Characterization of carotenoid biosynthetic pathway genes in the pea aphid(Acyrthosiphon pisum)revealed by heterologous complementation and RNA interference assays

Carotenoids are involved in many essential physiological functions and are produced from geranylgeranyl pyrophosphate through synthase,desaturase,and cyclase activities.In the pea aphid(Acyrthosiphon pisum),the duplication of carotenoid biosynthetic genes,including carotenoid synthases/cyclases(ApCscA-C)and desaturases(ApCdeA-D),through horizontal gene transfer from fungi has been detected,and ApCdeB has known dehydrogenation functions.However,whether other genes contribute to aphid carotenoid biosynthesis,and its specific regulatory pathway,remains unclear.In the current study,functional analyses of seven genes were performed using heterologous complementation and RNA interference assays.The bifunctional enzymes ApCscA-C were responsible for the synthase of phytoene,and ApCscC may also have a cyclase activity.ApCdeA,ApCdeC,and ApCdeD had diverse dehydrogenation functions.ApCdeA catalyzed the enzymatic conversion of phytoene to neurosporene(three-step product),ApCdeC catalyzed the enzymatic conversion of phytoene to ζ-carotene(two-step product),and ApCdeD catalyzed the enzymatic conversion of phytoene to lycopene(four-step product).Silencing of ApCscs reduced the expression levels of ApCdes,and silencing these carotenoid biosynthetic genes reduced the α-,β-,and γ-carotene levels,as well as the total carotenoid level.The results suggest that these genes were activated and led to carotenoid biosynthesis in the pea aphid.

Validation and Application of the Chinese Version of the M.D.Anderson Symptom Inventory in Breast Cancer Patients

Objective To validate and use the Chinese Version of the M.D.Anderson Symptom Inventory(MDASI-C)to assess the symptom burden of breast cancer patients in China.Methods A total of 342 breast cancer patients in China participated in this study.Their symptoms were investigated with the MDASI-C from November 2020 to February 2021,and the reliability and validity of this tool were evaluated,respectively.Cluster analysis and correlation analysis were also performed.Results The Cronbach’s alpha coefficient values of the symptom and interference items were 0.827 and 0.880,respectively.Construct validity revealed a four-factor structure.The Kaiser—Meyer—Olkin value was 0.760.The Karnofsky Performance Status,treatment phase,and cancer stage of the patients were grouped,and the differences of scores within the groups were significant.In addition,the employment status,education level,and age of the patients were significantly correlated with the symptoms.The correlation analysis of the education level of the patients showed that most of the symptoms and interference items were reduced as the education level was increased.The top three symptoms were disturbed sleep(3.10±2.52),difficulty remembering(2.54±2.30),and fatigue(2.24±2.13).The clinical and biochemical indicators such as body mass index and neutral granulocyte lymphocyte ratio had effects on many symptoms.As the patients’BMI increased,the patients’pain,disturbed sleep,and difficulty remembering were aggravated,and numbness was alleviated.Conclusion The MDASI-C is a reliable and effective assessment tool to evaluate patients with breast cancer in China.The symptoms are related to many clinical and biochemical indicators.

The Hengduan Mountains Region(HMR)is the largest“evolutionary frontier”of the northern temperate zone,and the origin and maintenance of species in this area is a research hotspot.Exploring species-specific responses to historical and contemporary environmental changes will improve our understanding of the role of this region in maintaining biodiversity.In this study,mitochondrial and microsatellite diversities were used to assess the contributions of paleogeological events,Pleistocene climatic oscillations,and contemporary landscape characteristics to the rapid intraspecific diversification of Liangshantriton taliangensis,a vulnerable amphibian species endemic to several sky-island mountains in the southeastern HMR.Divergence date estimations suggested that the East Asian monsoon,local uplifting events(Xigeda Formation strata),and Early-Middle Pleistocene transition(EMPT)promoted rapid divergence of L.taliangensis during the Pleistocene,yielding eight mitochondrial lineages and six nuclear genetic lineages.Moreover,population genetic structures were mainly fixed through isolation by resistance.Multiple in situ refugia were identified by ecological niche models and high genetic diversity,which played crucial roles in the persistence and divergence of L.taliangensis during glacial-interglacial cycles.Dramatic climatic fluctuations further promoted recurrent isolation and admixing of populations in scattered glacial refugia.The apparent mitonuclear discordance was likely the result of introgression by secondary contact and/or female-biased dispersal.Postglacial expansion generated two major secondary contact zones(Ganluo(GL)and Chuhongjue(CHJ)).Identification of conservation management units and dispersal corridors offers important recommendations for the conservation of this species.

To quantify the pore characteristics of various macerals in Chang 7 lacustrine shales,macerals were effectively identified according to their optical and morphological characteristics,and the nanoscale pore structure of macerals was observed by scanning electron microscope.Meanwhile,the reflectances of different positions in the same pieces of vitrinite or solid bitumen with heterogeneous pores development were measured.The results showed that the average contents of sapropelinite,liptinite,vitrinite,inertinite and solid bitumen are 42.7%,8.7%,13.6%,13.8% and 21.2%,respectively,which suggests that the source of the organic matter of the Chang 7 shales is a mixed source input.The organic pores of Chang 7 shales are enriched,and the pore shapes are mostly round or elliptical.The pore size of organic pores has a wide distribution,mainly concentrate in the range of 100-400 nm,and the average plane porosity of organic pores is 10.13%.The size order of the organic pores in various macerals is:solid bitumenfusinite>bituminite>solid bitumen>vitrinite>liptinite.OM pores are mainly contributed by bituminite,solid bitumen and fusinite.The plane porosity of bituminite increases with maturity.In the process of thermal evolution,the plane porosity of fusinite is distributed in the two ranges of 20%-28% and 1%-7%.The former is mainly the primary pores of the fusinite itself,and the latter is the secondary pores formed in the thermal evolution.As for the organic pores of other macerals,no obvious thermal evolution law was found.Meanwhile,the surface imperfections of vitrinite or solid bitumen is enhanced by the enrichment of organic pores(an increase in pore size or pore number),which may result in the underestimation of their reflectances.

