A cDNA library was constructed end characterized from the pulp of Cera Care navel orange （Citrus sinensis Osbeck） at different stages of ripening. Tittering results revealed that approximately 5.086×10^5 independent clones were included in this library. Electrophoresls gel results of 15 randomly selected clones revealed that the size of the insertion fragments ranged from 400 bp to 2 kb, with an average size of 900 bp. Sequencing results of 150 randomly picked clones showed that the recombination rate was 94%. During subsequent sequence analysis, 41 of 139 clones failed to be identified end the amino sequence of 71 clones shared less than 30% identity with related plants in GenBank. Of 27 clones whose amino sequences shared more than 60% identity with other related plants in GenBenk, 17 clones showed an 80% identity with the corresponding candidate genes of citrus. The clone recognized as the type Ⅲ metallothlonein-llke （MT） gene was observed to occur 13 tlmes, Indlcatlng that the protein may play an important role In frult development and rlpenlng.

Expressed sequence tag （EST） analysis of the pulp of the red-fleshed mutant ＇Cara Cara＇ navel orange provided a starting point for gene discovery and transcriptome survey during citrus fruit maturation. interpretation of the EST datasets revealed that the mutant pulp transcriptome held a high section of stress responses related genes, such as the type III metallothionein-like gene （6.0%）, heat shock protein （2.8%）, Cu/Zn superoxide dismutase （0.8%）, late embryogenesis abundant protein 5 （0.8%）, etc. 133 transcripts were detected to be differentially expressed between the red mutant and its orange-color wild genotype ＇Washington＇ via digital expression analysis. Among them, genes involved in metabolism, defense/stress and signal transduction were statistical overrepresented. Fifteen transcription factors, composed of NAM, ATAF, and CUC transcription factor （NAC）; myeloblastosis （MYB）; myelocytomatosis （MYC）; basic helix-loop-helix （bHLH）; basic leucine zipper （bZIP） domain members, were also included. The data reflected the distinct expression profile and the unique regulatory module associated with these two genotypes. Eight differently expressed genes analyzed in digital were validated by quantitative real- time polymerase chain reaction. For structural polymorphism, both simple sequence repeats and single nucleotide polymorphisms （SNP） loci were surveyed; dinucleotide presentation revealed a bias toward AG/GA/TC/CT repeats （52.5%）, against GC/CG repeats （0%）. SNPs analysis found that transitions （73%） outnumbered transversions （27%）. Seventeen potential cultivar-specific and 387 heterozygous SNP loci were detected from ＇Cara Cara＇ and ＇Washington＇ EST pool.

As the largest transposable element in the plant genome, retrotransposons are thought to be involved in citrus genetic instability and genome evolution, especially in sweet orange, which is prone to bud mutation. In the present study, the presence of copia-like retrotransposons, their heterogeneity, genomic distribution, and transcriptional activities in Citrus were investigated in 12 sweet orange （Citrus sinensis Osbeck） cultivars using a PCR assay designed to detect copia-like reverse transcriptase （RT） sequences. Twelve amplification products from each cultivar were cloned and sequenced. The cloned sequences showed great heterogeneity, except “Dream” navel and “Hamlin”, both of which shared the same sequence. Frame shifting, termination, deletion, and substitution accounted for the heterogeneity of RT sequences. Southern blot hybridization using the RT1 clone from the “Cara Cara” navel as a probe showed that multiple copies were integrated throughout the sweet orange genomes, which made the retrotransposon possible an effective molecular marker to detect citrus evolution events and to reveal its relationship with bud mutation. No transcriptional activities of the retrotransposon were detected by RT-PCR and Northern analysis in the fruits and leaves of either “Cara Cara” or “Seike” navels.

Delayed greening of young leaves is an unusual phenomenon of plants in *** are mostly evergreen tree ***,a natural mutant of“Guanxi”pummelo(Citrus maxima),which shows yellow leaves at the young stage,was characterized to identify the genes underlying the trait of delayed leaf greening in plants.A segregating population with this mutant as the seed parent and a normal genotype as the pollen parent was *** DNA pools respectively from the leaves of segregating seedlings with extreme phenotypes of normal leaf greening and delayed leaf greening were collected for *** segregant analysis(BSA)and In Del marker analysis demonstrated that the delayed leaf greening trait is governed by a 0.3 Mb candidate region on chromosome *** expression analysis further identified a key candidate gene(Citrus Delayed Greening gene 1,CDG1)in the 0.3 Mb region,which showed significantly differential expression between the genotypes with delayed and normal leaf greening *** was a 67 bp In Del region difference in the CDG1 promoter and the In Del region contains a TATA-box *** laser-scanning microscopy revealed that the CDG1-GFP fusion protein signals were co-localized with the chloroplast signals in the *** of CDG1 in tobacco and Arabidopsis led to the phenotype of delayed leaf *** results suggest that the CDG1 gene is involved in controlling the delayed leaf greening phenotype with important functions in chloroplast development.

