Yanggan Jiangmei Formula alleviates hepatic inflammation and lipid accumulation in non-alcoholic steatohepatitis by inhibiting the NF-κB/NLRP3 signaling pathway

The role of NF-κB and the NLRP3 inflammasome in the chronic inflammatory microenvironment of non-alcoholic steatohepatitis(NASH)has been posited as crucial.The Yanggan Jiangmei Formula(YGJMF)has shown promise in ameliorating hepatic steatosis in NASH patients,yet its pharmacological mechanisms remain largely unexplored.This study was conducted to investigate the efficacy of YGJMF in NASH and to elucidate its pharmacological underpinnings.To simulate NASH both in vivo and in vitro,high-fat-diet(HFD)rats and HepG2 cells stimulated with free fatty acids(FFAs)were utilized.The severity of liver injury and lipid deposition was assessed using serum indicators,histopathological staining,micro-magnetic resonance imaging(MRI),and the liver-tomuscle signal intensity ratio(SIRL/M).Furthermore,a combination of enzyme-linked immunosorbent assay(ELISA),immunohistochemistry(IHC),immunofluorescence,real-time quantitative polymerase chain reaction(RT-qPCR),and Western blotting analyses was employed to investigate the NF-κB/NLRP3 signaling pathway and associated cytokine levels.The results from liver pathology,MRI assessments,and biochemical tests in rat models demonstrated YGJMF’s significant effectiveness in reducing liver damage and lipid accumulation.Additionally,YGJMF markedly reduced hepatocyte inflammation by downregulating inflammatory cytokines in both liver tissue and serum.Furthermore,YGJMF was found to disrupt NF-κB activation,consequently inhibiting the assembly of the NLRP3 inflammasome in both the in vitro and in vivo models.The preliminary findings of this study suggest that YGJMF may alleviate hepatic steatosis and inhibit the NF-κB/NLRP3 signaling pathway,thereby exerting anti-inflammatory effects in NASH.

Identification and verification of aging-related lncRNAs for prognosis prediction and immune microenvironment in patients with head and neck squamous carcinoma

Aging is highly associated with tumor formation and progression.However,little research has explored the association of aging-related lncRNAs(ARLs)with the prognosis and tumor immune microenvironment(TIME)of head and neck squamous cell carcinoma(HNSCC).RNA sequences and clinicopathological data of HNSCC patients and normal subjects were downloaded from The Cancer Genome Atlas.In the training group,we used Pearson correlation,univariate Cox regression,least absolute shrinkage/selection operator regression analyses,and multivariate Cox regression to build a prognostic model.In the test group,we evaluated the model.Multivariate Cox regression was done to screen out independent prognostic factors,with which we constructed a nomogram.Afterward,we demonstrated the predictive value of the risk scores based on the model and the nomogram using time-dependent receiver operating characteristics.Gene set enrichment analysis,immune correlation analysis,and half-maximal inhibitory concentration were also performed to reveal the different landscapes of TIME between risk groups and to predict immuno-and chemo-therapeutic responses.The most important LINC00861 in the model was examined in HNE1,CNE1,and CNE2 nasopharyngeal carcinoma cell lines and transfected into the cell lines CNE1 and CNE2 using the LINC00861-pcDNA3.1 construct plasmid.In addition,CCK-8,Edu,and SA-β-gal staining assays were conducted to test the biofunction of LINC00861 in the CNE1 and CNE2 cells.The signature based on nine ARLs has a good predictive value in survival time,immune infiltration,immune checkpoint expression,and sensitivity to multiple drugs.LINC00861 expression in CNE2 was significantly lower than in the HNE1 and CNE1 cells,and LINC00861 overexpression significantly inhibited the proliferation and increased the senescence of nasopharyngeal carcinoma cell lines.This work built and verified a new prognostic model for HNSCC based on ARLs and mapped the immune landscape in HNSCC.LINC00861 is a protective factor for the development of HNSCC.

