Suppression of pancreatic carcinoma growth by activating peroxisome proliferator-activated receptor γ involves angiogenesis inhibition

AIM:To study the possible actions and mechanisms of peroxisome proliferator-activated receptorγ(PPARγ) ,a ligand-activated transcription factor,in pancreatic carcinogenesis,especially in angiogenesis. METHODS:Expressions of PPARγand retinoid acid receptor(RXRα) were examined by reverse-transcription polymerase chain reaction(RT-PCR) with immunocytochemical staining.Pancreatic carcinoma cells,PANC-1,were treated either with 9-cis-RA,a ligand of RXRα,or with 15-deoxy-Δ 12,14 prostaglandin J2(15d-PGJ2) ,a ligand of PPARγ,or both.Antiproliferative effect was evaluated by cell viability using methyltetrazolium(MTT) assay.A pancreatic carcinoma xenograft tumor model of nude mice was established by inoculating PANC-1 cells subcutaneously.Rosiglitazone,a specific ligand of PPARγ,was administered via water drinking in experimental group of nude mice.After 75 d,all mice were sacrificed.Expression of proliferating cell nuclear antigen(PCNA) in tumor tissue was examined with immunohistochemical staining.Expression of vascular endothelial growth factor(VEGF) mRNA in PANC-1 cells,which were treated with 15d-PGJ2 or 9-cis-RA at various concentrations or different duration,was detected by semi-quantitative RT-PCR.Effects of Rosiglitazone on changes of microvascular density(MVD) and VEGF expression were investigated in xenograft tumor tissue.Neovasculature was detected with immunohistochemistry staining labeled with anti-Ⅳcollagen antibody,and indicated by MVD. RESULTS:RT-PCR and immunocytochemical staining showed that PPARγand RXRαwere expressed in PANC-1 cells at both transcription level and translation level.MTT assay demonstrated that 15d-PGJ2,9-cis-RA and their combination inhibited the growth of PANC-1 cells in a dose-dependent manner.9-cis-RA had a combined inhibiting action with 15d-PGJ2 on the growth of pancreatic carcinoma.In vivo studies revealed that Rosiglitazone significantly suppressed the growth of pancreatic carcinoma as compared to control group(0.48±0.23 cm 3 vs 2.488±0.59 cm3,P<0.05) ,and the growth inhibition

Efficacy of intraductal ultrasonography in the diagnosis of non-opaque choledocholith

AIM: To evaluate the efficacy of intraductal ultrasonography (IDUS) in the diagnosis of non-opaque, common bile duct stones. METHODS: A total of 183 patients (102 males, mean age 73 years; 81 females, mean age 70 years) with suspected common bile duct stones diagnosed through abdominal computed tomography (CT), magnetic resonance imaging (MRI), and abdominal Type-B ultrasound were included in the study. The diagnosis was confirmed through endoscopic retrograde cholangiopancreatography (ERCP) followed by IDUS. RESULTS: A total of 183 patients with suspected common bile duct (CBD) stones were included in the study as follows: 36 patients with high-density CBD stones, 68 patients with sand-like stones, 44 patients with low-density stones, 21 patients with ampullary cancer, and 14 patients with pancreatic cancer. Conventional imaging revealed 124 cases of choledochectasia, and only 36 cases of suspected CBD stones; ERCP revealed 145 cases of CBD stones with three missed diagnoses. IDUS revealed 148 cases of CBD stones, 21 cases of ampullary tumors, and 14 cases of pancreatic cancer. CONCLUSION: IDUS was more effective in the diagnosis of bile duct stones than ERCP, upper abdominal CT or upper abdominal MRI.

