Intraocular pressure with rebound tonometry and effects of topical intraocular pressure reducing medications in guinea pigs

AIM： To investigate the intraocular pressure（IOP） of adult guinea pig eyes with rebound tonometry（RBT）,and assess the effects of four distinctive topical IOP reducing medications including Carteolol,Brimonidine,Brinzolamide and Latanoprost.METHODS： The IOPs of twenty-four 12-week-old guinea pigs（48 eyes） were measured every two hours in one day with RBT as baselines.All the animals were then divided into four groups（Carteolol,Brimonidine,Brinzolamide and Latanaprost groups,n=6）.The IOPs were measured and compared to the baseline 1,2,3,5,7,9,15 and 24 h after treatment.RESULTS： The mean baseline IOP of 24 guinea pigs（48 eyes） was 10.3±0.36 mm Hg（6-13 mm Hg） and no binocular significant differences of IOPs were observed（t=1.76,P〉0.05）.No significant difference of IOP in Carteolol group at each time point was observed before and after treatment（t=1.48,P〉0.05）.In Brimonidine group,IOP was 2.2±1.9 mm Hg lower than the baseline after one hour（t=3.856,P=0.003） and lasted for one hour.In Brinzolamide group,IOP was 1.4±1.1 mm Hg lower than the baseline after one hour（t=4.53,P=0.001） and lasted for 7h and the IOP declined most at 3h.In Latanaprost group,IOP was 2.1±1.3 mm Hg lower than the baseline after one hour（t=6.11,P=0.001） and lasted for one hour.CONCLUSION： The IOP of guinea pig eyes is relatively stable compared to human eyes.In four reducing IOP medications,no significant effect of Carteolol is observed.Brinzolamide has the longest duration,while the Brimonidine has the shortest duration and the maximum level of treatment.

Sodium valproate suppresses abnormal neurogenesis induced by convulsive status epilepticus

Status epilepticus has been shown to activate the proliferation of neural stem cells in the hippocampus of the brain, while also causing a large amount of neuronal death, especially in the subgranular zone of the dentate gyrus and the subventricular zone. Simultaneously, proliferating stem cells tend to migrate to areas with obvious damage. Our previous studies have clearly confirmed the effect of sodium valproate on cognitive function in rats with convulsive status epilepticus. However, whether neurogenesis can play a role in the antiepileptic effect of sodium valproate remains unknown. A model of convulsive status epilepticus was established in Wistar rats by intraperitoneal injection of 3 mEq/kg lithium chloride, and intraperitoneal injection of pilocarpine 40 mg/kg after 18–20 hours. Sodium valproate(100, 200, 300, 400, 500, or 600 mg/kg) was intragastrically administered six times every day(4-hour intervals) for 5 days. To determine the best dosage, sodium valproate concentration was measured from the plasma. The effective concentration of sodium valproate in the plasma of the rats that received the 300-mg/kg intervention was 82.26 ± 11.23 μg/mL. Thus, 300 mg/kg was subsequently used as the intervention concentration of sodium valproate. The following changes were seen: Recording excitatory postsynaptic potentials in the CA1 region revealed high-frequency stimulation-induced long-term potentiation. Immunohistochemical staining for BrdU-positive cells in the brain revealed that sodium valproate intervention markedly increased the success rate and the duration of induced long-term potentiation in rats with convulsive status epilepticus. The intervention also reduced the number of newborn neurons in the subgranular area of the hippocampus and subventricular zone and inhibited the migration of newborn neurons to the dentate gyrus. These results indicate that sodium valproate can effectively inhibit the abnormal proliferation and migration of neural stem cells and newborn neurons after convulsive status epileptic

Comparison of bone regeneration in alveolar bone of dogs on mineralized collagen grafts with two composition ratios of nanohydroxyapatite and collagen

To study the effect of two composition ratios of nano-hydroxyapatite and collagen(NHAC)composites on repairing alveolar bone defect of dogs.Eighteen healthy adult dogs were randomly divided into three groups.Two kinds of the NHAC composites were prepared according to the constituent ratios of 3:7 and 5:5;immediately after extraction of the mandibular second premolars,each kind of the NHAC composite was implanted into extraction socket,respectively:Group I,nHA/Col紏3:7;Group II,nHA/Col紏5:5 and Group III,blank control group.The bone-repairing ability of the two grafts was separately analyzed by morphometric measurement,X-ray tomography examination and biomechanical analysis at 1st,3rd and 6th month post-surgical,respectively.The NHAC composites were absorbed gradually after implanting into alveolar bone defect and were replaced by new bone.The ratios of new bone formation of Group I was significantly higher than that of Group II after 3 months(P<0.05).The structure and bioactive performance can be improved when the ratio between the collagen and the hydroxyapatite was reasonable,and the repairing ability and effect in extraction sockets are obviously better.

