Background The reliance on a solitary linear reference genome has imposed a significant constraint on our compre-hensive understanding of genetic variation in *** constraint is particularly pronounced for non-reference sequences(NRSs),which have not been extensively *** In this study,we constructed a pig pangenome graph using 2. pig assemblies and identified 2.,831 NRSs with a total length of 105 *** findings revealed that NRSs were more prevalent in breeds exhibiting greater genetic divergence from the reference ***,we observed that NRSs were rarely found within coding sequences,while NRS insertions were enriched in immune-related Gene Ontology ***,our investigation also unveiled a close association between novel genes and the immune capacity of *** observed substantial differences in terms of frequencies of NRSs between Eastern and Western pigs,and the heat-resistant pigs exhibited a substantial number of NRS insertions in an 11.6 Mb interval on chromosome ***,we discovered a 665 bp insertion in the fourth intron of the TNFRSF19 gene that may be associated with the ability of heat tolerance in South-ern Chinese *** Our findings demonstrate the potential of a graph genome approach to reveal important functional features of NRSs in pig populations.

Female hormone detection,particularly non-invasive monitoring progesterone(P4)levels in body fluids,plays a critical role in female health management and disease ***,the challenge still exists because of the ultralow abundance of P4(<100 pmol/L)in sweat and saliva,necessitating highly sensitive methods for wearable ***,we present a simple sandpaper-templated stretchable immunosensing electrode designed for ultra-sensitive detection of P4 at sub-picomolar level.A molding technique is employed to replicate the sandpaper textures to provide a microstructured elastomeric substrate for electrode *** microstructured surface coated with poly(3,4-ethylenedioxythiophene):poly(4-styrenesulfonate)(PEDOT:PSS)/LiTFSI[LiTFSI=lithium bis(trifluoromethanesulfonyl)imide]provided a stretchable polymer electrode with high conductivity,and further decoration with gold nanoparticles(AuNPs)enabled the immunosensing for P4 by electrochemical impedance spectroscopy(EIS)*** adjusting the AuNPs deposition conditions,ultrasensitive detection of P4 is realized with a low limit of detection(LOD)of ca.10 fmol/L and a tunable dynamic range up toµmol/***,the stretchable electrode exhibits stable electrochemical performance,enabling the detection of P4 at sub-picomolar levels even under mechanical strain of 30%.This innovative electrochemical sensor holds significant promise for non-invasive,on-site monitoring of P4 levels in healthcare,as well as hormone detection in food safety and environment surveillance.

Non-invasive bioelectronics,especially organic electrochemical transistors(OECTs),have drawn extensive attentions of academical and medical communities by virtue of their efficient bio-electronic interfacing,water-involved ionic transport,excellent ionic-electronic coupling,ultralow power consumption,wide detectable range,and outstanding detection *** structure diversity,low-temperature solution processability,facile bio/chemical functionalization,and excellent biocompatibility of organic mixed ionic-electronic conductors(OMIECs)render OECTs particularly suitable for non-invasive or minimally invasive healthcare analytical ***,we comprehensively review recent advances of the non-invasive analytical healthcare applications based on OECTs,especially on the detection of biomarkers or metabolites in the excretory biofluids,as well as the recording of electrophysiological signals.A brief introduction of OECT and its comparison with other organic thin-film transistors upon device configuration and working mechanism are firstly ***-of-the-art non-invasive OECT-based biosensors are summarized on their detection of ionic and molecular biomarkers,following with circuit design strategies of OECTs for real-time and in-situ electrophysiological recording from skin *** conclusion,remaining barriers and future challenges of non-invasive OECT-based bioelectronics towards lower detection limit,more accurate quantitative relationship between analyte concentrations and measured parameters,more intimate device-tissue interface,and long-term operation stability are deeply analyzed with a critical outlook.

Understanding interactions between viruses and their hosts is conducive to enabling better application of viruses as biocontrol *** viruses carried by parasitic wasps enhance the parasitic efficiency of wasp-larvae by protecting them against the immune system of their Lepidopteran ***,the relationship between prey pests and viruses found in predatory natural enemies remains ***,we report the interaction between Arma chinensis virus-1(AcV-1),originally isolated from a predatory natural enemy,Arma chinensis(Hemiptera:Pentatomidae),and one of its prey species,Spodoptera frugiperda(Lepidoptera:Noctuidae).The results showed that the AcV-1 virus appeared harmful to the novel host *** by inhibiting larval diet consumption and increasing pupal ***,sequencing data indicated that the virus altered the gene expression profiles of *** analysis showed that the proteasome and phagosome pathways related to protein degradation and immune response were significantly *** the expression levels of digestive enzyme genes did not change significantly,the total protease activity of AcV-1 virus-positive individuals was significantly decreased,suggesting that the virus inhibited diet consumption of *** via the down-regulation of digestive enzyme *** results indicate that a virus initially isolated in a predatory natural enemy can decrease the fitness of its prey *** virus was found to impact the host proteasome and phagosome pathways related to protein degradation and immunity,providing a potential mechanism to enhance controlling efficiency.

Wound repair is a complex physiological process that often leads to bacterial infections,which significantly threaten human ***,developing wound-healing materials that promote healing and prevent bacterial infections is *** this study,the coordination interaction between sulfhydryl groups on dithiothreitol(DTT)and MoS_(2.nanosheets is investigated to synthesize a MoS_(2.-DTT nanozyme with photothermal properties and an improved free-radical scavenging ***-bond-modified hyaluronic acid is used as a monomer and is cross-linked with a PF12.-DA *** is prepared in coordination with MoS_(2.-DTT as the functional *** hydrogel exhibits antioxidant and antibacterial properties,attributed to the catalytic activity of catalase-like enzymes and photothermal *** the near-infrared(NIR),it exhibits potent antibacterial effects against gram-positive(Staphylococcus aureus)and gram-negative bacteria(Escherichia coli),achieving bactericidal rates of 99.76%and 99.42.,***,the hydrogel exhibits remarkable reactive oxygen species scavenging and antioxidant capabilities,effectively countering oxidative stress in L92. ***,in an animal model,wounds treated with the PHMoD(2.0)and NIR laser heal the fastest,sealing completely within 10 *** results indicate the unique biocompatibility and bifunctionality of the PHMoD,which make it a promising material for wound-healing applications.

Subway tunnels often suffer from surface pathologies such as cracks,corrosion,fractures,peeling,water and sand infiltration,and sudden hazards caused by foreign object *** a mobile visual pathology sensing system at the front end of operating trains is a critical measure to ensure subway *** leakage as the typical pathology,a tunnel pathology automatic visual detection method based on Deeplabv3+(ASTPDS)was proposed to achieve automatic and high-precision detection and pixel-level morphology extraction of *** with similar methods,this approach showed significant advantages and achieved a detection accuracy of 93.12.,surpassing FCN and ***,it also exceeded the recall rates for detecting leaks of FCN and U-Net by 8.33%and 8.19%,respectively.
