Photoprotective Effects of D1 Protein Turnover and the Lutein Cycle on Three Ephemeral Plants under Heat Stress

To clarify the characteristics of photoinhibition and the primary defense mechanisms of ephemeral plant leaves against photodestruction under high temperature stress,inhibitors and the technology to determine chlorophyll fluorescence were used to explore the protective effects of D1 protein turnover and the lutein cycle in the high temperature stress of the leaves of three ephemeral plants.The results showed that the maximum light conversion efficiency(Fv/Fm)of the ephemeral plant leaves decreased,and the initial fluorescence(Fo)increased under 35℃±1℃ heat stress for 1-4 h or on sunny days in the summer.Both Fv/Fm and Fo could be recovered after 8 h of darkness or afternoon weakening of the external temperature.Streptomycin sulfate(SM)or dithiothreitol(DTT)accelerated the decrease of Fv/Fm and the photochemical quenching coefficient(qP)in the leaves of three ephemeral plants at high temperature,and the decrease was greater in the SM than in the DTT treatment.When the high temperature stress was prolonged,the Y(II)values of light energy distribution parameters of PSII decreased,and the Y(NPQ)and Y(NO)values increased gradually in all the treatment groups of the three ephemeral plants.The results showed that the leaves of the three ephemeral plants had their own highly advanced mechanisms to protect against photodamage,which inhibited the turnover of D1 protein and xanthophyll cycle.This can damage the PSII reaction center in the leaves of the three ephemeral plants under high temperature.The protective effect of D1 protein turnover on heat stress in Erodium oxyrrhynchum and Senecio subdentatus was greater than that of the lutein cycle,while the protective effect of lutein cycle was greater than that of D1 protein turnover in Heliotropium acutiflorum subjected to heat damage.

传统最小二乘法(LS)用于锂离子电池模型在线参数辨识精度低,通过带遗忘因子递推最小二乘算法能够有效地提高辨识精度,但固定的遗忘因子影响模型动态特性。遗忘因子的自适应处理能提高算法对动态系统的参数辨识能力,而目前的自适应方法容易忽略模型参数的稳定性,同时方法待定系数范围较大且难以确认。为了得到高精度且稳定性良好的模型参数,该文设计了一种精度和稳定性兼优且更简单的自适应遗忘因子递推最小二乘(AFFRLS)改进方法,并与其他AFFRLS、可变遗忘因子递推最小二乘(VFFRLS)进行仿真对比分析。结果表明,改进的AFFRLS能够在模型精度和参数稳定性取得更好的平衡,且对不同的在线工况具有良好的适用性。

为解决低碳铝镇静钢汽车板在磷化工艺下限条件下磷化膜结晶粗大及覆盖率不足问题,采用扫描电镜(SEM)、表面轮廓仪、辉光光谱仪(GDS)及透射电镜(TEM)研究了低碳铝镇静钢汽车板磷化性能关键影响因素,并提出了改善对策。结果表明:当磷化工艺处于下限条件即促进剂浓度、总酸浓度较小,而游离酸浓度较大时,钢板与磷化液界面磷酸盐不易饱和,磷化形核难度加大,导致磷化结晶粗大及覆盖率不足等问题。但当钢板表面存在均匀分布且数量较多的小尺寸氧化物颗粒时,氧化物与铁基体能够形成大量原电池,提升局部磷化酸蚀及沉积反应速度,进而改善磷化性能。通过对低碳铝镇静钢碳含量及退火快冷段、过时效段温度进行控制,成功在钢板表面析出尺寸较小且均分分布的碳化物。由于碳化物与铁基体之间存在电位差,同样增加了钢板表面原电池数量,促进了磷化形核,所获得的磷化膜晶粒尺寸在5μm以下,覆盖率为100%。

Oasis cold island effect and its influence on air temperature: a case study of Tarim Basin, Northwest China

Oasis effect can improve the regional climate and habitability of an arid region. In this study, we explored the cold island effects of oases distributed along the edge of Tarim Basin by analyzing the oasis cold island effect （OCIE） intensity, spatial-temporal variation of OCIE, factors influencing the OCIE and impacts of OCIE on air temperature using geographical statistics and GIS methods based on the MODIS land surface temperature, land use/cover change （LUCC） and observed air temperature data. Results showed that all the oases in the Tarim Basin exhibited cold island effects, with the OCIE intensity highest in summer （-9.08℃）, followed by autumn （-4.24℃） and spring （-3.85℃）. The total area of oasis cold island （OCI） and the comprehensive OCIE index showed the same seasonal change trend as the OCIE intensity. However, the changing trends in areas of OCI with strong, medium and weak OCIEs were inconsistent across different seasons. Farmland and water areas were found to be the key contributors that affected the OCIE, and the area and aggregation metrics of these two land use/cover types directly contributed to the OCIE. By contrast, natural vegetation, such as forest and grassland, almost had no contribution to the OCIE. Simulation of observed air temperature data showed that if farmland is replaced by forest or grassland in the oasis, the mean, maximum and minimum air temperatures will increase significantly. This heating effect will be higher in summer （reaching 1.14℃ to 2.08℃） and lower in spring and autumn. Moreover, the heating effect of farmland being replaced by forest will be higher than that of farmland being replaced by grassland. These results can provide a basis for understanding the cold island effect of oases in arid regions.

