Cerebral haemodynamics in symptomatic intracranial atherosclerotic disease:a narrative review of the assessment methods and clinical implications

Intracranial atherosclerotic disease(ICAD)is a common cause of ischaemic stroke and transient ischaemic attack(TIA)with a high recurrence rate.It is often referred to as intracranial atherosclerotic stenosis(ICAS),when the plaque has caused significant narrowing of the vessel lumen.The lesion is usually considered‘symptomatic ICAD/ICAS’(sICAD/sICAS)when it has caused an ischaemic stroke or TIA.The severity of luminal stenosis has long been established as a prognostic factor for stroke relapse in sICAS.Yet,accumulating studies have also reported the important roles of plaque vulnerability,cerebral haemodynamics,collateral circulation,cerebral autoregulation and other factors in altering the stroke risks across patients with sICAS.In this review article,we focus on cerebral haemodynamics in sICAS.We reviewed imaging modalities/methods in assessing cerebral haemodynamics,the haemodynamic metrics provided by these methods and application of these methods in research and clinical practice.More importantly,we reviewed the significance of these haemodynamic features in governing the risk of stroke recurrence in sICAS.We also discussed other clinical implications of these haemodynamic features in sICAS,such as the associations with collateral recruitment and evolution of the lesion under medical treatment,and indications for more individualised blood pressure management for secondary stroke prevention.We then put forward some knowledge gaps and future directions on these topics.

This paper initiates the formal study of attribute-based encryption within the framework of SM9,the Chinese National Cryptography Standard for Identity-Based Cryptography, by presenting two new faulttolerant identity-based encryption(FIBE) schemes. Our first scheme uses the same private-key/ciphertext structure as the original SM9 algorithm and operates in a small attribute universe. As a result, it can be effectively and smoothly integrated into the information systems using SM9. In the random oracle model,we prove that our scheme is ciphertext-indistinguishable against fuzzy selective-identity and chosen-plaintext attacks under the(k + 3)-DBDHI assumption. Our second design is a large universe FIBE scheme based on SM9 that is ciphertext-indistinguishable against chosen-plaintext attacks in the random oracle model under the(f, g)-GDDHE assumption. Finally, we compare the communication and computing costs of our schemes to those of other classical ones. The comparison shows that our schemes have comparable performance as others. We believe that our findings will accelerate the applications of SM9 in modern information systems such as cloud computing and blockchain.

El Ni?o–Southern Oscillation(ENSO) exhibits a distinctive phase-locking characteristic, first expressed during its onset in boreal spring, developing during summer and autumn, reaching its peak towards winter, and decaying over the next spring. Several studies have demonstrated that this feature arises as a result of seasonal variation in the growth rate of ENSO as expressed by the sea surface temperature(SST). The bias towards simulating the phase locking of ENSO by many state-of-the-art climate models is also attributed to the unrealistic depiction of the growth rate. In this study, the seasonal variation of SST growth rate in the Ni?o-3.4 region(5°S–5°N, 120°–170°W) is estimated in detail based on the mixed layer heat budget equation and recharge oscillator model during 1981–2020. It is suggested that the consideration of a variable mixed layer depth is essential to its diagnostic process. The estimated growth rate has a remarkable seasonal cycle with minimum rates occurring in spring and maximum rates evident in autumn. More specifically, the growth rate derived from the meridional advection(surface heat flux) is positive(negative) throughout the year. Vertical diffusion generally makes a negative contribution to the evolution of growth rate and the magnitude of vertical entrainment represents the smallest contributor. Analysis indicates that the zonal advective feedback is regulated by the meridional immigration of the intertropical convergence zone, which approaches its southernmost extent in February and progresses to its northernmost location in September, and dominates the seasonal variation of the SST growth rate.

Coordinated contraction of skeletal muscles relies on selective connections between the muscles and multiple classes of the spinal motoneuro ns.Howeve r,current research on the spatial location of the spinal motoneurons innervating differe nt muscles is limited.In this study,we investigated the spatial distribution and relative position of different motoneurons that control the deep muscles of the mouse hindlimbs,which were innervated by the obturator nerve,femoral nerve,inferior gluteal nerve,deep pe roneal nerve,and tibial nerve.Locations were visualized by combining a multiplex retrograde tracking technique compatible with three-dimensional imaging of solvent-cleared o rgans(3DISCO)and 3-D imaging technology based on lightsheet fluorescence microscopy(LSFM).Additionally,we propose the hypothesis that"messenger zones"exist as interlaced areas between the motoneuron pools that dominate the synergistic or antagonist muscle groups.We hypothesize that these interlaced neurons may participate in muscle coordination as messenger neurons.Analysis revealed the precise mutual positional relationships among the many motoneurons that innervate different deep muscles of the mouse.Not only do these findings update and supplement our knowledge regarding the overall spatial layout of spinal motoneurons that control mouse limb muscles,but they also provide insights into the mechanisms through which muscle activity is coordinated and the architecture of motor circuits.

