Exosomes derived from human induced pluripotent stem cell-derived neural progenitor cells protect neuronal function under ischemic conditions

Compared with other stem cells,human induced pluripotent stem cells-derived neural progenitor cells(iPSC-NPCs)are more similar to cortical neurons in morphology and ***,they have greater potential for promoting the survival and growth of neurons and alleviating the proliferation of *** of stem cell exosomes and stem cells themselves have both been shown to effectively repair nerve ***,there is no study on the protective effects of exosomes derived from iPSC-NPCs on oxygen and glucose deprived *** this study,we established an oxygen-glucose deprivation model in embryonic cortical neurons of the rat by culturing the neurons in an atmosphere of 95%N2 and 5%CO2 for 1 hour and then treated them with iPSC-NPC-derived exosomes for 30 *** results showed that iPSC-NPC-derived exosomes increased the survival of oxygen-and glucose-deprived neurons and the level of brain-derived neurotrophic factor in the culture ***,it attenuated oxygen and glucose deprivation-induced changes in the expression of the PTEN/AKT signaling pathway as well as synaptic plasticity-related proteins in the ***,it increased the length of the longest neurite in the oxygen-and glucose-deprived *** findings validate the hypothesis that exosomes from iPSCNPCs exhibit a neuroprotective effect on oxygen-and glucose-deprived neurons by regulating the PTEN/AKT signaling pathway and neurite *** study was approved by the Animal Ethics Committee of Sir Run Run Shaw Hospital,School of Medicine,Zhejiang University,China(approval ***20191010)on October 10,2019.

Arsenic Trioxide Combining Leflunomide Activates Nrf2-ARE-HO-1 Signaling Pathway and Protects Heart Xenografts

Objective:To investigate the molecular mechanisms underlying the effects of arsenic trioxide(As_(2)0_(3))in combination with leflunomide on the hamster-to-rat heart ***:Transplantation of LVG hamster hearts to Lewis rats was performed by anastomosis of vessels in the neck using end-to-end anastomosis with a non-suture cuff *** groups of recipient rats(n=6 in each)were treated with normal saline(control),As_(2)0_(3)[5 mg/(kg*day)intraperitoneally],leflunomide[5 mg/(kg*d)orally],or leflunomide[5 mg/(kg.d)+As_(2)0_(3)5 mg/(kg.d)]in *** hearts and/or rat spleens were harvested and analyzed 4 days after *** reverse-transcription polymerase chain reaction and Western blot analysis were performed to detect the expression of the nuclear factor erythroid-derived factor 2-related factor(Nrf2)and its target gene heme oxygenase-1(HO-1),Treg cell marker fork-head Box P3(FOXP3),apoptosis-associated proteins Bcl-2,Bax,and cleaved *** staining was used to detect the levels of inflammatory natural killer cell and macrophage infiltration,intercellular cell adhesion molecule-1(ICAM-1)and complement ***:Expression of Nrf2-ARE-HO-1 signaling pathway was upregulated in heart xenografts in rats treated with As_(2)0_(3) plus leflunomide compared with control rats or rats treated with either drug alone(P<0.01),and this was accompanied by an increased Treg cells in the recipient rat spleen(P<0.01).In contrast,the expressions of Bax,cleaved caspase-3,ICAM-1,and complement C3,and infiltration of inflammatory cells in the xenografts were inhibited by As_(2)0_(3) plus leflunomide treatment(P<0.01).Conclusion:Combination treatment with As_(2)0_(3) and leflunomide protected hamster heart-xenografts in recipient rats.

Temporary plugging agent(TPA)is widely used in many fields of petroleum reservoir drilling and production,such as temporary plugging while drilling and petroleum well stimulation by diverting in acidizing or fracturing *** commonly used TPA mainly includes hard particles,fibers,gels,and composite ***,current particles have many limitations in applications,such as insufficient plugging strength and slow degradation *** this paper,a degradable pre-formed particle gel(DPPG)was *** results show that the DPPG has an excellent static swelling effect and self-degradation *** a decrease in the concentration of total monomers or cross-linker,the swelling volume of the synthesized DPPG gradually ***,the entire self-degradation time gradually *** increase in 2-acrylamide-2-methylpropanesulfonic acid(AMPS)in the DPPG composition can significantly increase its swelling ratio and shorten the self-degradation ***,DPPG has excellent high-temperature resistance(150°C)and high-salinity resistance(200,000 mg/L NaCl).Core displacement results show that the DPPG has a perfect plugging effect in the porous media(the plugging pressure gradient was as high as 21.12 MPa),and the damage to the formation after degradation is incredibly ***,the DPPG can be used as an up-and-coming TPA in oil fields.