Uncertainty assessment of potential evapotranspiration in arid areas, as estimated by the Penman-Monteith method

The Penman-Monteith(PM)method is the most widely used technique to estimate potential worldwide evapotranspiration.However,current research shows that there may be significant errors in the application of this method in arid areas,although questions remain as to the degree of this estimation error and how different surface conditions may affect the estimation error.To address these issues,we evaluated the uncertainty of the PM method under different underlying conditions in an arid area of Northwest China by analyzing data from 84 meteorological stations and various Moderate Resolution Imaging Spectroradiometer(MODIS)products,including land surface temperature and surface albedo.First,we found that when the PM method used air temperature to calculate the slope of the saturation vapor pressure curve,it significantly overestimated the potential evapotranspiration;the mean annual and July–August overestimation was 83.9 and 36.7 mm,respectively.Second,the PM method usually set the surface albedo to a fixed value,which led to the potential evapotranspiration being underestimated;the mean annual underestimation was 27.5 mm,while the overestimation for July to August was 5.3 mm.Third,the PM method significantly overestimated the potential evapotranspiration in the arid area.This difference in estimation was closely related to the underlying surface conditions.For the entire arid zone,the PM method overestimated the potential evapotranspiration by 33.7 mm per year,with an overestimation of 29.0 mm from July to August.The most significant overestimation was evident in the mountainous and plain nonvegetation areas,in which the annual mean overestimation reached 5%and 10%,respectively;during July,there was an estimation of 10%and 20%,respectively.Although the annual evapotranspiration of the plains with better vegetation coverage was slightly underestimated,overestimation still occurred in July and August,with a mean overestimation of approximately 5%.In order to estimate potential evapotranspiration in the arid zone,it is

Hydrology and water resources variation and its response to regional climate change in Xinjiang

Based on the surface runoff, temperature and precipitation data over the last 50 years from eight representative rivers in Xinjiang, using Mann-Kendall trend and jump detection method, the paper investigated the long-term trend and jump point of time series, the surface runoff, mean annual temperature and annual precipitation. Meanwhile, the paper analyzed the relationship between runoff and temperature and precipitation, and the flood frequency and peak flow. Results showed that climate of all parts of Xinjiang conformably has experienced an increase in temperature and precipitation since the mid-1980s. Northern Xinjiang was the area that changed most significantly followed by southern and eastern Xinjiang. Affected by temperature and precipitation variation, river runoff had changed both inter-annually and intra-annually. The surface runoff of most rivers has increased significantly since the early 1990s, and some of them have even witnessed the earlier spring floods, later summer floods and increasing flood peaks. The variation characteristics were closely related with the replenishment types of rivers. Flood frequency and peak flow increased all over Xinjiang. Climate warming has had an effect on the regional hydrological cycle.

Mutations in epigenetic regulator KMT2C detected by liquid biopsy are associated with worse survival in prostate cancer patients