Receptor-like kinase OsCR4 controls leaf morphogenesis and embryogenesis by fixing the distribution of auxin in rice

Cell differentiation is a key event in organ development;it involves auxin gradient formation, cell signaling, and transcriptional regulation. Yet, how these processes are orchestrated during leaf morphogenesis is poorly understood. Here, we demonstrate an essential role for the receptor-like kinase Os CR4 in leaf development. oscr4 loss-of-function mutants displayed short shoots and roots, with tiny,crinkly, or even dead leaves. The delayed outgrowth of the first three leaves and seminal root in oscr4 was due to defects in plumule and radicle formation during embryogenesis. The deformed epidermal,mesophyll, and vascular tissues observed in oscr4 leaves arose at the postembryo stage;the corresponding expression pattern of pro Os CR4:GUS in embryos and young leaves suggests that Os CR4 functions in these tissues. Signals from the auxin reporter DR5 rev:VENUS were found to be altered in oscr4 embryos and disorganized in oscr4 leaves, in which indole-3-acetic acid accumulation was further revealed by immunofluorescence. Os WOX3 A, which is auxin responsive and related to leaf development,was activated extensively and ectopically in oscr4 leaves, partially accounting for the observed lack of cell differentiation. Our data suggest that Os CR4 plays a fundamental role in leaf morphogenesis and embryogenesis by fixing the distribution of auxin.

Use of high-resolution X-ray computed tomography and 3D image analysis to quantify mineral dissemination and pore space in oxide copper ore particles

Mineral dissemination and pore space distribution in ore particles are important features that influence heap leaching performance. To quantify the mineral dissemination and pore space distribution of an ore particle, a cylindrical copper oxide ore sample (I center dot 4.6 mm x 5.6 mm) was scanned using high-resolution X-ray computed tomography (HRXCT), a nondestructive imaging technology, at a spatial resolution of 4.85 mu m. Combined with three-dimensional (3D) image analysis techniques, the main mineral phases and pore space were segmented and the volume fraction of each phase was calculated. In addition, the mass fraction of each mineral phase was estimated and the result was validated with that obtained using traditional techniques. Furthermore, the pore phase features, including the pore size distribution, pore surface area, pore fractal dimension, pore centerline, and the pore connectivity, were investigated quantitatively. The pore space analysis results indicate that the pore size distribution closely fits a log-normal distribution and that the pore space morphology is complicated, with a large surface area and low connectivity. This study demonstrates that the combination of HRXCT and 3D image analysis is an effective tool for acquiring 3D mineralogical and pore structural data.

Neuroelectrophysiological characteristics of Hirayama disease： report of 14 cases

Background Hirayama disease is a juvenile muscular atrophy of the distal upper extremities and affects mainly young males. The present study aimed to investigate the neuroelectrophysiological characteristics of Hirayama disease. Methods We retrospectively analyzed the neural conduction velocity （NCV） parameters and needle-electrode electromyograms （EMG） of 14 patients with Hirayama disease. According to the clinical features of the patients, NCV was performed on affected upper-limb including median nerves and ulnar nerves, while EMG was selectively performed on upper and lower extremities, sternocleidomast and thoracic paraspinal muscles. Results The median nerves of all affected upper limbs of patients with Hirayama disease had normal conduction velocities and compound motor action potentials （CMAPs）. The ulnar nerves of all affected upper limbs also had normal conduction velocities. Of the 16 measured ulnar nerves of the affected upper limbs, eight had normal CMAPS, while the other eight showed CMAPs below the normal value by 〉20%. All patients had neurogenic injury on the affected side in muscles innervated by anterior horn cells at the lower cervical region （C7-8, T1）. Four patients had unilateral upper-limb muscle neurogenic injury on the affected side. Seven patients had bilateral upper-limb muscle neurogenic injury, while only two patients experienced bilateral upper-limb muscle atrophy / weakness. The other three patients showed extensive neurogenic injury （unilateral upper-limb muscle atrophy/weakness in one patient, bilateral symptoms in the other two patients）. Conclusions Electromyographic examination showed that the majority of Hirayama disease patients exhibited characteristic segmental injury in the anterior horn of the lower cervical region, while a few patients exhibited extensive neurogenic injury. These data suggest that the actual influence of Hirayama disease may be more extensive than indicated by the clinical presentations.

