Carbon dots/graphite carbon nitride(CDs/g-C_(3)N_(4)),a novel composite photocatalyst,has shown great po-tential for applications in energy regeneration and environmental remediation owing to its following advantages:metal-free,low cost,easily tunable,and excellent photocatalytic performance.Hence,we reviewed the development of synthetic strategies,photocatalytic enhancement mechanisms,and pho-tocatalytic applications of CDs/g-C_(3)N_(4) in this study.First,the three composite strategies of CDs and g-C_(3)N_(4)-self-assembly,solvothermal,and calcination polymerization are outlined,and their advantages and disadvantages are described in detail.Moreover,the photocatalytic enhancement mechanism of the com-posite strategies was elucidated according to the variation trends of CDs/g-C_(3)N_(4) band structure,electronic properties,light absorption range,and interfacial charge transfer.Then,the applications of CDs/g-C_(3)N_(4) in hydrogen evolution,pollutant degradation,CO_(2) reduction,and bacterial disinfection in recent years are reviewed systematically.Finally,the current obstacles and future research directions of CDs/g-C_(3)N_(4) are discussed from the perspective of preparation technology and practical applications,respectively.

SARS coronavirus papain-like protease inhibits the type I interferon signaling pathway through interaction with the STING-TRAF3- TBK1 complex

SARS coronavirus （SARS-CoV） develops an antagonis- tic mechanism by which to evade the antiviral activities of interferon （IFN）. Previous studies suggested that SARS-CoV papain-like protease （PLpro） inhibits activa- tion of the IRF3 pathway, which would normally elicit a robust IFN response, but the mechanism（s） used by SARS PLpro to inhibit activation of the IRF3 pathway is not fully known. In this study, we uncovered a novel mechanism that may explain how SARS PLpro effi- ciently inhibits activation of the IRF3 pathway. We found that expression of the membrane-anchored PLpro domain （PLpro-TM） from SARS-CoV inhibits STING/ TBKl/IKKE-mediated activation of type I IFNs and dis- rupts the phosphorylation and dimerization of IRF3, which are activated by STING and TBKI. Meanwhile, we showed that PLpro-TM physically interacts with TRAF3, TBK1, IKK~, STING, and IRF3, the key components that assemble the STING-TRAF3-TBK1 complex for activa- tion of IFN expression. However, the interaction between the components in STING-TRAF3-TBK1 complex is dis- rupted by PLpro-TM. Furthermore, SARS PLpro-TM reduces the levels of ubiquitinated forms of RIG-I, STING, TRAF3, TBK1, and IRF3 in the STING-TRAF3- TBK1 complex. These results collectively point to a new mechanism used by SARS-CoV through which PLpro negatively regulates IRF3 activation by interaction withSTING-TRAF3-TBK1 complex, yielding a SARS-CoV countermeasure against host innate immunity.

关于数字媒介与网红地点的塑造有诸多研究,但深入现场探究技术与人的互动,会发现这一议题仍有丰富的研究拓展空间。在对云南斗南花卉市场的田野观察中发现,短视频多元实践主体通过将花从实体物转变为虚拟物的打造过程,实现人、物和地点的多重连接与互动,最终达成斗南的网红化与意义的建构。透过物的视角理解网红地点生产的新意和重要价值在于:实体空间中的实存之物向虚拟空间中象征物的转化过程,是技术实践者将自身文化框架和情感表达注入人地关系的过程。这为我们理解人、物、技术与信息流动之间的复杂关系提供新的视角。

Coronavirus membrane-associated papain-like proteases induce autophagy through interacting with Beclinl to negatively regulate antiviral innate immunity

Autophagy plays important roles in modulating viral replication and antiviral immune response. Coronavirus infection is associated with the autophagic process, however, little is known about the mechanisms of autophagy induction and its contribution to coronavirus regulation of host innate responses. Here, we show that the membrane-associated papain-like protease PLP2 （PLP2-TM） of coronaviruses acts as a novel autophagy- inducing protein. Intriguingly, PLP2-TM induces incom- plete autophagy process by increasing the accumula- tion of autophagosomes but blocking the fusion of autophagosomes with lysosomes. Furthermore, PLP2- TM interacts with the key autophagy regulators, LC3 and Beclinl, and promotes Beclinl interaction with STING, the key regulator for antiviral IFN signaling. Finally, knockdown of Beclinl partially reverses PLP2-TM＇s inhibitory effect on innate immunity which resulting in decreased coronavirus replication. These results sug- gested that coronavirus papain-like protease induces incomplete autophagy by interacting with Beclinl, which in turn modulates coronavirus replication and antiviral innate immunity.

Magnetic Fields Affect Alcoholic Liver Disease by Liver Cell Oxidative Stress and Proliferation Regulation

It is well known that alcohol consumption leads to substantially increased free radical levels and health risks,which lacks effective treatment besides alcohol abstinence.Here,we compared different static magnetic field(SMF)settings and found that a downward quasi-uniform SMF of~0.1 to 0.2 T could effectively alleviate alcohol-induced liver damage and lipid accumulation and improve hepatic function.SMFs of two different directions can reduce the inflammation,reactive oxygen species levels,and oxidative stress in the liver,while the downward SMF had more obvious effects.Moreover,we found that the upward direction SMF of~0.1 to 0.2 T could inhibit DNA synthesis and regeneration in hepatocytes,which caused detrimental effects on the lifespan of"heavy drinking"mice.In contrast,the downward SMF prolongs survival of"heavy drinking"mice.On one hand,our study shows that~0.1 to 0.2 T moderate quasi-uniform SMFs with a downward direction have great promises to be developed into a physical method to reduce alcohol-induced liver damage;on the other hand,although the internationally recognized upper limit for SMF public exposure is 0.4 T,people should also pay extra attention to SMF strength,direction,and inhomogeneity that could generate harmful effects on specific severe pathological conditions.

