Icariin plus curcumol enhances autophagy through the mTOR pathway and promotes cathepsin B-mediated pyroptosis of prostate cancer cells

Objective:To examine the effect of icariin plus curcumol on prostate cancer cells PC3 and elucidate the underlying ***:We employed the Cell Counting Kit 8 assay and colony formation assay to assess cell viability and *** expression was analyzed using monodansylcadaverine *** and Western blot analyses were used to evaluate protein expressions related to autophagy,pyroptosis,and the mTOR *** damage was examined using the lactate dehydrogenase ***,cathepsin B and NLRP3 were detected by ***:Icariin plus curcumol led to a decrease in PC3 cell proliferation and an enhancement of *** levels of LC3-Ⅱ/LC3-Ⅰand beclin-1 were increased,while the levels of p62 and mTOR were decreased after treatment with icariin plus *** changes were reversed upon overexpression of ***,3-methyladenine resulted in a decrease in inflammatory cytokines,pyroptosis-related protein levels,and lactate dehydrogenase concentration,compared to the icariin plus curcumol *** cathepsin B reversed the regulatory effects of icariin plus ***:Icariin plus curcumol demonstrates great potential as a therapeutic agent for castration-resistant prostate cancer by enhancing autophagy via the mTOR pathway and promoting pyroptosis mediated by cathepsin *** findings provide valuable insights into the molecular mechanisms underlying the therapeutic potential of icariin and curcumol for prostate cancer treatment.

CCT Curves of Low-Carbon Mn-Si Steels and Development of Water-Cooled Bainitic Steels

CCT curves of Mn-Si steels with different manganese contents or carbon contents were determined. The results show that the transformation range of bainite can be separated from that of ferrite when the manganese content approaches a certain content, and the incubation period of ferrite increases more significantly than that of bainite transformation with the increase of carbon content in Mn-Si steels. Furthermore, water-cooled bainitic steels without adding expensive alloying element were developed. Granular bainite was obtained when a bar with diameter of 300 mm was cooled by water, and a mixed microstructure of granular bainite and martensite was obtained in watercooled plate with thickness of 40 mm. The developed water-cooled bainitic Steels containing no expensive alloying element showed a good combination of strength and toughness. The tensile strength, yield strength, and toughness （AKU at --20℃ ） of bar with diameter of 300 mm after water cooling were higher than 850 MPa, 620 MPa, and 65 J, respectively, and those of plate with thickness of 40mm after water cooling were higher than 1000 MPa, 800 MPa, and 50J, respectively.

Effects of GBE50 on hippocampal CA1 synaptic plasticity,learning and memory in an experimental rat model of aging

The content of total flavonoids in an extract of Ginkgo biloba, called GBE50, is 44% by weight. This is significantly greater than that in a standard extract of Ginkgo biloba, designated EGB761. To date, the mechanisms by which GBE50 and EGB761 function remain poorly understood. In the present study, an experimental rat model of aging was induced by intraperitoneal injection of D-galactose, followed by intragastric perfusion of GBE50 （30, 60 mg/kg）, or EGB761 （60 mg/kg）. The water maze scores and hippocampal CA1 synaptic plasticity were evaluated. In the place navigation test, the GBE50 group rats did better than EGB761, while similar scores were obtained in the spatial probe test, and in the platform-switched test. In addition, long-term potentiation was significantly enhanced following high-frequency stimulation in the GBE50 and EGB761 groups, compared with the model group. These results demonstrate that GBE50 and EGB761 improved the learning and memory of aging rats. In particular, GBE50 administered at the 60 mg/kg dose exhibited superior effects over EGB761 at the same 60 mg/kg dose. Furthermore, the enhancement of hippocampal synaptic plasticity may be an underlying mechanism.

Relationship between ADP-induced platelet-fibrin clot strength and anti-platelet responsiveness in ticagrelor treated ACS patients

Background Ticagrelor provides enhanced antiplatelet efficacy but increased risk of bleeding and dyspnea. This study aimed to display the relationship between ADP-induced platelet-fibrin clot strength （MAADP） and clinical outcomes in acute coronary syndrome （ACS） patients treated by ticagrelor. Methods Consecutive Chinese-Han patients with ACS who received maintenance dose ofticagrelor on top of aspirin were recruited. After 5-day ticagrelor maintenance treatment, MAADP measured by thrombelastography （TEG） were recorded for the evaluation of ticagrelor anti-platelet reactivity. Pre-specified cutoffs of MAADP 〉 47 mm for high on-treatment platelet reactivity （HTPR） and MAADP 〈 31 mm for low on-treatment platelet reactivity （LTPR） were applied for evaluation. The occurrences of primary ischemic cardiovascular events （including a composite of cardiac death, non-fatal myocardial infarction and stroke）, the Thrombolysis in Myocardial Infarction （TIMI） defined bleeding events, and ticagrelor related dyspnea were recorded after a follow-up of three months. Results Overall, 176 ACS patients （Male： 79.55%, Age： 59.91 ±10.54 years） under ticagrelor maintenance treatment were recruited. The value of MAADP ranged from 4.80% to 72.90% （21.27% ± 12.07% on average）, with the distribution higher skewed towards the lower values. Using the pre-specific cutoffs for HTPR and LTPR, seven patients （3.98%） were identified as HTPR and 144 patients （81.82%） as LTPR. After a follow-up of three months in 172 patients, major cardiovascular events occurred in no patient, but TIMI bleeding events in 81 （47.09%） with major bleedings in three patients. All patients with major bleedings were classified as LTPR. Ticagrelor related dyspnea occurred in 31 （18.02%） patients, with 30 （21.28%） classified as LTPR and no one as HTPR （P = 0.02）. Conclusions In ticagrelor treated ACS patients, MAADP measured by TEG might be valuable for the prediction of major bleeding and ticagrelor related dyspnea. Due to

