The most essential and crucial step during the pathogenesis of transmissible spongiformencephalopathy is the conformational change of cellular prion protein （PrPC） to pathologic isoform (PrPSc).Alot of data revealed that caveolae-like domains （CLDs） in the cell surface were the probable place where theconversion of PrP proteins *** E （ApoE） is an apolipoprotein which is considered toplay an important role in the development of Alzheimer's disease and other neurodegenerative diseases byforming protein complex through binding to the receptor located in the clathrin-coated pits of the cell *** this study,a 914-bp cDNA sequence encoding human ApoE3.was amplified from neuroblastoma cell *** human ApoE isomers were expressed and purified from Escherichia ***-specificantiserum was prepared by immunizing rabbits with the purified ***/His pull-down assay,immunoprecipitation and ELISA revealed that three full-length ApoE isomers interact with the recombinantfull-length PrP protein in *** regions corresponding to protein binding were mapped in the N-terminalsegment of ApoE （amino acid 1-194） and the N-terminal of PrP （amino acid 23.90）.Moreover,the recombinantPrP showed the ability to form a complex with the native ApoE from liver *** data provided directevidence of molecular interaction between ApoE and *** also supplied scientific clues for assessing thesignificance of CLDs on the surface of cellular membrane in the process of conformational conversion fromPrPCto PrPScand probing into the pathogenesis of transmissible spongiform encephalopathy.

Aim:To investigate the antinociceptive effect of adenosine agonist R-phenylisopropyl-adenosine（R-PIA）given to conscious rats by intracerebro-ventricular（ICV）and intrathecal（IT）,and identify the effect of R-PIA on minimumalveolar concentration（MAC）of halothane with pretreatment of A1receptor an-tagonist 8-cyclopentyl-1,3.dipropylxanthine（DPCPX）or K+channel blocker4-aminopyridine（4-AP）.Methods:Sprague-Dawley rats were implanted with 24-gauge stainless steel guide cannula using stereotaxic apparatus and ICV method,and an IT catheter（PE-10,8.5cm）was inserted into the lumbar subarachnoidspace,while the rats were under pentobarbital *** one week ofrecovery from surgery,rats were randomly assigned to one of the followingprotocols:MAC of halothane,or tail-flick *** measurements were per-formed after R-PIA（0.8-2.0μg）microinjection into ICV and IT with or withoutpretreatment of DPCPX or ***:Microinjection of adenosine agonist R-PIA in doses of 0.8-2.0μg into ICV and IT produced a significant dose-and time-dependent antinociceptive action as reflected by increasing latency times andICV administration of adenosine agonist R-PIA（0.8μg）reducing halothane anes-thetic requirements（by 29%）.The antinociception and reducing halothane re-quirements effected by adenosine agonist R-PIA was abolished by DPCPX ***:ICV and IT administration of adenosine agonist R-PIA pro-duced an antinociceptive effect in a dose-dependent manner and decreased hal-othane MAC with painful stimulation through activation of A1receptor subtype,and the underlying mechanism involves K+channel activation.

Methodology for the Assessment of Burden of Smear-positive Pulmonary TB and Its Infectivity

Objective The study investigated the burden of smear-positive pulmonary TB and itsinfectivity using DALY(disability-adjusted life year) as an indicator. Methods An assumedcohort of 2 000 cases was set up based on the age-specific incidence of 794 newly registeredsmear-positive cases in Beijing in 1994. Prognostic trees and model diagrams of infectivityunder natural history and DOTS(directly observed treatment, short-course) strategy wereestablished according to the epidemiological evidence. Results The results showed that 29. 6%of DALYs would be neglected if the burden caused by the infectivity was not *** DOTS strategy may reduce 97.3. of the number of potential cases infected,92.9% of DALYs related to TB-patients themselves, and 99.9% of DALYs caused by TB’sinfectivity as well.

Aim:To study the effects of long-term,low-dose sex hormone replacement therapy(HRT)on the volume and biochemical changes of the hippocampus in postmeno-pausal women carrying apolipoprotein E(apoE)gene ε3.or ε***:Eighty-three postmenopausal women who had used a low dose of HRT for over 4 yearswere selected as the HRT group,and 99 postmenopausal women with matchedage and education were enrolled as the control *** alleles were analyzedby *** resonance imaging was performed to determine the volume ofthe brain *** magnetic resonance spectroscopy was used todetect the biochemical changes in the anterior cingulate cortex and hippocampusin apoE ε4 and ε3.*** common cognitive tests were used to make anoverall evaluation of cognitive ***:Analysis with the apoE ε4carriers showed that the volume of the hippocampus of the control group weresignificantly lower than those of the HRT *** biochemical analysis showedthat there was an increase of N-acetylaspartate(NAA)/total creatine(tCr)and adecrease of myoinositol(mI)/tCr in the hippocampus of apoE ε4 carriers in theHRT group,compared with the control *** the apoE ε3.carriers,the leastsquares means(LSMEAN)of the HRT group was higher than that of the ***:This study showed that long-term,low dose HRT might bebeneficial for reducing the risk of AD development in vulnerable ***,HRT could increase the LSMEAN of apoE ε3.carriers.

