Elevated soluble 4-1BB is associated with serum markers of hepatitis B virus in patients with chronic hepatitis B

BACKGROUND Previous studies have suggested that the costimulatory molecule 4-1BB plays pivotal roles in regulating immunity during chronic viral ***,up to now,there are few studies about 4-1BB in chronic hepatitis B(CHB).AIM To clarify this issue,we report our comprehensive study results on the expression levels of 4-1BB in patients with *** From September 2018 to June 2019,a total of 64 patients with CHB were recruited from the Department of Hepatology,The First Hospital of Jilin *** blood samples were collected from 52 treatment-naïve and 12 entecavir-treated patients with CHB as well as 37 healthy donors(including 24 healthy adults and 13 healthy children).The levels of soluble 4-1BB(s4-1BB)in plasma were measured by ELISA.4-1BB mRNA expression in peripheral blood mononuclear cells was detected by real-time quantitative *** The s4-1BB levels in the plasma of patients with CHB were significantly higher than those in healthy adults(94.390±7.393 ng/mL vs 8.875±0.914 ng/mL,P<0.001).In addition,the s4-1BB level in plasma was significantly increased in patients with a higher viral load and a disease flare ***,there were no significant differences between treatment-naïve and entecavir-treated ***,among treatment-naïve patients with CHB,the levels of s4-1BB in plasma had a significant positive correlation with hepatitis B surface antigen,hepatitis B virus DNA,hepatitis B e antigen,and triglyceride levels(r=0.748,P<0.001;r=0.406,P=0.004;r=0.356,P=0.019 and r=-0.469,P=0.007,respectively).The 4-1BB mRNA expression was higher in the peripheral blood mononuclear cells of patients with CHB than in the peripheral blood mononuclear cells of healthy adults,but the difference was not statistically *** These results suggest that the levels of s4-1BB may be associated with pathogenesis of hepatitis B virus and therefore may be a promising biomarker for disease progression.

LC/MS/MS determination and pharmacokinetic studies of six compounds in rat plasma following oral administration of the single and combined extracts of Eucommia ulmoides and Dipsacus asperoides

AIM: To establish and apply a new LC/MS/MS method for the simultaneous, quantitative determination of six ingredients, aucubin(AU), geniposide(GP), geniposidic acid(GPA), pinoresinol diglucoside(PDG), secologanin(SLG), and loganin(LG) in single and combined extracts of Eucommia ulmoides and Dipsacus asperoides. METHOD: Using the LC/MS/MS-ESI--MRM mode to detect the six compounds, chromatographic separation was achieved on an Agilent Eclipse plus C18 column, and the mobile phase consisted of solvent A(CH3CN) and solvent B(H2O containing 0.01% CH3 COOH V/V). RESULTS: This method was successfully applied to quantify the six compounds in rat plasma after oral administration, and showed good precision, accuracy, reproducibility, and linear regression(r2 >0.99). CONCLUSION: The results showed that following the use of the two medicinal plants, for AU and GP, the values of tmax markedly increased, and the values of cmax markedly decreased. It was found that the compatibility of the medicinal plants might affect their pharmacokinetic properties of their constituents.

Isolation and Identification of Virus dsRNA from Strawberry Plants

The analysis of virus genome is based on nucleic acid isolation. The aims of this study were to develop a method for isolation and identification of virus double-stranded ribonucleic acid （dsRNA） and to elucidate the nucleotide sequences of strawberry virus. Using the modified method, virus dsRNA was extracted from strawberry virus indicator plants and cultivated strawberry plants and detected using agarose gel electrophoresis with ethidium bromide staining and reverse transcription-polymerase chain reaction （RT-PCR）. The quantity of virus dsRNA varied among strawberry cultivars. The quantity of dsRNA from in vitro plantlets was higher than that from the young leaves of field plants. For the field-grown plants, there was more dsRNA in the young leaves. Virus dsRNA extracted from strawberry plants was resistant to deoxyribonuclease Ⅰ （DNase Ⅰ）, but evidently, it became resistant to ribonuclease A （RNase A） only in the presence of 0.5 M NaCl. Its bands in agarose gel could be readily recycled using an agarose gel DNA purification kit. With RT-PCR, the segments of both strawberry mottle virus and Strawberry mild yellow edge virus genomes were amplified by using the virus dsRNA recycled from gel or treated with DNase Ⅰ /RNase A as templates. The system developed for dsRNA isolation and identification in strawberry plants laid a sound foundation for the work on genome analysis of strawberry virus isolates in China.

