Standard treatment options for breast cancer include surgery, chemotherapy, radiation, and targeted therapies, such as adjuvant hormonal therapy and monoclonal antibodies. Recently, the recognition that chronic inflammation in the tumor microenvironment promotes tumor growth and survival during different stages of breast cancer development has led to the development of novel immunotherapies. Several immunotherapeutic strategies have been studied both preclinically and clinically and already have been shown to enhance the efficacy of conven- tional treatment modalities. Therefore, therapies targeting the immune system may represent a promising next-generation approach for the treatment of breast cancers. This review will discuss recent findings that elucidate the roles of suppressive immune cells and proinflammatory cytokines and chemokines in the tumor-promoting microen- vironment, and the most current immunotherapeutic strategies in breast cancer.

An approach to optimize the settings of actuated signals

Traffic-actuated signal employs relatively complex control logic to regulate traffic flow. Introduction of control variables into the traffic-actuated system contributes to system operational flexibility and complexity, and also complicates the system with uncertainties. The paper proposes two tentative methods to optimize the actuated signal parameters： basic requirements of controller parameters and analytical model, and macroscopic computer simulation. It is concluded that when the actuated signal operates within the volume/capacity range of 0.4 to 0.6, it will create the most significant benefits; the research suggests that minimum green time in the main street shall be set long enough to meet the required demand, preferably at the 60% of the main street capacity. In order to ensure less control delay in a semi-actuated intersection, relatively small values of vehicle extension （e.g., 2.5 s） and maximum green time are recommended to be assigned to the less important street.

Macrophages are among the most abundant immune cells in the tumor microenvironment, where they are known as tumor-associated macrophages （TAMs）. Substantial evidence indicates that TAMs generally play pro＇tumoral roles in the primary as well as metastatic sites （Noy and Pollard, 2014）. TAMs are also known to interfere with several tumor therapeutic modalities, such as chemotherapy, irradiation, and immunotherapy. In general, depletion of M2-1ike TAMs or reprogramming them into M l-like pheno- type enhances the efficacy of these therapies （Rolny et al., 2011; de Palma and Lewis, 2013; Germano et al., 2013）. Chemotherapy has been shown to increase macrophage infiltration in human breast cancer tis- sues and the ratio of CD68/CD8 expression predicts the patient response to the treatment （DeNardo et al., 2011）. TAMs have been known to promote breast cancer progression and increase therapy resistance through mechanisms such as the enhancement of angiogenesis, tumor cell proliferation and invasive- ness by producing growth factors such as vascular endothelial growth factor （VEGF）, epidermal growth factor （EGF） and FGF2 （jiang, 2014）.

Effect of EGF1 peptides in directing nanoparticles to thrombi

In order to examine the effect of EGF1 peptides in directing nanoparticles to thrombi,the synthesized EGF1 peptide was conjugated to poly (ethyleneglycol)-poly (lactide) (PEG-PLA) nanoparticles (NP) to form EGF1-NP. A lipophilic fluorescent dye,coumarin-6,was incorporated into EGF1-NP so as to detect its loading and release capacity. The binding ability of EGF1-NP with TF-expressing cells was shown to be significantly higher than that of the non-conjugated NP. Following an intravenous administration,fluorescence was distributed along the vessel wall of the thrombosis regions in the model rats injected with coumarin-6-loaded EGF1-NP. The in vitro and in vivo results suggest that EGF1-NP is a promising drug delivery system for targeting cerebral thrombi.

Small interfering RNA delivery to the neurons near the amyloid plaques for improved treatment of Alzheimer’s disease

Gene therapy represents a promising treatment for the Alzheimer’s disease(AD). However,gene delivery specific to brain lesions through systemic administration remains big challenge. In our previous work, we have developed an siRNA nanocomplex able to be specifically delivered to the amyloid plaques through surface modification with both CGN peptide for the blood–brain barrier(BBB)penetration and QSH peptide for β-amyloid binding. But, whether the as-designed nanocomplex could indeed improve the gene accumulation in the impaired neuron cells and ameliorate AD-associated symptoms remains further study. Herein, we prepared the nanocomplexes with an siRNA against β-site amyloid precursor protein-cleaving enzyme 1(BACE1), the rate-limiting enzyme of Aβ production, as the therapeutic siRNA of AD. The nanocomplexes exhibited high distribution in the Aβ deposits-enriched hippocampus, especially in the neurons near the amyloid plaques after intravenous administration. In APP/PS1 transgenic mice, the nanocomplexes down-regulated BACE1 in both mRNA and protein levels,as well as Aβ and amyloid plaques to the level of wild-type mice. Moreover, the nanocomplexes significantly increased the level of synaptophysin and rescued memory loss of the AD transgenic mice without hematological or histological toxicity. Taken together, this work presented direct evidences that the design of precise gene delivery to the AD lesions markedly improves the therapeutic outcome.

为研究强制换道及冲突点分布对高速公路临时瓶颈交通流的影响,在NS(Na Sch)模型和STCA(symmetric two-lane cellular automata)模型的基础上,引入强制换道规则,根据瓶颈口上游驾驶员心理状态的变化,建立高速公路瓶颈交通流模型.在开口边界条件下,针对不同的安全换道概率、强制换道概率、冲突点距离和冲突区间长度参数,模拟得到瓶颈交通流量和换道频率与车辆到达率的关系.仿真结果表明,安全换道行为对系统流量影响小;强制换道行为是降低瓶颈系统最大流量的主要因素,当安全换道概率为0.5时,强制换道概率从0.0增加至0.1,最大流量下降了17%;冲突点距离的增加缓解了交通拥堵程度,当冲突点距离从1 cell增加至4 cell时,临界车辆到达率上升了4%;冲突区间长度对交通事故风险的影响较大,最大强制换道频率随冲突区间长度的增加而增加.

随机性是公交出行需求的重要特性.随机需求下的公交系统中,不同风险态度(中立风险、规避风险和寻求风险)的决策者会选择不同的设计方案,这在多阶段公交系统走廊设计中更为明显.为研究不同决策风险态度下随机特性对公交服务设计的影响,首先,基于均值-方差理论,提出不同风险态度下的决策方法,并构建对应的多阶段公交走廊设计模型,同时优化不同阶段的公交线路长度、站点选址和发车间隔;之后,利用局部分解法和最优化理论,得出模型的解析解;最后,对不同设计策略(全覆盖设计、单阶段和多阶段的公交线路非全覆盖设计)和风险决策态度下的公交走廊设计进行对比分析.结果表明:1)公交线路多阶段设计具有比全覆盖设计和单阶段设计更低的期望系统成本,以及比单阶段设计更稳定的方案;2)在不同设计策略中,决策风险态度对公交系统的参数配置(即公交线路长度、站点分布和发车间隔)有不同的影响,例如规避风险态度决策者在全覆盖设计中追求更密集的站点分布,而在单阶段设计中则会追求相对更低的站点密度,在多阶段设计中这就更为复杂.该研究从政府管理者和运营商的角度,提供基于需求随机增长下公交系统多阶段设计的风险投资决策方法.
