Peroxisome proliferator-activated receptor-γ is essential in the pathogenesis of gastric carcinoma

AIM： To investigate whether peroxisome proliferatoractivated receptor γ （PPAR-γ） is expressed in human gastric carcinoma and whether PPAR-γ, is a potential target for gastric carcinoma therapy. METHODS： PPAR-γ protein in gastric carcinoma was examined by immunohistochemistry. In the gastric carcinoma cell line MGCS03, PPAR-7, survivin, Skp2 and p27 protein and mRNA were examined by Western blotting and real-time reverse transcription-polymerase chain reaction, respectively; proliferation was examined by MTT; apoptosis was examined by chromatin staining with Hoechst 33342 and fluorescence activated cell sorting （FACS）. and cell cycle was examined by FACS; the knockdown of PPAR-γ was done by RNA interference. RESULTS： A high level of expression of PPAR-γ was observed in human gastric carcinoma and in a human gastric carcinoma cell line MGCS03. The PPAR-γ agonist 15-deoxy-△12,14-prostaglandin J2 （15d-PGJ2）inhibited growth, and induced apoptosis and G1/G0 cell cycle arrest in MGC803 cells in a concentration-dependent and time-dependent manner. The effect of 15d-PGJ2 on MGC803 cells was not reversed by the selective and irreversible antagonist GW9662 for PPAR-γ. Furthermore, survivin and Skp2 expression were decreased, whereas p27 expression was enhanced following 15d-PGJ2 treatment in a dose-dependent manner in MGC803 cells. Interestingly, we also found that small interfering RNA for PPAR-γ inhibited growth and induced apoptosis in MGC803 cells. The inhibition of PPAR-γ function may be a potentially important and novel modality for treatment and prevention of gastric carcinoma. CONCLUSION： A PPAR-γ agonist inhibited growth of human gastric carcinoma MGC803 cells by inducing apoptosis and G1/G0 cell cycle arrest with the involvement of survivin, Skp2 and p27 and not via PPAR-γ.

Relation of overexpression of S phase kinase-associated protein 2 with reduced expression of p27 and PTEN in human gastric carcinoma

AIM： TO investigate the significance of S phase kinase- associated protein 2 （Skp2） expression in human gastric carcinoma and the relation between expressions of Skp2, p27 and PTEN. METHODS： Immunohistochemical analysis was performed on 138 gastric carcinoma specimens, their paired adjacent mucosa specimens, 102 paired lymphatic metastatic carcinoma tissue specimens, 30 dysplasia specimens, 30 intestinal metaplasia specimens, 10 chronic superficial gastritis specimens and 5 normal gastric mucosa specimens for Skp2 expression and on 138 gastric carcinoma specimens for p27 and PTEN expression. RESULTS： Skp2 labeling frequency was significantly higher in intestinal rnetaplasia （12.68±0.86） and adjacent rnucosa （19.324±1.22） than in normal gastric rnucosa （0.53±0.13） and chronic superficial gastritis （0.47±0.19）（P = 0.000）, in dysplasia （16.74±0.82）than in intestinal metaplasia （P = 0.000）; in gastric primary carcinoma （31.34±2.17） than in dysplasia and adjacent rnucosa （P = 0.000）, in metastasis gastric carcinoma in lymph nodes （39.76±2.00） than in primary gastric carcinoma （P = 0.037）, respectively. Skp2 labeling frequency was positively associated with differentiation degree （rho = 0.315, P = 0.000）, vessel invasion （rho = 0.303, P = 0.000） and lymph node metastasis （rho = 0.254, P = 0.000） of gastric cancer. Expression of Skp2 was negatively associated with p27 （rho = -0.451, P = 0.000） and PTEN （rho = -0.480, P = 0.000） expression in gastric carcinoma, p27 expression was positively associated with PTEN expression in gastric carcinoma （rho = 0.642, P = 0.000）.CONCLUSION： Skp2 overexpression may be involvedin carcinogenesis and progression of human gastric carcinoma in vivo, possibly via p27 proteolysis. PTEN may regulate the expression of p27 by negatively regulating Skp2 expression.

Widely used spin-coated nickle oxide （NiOx） based perovskite solar cells often suffer from severe interfacial reactions between the NiOxand adjacent perovskite layers due to surface defect states,which inherently impair device performance in a long-term view,even with surface molecule *** this study,we developed high-quality magnetron-sputtered NiOxthin films through detailed process optimization,and compared systematically sputtered and spin-coated NiOxthin film surfaces from materials to *** sputtered NiOxfilms exhibit improved crystallinity,smoother surfaces,and significantly reduced Ni3+or Ni vacancies compared to their spin-coated ***,the interface between the perovskite and sputtered NiOxfilm shows a substantially reduced density of defect *** solar cells （PSCs） fabricated with our optimally sputtered NiOxfilms achieved a high power conversion efficiency （PCE） of up to 19.93%and demonstrated enhanced stability,maintaining 86.2% efficiency during 500 h of maximum power point tracking under one standard sun ***,with the surface modification using (4-(2,7-dibromo-9,9-dimethylacridin-10（9H）-yl)butyl)p hosphonic acid （DmacPA）,the device PCE was further promoted to 23.07%,which is the highest value reported for sputtered NiOxbased PSCs so far.