针对直方图方法分选脉间参差雷达信号能力不佳且无法解析其调制模式的问题,结合参差信号的交织特性,提出联合到达时间差值(Difference of Time Of Arrival,DTOA)的一阶差分曲线和相关分析的分选方法.该方法基于DTOA一阶差分曲线提取DTOA关注项,然后基于关注项的自相关和交叠率识别参差调制模式、基于关注项的互相关分析参差调制模式,最后结合参差分析结果进行序列检索.实验结果表明,所提方法所需计算量较少,可以有效分选参差信号并解析其完整的调制模式,对脉冲丢失表现出较强的适应性,对子周期分布的均匀程度和干扰脉冲有较好的稳健性.

The need for bladder reconstruction and side effects of cystoplasty have spawned the demand for the development of alternative material substitutes.Biomaterials such as submucosa of small intestine(SIS)have been widely used as patches for bladder repair,but the outcomes are not fully satisfactory.To capture stem cells in situ has been considered as a promising strategy to speed up the process of re-cellularization and functionalization.In this study,we have developed an anti-CD29 antibody-conjugated SIS scaffold(AC-SIS)which is capable of specifically capturing urine-derived stem cells(USCs)in situ for tissue repair and regeneration.The scaffold has exhibited effective capture capacity and sound biocompatibility.In vivo experiment proved that the AC-SIS scaffold could promote rapid endothelium healing and smooth muscle regeneration.The endogenous stem cell capturing scaffolds has thereby provided a new revenue for developing effective and safer bladder patches.

A diastereoselectivity-controllable formal[3+2]-cycloaddition of arylvinyl oxirane 2,2-diesters with cyclic N-sulfonyl imines is developed,affording the corresponding tricyclic oxazolidine derivatives in moderate to excellent yields with excellent diastereoselectivities in the presence of palladium(0)or scandium(III)triflate.This protocol allows selective synthesis of diastereomers of tricyclic oxazolidine derivatives under switchable and mild conditions.Further transformations of the obtained products were conducted by removing ester groups and arylvinyl moieties.

Overexpression of the apple expansin-like gene MdEXLB1 accelerates the softening of fruit texture in tomato

Fruit firmness is an important quality trait of apple fruit texture,and the pre-harvest ripening period is the key period for the formation of apple fruit texture.Expansin is a cell wall loosing protein family that has four subfamilies:α-expansin(EXPA),β-expansin(EXPB),expansin-like A(EXLA),and expansin-like B(EXLB).In this study,we investigated the key period of pre-harvest texture formation in‘Golden Delicious’apples based on fruit longitudinal diameter,transverse diameter,firmness,tissue structure,respiration intensity,ethylene release rate,and expansin activity.Within the 10 days before harvest,the fruit was found to reach maturity.Semi-quantitative RT-PCR revealed that most of the expansins were expressed at the ripening stage before harvest.The biological function of the EXLB subfamily gene,MdEXLB1,was further identified,and its subcellular localization on the cell wall was confirmed by transient transformation experiments.Compared with the wild type(WT),the transgenic tomato lines overexpressing MdEXLB1 had lower plant height,earlier fruiting period,fewer days for fruit ripening,higher fruit maturity,lower fruit firmness,higher fruit expansin activity,more discrete flesh cell structure,and accelerated fruit ripening process.Overall,this is the first study to propose that the apple EXLB subfamily gene,MdEXLB1,has biological functions and plays an important role in promoting fruit ripening and softening.

目的探究羊水染色体核型分析在产前诊断中的重要性。方法从2010年12月;017年11月妊娠17;1周具有羊水穿刺指征以及自愿要求的孕妇16896例,在B超引导下穿刺抽取20 ml,对胎儿的脱落细胞进行培养,收获,制片,G显带,上机扫片并进行核型分析。结果在16896例羊水染色体分析中,共检测出异常核型495例,检出率为2.92%,多态性1312例,检出率为7.76%;高龄孕妇6377例,异常检出率为2.47%,唐氏筛查高风险孕妇7746例,异常检出率为1.61%,18三体筛查高风险351例,异常检出率为3.13%,超声NT异常218例,异常检出率13.76%,鼻骨缺失50例,异常检出率4.00%,超声综合指标1168例,异常检出率6.25%,NIPT阳性141例,异常检出率58.15%,夫妻一方为染色体携带者或者异常者83例,异常检出率为34.93%,异常生育史448例,异常检出率为1.11%,不良接触史114例,异常检出率4.38%,要求羊水穿刺468例,异常检出率1.49%。结论在产前诊断的过程中,羊水染色体核型分析依然是重要的诊断方法 ,由于染色体条带分辨水平有限,超声和其它分子遗传学检测可以与之互补,提高胎儿异常检出率,减少出生缺陷。
