a dense microstructure in the cement mortar.

Mobile edge computing(MEC)-enabled satellite-terrestrial networks(STNs)can provide Internet of Things(IoT)devices with global computing ***,the network state information is uncertain or *** deal with this situation,we investigate online learning-based offloading decision and resource allocation in MEC-enabled STNs in this *** problem of minimizing the average sum task completion delay of all IoT devices over all time periods is *** decompose this optimization problem into a task offloading decision problem and a computing resource allocation problem.A joint optimization scheme of offloading decision and resource allocation is then proposed,which consists of a task offloading decision algorithm based on the devices cooperation aided upper confidence bound(UCB)algorithm and a computing resource allocation algorithm based on the Lagrange multiplier *** results validate that the proposed scheme performs better than other baseline schemes.

Wearable electronics powered by flexible energy storage devices have captured global *** low-temperature conditions,unfortunately,solidification of flexible hydrogel electrolyte and decreased pseudo-capacity of these devices largely hamper their practical *** this study,photothermally-active Prussian blue(PB)was introduced onto poly(3,4-ethylenedioxythiophene)/polyacrylamide(PEDOT/PAM)networks to address the challenges of electrolyte solidification and degraded pseudo-capacitance for flexible all-in-one device at low *** as-constructed PB/PEDOT/PAM hydrogel device delivers stable electrochemical performance and remarkable mechanical property with 1652%***,this hydrogel device well retains its flexibility in cold environment with a freezing point below−30℃.The incorporation of PB extends the voltage range to 1.5 V as a single device,thus significantly enhancing the electrochemical performance as an all-inone integrated *** from the outstanding photo-to-thermal conversion ability of embedded PB nanocubes,the temperature of the assembled all-in-one PB/PEDOT/PAM device increased from^(−2)0 to 17.7℃ after solar-light irradiation for only 5 ***,the degraded pseudo-capacitance was subsequently boosted to 287.1%of its original capacitance at−20℃.This study establishes a connection between flexible all-solid-state hydrogel devices and photothermally enhanced pseudocapacitors in freezing environments,thereby expanding the potential applications of multi-functional pseudo-capacitors.

Centennial Analysis of Human Activity Intensity and Associated Historical Events in Heilongjiang River Sino-Russo Watershed

Human activities in a transborder watershed are complex under the influence of domestic policies,international relations,and global *** the forces driving human activity change is important for the development of transborder *** this study,we used global historical land cover data,the hemeroby index model,and synthesized major historical events to analyze how human activity intensity changed in the Heilongjiang River(Amur River in Russia)watershed(HLRW).The results showed that there was a strong spatial heterogeneity in the variation of human activity intensity in the HLRW over the past century(1900-2016).On the Chinese side,the human activity intensity change shifted from the plain areas for agricultural reclamation to the mountainous areas for timber *** the Russian side,human activity intensity changes mostly concentrated along the Trans-Siberian Railway and the Baikal-Amur *** variation of human activity intensity tended to respond to regional events while regionalized variation tends to reflect national policy change or broad international *** similarities and differences between China and Russia in policies and positions in international events resulted in synchronous and asynchronous changes in human activity ***,policy shifts were often confined by the natural features of the *** results reveal the historical origins and fundamental connotations of watershed development and contribute to formulating regional management policies that coordinate population,eco-nomic,social,and environmental activities.