Provitamin A carotenoids in staple crops are not very stable during storage and their loss compromises nutritional quality. To elucidate the fundamental mechanisms underlying carotenoid accumulation and stability, we investigated transgenic potato tubers that expressed the cauliflower Orange （Or） gene. We found that the Or transgene not only promoted retention of 13-carotene level, but also continuously stimulated its accumulation during 5 months of cold storage. In contrast, no increased levels of carotenoids were observed in the tubers of vector-only controls or a yellow- flesh variety during the same period of storage. The increased carotenoid accumulation was found to be associated with the formation of lipoprotein-carotenoid sequestering structures, as well as with the enhanced abundance of phytoene synthase, a key enzyme in the carotenoid biosynthetic pathway. Furthermore, the provitamin A carotenoids stored were shown to be stable during simulated digestion and accessible for uptake by human intestinal absorptive cells. Proteomic analysis identified three major functional groups of proteins （i.e. heat shock proteins, glutathione-S-transferases, and carbohydrate metabolic proteins） that are potentially important in the Or-regulated carotenoid accumulation. Our results show that regulation of carotenoid sequestration capacity is an important mechanism by which carotenoid stability is regulated. Our findings suggest that induction of a proper sink structure formation in staple crops may provide the crops with a unique ability to promote and/or stabilize provitamin A accumulation during plant growth and post-harvest storage.

＜正＞Isolation of two kinds of subprotoplasts via ulwacentrifugation in discontinuous sucrose gradient was conducted from Satsuma mandarin （Citrus unshiu Marc.）.Cytoplasts were successfully obtained when embryogenic protoplasts of Satsuma mandarin was centrifugated for 20 min at 38000 rpm at 25℃.DAPI staining did not detect nuclei that were present in normal protoplasts.

Nerve regeneration following traumatic peripheral nerve injuries and neuropathies is a complex process modulated by diverse factors and intricate molecular *** studies have focused on factors that stimulate axonal outgrowth and myelin ***,recent studies have highlighted the pivotal role of autophagy in peripheral nerve regeneration,particularly in the context of traumatic ***,autophagy-targeting modulation has emerged as a promising therapeutic approach to enhancing peripheral nerve *** current understanding suggests that activating autophagy facilitates the rapid clearance of damaged axons and myelin sheaths,thereby enhancing neuronal survival and mitigating injury-induced oxidative stress and *** actions collectively contribute to creating a favorable microenvironment for structural and functional nerve regeneration.A range of autophagyinducing drugs and interventions have demonstrated beneficial effects in alleviating peripheral neuropathy and promoting nerve regeneration in preclinical models of traumatic peripheral nerve *** review delves into the regulation of autophagy in cell types involved in peripheral nerve regeneration,summarizing the potential drugs and interventions that can be harnessed to promote this *** hope that our review will offer novel insights and perspectives on the exploitation of autophagy pathways in the treatment of peripheral nerve injuries and neuropathies.

The red-flesh mutant Hong Anliu sweet orange is of high nutritional value due to its lycopene *** previous studies on this mutant fruits suggested that photosynthesis and oxidative stress could promote the formation of mutation ***,leaf rather than fruit is the major part for some important biological processes such as *** this study,we analyzed the proteomic alteration in leaves of the red-flesh mutant Hong Anliu *** wild type （WT）.Ten differentially expressed proteins were identified,of which two were involved in photosynthesis,three in oxidative stress,two in defense,and three in *** high up-regulation of photosynthetic proteins proved the hypothesis that enhanced photosynthesis could provide and transport more substrates into mutant fruits for carotenoid *** to the previous results in fruits,anti-oxidative proteins were highly up-regulated in leaves,suggesting the whole plant of Hong Anliu suffered from enhanced oxidative *** involved in defense and metabolism were also identified,and their possible roles in the mutation were discussed.