In exploring persistent infections and malignancies, a distinctive subgroup of CD8+T cells, progenitor exhausted CD8+T（Tpex） cells, has been identified. These Tpex cells are notable for their remarkable self-renewal and rapid proliferation abilities. Recent strides in immunotherapy have demonstrated that Tpex cells expand and differentiate into responsive exhausted CD8+T cells, thus underscoring their critical role in the immunotherapeutic retort. Clinical examinations have further clarified a robust positive correlation between the proportional abundance of Tpex cells and enhanced clinical prognosis. Tpex cells have found noteworthy applications in the formulation of inventive immunotherapeutic approaches against tumors. This review describes the functions of Tpex cells in the tumor milieu, particularly their potential utility in tumor immunotherapy. Precisely directing Tpex cells may be essential to achieving successful outcomes in immunotherapy against tumors.

With the rapid development of histological techniques and the widespread applica-tion of single-cell sequencing in eukaryotes,researchers desire to explore individual microbial.genotypes and functional expression,which deepens our understanding of microorganisms.In this review,the history of the development of microbial detection technologies was revealed and the difficulties in the application of single-cell sequencing in microorganisms were dissected as well.Moreover,the characteristics of the currently emerging microbial single-cell sequencing(Microbe-seq)technology were summarized,and the prospects of the application of Microbe-seq in microorganisms were distilled based on the current development status.Despite its mature development,the Microbe-seq technology was still in the optimization stage.A retrospective study was conducted,aiming to promote the widespread application of single-cell sequencing in microorganisms and facilitate further improvement in the technol-ogy.

A Novel Sensing Imaging Equipment Under Extremely Dim Light for Blast Furnace Burden Surface:Starlight High-Temperature Industrial Endoscope

Blast furnace(BF)burden surface contains the most abundant,intuitive and credible smelting information and acquiring high-definition and high-brightness optical images of which is essential to realize precise material charging control,optimize gas flow distribution and improve ironmaking efficiency.It has been challengeable to obtain high-quality optical burden surface images under high-temperature,high-dust,and extremelydim(less than 0.001 Lux)environment.Based on a novel endoscopic sensing detection idea,a reverse telephoto structure starlight imaging system with large field of view and large aperture is designed.Combined with a water-air dual cooling intelligent self-maintenance protection device and the imaging system,a starlight high-temperature industrial endoscope is developed to obtain clear optical burden surface images stably under the harsh environment.Based on an endoscope imaging area model,a material flow trajectory model and a gas-dust coupling distribution model,an optimal installation position and posture configuration method for the endoscope is proposed,which maximizes the effective imaging area and ensures large-area,safe and stable imaging of the device in a confined space.Industrial experiments and applications indicate that the proposed method obtains clear and reliable large-area optical burden surface images and reveals new BF conditions,providing key data support for green iron smelting.

Objective: Abnormal metabolism is the underlying reason for breast cancer progression. Decreased lactate dehydrogenase B(LDHB) has been detected in breast cancer but the function of LDHB remains unknown.Methods: Western blot was used to analyze LDHB expression in breast cancer cells. The impact of LDHB on tumor cell migration and invasion was determined using Transwell assays, wound healing assays, and a mouse lung metastasis model. Subcutaneous tumor formation, a natural killer(NK) cell cytotoxicity assay, and flow cytometry evaluated NK cell activation. Immunofluorescence and quantitative real-time PCR detected NK cell activation markers. Kaplan-Meier analysis evaluated the effect of immune cell infiltration on prognosis. Single-sample gene set enrichment analysis determined NK cell activation scores. A support vector machine predicted the role of LDHB in NK cell activation.Results: In this study we showed that LDHB inhibits the breast cancer cell metastasis and orchestrates metabolic reprogramming within tumor cells. Our results revealed that LDHB-mediated lactic acid clearance in breast cancer cells triggers NK cell activation within the tumor microenvironment. Our findings, which were confirmed in a murine model, demonstrated that LDHB in tumor cells promotes NK cell activation and ultimately results in the eradication of malignant cells. Clinically, our study further validated that LDHB affects immune cell infiltration and function. Specifically, its expression has been linked to enhanced NK cell-mediated cytotoxicity and improved patient survival. Furthermore, we identified LDHB expression in tumors as an important predictor of NK cell activation, with strong predictive ability in some cancers.Conclusions: Our results suggest that LDHB is a promising target for activating the tumor immune microenvironment in breast cancer, where LDHB-associated lactic acid clearance leads to increased NK cell activity. Synthetic chemistry plays an indispensable role in drug discovery,contributing to hit compounds identification,lead compounds optimization,candidate drugs preparation,and so on.As Nobel Prize laureate James Black emphasized,“the most fruitful basis for the discovery of a new drug is to start with an old drug”1.Late-stage modification or functionalization of drugs,natural products and bioactive compounds have garnered significant interest due to its ability to introduce diverse elements into bioactive compounds promptly.Such modifications alter the chemical space and physiochemical properties of these compounds,ultimately influencing their potency and druggability.To enrich a toolbox of chemical modification methods for drug discovery,this review focuses on the incorporation of halogen,oxygen,and nitrogen—the ubiquitous elements in pharmacophore components of the marketed drugs—through late-stage modification in recent two decades,and discusses the state and challenges faced in these fields.We also emphasize that increasing cooperation between chemists and pharmacists may be conducive to the rapid discovery of new activities of the functionalized molecules.Ultimately,we hope this review would serve as a valuable resource,facilitating the application of late-stage modification in the construction of novel molecules and inspiring innovative concepts for designing and building new drugs.