An improved formula for standard hypoxia tolerance time to evaluate hypoxic tolerance in mice

Background: Hypoxia is a primary cause of mountain sickness and a common pathological condition in patients with heart failure, shock, stroke, and chronic obstructive pulmonary disease(COPD). Thus far, little advancement in countering hypoxic damage has been achieved, and one of the main reasons is the absence of an ideal algorithm or calculation method to normalize hypoxia tolerance scores when evaluating an animal model. In this study, we improved a traditional calculation formula for assessment of hypoxia ***: We used a sealed bottle model in which the oxygen is gradually consumed by a mouse inside. To evaluate the hypoxia tolerance of mice, the survival time(ST) of the mouse is recorded and was used to calculate standard hypoxia tolerance time(STT) and adjusted standard hypoxia tolerance time(ASTT). Mice administered with methazolamide and saline were used as positive and negative controls, ***: Since mice were grouped according to either body weight(BW) or bottle volume, we found a strongly negative correlation between STT and BW instead of between STT and bottle volume, suggesting that different BWs could cause false positive or negative errors in the STT results. Furthermore, both false positive and negative errors could be rectified when ASTT was used as the evaluation index. Screening for anti-hypoxic medicines by using mice as the experimental subjects would provide more credible results with the improved ASTT method than with the STT ***: ASTT could be a better index than STT for the evaluation of hypoxia tolerance abilities as it could eliminate the impact of animal BW.

Dear Editor,The Hippo tumor suppressor pathway plays an important role in development and ***-associated protein(YAP)is a major effector of the Hippo pathway regulating the expression of genes involved in cell proliferation,cell death,and cell differentiation(Pan,2010;Johnson and Haider,2014;Piccolo etal.,2014;Yu etal.,2015).In intestine,YAP is mainly expressed in leucine rich repeat containing G protein-coupled receptor 5(LGR5)marked stem cells,and is required for regeneration following tissue damage and the development of colon cancer in mice(Cai etal.,2010;Barry etal.,2013;Wang etal.,2017).Moreover,in colorectal cancer(CRC)specimens,elevated YAP expression and activity is frequently observed(Steinhardt et al.,2008).It has been shown that high YAP expression is associated with cancer metastasis,poor prognosis,resistance to chemotherapy,and greater possibility of relapse(Johnson and Haider,2014;Yu etal.,2015).

We present high angular resolution images of both NH3 (1, 1) and (2, 2) lines toward NGC 7538 IRS 1. The density and velocity-position plots have been used to study the interaction among the outflows, winds and their environment. For the first time we have found an expanding half-shell of molecular gas around the HII region associated with IRS 1, which may be produced by the interaction of the bipolar outflows and the winds originating in IRS 1-3, and optical HII region NGC 7538 with ambient molecular gas.

BACKGROUND Rebleeding after recovery from esophagogastric variceal bleeding(EGVB)is a severe complication that is associated with high rates of both incidence and *** its clinical importance,recognized prognostic models that can effectively predict esophagogastric variceal rebleeding in patients with liver cirrhosis are *** To construct and externally validate a reliable prognostic model for predicting the occurrence of esophagogastric variceal *** This study included 477 EGVB patients across 2 cohorts:The derivation cohort(n=322)and the validation cohort(n=155).The primary outcome was rebleeding events within 1 *** least absolute shrinkage and selection operator was applied for predictor selection,and multivariate Cox regression analysis was used to construct the prognostic *** validation was performed with bootstrap *** assessed the discrimination,calibration and accuracy of the model,and performed patient risk *** Six predictors,including albumin and aspartate aminotransferase concentrations,white blood cell count,and the presence of ascites,portal vein thrombosis,and bleeding signs,were selected for the rebleeding event prediction following endoscopic treatment(REPET)*** predicting rebleeding within 1 year,the REPET model ex-hibited a concordance index of 0.775 and a Brier score of 0.143 in the derivation cohort,alongside 0.862 and 0.127 in the validation ***,the REPET model revealed a significant difference in rebleeding rates(P<0.01)between low-risk patients and intermediate-to high-risk patients in both *** We constructed and validated a new prognostic model for variceal rebleeding with excellent predictive per-formance,which will improve the clinical management of rebleeding in EGVB patients.

Identification and expression patterns of alcohol dehydrogenase genes involving in ester volatile biosynthesis in pear fruit

Alcohol dehydrogenase （ADH） catalyzes the interconversion of aldehydes and their corresponding alcohols, and is a key enzyme in volatile ester biosynthesis. However, little is known regarding ADH and ADH encoding genes （ADHs） in pear. We identified 8 ADHs in the pear＇s genome （PbrADHs） by multiple sequences alignment. The PbrADHs were highly ho- mologous in their coding regions, while were diversiform in structure. 9 introns were predicted in PbrADH3-PbrADH8, while 8 introns, generated through exon fusion and intron loss, were predicted in PbrADH1 and PbrADH2. To study the genetic regulation underlying aroma biogenesis in pear fruit, we determined the PbrADH transcripts, ADH activities and volatile contents of fruits during ripening stage for Nanguoli and Dangshansuli, two cultivars having different aroma characteristics. ADH activity was strongly associated with the transcription of ADH~ in the two cultivars during fruit ripening stage. The higher ester content paralleling to a higher ADH activity was detected in Nanguoli than in Dangshansuli, so it is induced that the lower ester content in Dangshansuli fruit may be the result of weak ADH activity. The present study revealed that total ADH activity and volatile ester production correlated with increased PbrADHstranscript levels. PbrADH6 may contribute to ADH activity catalyzing aldehyde reduction and ester formation in pear fruit.