1 T moderate intensity static magnetic field affects Akt/mTOR pathway and increases the antitumor efficacy of m TOR inhibitors in CNE-2Z cells

Static magnetic field(SMF) has been known to affect cell proliferation in a cell-type-dependent manner,while the mechanism still remains unclear. We found that 1T moderate intensity SMF inhibits cell proliferation of nasopharyngeal carcinoma CNE-2Z cells and the Akt/m TOR signaling pathway, which is upregulated in many cancers. m TOR inhibitors are potential chemodrugs, but their clinical effects are limited by the feedback reactivation of other signaling components such as EGFR and Akt.We showed that 1 T SMF increases the antitumor efficacy of m TOR inhibitor Torin 2. In addition, 1 T SMF increases the inhibition efficiency on m TOR substrates phosphorylation and represses the m TOR inhibitor-induced feedback reactivation of EGFR and Akt. Our study not only demonstrates that m TOR/Akt pathway is one of the molecular targets of SMFs in cells, but also reveals the clinical potentials of combinations of m TOR inhibitors and SMFs in cancer treatment.

目的探究基于磁共振第2.1版前列腺影像报告和数据系统(PI-RADS V2.1)评分联合游离前列腺特异性抗原(PSA)/总PSA在前列腺癌早期诊断中的价值。方法筛选2018年6月至2022年6月江苏省南通市第二人民医院收诊的82例早期前列腺癌患者为癌变组;另选取同期收诊的82例良性前列腺增生症患者为良性组。收集两组血清PSA检测结果、磁共振扫描结果等,分析早期前列腺癌发生的影响因素;评估游离PSA/总PSA和PI-RADS V2.1评分对早期前列腺癌的诊断效能。结果癌变组总PSA水平高于良性组,游离PSA/总PSA低于良性组(P<0.05)。癌变组移行带PI-RADS V2.1评分高于良性组(P<0.05)。总PSA、游离PSA/总PSA、移行带PI-RADS V2.1评分是早期前列腺癌发生的影响因素(P<0.05)。游离PSA/总PSA、移行带PI-RADS V2.1评分联合诊断早期前列腺癌的曲线下面积高于各指标单独检测。结论游离PSA/总PSA、PI-RADS V2.1可用于辅助诊断早期前列腺癌,且诊断效能良好。

目的研究肾阳虚慢性阻塞性肺疾病(COPD)合并骨质疏松大鼠肠道菌群、骨保护素(OPG)/核因子κB受体活化因子配体(RANKL)/核因子κB受体活化因子(RANK)通路变化及益生菌的治疗作用。方法雄性SD大鼠(8~10周龄、体重180~200 g)随机分为对照组、模型组、益生菌组,每组8只。模型组和益生菌组大鼠进行肾阳虚COPD合并骨质疏松建模,建模成功后模型组给予生理盐水灌胃、益生菌组给予益生菌[0.9×10^(9)CFU/(g•kg•d)]灌胃,持续28 d。检测肺功能用力肺活量(FVC)、第0.2秒用力呼气量(Fev 0.2),股骨的骨密度(BMD),血清中干扰素-γ(IFN-γ)、白介素-4(IL-4)、碱性磷酸酶(ALP)、Ⅰ型前胶原氨基端原肽(PINP)、抗酒石酸酸性磷酸酶(TRAP)、Ⅰ型胶原羧基端肽β特殊序列(β-CTX)的水平,肺组织和骨组织中OPG、RANK、RANKL的mRNA表达水平。结果模型组的FVC、Fev 0.2、BMD、血清ALP和PINP水平以及肠道菌群丰富度的Ace指数、Chao1指数、Shannon指数、肺组织和骨组织中OPG的mRNA表达水平均低于对照组,血清IFN-γ/IL-4比例、IFN-γ水平、TRAP水平、β-CTX水平、肺组织和骨组织中RANK与RANKL的mRNA表达水平均高于对照组(均P<0.05);益生菌组的FVC、Fev 0.2、BMD、血清ALP和PINP水平以及肠道菌群丰富度的Ace指数、Chao1指数、Shannon指数、肺组织和骨组织中OPG的mRNA表达水平均高于模型组,血清IFN-γ/IL-4比例、IFN-γ水平、TRAP水平、β-CTX水平、肺组织和骨组织中RANK与RANKL的mRNA表达水平均低于模型组(均P<0.05)。结论肾阳虚COPD合并骨质疏松大鼠存在肠道菌群失调和OPG/RANKL/RANK通路异常,补充益生菌显著纠正肠道菌群失调、OPG/RANKL/RANK通路异常并改善肺功能和骨密度。

建立一种3-氨基-2-恶唑烷酮(3-amino-2-oxazolidinone,AOZ)夹心免疫检测方法。通过1,6-己二醇连接2-硝基-4-羧基苯甲醛和生物素合成针对AOZ的新型衍生试剂。在样品前处理时加入衍生试剂可对AOZ进行衍生,衍生产率为89%。在酶标板上包被抗AOZ单克隆抗体并以辣根过氧化物酶标记的亲和素或抗生物素抗体作为第二结合物可实现AOZ的夹心酶联免疫吸附检测(enzyme-linked immunosorbnent assay,ELISA)。从实际应用的角度,得到双抗夹心和抗体-亲和素夹心模式下两个表位的极限距离,分别为12Å和13Å,理想距离分别为16Å和17Å。在双抗夹心和抗体-亲和素夹心模式下的检出限分别达到1.8 pg/mL和0.8 pg/mL(以AOZ质量浓度计),相对于竞争ELISA,其灵敏度最高提高了25倍。平均回收率为73%~85%,平均相对标准偏差为9.0%。
