MMAE-loaded PLGA nanomedicine with improved biosafety to achieve efficient antitumor treatment

Monomethyl auristatin E(MMAE)is a derivative of the marine peptide Dolastatin 10,which has therapeutic effects against various cancers according to its antimitotic activity in multiple clinical trials.The antibody drug conjugate(ADC)of MMAE is currently used in clinical practice.However,the safety issues of MMAE-based ADC,such as high drug toxicity and poor bioavailability,still exist when using it for anticancer therapy.A sustained release of drug delivery approach should be used to reduce toxicity and achieve sufficient anticancer effects.Herein,PLGA-b-PEG 2000 with excellent biocompatibility and slow degradation ability was adopted to construct MMAE-loaded nanoparticles for safe and effective chemotherapy.The sustained release effect and the immunogenic cell death(ICD)effect of PLGA-MMAE nanoparticles were assessed by in vitro experiments.The PLGA-MMAE nanoparticles effectively accumulated in the tumor through the enhanced permeability and retention(EPR)effect,inducing cell apoptosis and causing a certain degree of immune response.The sustained drug release of PLGA-MMAE improved the bioavailability and effectively reduced the toxicity and development of the tumor compared to the effect of free MMAE or ADC.Overall,this study provides a safe and effective chemotherapeutic approach,as well as a simple and effective synthetic process for MMAE-based nanoparticles,improving their therapeutic efficacy and safety.

Triphenylamine(TPA)-based aggregation-induced emission luminogens(TPA-AIEgens),a type of photoactive material utilizing the typical TPA moiety,has recently attracted increasing attention for the diagnostics and treatment of tumors due to their remarkable chemo-physical performance in optoelectronic research.TPA-AIEgens are distinguished from other photoactive agents by their strong fluorescence,good sensitivity,high signal-to-noise ratio,resistance to photobleaching,and lack of high concentration or aggregation-caused fluoresce quenching effects.In this review,we summarize the current advancements and the biomedical progress of TPA-AIEgens in tumor theranostics.First,the design principles of TPAAIEgens photoactive agents as well as the advanced targeting strategies for nuclei,cell membranes,cell organelle and tumors were introduced,respectively.Next,the applications of TPA-AIEgens in tumor diagnosis and therapeutic techniques were reviewed.Last,the challenges and prospects of TPA-AIEgens for cancer therapy were performed.The given landscape of the TPA-AIEgens hereby is meaningful for the further design and utilization of the novel photoactive material,which could be beneficial for the development of clinic applications.

Analysis of the Impact of Source Region Structures on Seismological Parameter Scanning

By taking moderate-strong earthquakes in South,North and West China as the research subjects and taking into consideration the fault strikes in these regions,this paper selects 8 kinds of seismology indexes with clear physical significance and strong independence to carry out spatial scanning of the parallel,vertical and oblique slip of fault along the fault strike.Based on the size of correlation coefficients between the scanning curve and source region curve we quantitatively analyze the difference between scan results among different slip modes and study the impact of fault strike in different tectonic divisions on scanning results and variation rules of seismological parameters.The results show that not only does the change of spatial parameters have a great influence on seismological parameter scanning,but so does the fault strike in the source region.This paper presents the optimum condition parameters with least spatial influencing scanning scope for different magnitude seismology indexes and analyzes the possible influence of fault strike on seismological parameter scanning results.

目的探讨高分辨率CT对成人麻疹肺炎的诊断价值及优势。方法回顾性分析临床诊断的65例麻疹肺炎患者,所有患者均采用薄层CT进行检查,观察并分析其CT表现。结果薄层CT检查成人麻疹肺炎以间质性病变为主,主要特征为小叶内结节、网格状阴影、磨玻璃影、支气管血管束增粗,将以上特征分三型,本组中Ⅱ型最多见,占40例(61.5%);Ⅰ型14例(21.5%);Ⅲ型11例(17%)。结论高分辨率CT能清晰显示成人麻疹肺炎的主要影像特征,为临床的诊断和鉴别诊断提供价值的依据。

Analysis of therapeutic targets for SARS-CoV-2 and discovery of potential drugs by computational methods

