Engineered tumor cell-derived vaccines against cancer: The art of combating poison with poison

Tumor vaccination is a promising approach for tumor immunotherapy because it presents high specificity and few side effects.However,tumor vaccines that contain only a single tumor antigen can allow immune system evasion by tumor variants.Tumor antigens are complex and heterogeneous,and identifying a single antigen that is uniformly expressed by tumor cells is challenging.Whole tumor cells can produce comprehensive antigens that trigger extensive tumor-specific immune responses.Therefore,tumor cells are an ideal source of antigens for tumor vaccines.A better understanding of tumor cell-derived vaccines and their characteristics,along with the development of new technologies for antigen delivery,can help improve vaccine design.In this review,we summarize the recent advances in tumor cell-derived vaccines in cancer immunotherapy and highlight the different types of engineered approaches,mechanisms,administration methods,and future perspectives.We discuss tumor cell-derived vaccines,including whole tumor cell components,extracellular vesicles,and cell membrane-encapsulated nanoparticles.Tumor cell-derived vaccines contain multiple tumor antigens and can induce extensive and potent tumor immune responses.However,they should be engineered to overcome limitations such as insufficient immunogenicity and weak targeting.The genetic and chemical engineering of tumor cell-derived vaccines can greatly enhance their targeting,intelligence,and functionality,thereby realizing stronger tumor immunotherapy effects.Further advances in materials science,biomedicine,and oncology can facilitate the clinical translation of tumor cell-derived vaccines.

mHealth as a health system strengthening tool in China

Over the past 70 years,there was a rapid epidemiological transition in disease burden in China,from infectious disease to non-communicable diseases(NCDs),which requires long-term prevention and management.Rapid growth in mobile phone use,internet connectivity and digital health technology,presents new opportunities for improvement in NCD healthcare delivery and population-based outcomes.Although there were a growing number of research to evaluate the feasibility and effectiveness of the mobile health(mHealth)interventions for NCD management,the extent to which mHealth contributes towards the health system strengthening in China remains unknown.In this paper,we provided a high-level overview of mHealth in China and its role for Chinese health system strengthening.We conclude with several recommendations for the future of mHealth research in China based on existing evidence identified.

Breed identification using breed‑informative SNPs and machine learning based on whole genome sequence data and SNP chip data

Background Breed identification is useful in a variety of biological contexts.Breed identification usually involves two stages,i.e.,detection of breed-informative SNPs and breed assignment.For both stages,there are several methods proposed.However,what is the optimal combination of these methods remain unclear.In this study,using the whole genome sequence data available for 13 cattle breeds from Run 8 of the 1,000 Bull Genomes Project,we compared the combinations of three methods(Delta,FST,and In)for breed-informative SNP detection and five machine learning methods(KNN,SVM,RF,NB,and ANN)for breed assignment with respect to different reference population sizes and difference numbers of most breed-informative SNPs.In addition,we evaluated the accuracy of breed identification using SNP chip data of different densities.Results We found that all combinations performed quite well with identification accuracies over 95%in all scenarios.However,there was no combination which performed the best and robust across all scenarios.We proposed to inte-grate the three breed-informative detection methods,named DFI,and integrate the three machine learning methods,KNN,SVM,and RF,named KSR.We found that the combination of these two integrated methods outperformed the other combinations with accuracies over 99%in most cases and was very robust in all scenarios.The accuracies from using SNP chip data were only slightly lower than that from using sequence data in most cases.Conclusions The current study showed that the combination of DFI and KSR was the optimal strategy.Using sequence data resulted in higher accuracies than using chip data in most cases.However,the differences were gener-ally small.In view of the cost of genotyping,using chip data is also a good option for breed identification.

3D electrodes have shown extraordinary promise for electrochemical energy storage devices in wearable electronics.However,it is still a significant challenge to rationally design 3D electrode that has the characteristics of lightweight,flexibility,low cost,high performance and miniaturization.In this work,we present a novel design of 3D electrode by directly growing the nanocrystalline TiO_(2) film on carbonized eggshell membrane.The unique architecture can supply not only a continuous electron transport pathway through an interconnected carbon fibrous network but also an efficient electrolyte flux via an interpenetrating porous network.Besides,nanocrystalline TiO_(2) film on carbonized eggshell membrane offers a short ion diffusion path in the solid-phase,leading to a rapid kinetic of electrochemical reaction.When tested as an anode for Li-ion battery,TiO_(2) film in 3D electrode demonstrated excellent electrochemical performance with a large reversible capacity,excellent rate capacity and long life cycling property.