中药的减毒配伍是保障中药安全性的有效措施。但中药减毒配伍研究面临诸多难点,涉及因素包括品种基原、炮制方法、配伍模式、用药剂量等,呈现出“毒性物质的不确定性”“毒性发生的隐匿性”“安全剂量的模糊性”“毒-效关系的复杂性”和“个体差异的特殊性”等特征,使得“分子-细胞-器官-整体”系统评价模型的一致性、“多药多物质”的联合暴露、“毒性-功效-配伍-证候”的关联、“量-时-毒-效”转化规律等成为影响中药临床安全研究的关键科学问题。针对上述关键科学问题,应遵循“始于临床、证于基础、终于临床”的模式,通过构建“分子-细胞-器官-整体”多水平毒性确证、多药多物质相互作用和联合暴露、多层面循证、多“量-时-毒-效”转换、基础与临床生物标志物一致性求证等“五多一证”技术体系,系统研究其减毒增(存)效配伍的作用特征、单药和多药体内命运、毒性发生分子机制、“量-时-毒-效”转化规律及病证结合的有毒中药临床特征。通过研究“两类三种机制”赋予中医七情和合配伍理论新内涵,最终提出安全用药策略。

Protective effects of upregulated HO-1 gene against the apoptosis of human retinal pigment epithelial cells in vitro

AIM:To investigate the protective effect of heme oxygenase-1(HO-1)against H_(2)O_(2)-induced apoptosis in human ARPE-19 ***:The lentiviral vector expressing HO-1 was prepared and transfected into apoptotic ARPE-19 cells induced by H_(2)O_(2).Functional experiments including cell counting kit-8(CCK-8)assay,flow cytometry(FCM)and mitochondrial membrane potential assay were ***:The ultrastructure of ARPE-19 cells was observed using transmission electron microscope(TEM).It was found that exogenous HO-1 significantly ameliorated H_(2)O_(2)-induced loss of cell viability,apoptosis and intracellular levels of reactive oxygen species(ROS)in ARPE-19 *** overexpression of HO-1 facilitated the transfer of nuclear factor erythroid-2-related factor 2(Nrf2)from cytoplasm to nucleus,which in turn upregualted expressions HO-1 and B-cell lymphoma-2(Bcl-2).Furthermore,HO-1 upregulation further inhibited H_(2)O_(2)-induced release of cysteinyl aspartate specific proteinase-3(caspase-3).CONCLUSION:Exogenous HO-1 protect ARPE-19 cells against H_(2)O_(2)-induced oxidative stress by regulating the expressions of Nrf2,HO-1,Bcl-2,and caspase-3.

The construction of a new beamline,BL10U1,was completed at the Shanghai synchrotron radiation facility in *** multipurpose beamline was designed to provide X-ray scattering techniques such as ultra-small-angle X-ray scattering(USAXS),small-angle X-ray scattering(SAXS),wide-angle X-ray scattering,and microfocus SAXS(μSAXS)for a broad user *** realize fast time-resolved USAXS experiments,the beamline adopted an in-vacuum undulator with a total length of 1.6 m as the photon *** in-house cryogenic-cooled double multilayer monochromator was installed to deliver a photon flux of approximately 10^(13) photons/s at a photon energy of 10 *** three-year successful operation of this beamline demonstrated that the monochromator operated smoothly,as ***10U1 has three end stations in succession:USAXS end station,μSAXS end station,and end station for industrial *** minimum scattering vector q~0.0042 nm^(-1) at 10 keV can be achieved at the USAXS end station equipped with a 28 m-long and 1.8 m-diameter vacuum flight *** theμSAXS end station,a beam spot of less than 10×8μm was achieved for micro-SAXS *** contrast,in situ experimental instruments up to 5 m high and 8 m wide can be mounted at the industrial application end station,which offers industrial scientists the opportunity to use their large industrial ***10U1 opens up a new capability to investigate phenomena such as non-equilibrium and dynamic processes of materials with a wide length scale from angstroms to micrometers with millisecond time *** this paper,we also report beamline design considerations and commissioning results.

针对复杂协同攻击下一类非线性系统的状态估计问题,提出一种分布式一致性递推滤波算法.首先,将拒绝服务攻击(denial of service, DoS)和虚假数据注入攻击(false data injection, FDI)现象描述为两个随机Bernoulli序列,并利用统一的框架建立包含DoS和FDI的复杂协同攻击模型;然后,基于一致性理论设计具有分布式结构的递推滤波器,计算最优滤波器增益,并推导该滤波器估计误差满足均方有界的充分必要条件;最后,利用室内机器人的定位问题进行验证,仿真结果验证了所提出滤波器算法的有效性.

Yang-tonifying traditional Chinese medicinal plants and their potential phytoandrogenic activity

The concept of phytoandrogens, plants that contain androgens or those that stimulate androgenic activity in men, is relatively new. In traditional Chinese medicine a number of phytoandrogens are classified in medicinal plant restoratives for reinforcing yang, and they find their application in the treatment of the kidney yang deficiency diseases. In this review, the phytoandrogens used in traditional Chinese medicine are listed, and their proven applications in the treatment of kidney yang deficiency diseases, such as sexual disorders, cancer, and obesity and associated metabolic syndromes are presented. As a background, the mechanism of action of androgens, their synthesis and metabolism, the interrelations of androgens and estrogens, as well as the state of art methods to detect and analyze these hormonal activities in medicinal plants are discussed.