Gene expression profile of amyloid beta protein-injected mouse model for Alzheimer disease

Aim: To investigate the gene expression profile changes in the cerebral cortex of mice injected icv with amyloid beta-protein fragment 25-3. using cDNA microarray. Methods: Balb/c mice were randomly divided into a control group and’ A(3.treated group. The Morris water maze test was performed to detect the effect of A-injection on the learning and memory of mice. Atlas Mouse 1.2 Expression Arrays containing 1176 genes were used to investigate the gene expression pattern of each group. Results: The gene expression profiles showed that 19 genes including TBXl, NF-kB, AP-1/c-Jun, cadherin, integrin, erb-B2, and FGFRl were up-regulated after 2 weeks of icv administration of A; while 12 genes were down-regulated, including NGF, glucose phosphate isomerase 1, AT motif binding factor 1, Na~+/K~+-ATPase, and Akt. Conclusions: The results provide important leads for pursuing a more complete understanding of the molecular events of Ap-injection into mice with Alzheimer disease.

近年来,细菌外膜囊泡(OMVs)在抗肿瘤治疗中展现出的巨大潜力引起了广泛的关注.为探究伤寒沙门菌外膜囊泡(***-OMVs)对人结直肠癌细胞株HT-29增殖的影响及可能的作用机制,通过超速离心法提取不同细菌的OMVs,细胞增殖实验(CCK-8)检测各细菌OMVs对细胞活力的影响;转录组测序(RNA-seq)分析处理后细胞基因表达水平的变化;试剂盒检测铁死亡相关标志物的含量变化;实时荧光定量PCR(qRT-PCR)和蛋白质免疫印迹法(western blot)分别检测相关基因mRNA和蛋白质的表达变化.结果显示,在提取的6种细菌OMVs中,***-OMVs对HT-29细胞的增殖产生了最明显的抑制作用,并且呈现出浓度和时间梯度依赖性;RNA-seq显示HT-29细胞可能发生了铁死亡;***-OMVs作用后,细胞内发生了铁沉积,氧化产物增多,抗氧化剂减少,符合铁死亡生化特征;SAT1是***-OMVs处理后HT-29胞内mRNA表达量变化最大的基因;p53.SAT1-ALOX15是铁死亡的信号通路之一,***-OMVs处理后胞内p53.SAT1、ALOX15的mRNA与蛋白表达均增高.以上结果表明,***-OMVs能够抑制结直肠癌细胞HT-29的增殖,其机制可能与通过p53.SAT1-ALOX15信号通路诱导HT-29发生铁死亡有关.

Autophagy is a major cellular pathway for the degradation of long-lived pro-teins and cytoplasmic organelles in eukaryotic cells.A large number of intracellu-lar/extracellular stimuli,including amino acid starvation and invasion ofmicroorganisms,are able to induce the autophagic response in *** discov-ery of the ATG genes in yeast has greatly advanced our understanding of themolecular mechanisms participating in autophagy and the genes involved in regu-lating the autophagic *** yeast genes have mammalian homologs,suggesting that the basic machinery for autophagy has been evolutionarily con-served along the eukaryotic *** regulation of autophagy is a very com-plex *** signaling pathways,including target of rapamycin(TOR)ormammalian target of rapamycin(mTOR),phosphatidylinositol 3.kinase-I(PI3.-I)/PKB,GTPases,calcium and protein synthesis all play important roles in *** molecular mechanisms and regulation of autophagy are dis-cussed in this review.

This cohort study was performed to explore the influence of intensive care unit(ICU)quality on in-hospital mortality of veno-venous(V-V)extracorporeal membrane oxygenation(ECMO)-supported patients in *** study involved all V-V ECMO-supported patients in 3.8 of 1700 tertiary hospitals from 2017 to 2019,using data from the National Clinical Improvement System and china National Critical Care Quality Control *** quality was assessed by quality control indicators and capacity *** the 2563.V-V ECMO-supported patients in 3.8 hospitals,a significant correlation was found between ECMO-related complications and *** reintubation rate within 48 hours after extubation and the total ICU mortality rate were independent risk factors for higher in-hospital mortality of V-V ECMO-supported patients(cutoff:1.5%and 7.0%;95%confidence interval:1.05–1.48 and 1.04–1.45;odds ratios:1.25 and 1.23.P=0.012 and P=0.015,respectively).Meanwhile,the V-V ECMO center volume was a protective factor(cutoff of≥50 cases within the 3.year study period;95%confidence interval:0.57–0.83.odds ratio:0.69,P=0.0001).The subgroup analysis of 864 patients in 11 high-volume centers further strengthened these ***,ICU quality may play an important role in improving the prognosis of V-V ECMO-supported patients.
