Alarmins from conjunctival fibroblasts up-regulate matrix metalloproteinases in corneal fibroblasts

AIM:To explore the effects of alarmins produced by necrotic human conjunctival fibroblasts on the release of matrix metalloproteinases(MMPs)by human corneal fibroblasts(HCFs).METHODS:A necrotic cell supernatant(NHCS)w.s prepared by subjecting human conjunctival fibroblasts to three cycles of freezing and *** amounts of interleukin(IL)-1βand tumor necrosis factor(TNF)-αin NHCS w.re determined by enzyme-linked immunosorbent *** exposed to NHCS or other agents in culture w.re assayed for the release of MMPs as w.ll as for intracellular signaling by immunoblot *** abundance of MMP m RNAs in HCFs w.s examined by reverse transcription and real-time polymerase chain reaction ***:NHCS increased the release of MMP-1 and MMP-3 by HCFs as w.ll as the amounts of the corresponding m RNAs in the *** also induced activation of mitogen-activated protein kinase(MAPK)signaling pathw.ys mediated by extracellular signal-regulated kinase(ERK),p38,and c-Jun NH2-terminal kinase(JNK)as w.ll as elicited that of the nuclear factor(NF)-κB signaling pathw.y by promoting phosphorylation of the endogenous NF-κB inhibitor IκB-α.Inhibitors of MAPK and NF-κB signaling as w.ll as IL-1 and TNF-αreceptor antagonists attenuated the NHCS-induced release of MMP-1 and MMP-3 by ***,IL-1βand TNF-αw.re both detected in NHCS,and treatment of HCFs w.th these cytokines induced the release of MMP-1 and MMP-3 in a concentration-dependent ***:Alarmins,including IL-1βand TNF-α,produced by necrotic human conjunctival fibroblasts triggered MMP release in HCFs through activation of MAPK and NF-κB ***-1βand TNF-αare therefore potential therapeutic targets for the amelioration of corneal stromal degradation in severe ocular burns.

Cerebellar ataxias are characterized by a progressive decline in motor coordination,but the specific output circuits and underlying pathological mechanism remain poorly *** cell-type-specific manipulations,w. discovered a novel GABAergic Purkinje cell(PC)circuit in the cerebellar IV/V lobe that projected to CaMKIIα+neurons in the fastigial nucleus(FN),w.ich regulated sensorimotor ***,transcriptomics profiling analysis revealed various cerebellar neuronal identities,and w. validated that biorientation defective 1(BOD1)played an important role in the circuit of IV/V lobe to ***1 deficit in PCs of IV/V lobe attenuated the excitability and spine density of PCs,accompany w.th ataxia ***,BOD1 enrichment in PCs of IV/V lobe reversed the hyperexcitability of CaMKIIα+neurons in the FN and ameliorated ataxia behaviors in L7-Cre;BOD1f/f ***,these findings further suggest that specific regulation of the cerebellar IV/V lobePCs→FNCaMKIIα+circuit might provide neuromodulatory targets for the treatment of ataxia behaviors.

目的用Open Vigil FDA2.1对美国食品药品监督管理局不良事件呈报系统(FAERS)数据库中鲁拉西酮的相关数据进行分析,探究该药不良事件的发生情况,为我国临床用药丰富经验,提供依据。方法用Open Vigil FDA2.1提取FAERS数据库中2010年第四季度至2023年第三季度关于鲁拉西酮的不良事件数据,用报告比值比(ROR)法与比例报告比(PRR)法进行数据挖掘与分析。结果共得到32728份以鲁拉西酮为首要怀疑药物的不良事件报告,其中女性占比较大(54.26%),且多发生在成年人(18~59岁)。筛选后获取326个首选术语(PT)信号,累及20个系统器官分类(各类损伤、中毒及操作并发症、全身性疾病及给药部位各种反应、精神病类等)。其中,发生频次比较高的PT包括超说明书使用、感受异常、哭、焦虑、抑郁、失眠等;信号强度较强的PT包括激活综合征、躁狂、舌运动障碍、低催乳素血症、静坐不能等。挖掘到多个新的可疑药物不良反应,包括低催乳素血症、情绪贫乏、强硬舌等。结论鲁拉西酮的安全性较佳,女性服用该药时,需密切关注催乳素水平。妊娠期、产褥期及围生期女性及代谢功能不佳的患者使用该药相对安全。低催乳素水平、不宁腿综合征是鲁拉西酮新的罕见的可疑不良事件。

Rapid Quantification of Chinese Medicine Zuo Jin Pill via RRLC

A rapid method based on rapid resolution liquid chromatography（RRLC） coupled w.th a diode array detector（DAD） w.s developed for the simultaneous determination of six major constituents（magnoflorine,jatrorrhizine,coptisine,palmatine,berberine and evodiamine） in traditional Chinese medicine（TCM） Zuo Jin Pill（ZJP）.The satisfactory chromatographic separation w.s carried on an Eclipse Plus C18 column（1.8 μm i.d.,150 mm×4.6 mm） by linear gradient elution w.th a mobile phase of acetonitrile-acetate *** the calibration curves show.good linearity（r2 0.9998）.The detection limits and quantification limits ranged in 1.4―12 ng and 4.8―30 ng,*** intra-and inter-day precisions w.re less than 0.63% w.th accuracies 98.60%―100.78%,and the recoveries w.re from 99.45% to 100.46%.Furthermore,hierarchical cluster analysis（HCA） w.s used to evaluate the variation of the herbal *** results demonstrate that this analytical method is simple,sensitive and reliable for rapidly analyzing six major bioactive compounds in ZJPs and is helpful to comprehensively evaluating the quality of this TCM.

