本文简要综述了时域连续Galerkin(TCG),改进TCG,以及时域间断Galerkin(TDG)以及高阶单步法等几种典型的高阶时域积分算法。若采用p阶多项式对基本未知量进行插值近似,采用单场格式的常规TCG算法、改进TCG算法、两场格式的TDG算法求得的位移和速度分别具有p、2p、2p+1阶收敛精度,在计算成本相当的情况下,改进TCG算法具有最高收敛精度。高阶单步法多为后处理的算法,大多具有4阶精度。相比最常用的二阶算法,高阶单步法操作简单,易于实施,保留二阶算法无条件稳定、计算效率高等优点,可对二阶算法进行误差估计,并可根据精度需求,指导时间步长自适应调整。

In the realm of autoimmune and inflammatory diseases,the cyclic GMP-AMP synthase(cGAS)stimulator of interferon genes(STING)signaling pathway has been thoroughly investigated and *** this,the clinical approval of drugs targeting the cGAS-STING pathway has been *** Total glucosides of paeony(TGP)is highly anti-inflammatory and is commonly used in the treatment of rheumatoid arthritis(RA),emerged as a subject of our *** found that the TGP markedly reduced the activation of the cGAS-STING signaling pathway,triggered by various cGAS-STING agonists,in mouse bone marrow-derived macrophages(BMDMs)and Tohoku Hospital Pediatrics-1(THP-1)*** inhibition was noted alongside the suppression of interferon regulatory factor 3(IRF3)phosphorylation and the expression of interferon-beta(IFN-β),C-X-C motif chemokine ligand 10(CXCL10),and inflammatory mediators such as tumor necrosis factor-alpha(TNF-α)and interleukin-6(IL-6).The mechanism of action appeared to involve the TGP’s attenuation of the STING-IRF3 interaction,without affecting STING oligomerization,thereby inhibiting the activation of downstream signaling *** vivo,the TGP hindered the initiation of the cGAS-STING pathway by the STING agonist dimethylxanthenone-4-acetic acid(DMXAA)and exhibited promising therapeutic effects in a model of acute liver injury induced by lipopolysaccharide(LPS)and D-galactosamine(D-GalN).Our findings underscore the potential of the TGP as an effective inhibitor of the cGAS-STING pathway,offering a new treatment avenue for inflammatory and autoimmune diseases mediated by this pathway.

Lipotoxicity is a pivotal factor that initiates and exacerbates liver injury and is involved in the development of metabolic-associated fatty liver disease(MAFLD).However,there are few reported lipotoxicity ***,we identified a natural anti-lipotoxicity candidate,HN-001,from the marine fungus Aspergillus ***-001 dose-and time-dependently reversed palmitic acid(PA)-induced hepatocyte *** protection was associated with IRE-1a-mediated XBP-1 splicing inhibition,which resulted in suppression of XBP-1s nuclear translocation and transcriptional *** of XBP-1s attenuated lipotoxicity,but no additional ameliorative effect of HN-001 on lipotoxicity was observed in XBP-1s knockdown ***,the ER stress and lipotoxicity amelioration was associated with *** HN-001 and the PLA2 inhibitor MAFP inhibited PLA2 activity,reduced lysophosphatidylcholine(LPC)level,subsequently ameliorated *** contrast,overexpression of PLA2 caused exacerbation of lipotoxicity and weakened the anti-lipotoxic effects of ***,HN-001 treatment suppressed the downstream pro-apoptotic JNK *** vivo,chronic administration of HN-001(i.p.)in mice alleviated all manifestations of MAFLD,including hepatic steatosis,liver injury,inflammation,and *** effects were correlated with PLA2/IRE-1a/XBP-1s axis and JNK signaling *** data indicate that HN-001 has therapeutic potential for MAFLD because it suppresses lipotoxicity,and provide a natural structural basis for developing anti-MAFLD candidates.

Objective:Based on the transcriptome sequencing data of Chinese patients with liver cancer,we found new subpopulations of liver cancer and identified specific markers,so as to provide new ideas for accurate diagnosis and individualized treatment of liver ***:High throughput transcriptome sequencing was performed on 296 samples of cancer tissues and paracancerous tissues from patients with liver cancer from Run Run Shaw Hospital Affiliated to Zhejiang University,and the obtained data were subjected to gene differential expression analysis,functional enrichment analysis,immune infiltration,lineage analysis of metabolic pathways,co-expression network analysis and so *** analyzed the specific differentially expressed genes of HCC,and verified in the tissue microarray of liver cancer patients treated with sorafenib to identify the HCC subgroup marker genes and efficacy marker ***:We found a new subgroup of hepatocellular carcinoma—HCC,that is very similar to *** sequencing data analysis showed that HCC and ICC are highly similar in terms of prognosis,immune infiltration,metabolic pathway and hypoxia *** the tissue microarray of liver cancer patients treated with sorafenib,the subgroup marker gene CEACEM5/6 of HCC and the efficacy marker gene FGFR1 were ***:HCC is a new subgroup of hepatocellular carcinoma which is very similar to cholangiocarcinoma and has a similar expression ***5/6 can be used as a marker gene of HCC to distinguish it from other HCC samples,and FGFR1 can predict the response to sorafenib in liver cancer patients.

