China experienced significant flooding in the summer of 2020 and multiple extreme cold surges during the winter of 2020/21.Additionally,the 2020 typhoon season had below average activity with especially quiet activity during the first half of the season in the western North Pacific(WNP).Sea surface temperature changes in the Pacific,Indian,and Atlantic Oceans all contributed to the heavy rainfall in China,but the Atlantic and Indian Oceans seem to have played dominant roles.Enhancement and movement of the Siberian High caused a wavier pattern in the jet stream that allowed cold polar air to reach southward,inducing cold surges in China.Large vertical wind shear and low humidity in the WNP were responsible for fewer typhoons in the first half of the typhoon season.Although it is known that global warming can increase the frequency of extreme weather and climate events,its influences on individual events still need to be quantified.Additionally,the extreme cold surge during 16–18 February 2021 in the United States shares similar mechanisms with the winter 2020/21 extreme cold surges in China.

The China Meteorological Administration(CMA)has recently developed a new global monthly homogenized land-surface air temperature data set.Based on this data set,we reanalyzed the change in global annual mean land-surface air temperature(LSAT)during three time periods(1901–2014,1979–2014 and 1998–2014).The results show that the linear trends of global annual mean LSAT were 0.104°C/decade,0.247°C/decade and 0.098°C/decade for the three periods,respectively.The trends were statistically significant except for the period 1998–2014,the period that

The programmed cell death-1(PD-1)/programmed cell death ligand 1(PD-L1)signaling pathway is an important mechanism in tumor immune escape,and expression of PD-L1 on tumor cells has been reported more frequently.However,accumulating evidence suggests that PD-1/PD-L1 is also widely expressed on immune cells,and that regulation is also critical for tumor immune responses.In this review,we emphasized that under solid tumor conditions,the immunoregulatory effects of immune cells expressing PD-1 or PD-L1,affected the prognoses of cancer patients.Therefore,a better understanding of the mechanisms that regulate PD-1 or PD-L1 expression on immune cells would provide clear insights into the increased efficacy of anti-PD antibodies and the development of novel tumor immunotherapy strategies.

Diff erent sea cucumbers have diff erent preferred habitats and seasonal changes,which is still lack of detailed research.This study selects two common tropical sea cucumbers(Holothuria edulis and Stichopus chloronotus)to explore the reasons that aff ect their habitat selection and seasonal changes,so as to provide support for the spatial planning of sea cucumbers reserve in coral reef area.The study area is a compound distribution area of living coral reefs and sandy bottom in the north of Wuzhizhou Island.The survey time is January,April-September,2019.Three fi xed transects(G,H,and Q)were selected to reveal the population ecological characteristics of them in the typical dry season(January,April,May,and June)and rainy season(July,August,and September).The variation range of density for H.edulis and S.chloronotus were 2.0-8.9 inds./10 m^(2) and 0.2-0.7 inds./10 m^(2),respectively.The population density in rainy season was signifi cantly higher than that in dry season(P<0.05).H.edulis tends to be distributed in deeper water.The distribution of H.edulis was signifi cant positively correlated with the sand coverage and sand continuity in G transect(P<0.05),and signifi cant negatively correlated with live coral coverage in three transects(P<0.01).The shift from dry season to rainy season is a key factor aff ecting their habitat preference.In dry season,the preferred habitats of S.chloronotus are small rock(SR),sand(S),and small coral reef(SCR),while the H.edulis is S,SR,and dead coral rubbles(DCR).In rainy season,the preferred habitats of S.chloronotus are shift to SCR and SR,while the H.edulis is SCR,DCR,and sand.

At the single-cell level,cellular parameters,gene expression and cellular function are assayed on an individual but not population-average basis.Essential to observing and analyzing the heterogeneity and behavior of these cells/clusters is the ability to prepare and manipulate individuals.Here,we demonstrate a versatile microsystem,a stereo acoustic streaming tunnel,which is triggered by ultrahigh-frequency bulk acoustic waves and highly confined by a microchannel.We thoroughly analyze the generation and features of stereo acoustic streaming to develop a virtual tunnel for observation,pretreatment and analysis of cells for different single-cell applications.3D reconstruction,dissociation of clusters,selective trapping/release,in situ analysis and pairing of single cells with barcode gel beads were demonstrated.To further verify the reliability and robustness of this technology in complex biosamples,the separation of circulating tumor cells from undiluted blood based on properties of both physics and immunity was achieved.With the rich selection of handling modes,the platform has the potential to be a full-process microsystem,from pretreatment to analysis,and used in numerous fields,such as in vitro diagnosis,high-throughput single-cell sequencing and drug development.