Visualizing Aβdeposits in live young AD model mice with a simple red/near-infrared-fluorescent AIEgen

Precise and early detection ofβ-amyloid(Aβ)deposits in situ and in real time is pivotal to the diagnosis and early intervention of Alzheimer’s disease(AD).Optical imaging stands out to be a promising technique for such a task;however,it still remains a big challenge,due to the lack of high-performance imaging contrast agent.Restricted by poor blood-brain barrier(BBB)penetrability,short-wavelength excitation and emission,as well as the aggregation-caused quenching effect,the widely used goldstandard probes cannot be used for early in-vivo imaging of Aβdeposits.Herein,we integrate the Aβdeposits-favored geometry,amphiphilic and zwitterionic molecular structure,extended D-π-A electronic structure,and 3 D conformation into one molecule,facilely establishing a simple and economic imaging contrast agent that enjoys high specificity and affinity to Aβdeposits,good BBB penetrability,bright red/near-infrared fluorescence,low interference from autofluorescence,aggregation-induced emission(AIE)feature,high signal-to-noise ratio(SNR),and high contrast.In-vitro,ex-vivo,and in-vivo experiments with different strains of mice indicate that AIE-CNPy-AD holds the universality to Aβdeposits identification.Noteworthily,AIE-CNPy-AD is even able to precisely trace the small and sparsely-distributed Aβdeposits in AD model mice as young as 4-month-old APP/PS1 mice,the youngest having Aβdeposits.Moreover,the present probe could clearly reveal the increase and enlargement of Aβdeposits as the mice grow.Therefore,AIE-CNPy-AD might be an ideal alternative for early AD diagnosis and highly reliable monitoring of AD progression.

Osteosarcoma is the most common primary malignant bone tumor among adolescents and children,with high level of lung metastasis and poor prognosis.The treatment of metastatic cases remains a challenge in the clinic,leading to the dramatically decreased survival rate.However,the molecular mechanisms are still unclear.A recent article by Kang et al.demonstrated how chromosomal translocations in osteosarcoma facilitated lung metastasis,and shed new light on its therapeutic strategies1(Fig.1).

The generation of methylsufonyl-containing thioflavones through a radical relay reaction of methylthiolated phenylalkynones and potassium metabisulfite in the presence of a catalytic amount of sodium methylsulfinate as an initiator under photoinduced conditions is developed.This photocatalytic reaction proceeds smoothly with good functional group tolerance under visible light irradiation.Preliminary mechanistic investigations indicate that this reaction occurs through a radical relay pathway.

Corn ear test using SIFT-based panoramic photography and machine vision technology

Corn ear test is important to modern corn breeding.The test indexesmainly include lengths,radiuses,rows and numbers of corn ears and the kernels they bear,which can benefit the study on breeding new and fine corn varieties.These corn traits are often collected by traditional manual measurement,which is difficult to meet the needs of high throughput corn ear test.In this study,image sequences of corn ear samples were captured by building a panoramic photography collecting system.And then,to get the lengths and radiuses indexes,the corn area images were processed based on Lab color space and adaptive threshold segmentation.The sequence images were then matched and the panoramic image of a corn surface were extracted using Scale-invariant feature transform(SIFT).Finally,by using Exponential transformation(ETR)and Sobel-Hough algorithm,ears and rows indexes were acquired.Test results showed that the accuracy of the radiuses and lengths were 93.84%and 94.53%,respectively.Meanwhile,the accuracy of kernels and rows indexes were 98.12%and 96.14%,whichwere 4.03%and 7.25%higher than that of common mosaiced panoramic image.And the accuracy of kernel area and length-width ratio were 95.36%and 97.42%,respectively.All the results showed that the proposed method can be used for corn ear test effectively.

Point mutations of homologs as an adaptive solution to the gene loss

Gene loss is common and influences genome evolution trajectories.Multiple adaptive strategies to compensate for gene loss have been observed,including copy number gain of paralogous genes and mutations in genes of the same pathway.By using the Ubl-specific protease 2(ULP2)eviction model,we identify compensatory mutations in the homologous gene ULP1 by laboratory evolution and find that these mutations are capable of rescuing defects caused by the loss of ULP2.Furthermore,bioinformatics analysis of genomes of yeast gene knockout library and natural yeast isolate datasets suggests that point mutations of a homologous gene might be an additional mechanism to compensate for gene loss.

Semi-quantitative design of synergetic surficial/interfacial sites for the semi-continuous oxidation of glycerol

Qualitatively identifying the dominant catalytic site for each step of a semi-continuous reaction and semi-quantitatively correlating such different sites to the catalytic performance is of great significance toward the integration of multiple well-optimized sites on a heterogeneous catalyst.Herein,a series of structurally defined TiO_(x)-based catalysts were synthesized to provide a feasible approach to investigate the aforementioned issues using the semi-continuous oxidation of glycerol as a model reaction.Detailed investigations have verified the simultaneous presence of two kinds of Pt active sites:1)Negatively charged Pt bound to the oxygen vacancies of modified TiO_(x)in the form of Pt^(δ−)-O_(v)-Ti^(3+) sites and 2)metallic Pt(Pt_(0)site)located away from the inter-face.Meanwhile,the proportion of surficial and interfacial sites varies over this series of catalysts.Combined in situ FTIR experiments revealed that the reaction network was well-tuned via a site cooperation mechanism:The surficial Pt_(0)sites dissociatively adsorb the OH group of glycerol with a monodentate bonding geometry and the Pt^(δ−)-O_(v)-Ti^(3+) sites dissociate the C=O bond of the aldehyde group in a bidentate form.Furthermore,CO-FTIR spectroscopy confirmed a correlation between the reaction rate/product selectivity and the fraction of surficial/interfacial sites.A rational proportion of surficial and interfacial sites is key to enabling a high yield of glyceric acid.The most active catalyst with 32%surface sites and 68%interfacial sites exhibited 90.0%glycerol conversion and 68.5%GLYA selectivity.These findings provide a deeper understanding of the structure-activity relationships using qualitative identification and semi-quantitative analysis.