[目的]本试验以舍饲育肥牦牛为研究对象,探究日粮精粗比对舍饲育肥牦牛瘤胃代谢的影响,期望为青海牦牛规模化、产业化和现代化养殖过程中日粮精粗比调控提供参考。[方法]对15头试验牛群进行分组,分别饲喂精粗(质量)比为3∶7(A组)、5∶5(B组)、7∶3(C组)的全混合日粮(total mixed ration,TMR)。饲喂90 d后于凌晨空腹屠宰,采集瘤胃液和瘤胃上皮组织,通过16S核糖体RNA(16S ribosomal RNA,16S rRNA)高通量测序、气相色谱、RT-qPCR以及Western blot试验方法,探究不同精粗比日粮饲喂下,舍饲育肥牦牛瘤胃细菌菌群结构、瘤胃发酵参数以及瘤胃上皮中挥发性脂肪酸(VFA)转运载体表达量的变化。[结果]日粮精粗比显著影响瘤胃细菌菌群结构,随着精料占比升高,在门水平上,拟杆菌门(Bacteroidetes)、螺旋体门(Spirochaetes)和黏胶球形菌门(Lentisphaerae)相对丰度显著上升(P<0.05),厚壁菌门(Firmicutes)和软壁菌门(Tenericutes)相对丰度显著下降(P<0.05);属水平上,随着精料比例提高,理研菌科RC9肠道群(Rikenellaceae RC9 gut group)、克里斯腾森菌科R7群(Christensenellaceae R7 group)、Saccharofermentans、产琥珀酸菌属(Succiniclasticum)和Lachnoclostridium相对丰度显著升高(P<0.05),C组牦牛瘤胃球菌属(Ruminococcus)和拟杆菌属(Bacteroides)相对丰度显著低于A组(P<0.05),B组和C组牦牛普氏菌属(Prevotella)、假丁酸弧菌(Pseudobutyrivibrio)和密螺旋体属(Treponema)相对丰度显著高于A组(P<0.05)。日粮精粗比对瘤胃pH值及VFA产量影响显著,精粗比上升显著下调瘤胃内容物pH值(P<0.05),乙酸浓度、乙酸浓度/丙酸浓度显著降低(P<0.05),丙酸、戊酸和总VFA浓度显著升高(P<0.05),与A、B组相比,C组丁酸和异丁酸浓度显著升高(P<0.05)。日粮精粗比对瘤胃上皮VFA转运载体表达量影响显著。随着日粮精料比例升高,腺瘤下调蛋白基因(DRA)、假定阴离子转运载体1基因(PAT1)、单羧酸转运载体1基因(MCT1)和单羧酸转运载体4基因(MCT4)在mRNA水平的相对表达量显著升高(P<0.05),而阴离子交换蛋白2基因(AE2)在mRNA水平和蛋白水平的相对表达量则显著下降(P<0.05),上述基因的蛋白表达趋势与mRNA的一致。随着日粮精粗比例升高,牦牛的生产性能,即平均日增重、饲料转化率、屠宰率也随之提高,且具有显著差异(P<0.05)。[结论]饲喂高精料日粮可提升牦牛瘤胃中非纤维素降解菌的相对丰度,使大量能量饲料和蛋白饲料快速转化为VFA,为机体供能。激活瘤胃上皮细胞中VFA转运载体,提高其表达量,加快VFA转运入血的速度,以提高脂肪代谢底物,最终达到快速育肥的效果。

稀土永磁材料是当今人类社会不可缺少的组成部分,自20世纪60年代问世以来一直在生产和生活中扮演着重要的角色。随着科技的进步和生活水平的提升,人们对工业产品及功能器件等提出了更高的期望,因此对稀土永磁材料的性能也有了更高的要求,包括能量密度、高温性能等。近年来,化学法成为合成稀土永磁材料常见的方法,通过"自下而上"的过程实现对产物形貌、尺寸等方面的精确调控,为制备高性能磁体提供了新的思路。化学合成对产物尺寸的控制在交换耦合磁体方面也有着独特的优势,可以通过精确控制双相耦合磁体中软磁相的尺寸以充分实现交换耦合效应,在提升磁体能量密度方面有着广阔的前景。从化学合成出发,介绍了近年来不同稀土永磁材料制备方面的工作,同时也总结了稀土永磁基耦合磁体在化学制备方面的进展和挑战,对未来化学合成稀土永磁材料的发展做出了分析和展望。

Aging is a contributor to liver ***,the concept of liver aging has become prominent and has attracted considerable interest,but its underlying mechanism remains poorly *** our study,the internal mechanism of liver aging was explored via multi-omics analysis and molecular experiments to support future targeted *** aged rat liver model was established with D-galactose,and two other senescent hepatocyte models were established by treating HepG2 cells with D-galactose and *** then performed transcriptomic and metabolomic assays of the aged liver model and transcriptome analyses of the senescent hepatocyte *** livers,genes related to peroxisomes,fatty acid elongation,and fatty acid degradation exhibited down-regulated expression with aging,and the hepatokine Fgf21 expression was positively correlated with the down-regulation of these *** senescent hepatocytes,similar to the results found in aged livers,FGF21 expression was also ***,the expressions of cell cycle-related genes were significantly down-regulated,and the down-regulated gene E2F8 was the key cell cycle-regulating transcription *** then validated that FGF21 overexpression can protect against liver aging and that FGF21 can attenuate the declines in the antioxidant and regenerative capacities in the aging *** successfully validated the results from cellular and animal experiments using human liver and blood *** study indicated that FGF21 is an important target for inhibiting liver aging and suggested that pharmacological prevention of the reduction in FGF21 expression due to aging may be used to treat liver aging-related diseases.