Background:KMT2(lysine methyltransferase)family enzymes are epigenetic regulators that activate gene transcription.KMT2C is mainly involved in enhancer-associated H3K4me1,and is also one of the top mutated genes in cancer(6.6%in pan-cancer).Currently,the clinical significance of KMT2C mutations in prostate cancer is understudied.Methods:We included 221 prostate cancer patients diagnosed between 2014 and 2021 in West China Hospital of Sichuan University with cell-free DNA-based liquid biopsy test results in this study.We investigated the association between KMT2C mutations,other mutations,and pathways.Furthermore,we evaluated the prognostic value of KMT2C mutations,measured by overall survival(OS)and castration resistance-free survival(CRFS).Also,we explored the prognostic value of KMT2C mutations in different patient subgroups.Lastly,we investigated the predictive value of KMT2C mutations in individuals receiving conventional combined anti-androgen blockade(CAB)and abiraterone(ABI)as measured by PSA progression-free survival(PSA-PFS).Results:The KMT2C mutation rate in this cohort is 7.24%(16/221).KMT2C-mutated patients showed worse survival than KMT2C-wild type(WT)patients regarding both CRFS and OS(CRFS:mutated:9.9 vs.WT:22.0 months,p=0.015;OS:mutated:71.9 vs.WT 137.4 months,p=0.012).KMT2C mutations were also an independent risk factor in OS[hazard ratio:3.815(1.461,9.96),p=0.006]in multivariate analyses.Additionally,we explored the association of KMT2C mutations with other genes.This showed that KMT2C mutations were associated with Serine/Threonine-Protein Kinase 11(STK11,p=0.004)and Catenin Beta 1(CTNNB1,p=0.008)mutations.In the CAB treatment,KMT2C-mutated patients had a significantly shorter PSA-PFS compared to KMT2C-WT patients.(PSA-PFS:mutated:9.9 vs.WT:17.6 months,p=0.014).Moreover,KMT2C mutations could effectively predict shorter PSA-PFS in 10 out of 23 subgroups and exhibited a strong trend in the remaining subgroups.Conclusions:KMT2C-mutated patients showed worse survival compared to KMT2C-WT patients i

蒸散是水—能—碳循环之间的纽带,了解蒸散的动态过程及其驱动因素对中亚干旱区水资源稳定、生态环境安全及农业水资源管理等方面具有重要意义。本研究基于Priestley-Taylor方法,估算并分析了2000—2019年中亚干旱区蒸散的时空变化,采用Lindeman-Merenda-Gold方法定量评估了不同驱动因素对蒸散各组分的绝对贡献,并以各组分对蒸散变化的贡献加权,评估了各驱动因素对蒸散的贡献。结果表明:中亚干旱区蒸散整体以1.45mm·a^(-1)的速度波动上升,其变化趋势呈“东升西降”的空间分布;植被蒸腾、蒸发、冠层截留蒸发的变化趋势分别是2.46 mm·a^(-1)、-1.03 mm·a^(-1)、0.02 mm·a^(-1),三者对蒸散变化的贡献分别是70.09%、29.34%、0.57%;植被蒸腾和冠层截留蒸发的主导因素是NDVI,蒸发的主导因素是气温;总体上,NDVI是中亚干旱区蒸散变化的主导因素,其绝对贡献为28.16%。

The origin of Wx^(la) provides new insights into the improvement of grain quality in rice

Appearance and taste are important factors in rice(Oryza sativa) grain quality. Here, we investigated the taste scores and related eating-quality traits of533 diverse cultivars to assess the relationships between—and genetic basis of—rice taste and eating-quality. A genome-wide association study highlighted the Wx gene as the major factor underlying variation in taste and eating quality. Notably, a novel waxy(Wx) allele, Wx^(la), which combined two mutations from Wx^(b) and Wx^(in), exhibited a unique phenotype. Reduced GBSSI activity conferred Wx^(la) rice with both a transparent appearance and good eating quality. Haplotype analysis revealed that Wx^(la) was derived from intragenic recombination. In fact,the recombination rate at the Wx locus was estimated to be 3.34 kb/c M, which was about 75-fold higher than the genome-wide mean, indicating that intragenic recombination is a major force driving diversity at the Wx locus. Based on our results, we propose a new network for Wx evolution, noting that new Wx alleles could easily be generated by crossing genotypes with different Wx alleles. This study thus provides insights into the evolution of the Wx locus and facilitates molecular breeding for quality in rice.

Genetic architecture and key genes controlling the diversity of oil composition in rice grains

Rice grain oil is a valuable nutrient source.However,the genetic basis of oil biosynthesis in rice grains remains unclear.In this study,we performed a genome-wide association study on oil composition and oil concentration in a diverse panel of 533 cultivated rice accessions.High variation for 11 oil-related traits was observed,and the oil composition of rice grains showed differentiation among the subpopulations.We identified 46 loci that are significantly associated with grain oil concentration or composition,16 of which were detected in three recombinant inbred line populations.Twenty-six candidate genes encoding enzymes involved in oil metabolism were identified from these 46 loci,four of which(PAL6,LIN6,MYR2,and ARA6)were found to contribute to natural variation in oil composition and to show differentiation among the subpopulations.Interestingly,population genetic analyses revealed that specific haplotypes of PAL6 and LIN6 have been selected in japonica rice.Based on these results,we propose a possible oil biosynthetic pathway in rice grains.Collectively,our results provide new insights into the genetic basis of oil biosynthesis in rice grains and can facilitate marker-based breeding of rice varieties with enhanced oil and grain quality.