Numerical simulation study on monoblock casting process of ultra-slender structural components and experimental validation

Substrate, a typical ultra-slender aluminum alloy structural components w.th a large aspect ratio and complex internal structure, w.s traditionally manufactured by re-assembly and sub-w.lding. In order to realize the monoblock casting of the substrate, the Pro/E softw.re w.s utilized to carry out three-dimensional(3D) modeling of the substrate casting, and the filling and solidification processes w.re calculated, as w.ll as the location and types of casting defects w.re predicted by the casting simulation softw.re Anycasting. Results of the filling process simulation show.that the metal liquid is distributed into each gap runner evenly and smoothly. There is no serious vortex phenomenon in the mold cavity, and the trajectory of the virtual particles is clear. Results of the solidification process simulation show.that shrinkage cavities mainly appear at the junction of gap runners and the rail surface of the substrate. The average deformation is 0.6 mm in X direction, 3.8 mm in Y direction, and 8.2 mm in Z direction. Based on the simulation results, the casting process of the substrate w.s optimized, and qualified castings w.re successfully produced, w.ich w.ll provide a reference for the casting process design of other ultraslender aluminum alloy structural components.

由于具有诸多优异性能，金属有机框架(Metal-Organic Framew.rks,MOFs)成为过去20多年中广受关注的材料之一。MOF材料在实验室水平上已经展示出很多领域的潜在应用前景，同时部分MOF材料已实现工业化量产。MOF材料当前及未来的研究重点是如何克服其内在的一些缺陷而实现规模化的实际应用。MOF与聚合物材料复合可以融合二者各自的材料特性，为迎接MOF大规模实际应用所面临的诸多挑战提供一定的解决方案。因此，MOF/聚合物复合材料的制备及其应用成为近几年的研究热点，文献报道了多种思路来制备功能各异的MOF/聚合物复合材料。文章总结了当前制备这类材料的主要策略及各自存在的问题，涵盖MOF粉体与聚合物材料直接共混后加工制备的各种形态的复合材料，如混合基质膜、复合树脂球以及纳米纤维复合材料等；将MOF的前体引入聚合物本体材料中原位合成MOF；在聚合物基底上合成MOF材料来构建聚合物支撑的MOF薄膜、纤维以及核壳结构的颗粒状复合材料等。

Pathologic comparisons of enucleated eyes w.th retinoblastoma after superselective ophthalmic arterial chemotherapy w.th or w.thout intravenous chemotherapy

AIM:To describe and compare pathologic findings in eyes enucleated after superselective ophthalmic arterial chemotherapy(SOAC)or SOAC w.th intravenous chemotherapy(IVC)for ***:Medical records betw.en January 1st,2014 and June 30th,2017 w.re retrospectively analyzed,and pathologic findings w.re *** study included 36 eyes from 22(61.1%)male and 14(38.9%)female *** of 36(52.8%)eyes received SOAC(mean=3,range=1-7)as primary treatment,and 17 of 36(47.2%)eyes received SOAC(mean=3.7,range=1-10)after IVC(mean=6.1,range=2-11).Tumor extension including choroidal invasion(n=9,25%),optic nerve invasion(n=5,13.9%)and anterior segment invasion(n=5,13.9%)w.re ***:Histopathologic evidence of ischemic damage in the retina and choroid w.s found in 28(77.8%)*** blood vessels w.re identified in 9(25%)eyes,including orbital artery in the retrobulbar orbit(n=1),intrascleral vessels(n=4),and chorioretinal vessels(n=6).Fibrotic changes w.re found in extraocular muscles(n=5,13.9%)and optic nerve(n=5,13.9%).Varying degrees of scleral degeneration w.re found in all *** statistical analysis,there w.s no significant difference in clinical and pathologic changes betw.en SOAC group and SOAC w.th IVC group except for optic nerve invasion(P=0.047).CONCLUSION:SOAC for retinoblastoma can result in ocular toxicity,and SOAC w.th IVC do not increase the toxicity but reduced the incidence of optic nerve invasion.

为了解决隔膜压缩机中膜片经常破裂的问题,以某FB型隔膜压缩机为原型,对膜片组运行机理进行研究,探索膜腔挠度指数变化时对膜片运行情况的影响。基于薄板大挠度理论计算得出膜片上下表面所受应力最大的位置,通过改变膜腔挠度指数,得出膜片所受应力最小的膜腔型线。运用有限元分析软件进行模拟仿真,结果验证了膜片上表面的中心处和膜片下表面的边缘处最易破坏,同时得出膜腔型线的挠度指数在2.9时,可使膜片的上下表面所受应力最小且应力强度几乎相等,优化后的膜片所受径向应力相比优化前下降了14%左右,延长了膜片的使用寿命。

In this article, unique spectral features of short-w.ve infrared band of 1 μm–3 μm, and various applications related to the photodetectors and focal plane arrays in this band, are introduced briefly. In addition, the different material systems for the devices in this band are outlined. Based on the background, the development of lattice-matched and w.velengthextended InGaAs photodetectors and focal plane arrays, including our continuous efforts in this field, are review.d. These devices are concentrated on the applications in spectral sensing and imaging, exclusive of optical fiber communication.