Objective To investigate the occurrence of important foodbome pathogens in shellstock Pacific oysters in the food markets in South China. Methods From July 2007 to June 2008, retail oysters were collected in different seasons from South China and analyzed for the prevalence and levels of Listeria monocytogenes, Vibrio vulnificus and Vibrio parahaemolyticus. Results None of L. monocytogenes could be detected in any of the 202 oyster samples tested, while E vulnificus and E parahaemolyticus could be detected in 67 （54.9%） and 109 （89.3%） of the 122 oyster samples analyzed, respectively, with an MPN （most probable number） value greater than or equal to 3. V. wdnificus and Eparahaemolyticus with a more than 102 MPN/g were found in 36 （29.5%） and 59 （48.4%） of the 122 oyster samples, respectively. The tdh and trh genes were detected in 4 （0.3%） and 8 （0.6%） of the 1 349 V. parahaemolyticus isolates, respectively. Of the 122 samples, 4 （3.3%） was positive for either tdh or trh. The levels of E vulnificus and total V. parahaemolyticus in oysters in South China varied in different seasons. Conclusion V. vulnificus and pathogenic V. parahaemolyticus are frequently found in oysters in south China, which may pose a potential threat to public health. Data presented here will be useful for the microbiological risk assessment in oysters in China.

Transgenic japonica rice expressing the cry1C gene is resistant to striped stem borers in Northeast China

Rice production and quality are seriously affected by the lepidopteran pest,striped stem borer(SSB),in Northeast *** this study,a synthetic cry1 C gene encoding Bacillus thuringiensis(Bt)δ-endotoxin,which is toxic to lepidopteran pest,was transformed into a japonica rice variety(Jigeng 88)in Northeast China by Agrobacterium-mediated *** molecular detection and the Basta resistance germination assay,a total of 16 single-copy homozygous transgenic lines were obtained from 126 independent transformants expressing cry1 ***,four cry1 C-transgenic lines(JL16,JL23,JL41,and JL42)were selected by evaluation of the Cry1 C protein level,insect-resistance and agronomic *** cry1 C-transgenic lines had higher resistance to SSB and higher yield compared with non-transgenic(NT)control plants.T-DNA flanking sequence analysis of the transgenic line JL42 showed that the cry1 C gene was inserted into the intergenic region of chromosome 11,indicating that its insertion may not interfere with the genes near insertion *** summary,this study developed four cry1 C-transgenic japonica rice lines with high insect resistance and high *** can be used as insect-resistant germplasm materials to overcome the problem of rice yield reduction caused by SSB and reduce the use of pesticides in Northeast China.

Asymptotic Behavior of Solutions of a Free Boundary Problem Modeling Multi-layer Tumor Growth in Presence of Inhibitor

In this paper we study well-posedness and asymptotic behavior of solution of a free boundary problem modeling the growth of multi-layer tumors under the action of an external inhibitor. We first prove that this problem is locally well-posed i[n little H61der spaces. Next we investigate asymptotic behavior of the solution. By computing the spectrum of the linearized problem and using the linearized stability theorem, we give the rigorous analysis of stability and instability of all stationary fiat solutions under the non-fiat perturbations. The method used in proving these results is first to reduce the free boundary problem to a differential equation in a Banach space, and next use the abstract well-posedness and geometric theory for parabolic differential equations in Banach spaces to make the analysis.

Reduced nonlocal integrable mKdV equations of type(-λ, λ) and their exact soliton solutions

We conduct two group reductions of the Ablowitz-Kaup-Newell-Segur matrix spectral problems to present a class of novel reduced nonlocal reverse-spacetime integrable modified Korteweg-de Vries equations. One reduction is local, replacing the spectral parameter with its negative and the other is nonlocal, replacing the spectral parameter with itself. Then by taking advantage of distribution of eigenvalues, we generate soliton solutions from the reflectionless Riemann-Hilbert problems, where eigenvalues could equal adjoint eigenvalues.

A class of trilinear differential operators is introduced through a technique of assigning signs to derivatives and used to create trilinear differential equations. The resulting trilinear differential operators and equations are characterized by the Bell polynomials, and the superposition principle is applied to the construction of resonant solutions of exponential waves. Two illustrative examples are made by an algorithm using weights of dependent variables.

A combined Liouville integrable hierarchy associated with a fourth-order matrix spectral problem

This paper aims to propose a fourth-order matrix spectral problem involving four potentials and generate an associated Liouville integrable hierarchy via the zero curvature formulation.A bi-Hamiltonian formulation is furnished by applying the trace identity and a recursion operator is explicitly worked out,which exhibits the Liouville integrability of each model in the resulting *** specific examples,consisting of novel generalized combined nonlinear Schrodinger equations and modified Korteweg-de Vries equations,are given.
