Construction of human liver cancer vascular endothelium cDNA expression library and screening of the endothelium-associated antigen genes

AIM: To gain tumor endothelium associated antigen genes from human liver cancer vascular endothelial cells (HLCVECs)cDNA expression library, so as to find some new possible targets for the diagnosis and therapy of liver ***: HLCVECs were isolated and purified from a fresh hepatocellular carcinoma tissue sample, and were cultured and proliferated in vitro. A cDNA expression library was constructed with the mRNA extracted from ***-sera were prepared from immunized BALB/c mice through subcutaneous injection with high dose of fixed HLCVECs, and were then tested for their specificity against HLCVECs and angiogenic effects in vitro, such as inhibiting proliferation and inducing apoptosis of tumor endothelial cells, using immunocytochemistry, immunofluorescence,cell cycle analysis and M-I-l- assays, etc. The identified xenogeneic sera from immunized mice were employed to screen the library of HLCVECs by modified serological analyses of recombinant cDNA expression libraries (SEREX).The positive clones were sequenced and analyzed by ***: The primary cDNA library consisted of 2×10^6 recombinants. Thirty-six positive clones were obtained from 6× 10^5 independent dones by immunoscreening. Bio-informatics analysis of cDNA sequences indicated that 36 positive clones represented 18 different genes. Among them, 3 were new genes previously unreported, 2 of which were hypothetical genes. The other 15 were already known ones. Series analysis of gene expression (SAGE) database showed that ERP70,GRP58, GAPDH, SSB, $100A6, BMP-6, DVS27, HSP70 and NAC alpha in these genes were associated with endothelium and angiogenesis, but their effects on HLCVECs were still unclear. NUMERICAL SIMULATIONS OF POLARIZED SCATTERING FROM RANDOM CLUSTERS OF SPATIALLY-ORENTED, NON-SPHERICAL SCATTERS

Employing multiple scattering formulation of T-matrix method, numerical simulations are developed and applied to polarized scattering from random clusters of spatially-oriented, non-spherical particles. Polarized scattering is numerically presented for the functional dependence on particle shape, size, spatial distribution and orientation, and other physical parameters. Numerical calculations of backscattering from randomly clustered particles are well compared with that from independent particles and clusters. It can be seen that spatial distribution and orientation of non-spherical particles can have significant effect on scattering.

Prevention of Regional Ischemic-Reperfused Myocardial Damages in the Isolated Working Rat Hearts by Puerarin

The performances of the isolated working rat hearts perfusedwith modified K-H solution were stable for 90 min of perfusion ***, the contractility and the total cardiac output decreasedsignificantly after 120 min. The acute regional ischemia produced by theligation of left main coronary artery in the isolated working rat hearts wasfollowed by decrease in cardial power production and increase in creatinephosphokinase release. Coronary flow restored to the preligation valueimmediately after the beginning of reperfusion following 30 min regionalischemia, but the cardiac function （as measured by total cardiac output andpower production） did not recover significantly and the creatinephosphokinase release even further increased during 15 min reperfusion.1.4 × 10-4 M puerarin significantly increased coronary flow and did notaffect other mechanical performances of the normal isolated working rathearts. However, 10-5M propranolol as well as 1.4 × 10-4 M puerarinreduced significantly the creatine phosphokinase release from the regionalischemic-reperfused myocardium and accelerated the recovery of the cardiacfunction during reperfusion time in the isolated working rat hearts. Theresults suggested that both puerarin and propranolol have direct benificialeffects on the ischemic-reperfused myocardium.

Network pharmacology-based study on the mechanism of Tangfukang formula(糖复康方) against type 2 diabetes mellitus

OBJECTIVE: To explore the mechanism of Tangfukang formula(糖复康方, TFK) in treating type 2 diabetes mellitus(T2DM). METHODS: We employed network pharmacology combined with experimental validation to explore the potential mechanism of TFK against T2DM. Initially, we filtered bioactive compounds with the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP) and Symptom Mapping(Sym Map), and gathered targets of TFK and T2DM. Subsequently, we constructed a protein-protein interaction(PPI) network, enriched core targets through Gene Ontology(GO) and Kyoto Encyclopedia of Genes and Genomes(KEGG), and adopted molecular docking to study the binding mode of compounds and the signaling pathway. Finally, we employed a KKAy mice model to investigate the effect and mechanism of TFK against T2DM. Biochemical assay, histology assay, and Western blot(WB) were used to assess the mechanism. RESULTS: There were 492 bioactive compounds of TFK screened, and 1226 overlapping targets of TFK against T2DM identified. A compound-T2DM-related target network with 997 nodes and 4439 edges was constructed. KEGG enrichment analysis identified some core pathways related to T2DM, including adenosine 5-monophosphate-activated protein kinase(AMPK) signaling pathway. Molecular docking study revealed that compounds of TFK, including citric acid, could bind to the active pocket of AMPK crystal structure with free binding energy of -4.8,-8 and -7.9, respectively. Animal experiments indicated that TFK decreased body weight, fasting blood glucose, fasting serum insulin, homeostasis model of insulin resistance, glycosylated serum protein, total cholesterol, triglyceride, and low-density lipoprotein cholesterol, and improve oral glucose tolerance test results. TFK reduced steatosis in liver tissue, and infiltration of inflammatory cells, and protected liver cells to a certain extent. Background:The role of vitamin D in ankylosing spondylitis(AS) is largely *** paper aims to examine the association between serum vitamin D levels and susceptibility and disease activity of ***:We searched the relevant literatures in Pub Med,Elsevier Science Direct,Chinese Biomedical Database(CBM),Chinese National Knowledge Infrastructure(CNKI) and Wanfang(Chinese) Database published before June *** independent case–control studies with a total of 533 AS patients and 478 matching controls were selected into this *** mean differences(SMDs) with 95% confidence intervals(CIs)were used to assess the levels of serum vitamin D,parathyroid hormone(PTH),serum calcium and alkaline phosphatase(ALP) in cases and controls,*** coefficients(CORs) have been performed to value the correlationship between vitamin D and disease activity(erythrocyte sedimentation rate(ESR) and Creactive protein(CRP) and Bath Ankylosing Spondylitis Disease Activity Index(BASDAI)) of AS ***:Meta-analysis results suggested that vitamin D may play a protective role in AS(for total vitamin D:SMD =-0.71,P< 0.001;for 25OHD:SMD =-0.66,P = 0.002;for 1,25OHD:SMD =-0.72,P = 0.19).Differences in PTH and serum calcium levels were not significant in AS(SMD =-0.10,P = 0.67;SMD = 0.12,P =0.17 respectively),while ALP was associated with AS susceptibility(SMD = 0.20,P = 0.04).The relationship between serum vitamin D levels and disease activity was statistically significant except for 25 OHD versus(vs.) CRP or BASDAI(for CRP vs.25OHD:COR =-0.22,P = 0.08;for BASDAI vs.25OHD:COR =-0.20,P = 0.06,respectively).Conclusion:The higher levels of serum vitamin D were associated with a decreased risk of AS,and showed an inverse relationship with AS activity.