Energetic Semiconductor bridge(ESCB)based on reactive multilayered films(RMFs)has a promising application in the miniature and intelligence of initiator and pyrotechnics *** the ignition enhancement mechanism of RMFs on semiconductor bridge(SCB)during the ignition process is crucial for the engineering and practical application of advanced initiator and pyrotechnics *** this study,a one-dimensional(1D)gas-solid two-phase flow ignition model was established to study the ignition process of ESCB to charge particles based on the reactivity of Al/MoO_(3) *** order to fully consider the coupled exothermic between the RMFs and the SCB plasma during the ignition process,the heat release of chemical reaction in RMFs was used as an internal heat source in this *** is found that the exothermal reaction in RMFs improved the ignition performance of *** the process of plasma rapid condensation with heat release,the product of RMFs enhanced the heat transfer process between the gas phase and the solid charge particle,which accelerated the expansion of hot plasma,and heated the solid charge particle as well as gas phase region with low *** addition,it made up for pressure loss in the gas *** the plasma dissipation process,the exothermal chemical reaction in RMFs acted as the main heating source to heat the charge particle,making the surface temperature of the charge particle,gas pressure,and gas temperature rise *** result may yield significant advantages in providing a universal ignition model for miniaturized ignition devices.

Increasing effective panicle number per plant(EPN)is one approach to increase yield potential in ***,molecular mechanisms underlying EPN remain *** this study,we integrated mapbased cloning and genome-wide association analysis to identify the EPN4 gene,which is allelic to NARROW LEAF1(NAL1).Overexpression lines containing the Teqing allele(TQ)of EPN4 had significantly increased ***-EPN4^(TQ) in japonica(geng)cultivar Lemont(LT)exhibited significantly improved EPN but decreased grain number and flag leaf size relative to *** analysis indicated that accessions with EPN4-1 had medium EPN,medium grain number,and medium grain weight,but had the highest grain yield among seven haplotypes,indicating that EPN4-1 is an elite haplotype of EPN4 for positive coordination of the three components of grain ***,accessions carrying the combination of EPN4-1 and haplotype GNP1-6 of GNP1 for grain number per panicle showed higher grain yield than those with other allele ***,pyramiding of EPN4-1 and GNP1-6 could be a preferred approach to obtain high yield potential in breeding.

MPI/OpenMP-Based Parallel Solver for Imprint Forming Simulation

In this research,we present the pure open multi-processing(OpenMP),pure message passing interface(MPI),and hybrid MPI/OpenMP parallel solvers within the dynamic explicit central difference algorithm for the coining process to address the challenge of capturing fine relief features of approximately 50 *** such precision demands the utilization of at least 7 million tetrahedron elements,surpassing the capabilities of traditional serial programs previously *** mitigate data races when calculating internal forces,intermediate arrays are introduced within the OpenMP *** helps ensure proper synchronization and avoid conflicts during parallel ***,in the MPI implementation,the coins are partitioned into the desired number of *** division allows for efficient distribution of computational tasks across multiple *** simulation examples are conducted to compare the three solvers with serial programs,evaluating correctness,acceleration ratio,and parallel *** results reveal a relative error of approximately 0.3%in forming force among the parallel and serial solvers,while the predicted insufficient material zones align with experimental ***,speedup ratio and parallel efficiency are assessed for the coining process *** pureMPI parallel solver achieves a maximum acceleration of 9.5 on a single computer(utilizing 12 cores)and the hybrid solver exhibits a speedup ratio of 136 in a cluster(using 6 compute nodes and 12 cores per compute node),showing the strong scalability of the hybrid MPI/OpenMP programming *** approach effectively meets the simulation requirements for commemorative coins with intricate relief patterns.

Understanding livelihood vulnerability:a perspective from Western Sichuan’s ethnic rural settings

To explore the livelihood status and key influencing factors of rural households in the minority areas,we collected flat data from 284 rural households in 32 villages across 12 counties of Western Sichuan from 2021 to *** conducted participatory household survey on the livelihood status of the rural households and try to identify the key factors to influence their livelihood vulnerability using multiple linear *** results showed that:the livelihood situation of the rural households is relatively *** vulnerability varies significantly with the income levels,education levels,and income *** vulnerability of farm households,categorized from low to high livelihood types,follows the sequence:non-agricultural dominant households,non-agricultural households,agricultural dominant households,and pure agricultural *** degree of damage to the natural environment,education costs,loan opportunities,the proportion of agricultural income to annual household income,and the presence of sick people in the household have significant positive effects on the livelihood vulnerability index(LVI)of rural households;while help from relatives and friends,net income per capita,household size,household education,agricultural land area,participation in industrial organizations,number of livestock,purchase of commercial houses,drinking water source,and self-supply of food have significant negative *** on the findings,we believe that local rural households operate in a complex livelihood system and recommend continuous interventions targeting key influences to provide empirical research support for areas facing similar situations.