Effect of L-cysteine on remote organ injury in rats with severe acute pancreatitis induced by bile-pancreatic duct obstruction

BACKGROUND: Remote organ failure occurs in cases of acute pancreatitis (AP); however, the reports on AP induced by pancreatic duct obstruction are rare. In this study we determined the effect of L-cysteine on pancreaticobiliary inflammation and remote organ damage in rats after pancreaticobiliary duct ligation (PBDL). METHODS: AP was induced by PBDL in rats with 5/0 silk Sixty rats were randomly divided into 4 groups. Groups A and B were sham-operated groups that received injections of saline or L-cysteine (10 mg/kg) intraperitoneally (15 rats in each group). Groups C and D were PBDL groups that received injections of saline or L-cysteine (10 mg/kg) intraperitoneally (15 rats in each group). The tissue samples of the pancreas and remote organs such as the lung, liver, intestine and kidney were subsequently examined for pathological changes under a light microscope. The samples were also stored for the determination of malondialdehyde and glutathione levels. Blood urea nitrogen (BUN), plasma amylase, ALT and AST levels were determined spectrophotometrically using an automated analyzer. Also, we evaluated the effect of L-cysteine on remote organ injury in rats with AP induced by retrograde infusion of 3.5% sodium taurocholate (NaTc) into the bile-pancreatic duct. RESULTS: Varying degrees of injury in the pancreas, lung, liver intestine and kidney were observed in the rats 24 hours after PBDL. The severity of injury to the lung, liver and intestine was attenuated, while injury status was not changed significantly in the pancreas and kidney after L-cysteine treatment. Oxidativestress was also affected by L-cysteine in PBDL-treated rats. The concentration of tissue malondialdehyde decreased in the pancreas and remote organs of PBDL and L-cysteine administrated rats, and the concentration of glutathione increased more significantly than that of the model control group. However, L-cysteine administration reduced the severity of injury in remote organs but not in the pancreas in rats with NaTc-induced AP. CONCLUSION: L-c

Data mining of microarray for differentially expressed genesin liver metastasis from gastric cancer

Tumor metastasis is the leading cause of death for gastric cancer.Metastasis is the main reason for the failure of clinical treatment for gastric cancer.In order to find metastasis-related genes and abnormal signal transduction pathway of high-invasive gastric cancer,samples of gastric cancer with liver metastasis were collected for microarray detection;up-regulated or down-regulated genes in all three cases were simultaneously screened out.Subsequently,from the preliminary screened genes,molecular pathways possibly impacting liver metastasis from gastric cancer were investigated by the Gene Cluster with Literature Profiles(GenCLip)analysis software.Many biological effects including apoptosis have been validated.Functional analysis of differentially expressed genes revealed that a variety of biological pathways,such as blood circulation and gas exchange,vasodilation and vasoconstriction regulation,and immune defense,could be significantly activated.Besides,gene sequences,specific keywords or gene regulatory networks were further searched by GenCLiP.We conclude that data mining allows to quickly identify a series of special signal transduction pathways involving abnormally expressed genes.

MicroRNA-155 promotes the pathogenesis of experimental colitis by repressing SHIP-1 expression

AIM To explore the mechanism by which micro RNA-155(miR-155) regulates the pathogenesis of experimental colitis.METHODS A luciferase assay was performed to confirm the binding of miR-155 to the SHIP-1 3'-UTR. MiR-155 mimics, negative controls and SHIP-1 expression/knockdown vectors were established and then utilized in gain-and loss-of-function studies performed in raw264.7 cells and primary bone marrow-derived macrophages (BMDMs). Thereafter, dextran sulfate sodium(DSS)-induced colitis mouse model with or without antagomiR-155 treatment was established, and the levels of miR-155 and SHIP-1, as well as the pro-inflammatory capabilities, were measured by western blot, quantitative polymerase chain reaction, and immunohistochemistry.RESULTS MiR-155 directly bound to the 3'-UTR of SHIP-1 mRNA and induced a significant decrease in SHIP-1 expression in both raw264.7 cells and primary BMDMs. MiR-155 markedly promoted cell proliferation and proinflammatory secretions including IL-6, TNF-α, IL-1β, and IFN-γ, whereas these effects could be reversed by the restoration of SHIP-1 expression. In vivo studies showed that antagomiR-155 administration could alleviate DSS-induced intestinal inflammation in Balb/c mice. Moreover, significantly increased SHIP-1 expression, as well as decreased Akt activation and inflammatory response, were observed in the antagomiR-155-treated mice.CONCLUSION MiR-155 promotes experimental colitis by repressing SHIP-1 expression. Thus, the inhibition of miR-155 might be a promising strategy for therapy.