配网不停电作业已成为保障电力系统安全、可靠、稳定运行的重要检修手段。在旁路变压器与待检修变压器短时并联过程中可能出现损害作业设备的环流,为解决此类工程问题,对旁路变压器并联运行的稳态过程进行理论分析,并在电磁暂态仿真软件中搭建10 kV配电网变压器并联模型。仿真结果表明,影响并联运行变压器是否产生环流的主要因素是变压器联结组别与变比差异,变压器容量及短路阻抗百分比为次要因素。进一步给出了保证变压器稳态运行的并联方案,为不停电作业旁路变压器的参数选型提供了理论基础。

钛合金因具有高比强度、低密度、耐腐蚀性、抗氧化性、高温稳定性以及低中子截面等特点,逐渐被用作船舶和空间核动力装置的关键部件。为提高钛合金抗辐照性能,推进钛合金在核工程领域广泛应用,不少研究人员在钛合金辐照效应等关键问题研究上做出了很大努力。本文回顾了钛及钛合金在核领域的发展与辐照效应研究,全面综述了不同粒子辐照(中子、离子等)下,多种先进钛合金中辐照缺陷演变及相互作用机制,还总结了服役条件(温度、应力、辐照)对钛合金的硬度、拉伸、疲劳以及蠕变等力学性能的影响规律。最后,基于目前核用钛合金研究现状,展望了未来钛合金辐照效应的研究方向和改善抗辐照性能的发展趋势。

采用阴极电弧离子镀技术在退火态合金钢表面沉积TiN和TiAlN涂层,使用扫描电镜和X射线衍射仪分析了所得涂层的结构、成分与物相组成,利用显微维氏硬度计测试了涂层的硬度,通过压痕法评价了涂层的结合性能,并采用往复式球-盘摩擦磨损试验测试了涂层的耐磨性能。结果表明,TiN和TiAlN两种涂层表面均平整,但TiAlN涂层表面有明显“液滴”缺陷。退火态合金钢表面TiN和TiAlN涂层均发生了择优生长,其中TiN涂层择优生长的晶面为(111),而TiAlN涂层择优生长的晶面为(200)。沉积TiN和TiAlN涂层后,退火态合金钢的表面硬度分别提高3倍和6倍以上。尽管TiAlN涂层在硬度及摩擦因数上均优于TiN涂层,但由于TiAlN涂层与退火态合金钢结合性能差,导致其在磨损过程中发生剥落,使合金钢耐磨性降低。相比之下,TiN涂层为退火态合金钢提供了更为可靠的保护,显著提高了其表面的耐磨性。

INFERENCE FOR REPEATED MEASURES MODELS UNDER HETEROSCEDASTICITY

This paper considers a widely used mixed effects model in repeated measures under het- eroscedasticity. Hypotheses of the equality of the fixed effects and the simultaneous confidence intervals for all pair-wise differences are discussed. A generalized F-test has been proposed to test the equality of the fixed effects in the model, but simulation results for evaluating its performance have not been shown in the literature. Moreover, the generalized F-test cannot be used to deduce the simultaneous confidence intervals for all pair-wise differences of the fixed effects. The authors propose two new p-values to test the hypotheses of equality of the fixed effects and simultaneous confidence intervals of the differences of the effects based on the generalized pivotal quantities derived in this paper. The authors also compare the empirical performances of the proposed tests and the generalized F-test. The type I error rates and powers of these tests are evaluated using the Monte Carlo simulation. The simulation studies show that the generalized F-test does not perform well in terms of type I error rate under various sample size and parameter combinations. However, the type I error probabilities of the proposed tests are always close to the nominal value. It can also be seen that the simultaneous confidence intervals perform well.

A Bijective Map on Normal Fusion Subsystems

Let F be a saturated fusion system over a finite p-group S, and let Z ≤ Zn （F）, which is strongly closed in S with respect to F. In this paper, we prove that there is a natural bijection from the set of normal subsystems of 7 containing Z to the set of normal subsystems of the factor system F＋ =F/Z. This generalizes a result of Aschbacher.