To the Editor： A 50-year-old man presented with recurrent episodes of bullae, erosions, and crust on sun-exposed areas for 1 year. Lesions healed with scarring and hyperpigmentation. The skin lesions were aggravated after sun exposure. The patient had a 30-year alcohol abuse （100-200 ml/day） history. Family heredity history was unremarkable. Physical examination revealed multiple erosions with erythema, hyperpigmentation, bullae, and crusting on his lace, neck, and dorsum of the hands [Figure 1a].

Ziyin Huatan Recipe, a Chinese herbal compound, inhibits migration and invasion of gastric cancer by upregulating RUNX3 expression

Objectives: Ziyin Huatan Recipe(ZYHT), a traditional Chinese medicine comprised of Lilii Bulbus, Pinelliae Rhizoma, and Hedyotis Diffusa, has shown promise in treating gastric cancer(GC). However, its potential mechanism has not yet been clearly addressed. This study aimed to predict targets and molecular mechanisms of ZYHT in treating GC by network pharmacology analysis and to explore the role of ZYHT in GC both in vitro and in vivo.Methods: Targets and molecular mechanisms of ZYHT were predicted via network pharmacology analysis. The effects of ZYHT on the expression of metastasis-associated targets were further validated by Western blot and quantitative real-time polymerase chain reaction. To explore the specific molecular mechanisms of the effects of ZYHT on migration and invasion, the runt-related transcription factor 3(RUNX3) gene was knocked out by clustered regularly interspaced short palindromic repeats/Cas9, and lentiviral vectors were transfected into SGC-7901 cells. Then lung metastasis model of GC in nude mice was established to explore the anti-metastasis effect of ZYHT. Western blot and immunohistochemistry were used to explore the impact of ZYHT on the expression of metastasis-related proteins with or without RUNX3 gene.Results: The network pharmacology analysis showed that ZYHT might inhibit focal adhesion, migration,invasion and metastasis of GC. ZYHT inhibited the proliferation, migration and invasion of GC cells in vitro via regulating the expression of metastasis-associated targets. Knocking out RUNX3 almost completely reversed the cell phenotypes(migration and invasion) and protein expression levels elicited by ZYHT.In vivo studies showed that ZYHT inhibited the metastasis of GC cells to the lung and prolonged the survival time of the nude mice. Knocking out RUNX3 partly reversed the metastasis of GC cells to the lung and the protein expression levels elicited by ZYHT.Conclusion: ZYHT can effectively inhibit the invasion and migration of GC in vitro and in vivo, and its molecular mechanism m

A high-precision hydrodynamic model coupled with the hydrological habitat suitability model to reveal estuarine vegetation distribution

In this paper,based on the finite volume method,a high-precision hydrodynamic model coupled with the habitat suitability model is established,and the computational efficiency of the coupled model is improved by a graphics processing unit(GPU)-accelerated technology.The coupled model is used to solve the problem of the non-conservation of mass that may be caused by the nearshore hydrodynamic model in the processing of wetting and drying,while avoiding the unphysical high velocities at the wetting and drying boundaries.The coupled model is applied to simulate the high-precision hydrodynamic process of the Liao River estuary(LRE)and the hydrological habitat suitability of the estuarine vegetation(Suaeda heteroptera)growing in the LRE.The simulated values of the hydrological variables(the water level,the water depth,the current velocity and direction)are highly consistent with the measured values.The root mean square errors(RMSE)of the hydrological variables are 0.10m,0.12m/s and 17.24°,respectively.Furthermore,the simulated combined suitability index(CSI)distribution of Suaeda heteroptera(S.heteroptera)matches with the distribution of S.heteroptera obtained from the high-resolution remote sensing satellite images during the same time period.The ratio of the simulated weighted usable area(WUA)of S.heteroptera to the area obtained from the remote sensing satellite images during the same period is 81.9%.This study reveals the phenomenon that the distribution of S.heteroptera in the LRE is highly correlated with the high-precision hydrodynamic processes,and provides a scientific basis and a valuable reference for the conservation and the restoration of the estuarine vegetation.