Effect of Lianshu preparation on lipopolysaccharide-induced diarrhea in rats

AIM:To investigate the effect of Lianshu preparation on lipopolysaccharide (LPS)-induced diarrhea in rats.METHODS: A diarrhea model was established in Sprague Dawley rats via injection of 1 mL of 30 mg/kg LPS. A total of 40 rats were randomly divided into normal group, LPS group, LPS + Lianshu group, LPS + berberine group (n=10 in each group). Their intestinal mucosal barrier and frequency of diarrhea were observed. Levels of glucose, serum Na+, K+, Cl- and hematocrit, plasma nitrogen monoxide (NO), diamine oxidase (DAO), and D (-)-lactate were measured. The number of IgA+ plasma cells in small intestine was detected and SIgA levels in the intestinal fluid were measured. The antipyretic activity of Lianshu preparation in rats was evaluated using Brewer's yeast-induced pyrexia (10 mL/kg of 20% aqueous suspension). Acetaminophen (250 mg/kg, intragastric administration, bid) was used as a standard drug for comparison. Temperature was recorded 1 h before and 6 h after Brewer's yeast injection. Finally, small intestinal transmission in mice treated with Lianshu was detected after intraperitoneal injection of methyl prostigmin (2 mg/kg). Atropine (10 g/kg) was used as a control. The ink content in intestine was determined and the total length of intestine was measured.RESULTS: The frequency of diarrhea was higher in LPS group than in LPS + Lianshu group and LPS + berberine group (36.70±5.23 vs 28.50 ±4.06 and 32.70±9.30 respectively, P<0.01), and lower in LPS + Lianshu group than in LPS + berberine group (P=0.03). The levels of Na+, glucose, Cl-, K+ were significantly lower in LPS + Lianshu group than in LPS + berberine group (140.35±3.19 mmol/L vs 131.99±4.86 mmol/L, 8.49 ±1.84 mmol/L vs 6.54±2.30 mmol/L, 106.29±4.41 mmol/L vs 102.5±1.39 mmol/L, 5.08±0.66 mmol/L vs 4.32±0.62 mmol/L respectively, p<0.05). The level of hematocrit was lower in LPS + Lianshu group than in LPS + berberine group (0.50% ± 0.07% vs 0.59% ± 0.10% respectively, P<0.05). Suppression of angiotensin Ⅱ stimulated responses in aortic vascular smooth muscle cells of experimental cirrhotic rats

Functional responses to angiotensin Ⅱ (AT-Ⅱ) were determined in aortic vascular smooth muscle cells (VSMCs) from experimental cirrhotic rats. Our data showed that AT-Ⅱ-stimulated extracellular acidification rate (ECAR), which was measured by Cytosensor microphysiometry, was significantly reduced in the aortic VSMCs from the cirrhotic rats as compared to those from the control animals. The ability of AT-Ⅱ to promote formation of inositol phosphates, the second messenger produced by the activation of Gq-coupled receptors, was also considerably suppressed in the cirrhotic VSMCs. Furthermore, the maximal p42/44 MAPK phosphorylation stimulated by AT-Ⅱ was significantly reduced in the cirrhotic VSMCs in contrast to that in the normal VSMCs. Taken together, our data clearly demonstrated that the functional responses to AT-Ⅱ was severely suppressed in aortic VSMCs in cirrhosis, indicating the impairment of general Gq-coupled receptor signaling and subsequent biological function in the cirrhotic VSMCs.

