circRNA_0046366 inhibits hepatocellular steatosis by normalization of PPAR signaling

AIM To investigate micro(mi)R-34 a-antagonizing circular(circ)RNA that underlies hepatocellular *** The effect of circ RNA on mi R-34 a was recognized by the mi RNA response element(MRE), and validated by the dual-luciferase reporter assay. Its association with hepatocellular steatosis was investigated in Hep G2-based hepatocellular steatosis induced by free fatty acids(FFAs; 2:1 oleate:palmitate) stimulation. After normalization of the steatosis-related circRNA by expression vector, analysis of mi R-34 a activity,peroxisome proliferator-activated receptor(PPAR)α level, and expression of downstream genes were carried out so as to reveal its impact on the mi R-34 a/PPARα regulatory system. Both triglyceride(TG) assessment and cytopathological manifestations uncovered the role of circRNA in miR-34 a-dependent *** Bioinformatic and functional analysis verified circRNA_0046366 to antagonize the activity of mi R-34 a via MRE-based complementation. In contrast to its lowered level during FFA-induced hepatocellular steatosis, circ RNA_0046366 up-regulation abolished the mi R-34 a-dependent inhibition of PPARα that played a critical role in metabolic signaling pathways. PPARα restoration exerted transcriptional improvement to multiple genes responsible for lipid metabolism. TGspecific lipolytic genes [carnitine palmitoyltransferase 1 A(CPT1 A) and solute-carrier family 27 A(SLC27 A)] among these showed significant increase in their expression levels. The circ RNA_0046366-related rebalancing of lipid homeostasis led to dramatic reduction of TG content, and resulted in the ameliorated phenotype of hepatocellular *** Dysregulation of circ RNA_0046366/mi R-34 a/PPARα signaling may be a novel epigenetic mechanism underlying hepatocellular steatosis. circ RNA_0046366 serves as a potential target for the treatment of hepatic steatosis.

Distributed optimal consensus control for multiagent systems based on event-triggered and prioritized experience replay strategies

This study uses event-triggered(ET) and reinforcement learning methods to investigate the optimal consensus control problem for cooperative-competitive multiagent systems. It proposes a novel distributed ET control strategy, which relies on a prioritized experience replay(PER) policy. This strategy not only conserves communication resources but also ensures acceptable system performance. To implement the proposed method, actor-critic(AC) dual-structured neural networks(NNs) are used to approximate the value function and control policy. In the AC NNs, the weight estimates for the NNs are updated at the moment of event triggering, resulting in a nonperiodic weight adjustment pattern. This approach decreases the computational cost in comparison with the traditional ET mechanism. The PER-based ET mechanism makes full use of valid historical data and effectively establishes a balance between system performance and communication resource ***, it does not require the following two conditions in most existing studies:(1) requirement of the system dynamics model to be known, and(2) persistent excitation. In addition, Zeno behavior is excluded from this study. Finally, a simulation is conducted to confirm the validity of the suggested approach.

The textured roll and polished roll were applied instead of the ground roll in a 20-high mill to conduct two-pass rolling of 316L stainless steel strip with thickness of 0.027 *** the two-pass rolling with the textured roll and polished roll(TPR),the surface roughness of the strip is dramatically reduced,and the surface topographical anisotropy index is diminished to 30.9%of the initial *** with the strip rolled using the ground roll in both passes(GGR),the elongation of TPR rolled strip is obviously improved,and the mechanical property anisotropy is greatly *** anisotropy index of tensile strength and elongation are 42.58%and 52.59%of that of GGR rolled strip,which is mainly attributed to the significant decrease of the texture intensity of the strip by TPR *** results indicate that TPR process can obtain the stainless steel ultra-thin strip with smooth and uniform surface topography and good mechanical properties.

3D morphology of crab(Macrophthalmus japonicus) burrows from the Pearl River Delta front,China:The physicochemical factors,with implications for the rock record

Neoichnological characterization of modern depositional settings addresses the response of benthic animals to ecological conditions and their fluctuations, usually on a relatively short time scale. In this way, analogue models for interpretation of sedimentary strata in the geological record can be *** japonicus(De Haan, 1835), a crab, is commonly found in the Pearl River Delta front of southern China. The burrows of M. japonicus are vertical or inclined, I-, U-, Y-, J-and L-shaped tubes, and these are imaged in 7.5-cm-diameter sediment cores. The cores were studied by X-ray radiography, computed tomography, and VG Studio MAX reconstruction to obtain three-dimensional images for elaborate morphological study. This crab's burrows are analogous to the trace fossil Psilonichnus. Several environmental parameters, i.e., sediment grain size and its total organic carbon(TOC) content, and turbidity and salinity of water, have been measured. The results indicated that M. japonicus prefers to live in mesohaline water with turbidity levels of 20-170 NTU, where bioturbation is more abundant. It prefers to occur in mud and silty sand substrates, with TOC content decreasing as particle size increases. The sedimentary environment, grain size and ichnofacies of analogical trace fossils Psilonichnus are discussed which supports the interpretation that Psilonichnus is a good indicator of delta front environments.

To elucidate the process of adaptation, particularly the traits subject to natural selection and the molecular mechanisms underlying their natural variation, is one of the primary objectives of evolutionary biology. The uplifted landscape offers an excellent framework for understanding how organisms adapt to dramatic climatic gradients. To investigate the genetic basis of plant adaptation to the extremely high altitude, we first compared the genomic and phenotypic variations of two closely related Arabidopsis thaliana accessions from high altitude(Xizang, also known as “Tibet”) and low altitude(Yunnan), respectively. The Xizang population represents a relict group characterized by a small effective population size. Notably, the Xizang genome has more transposable elements(TEs) and more gene loss-offunction(LoF) mutations. Differentially expressed genes were enriched in biological processes of cellular response to oxygen-containing compound, regulation of defense response, and response to light intensity. Intriguingly, the phenotypic selection analysis revealed that silique density was under natural selection. Furthermore, we genetically mapped and validated that the LoF mutation of GA20ox1, the homologous gene of green revolution in rice, resulted in a higher silique density in Xizang Arabidopsis. Given that GA20ox1 is linked to Arabidopsis adaptation to the Alps Mountains, its parallel evolution plays an important role in the adaptation to Alpine habitats. Overall,our results highlight that identifying adaptive traits and elucidating the molecular mechanisms underlying natural variation of these traits is crucial for unraveling the mystery of adaptive evolution and has significant implications for crop breeding.