SARS-CoV-2 has caused tens of thousands of infections and more than one thousand deaths.There are currently no registered therapies for treating coronavirus infections.Because of time consuming process of new drug development,drug repositioning may be the only solution to the epidemic of sudden infectious diseases.We systematically analyzed all the proteins encoded by SARS-CoV-2 genes,compared them with proteins from other coronavirnses,predicted their structures,and built 19 structures that could be done by homology modeling.By performing target-based virtual ligand screening,a total of21 targets(including two human targets)were screened against compound libraries including ZINC drug database and our own database of natural products.Structure and screening results of important targets such as 3-chymotrypsin-like protease(3 CLpro),Spike,RNA-dependent RNA polymerase(RdRp),and papain like protease(PLpro)were discussed in detail.In addition,a database of 78 commonly used antiviral drugs including those currently on the market and undergoing clinical trials for SARS-CoV-2 was constructed.Possible targets of these compounds and potential drugs acting on a certain target were predicted.This study will provide new lead compounds and targets for further in vitro and in vivo studies of SARS-CoV-2,new insights for those drugs currently ongoing clinical studies,and also possible new strategies for drug repositioning to treat SARS-CoV-2 infections.

New Activity of the Tanlu Fault Zone in the South of Huaihe to the Nvshanhu Segment since the Late Quaternary

Taking the Huaihe to the Nvshanhu segment of the Tanlu( Tancheng-Lujiang) fault zone as the main research target to explore whether there has been new activity since the late Quaternary,and based on the interpretation of remote sensing images and repeated surface investigations,we excavated trenches at the sections where the tectonic landform is significant,identified and recorded the deformation patterns of the fault and analyzed the activity behavior. Samples of new activity and deformation were collected and oriented slices were ground based on the samples ' original state to make the micro structural analysis and demonstration. All of the above research shows very clear linear tectonic geomorphology along the fault,three trenches across the fault zone all revealed new deformation traces since late Quaternary. The latest stratum dislocated by the fault is the late Quaternary and Holocene. The main slip mode is stick slip,as represented typically by fault scarps,wedge accumulation,the faults and the filled cracks and so on. In general,it shows the characteristics of brittle high-speed deformation and belongs to the prehistoric earthquake ruins. The above understanding was confirmed partially by microscopic analysis. In addition,the similarities and differences and the possible reasons for the characteristics of the latest activities of the Tancheng-Lujiang fault zone in the north and south of the Huaihe River regions are also discussed in this paper.

1901年中国境内最早的工程师组织——中华国际工程学会成立于上海,由在华的外国人创立,旨在促进中国的工程近代化和提升工程师的专业能力。学会出版发行了机关刊物《中华国际工程学会会刊》(1901—1939年),后短暂改版为《中华国际工程学会会报》(1940—1941年),讨论涵盖建筑技术在内的广泛工程议题,记录了学会推进中国城市与建筑走向技术层面现代化的过程。本文借鉴统计学方法,尝试以当代视角梳理学会会刊探讨的建筑技术内容与关切,探讨学会以会刊为载体推进学术研讨、引介新兴钢筋混凝土结构技术、探索地基基础技术的本土化应用、影响公共租界建筑控制活动的过程,以期挖掘其在中国建筑技术近代化过程中的影响与意义,扩展中国近代建筑技术史的历史维度。

A Study on the Prediction Method of Strong Earthquakes Based on Dynamic Seismicity Patterns with Dynamic Implications

Seismic gap method is one of the effective earthquake prediction methods using seismicity patterns. However, this method has some limitations and uncertainty when using it singly in predicting earthquakes. This paper puts forward the prediction method using the dynamic seismicity pattern with dynamic implications. This method considers the formation and evolution of the seismic gap on the basis of plate movement and structural characteristics. Through analysis of 26 cases of earthquakes of MS≥5.0 occurring in East China and South China, this paper obtains the relationship between the main shock with seismic gap and active fault's location, as well as the relationship between the seismic gap and location and strike of active faults. Meanwhile, this paper provides a dynamic explanation of the differences in the formation and evolution patterns of the seismic gap between the two regions, thus providing the physical basis for and reducing the uncertainty of predicting earthquakes using the seismic gap method.