Anti-tumor activity of erlotinib in the BxPC-3 pancreatic cancer cell line

AIM: To investigate the effect and mechanism of action of erlotinib, an epidermal growth factor receptor (EGFR) small molecule tyrosine kinase inhibitor (TKI), in the human pancreatic cancer cell line BxPC-3 both in vitro and in vivo. METHODS: In vitro, human pancreatic cancer cel line BxPC-3 was exposed to varying concentrations of erlotinib, and its effects on proliferation, cell cycle distribution, apoptosis and the expression of proand antiapoptotic factors such as bcl-2, bcl-xl, bax and bak, and the expression of vascular endothelia cell growth factor (VEGF) were measured with 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay, flow cytometric analysis, terminal deoxynucleotidyl transferase-mediated nick end labeling assay (TUNEL), and reverse transcription- polymerase chain reaction (RT-PCR). Potential effect of erlotinib on angiogenesis was examined by tube formation assay. Tumor growth suppression was observed in xenografted nude mice with pancreatic cancer in vivo. Immunohistochemical (IHC) staining for EGFR and factor Ⅷ-related antigen was undertaken to detect the microvessel density and VEGF expression in tumor tissue in xenograft nude mice. RESULTS: Erlotinib, as a single agent, repressed BxPC-3 cell growth in a dose-dependent manner, triggered G1 arrest and induced cell apoptosis, and suppressed capillary formation of endothelium in vitro. Expressions of VEGF were significantly down-regulatedat a high concentration of 200 μmol/L, however, the expressions of bcl-2 and bcl-xl were decreased at 50 μmol/L. In vivo , Erlotinib-treated mice demon- strated a reduced tumor volume, weight and microvessel density as compared to the control. IHC staining showed decreased expression of EGFR and RT-PCR had lower VEGF expression in treated mice. CONCLUSION: The in vitro and in vivo findings provide evidence that BxPC-3 cells are inhibited with erlotinib treatment. Inhibition of EGFR may be a promising adjuvant chemotherapy strategy in pancreatic cancer treatment.

Effects of combustion temperature on the optical properties of brown carbon from biomass burning

Biomass burning has been known as one of main sources of Brown Carbon(BrC)in atmosphere.In this study,by controlling the combustion temperature at 250℃,350℃,and 450℃,the methanol soluble organic carbon(MSOC)and methanol insoluble carbon(MISC)from pine wood burning was collected by impinger.UV–Vis,excitation emission matrix(EEM),TEM and FTIR spectra were applied to investigate the properties of BrC collected.For MSOC at 250℃ and 350℃,all the spectral profiles of UV–Vis absorption and excitation emission matrix are almost the same,while the EEM of MSOC at 450℃ are different from that of the other two.For MISC fuorescence was observed only in the case of 450℃.In the FTIR spectra,with the temperature increasing the peaks associated to the oxygen-contained functions was weakened,indicating the formation of the fuorophores with larger conjugated system,especially aromatic hydrocarbons.Our results show that biomass combustion at low temperature produces more oxygen-riched BrC,which possesses relatively lower light absorption,while at high temperature produces more aromatics hydrocarbons with relatively strong light absorption.The results of this work are helpful to trace the source of brown carbon and optimize biomass energy utilization.

The development of tellurium(Te)-based semiconductor nanomaterials for efficient light-to-heat conversion may offer an effective means of harvesting sunlight to address global energy concerns.However,the nanosized Te(nano-Te)materials reported to date suffer from a series of drawbacks,including limited light absorption and a lack of surface structures.Herein,we report the preparation of nano-Te by electrochemical exfoliation using an electrolyzable room-temperature ionic liquid.Anions,cations,and their corresponding electrolytic products acting as chemical scissors can precisely intercalate and functionalize bulk Te.The resulting nano-Te has high morphological entropy,rich surface functional groups,and broad light absorption.We also constructed foam hydrogels based on poly(vinyl alcohol)/nano-Te,which achieved an evaporation rate and energy efficiency of 4.11 kg m^(−2)h^(−1)and 128%,respectively,under 1 sun irradiation.Furthermore,the evaporation rate was maintained in the range 2.5-3.0 kg m^(−2)h^(−1)outdoors under 0.5-1.0 sun,providing highly efficient evaporation under low light conditions.
