Exploring the N-terminus region： Synthesis, bioactivity and 3D-QSAR of allatostatin analogs as novel insect growth regulators

Allatostatins （ASTs）, a family of insect neuropeptide, can inhibit juvenile hormone （JH） biosynthesis by the corpora allata （CA） in Diploptera punctata, and therefore be regarded as potential leads for the discovery of new insect growth regulators （1GRs）. But several shortcomings, such as their sensitivity to peptidases and high cost, impeded their practical application in pest management. In order to discover new IGRs, one AST analog B1 possessing non-peptide group was discovered with high ability to inhibit JH biosynthesis in vitro （IC50： 0.09 μmol/L） in our previous studies. In the present work, two series of B1 analogs with different substituents on the N-terminus region were designed and synthesized. The result suggested that benzene showed better activity than other heterocycles, and the para-substitution on the benzene was beneficial for activity. Moreover, analogs with logP value over 2.0 exhibited good activity, which indicated the hydrophobicity is important to the bioactivity. Three dimension quantitative structure-activity relationship （3D-QSAR） studies were performed to highlight the structural require- ments of ASTanalogs, which demonstrated introduction of bulkier substituents on the N-terminus would increase the activity. Analog Ⅱ12 （IC50： 0.08 μmol/L） exhibited similar inhibitory activity to the lead B1, but its synthetic route was simpler than B1. Therefore, Ⅱ12 could be used as a new lead compound for the discovery eco-friendly IGRs.

Ti6Al4V alloy‒CoCrMo alloy pair is commonly applied for modular head‒neck interfaces for artificial hip ***,the fretting corrosion damage at this interface seriously restricts its *** work studied the fretting corrosion of Ti6Al4V‒CoCrMo pair in calf serum *** established this material pair’s running condition fretting map(RCFM)regarding load and displacement,and revealed the damage mechanism of this material pair in various fretting regimes,namely partial slip regime(PSR),mixed fretting regime(MFR),and gross slip regime(GSR).The damage mechanism of Ti6Al4V alloy was mainly abrasive wear induced by CoCrMo alloy and *** wear(material transfer)also existed in *** damage mechanism of CoCrMo alloy was mainly abrasive wear induced by metal oxides and tribocorrosion in GSR and MFR,while no apparent damage in ***,a dense composite material layer with high hardness was formed in the middle contacting area in GSR,which reduced the corrosion and wear of Ti alloys and exacerbated damage to Co ***,the ion concentration maps for Ti and Co ions were constructed,which displayed the transition in the amount of released Ti and Co ions under different displacements and loads.

A three-dimensional finite element dynamic simulation platform of the ground source heat pump system(GSHPS)is *** to the outlet temperature of ground heat exchangers(GHEs)required by the code in summer and winter,the calculated minimum buried depth of GHEs meeting the requirements is 60 m,when the number of borehole is *** using the established platform,the annual operation performance and cost of the GSHPS under different buried pipe depths are *** results show that the deeper the buried depth of GHEs is,the better the heat exchange effect of GHEs *** with the GHEs with 60 m buried depth,when the buried depth of GHEs is 65 m,70 m,75 m and 80 m,the average coefficient of performance(COP)of the unit increases by 4.1%,6.3%,7.7%and 8.2%in cooling period and 1.0%,1.6%,1.8%and 1.9%in heating period,*** the performance and initial investment of the GHSPS comprehensively,the optimal buried depth of GHEs is 60 ***,considering the performance the system and the total cost of the system running for 20 years comprehensively,the optimal buried depth of GHEs is 70 m.

High amylose starch can be produced by plants deficient in the function of branching enzymes(BEs).Here we report the production of transgenic cassava(Manihot esculenta Crantz)with starches containing up to 50%amylose due to the constitutive expression of hair-pin ds RNAs targeting the BE1 or BE2 *** BE1-RNAi plant lines(BE1i)and BE2-RNAi plant lines(BE2i)were grown up in the field,but with reduced total biomass *** high amylose content in the storage roots of BE2i plant lines was *** starch granules of BE1i and BE2i plants had similar morphology as wild type(WT),however,the size of BE1i starch granules were bigger than that of *** of amylograms and thermograms of all three sources of storage starches revealed dramatic changes to the pasting properties and a higher melting temperature for BE2i *** chain length distribution analysis showed a slight increase in chains of DP>36 in BE1i lines and a dramatic increase in glucan chains between DP 10-20 and DP>40 in BE2i ***,BE2i starches displayed a B-type X-ray diffraction pattern instead of the A-type pattern found in BE1i and WT ***,cassava BE1 and BE2 function differently in storage root starch biosynthesis.

In the present study,we introduced the H2O2-sensitive thiazolidinone moiety at the 4th amino group of gemcitabine(GEM)to synthesize a new target compound named GEM-ZZQ,and then we confirmed its chemical structure by nuclear magnetic resonance *** further confirmed that GEM-ZZQ had a good chemical stability in different pH solutions in vitro and that it could be activated by H2O2 to release *** studies revealed that the growth inhibition of human normal epithelial cells was weaker by GEM-ZZQ than by GEM treatment and that the inhibition of various lung cancer cell lines by GEM-ZZQ was similar to that of *** the lung cancer cell lines that are resistant to the epidermal growth factor receptor(EGFR)-targeting inhibitor osimertinib,GEM-ZZQ showed less growth inhibition than GEM;however,GEM-ZZQ in combination with cisplatin showed better synergistic effects than GEM in the low-dose *** summary,we provided a new anti-cancer compound GEM-ZZQ for treating lung cancer by modifying the GEM structure.

Decision-making under changing climate has become uncertain and complex,which brings great challenges to traditional risk governance *** areas in China have been highly urbanized with increasing population and physical assets during the past *** on a simplified risk governance framework,a case of Chinese coastal megacity is evaluated with key functions of risk assessment,risk management,and risk *** finds that the case of "Comprehensive Coordination Centre" in Shanghai Fengxian District can play a positive role to improve effeectiveness,efficiency,and accountability of risk governance in the Chinese context,especially a potential to reduce "organized irresponsibility" and "systematic risks".Adaptive and integrated risk governance can be improved from policy experiments and experience learnt from *** paper concludes that the innovation in the existing disaster risk reduction system will promote the ability of adaptation to climate change in the long run.