Objective:Patients with non–small cell lung cancer(NSCLC)respond differently to cytokine-induced killer cell(CIK)treatment.Therefore,potential prognostic markers to identify patients who would benefit from CIK treatment must be elucidated.The current research aimed at identifying predictive prognostic markers for efficient CIK treatment of patients with NSCLC.Methods:Patients histologically diagnosed with NSCLC were enrolled from the Tianjin Medical University Cancer Institute and Hospital.We performed whole-exome sequencing(WES)on the tumor tissues and paired adjacent benign tissues collected from 50 patients with NSCLC,and RNA-seq on tumor tissues of 17 patients with NSCLC before CIK immunotherapy treatment.Multivariate Cox proportional hazard regression analysis was used to analyze the association between clinical parameters and prognostic relevance.WES and RNA-seq data between lung squamous cell carcinoma(SCC)and adenocarcinoma(Aden)were analyzed and compared.Results:The pathology subtype of lung cancer was the most significantly relevant clinical parameter associated with DFS,as analyzed by multivariate Cox proportional hazard regression(P=0.031).The patients with lung SCC showed better CIK treatment efficacy and extended DFS after CIK treatment.Relatively low expression of HLA class II genes and checkpoint molecules,and less immunosuppressive immune cell infiltration were identified in the patients with lung SCC.Conclusions:Coordinated suppression of the expression of HLA class II genes and checkpoint molecules,as well as less immune suppressive cell infiltration together contributed to the better CIK treatment efficacy in lung SCC than lung Aden.

季节是全球和区域人类农业生产活动遵循的重要气候年内变化规律。近半个世纪以来全球地表气温明显升高,与地表气温相关联的季节起讫和持续时间等参数也随之发生显著变化。首先,论文总结了目前广泛应用的三大类季节定义方式,包括从行星角度出发的固定长度季节定义法、气温的绝对和相对阈值季节定义法、区域地理因素和天气平均态转变的季节定义法;其次,回顾了当前利用不同动态指标定义季节主要获得的历史时期冷季缩短、暖季变长的时空规律,以及气候变暖背景下自然变率和外强迫对季节长度变化原因的解释;最后,从季节定义方法的应用领域和空间上通用性角度评述了各个方法的优缺点,并从数据偏差影响、通用指数发展、气候变暖背景下变化机理三方面对未来进行了展望。研究结果明确了季节划分方式间的差别、季节变化特征及原因,不仅有利于指导农业生产,也可以为妥善应对气候变化提供可靠科学信息。

Complex congenital disorders may be caused by multiple genetic alterations and/or environmental hazards. Diagnosis and management of these diseases are usually difficult. Robust next-generation sequencing （NGS） technologies provide unprecedented opportunities to maximize mutation detection and improve genetic counseling and clinical management. Targeted or whole exome sequencing （WES） mainly detects protein-coding DNA sequence aberrations and is the major DNA sequencing technology that is entering clinical practice （Liu et al., 2014）.

Objective:Trained immunity of natural killer(NK)cells has shown great potential in the treatment of cancers by eliciting enhanced effector responses to restimulation by cytokines or cancer cells for long time periods after preactivation.However,the human NK cells responsible for the generation and maintenance of trained immunity are largely unknown.We hypothesized that heterogeneous human NK cells would respond differentially to stimulation with a combination of IL-12,IL-15,and IL-18,and that an NK cell subset might exist that is mainly responsible for the induction of trained immunity.On the basis of our hypothesis,we aimed to identify the subset from which cytokine-trained human NK cells originate and to explore possible regulatory targets for drug intervention.Methods:Flow cytometry assays were performed to analyze the functions of cytokine-trained NK cells and examine cell division and protein expression in NK cell subsets.Single-cell RNA sequencing(scRNA-seq)plus TotalSeq™technology was used to track the heterogeneity of NK cells during the induction of trained immunity.Results:Traditional developmental markers for peripheral NK cells were unable to identify the precursors of human NK cells with trained immunity.Therefore,we used scRNA-seq plus TotalSeq™technology to track the heterogeneity of NK cells during the induction of trained immunity and identified a unique cluster of CD57−NKG2A+EZH2+IFNG+MKI67+IL12R+IL15R+IL18R+NK cells.Enrichment and pseudotime trajectory analyses suggested that this cluster of NK cells contained the precursor of trained NK cells.We then used flow cytometry to further investigate the role of EZH2 in trained NK precursors and found that CD57−NKG2A+EZH2+NK cells had faster cell cycles and an enhanced trained phenotype,and EZH2 inhibition significantly impaired the induction of trained immunity in NK cells.These results suggested that EZH2 is a unique epigenetic marker of precursors of human NK cells with trained immunity.Conclusions:Our work revealed human NK heterogeneity in the in