The creation of universal strategies to affect the reaction route of the electroreduction of CO_(2) is critical.Here,we report the first work to introduce cations into diverse metals such as Cu,Bi,In,and Sn via the electroreduction of related metallic oxides in quaternary ammonium surfactant solutions.Compared to their physical adsorption,cations embedded into the electrodes have a more pronounced impact on the electrical field,which effectively influences the adsorption state of intermediates.With the increase of surface field,the hydrogen evolution reaction and*COOH route are significantly reduced,favouring the*OCHO pathway instead.As a result,hydrogen,CO,and C_(2+)products almost completely vanish at−0.5 V versus RHE in 0.1 M Na_(2)SO_(4)in an H-type cell after enough cations are embedded into the Cu electrode,and the faradaic efficiency of formate rises from 18.0%to 99.5%simultaneously.

Inhibitory Effect of Flavonoid Glycosides from Chlorophytum comosum on Nasopharyngeal Carcinoma 5-8F Cells and Its Mechanism

[Objectives]To study the inhibitory activity of two flavonoid glycosides isolated from Chlorophytum comosum Laxum R.Br on human nasopharyngeal carcinoma(NPC)cell line 5-8F in vitro and its mechanism.[Methods]The flavonoid glycosides were isolated and purified from the ethanol alcoholic extract of the roots of Liliaceae plant Chlorophytum comosum by silica gel column chromatography,macroporous resin column chromatography,Sephadex LH-20,and reverse column chromatography(ODS).The inhibitory activity of flavonoid glycosides on human nasopharyngeal carcinoma cells was analyzed by CCK-8 method,and the potential mechanism was preliminarily analyzed by molecular docking.[Results]Two flavonoid glycosides were identified as isovitexin 2″-0-rhamnoside and 7-2″-di-O-β-glucopyranosylisovitexin.Two flavonoid glycosides showed promising inhibitory effect on human nasopharyngeal carcinoma cell line 5-8F,with IC_(50) values of 24.8 and 27.5μmol/L,respectively.Molecular docking results showed that the potential targets of two flavonoid glycosides include CyclinD1,Bcl-2β-Catenin,ILK,TGF-β,in addition,two glycosides showed higher predicted binding affinity towards CyclinD1,which verifies the cytotoxicity of the two compounds on human nasopharyngeal carcinoma cell line 5-8F in vitro.[Conclusions]Two flavonoid glycosides are the active molecules in Chlorophytum comosum that can inhibit the proliferation of human nasopharyngeal carcinoma cells,and have the potential to be used in the research and development of anti nasopharyngeal carcinoma drugs.

Report of China's innovation increase and research growth in radiation oncology

Aims： To investigate the research status of radiation oncology in China through survey of literature in international radiation oncology journals and retrospectively compare the outputs of radiation oncology articles of the three major regions of China--Mainland （ML）, Taiwan （TW） and Hong Kong （HK）. Methods： Radiation oncology journals were selected from ＂oncology＂ and ＂radiology, nuclear ＆ medical image＂ category from Science Citation Index Expand （SCIE）. Articles from the ML, TW and HK were retrieved from MEDLINE. The number of total articles, clinical trials, case reports, impact factors （IF）, institutions and articles published in each journals were conducted for quantity and quality comparisons. Results： A total 818 articles from 13 radiation oncology journals were searched, of which 427 are from ML, 259 from TW, and 132 from HK. Ninety-seven clinical trials and 5 case reports are reported in China. Accumulated IF of articles from ML （1,417.11） was much higher than that of TW （1,003.093） and HK （544.711）, while the average IF of articles from ML is the lowest. Conclusions： The total number of articles from China especially ML increased significantly in the last decade. The number of articles published from the ML has exceeded those from TW and HK. However, the quality of articles from TW and HK is better than that from ML.

Targets and molecular mechanisms of triptolide in cancer therapy

Triptolide（TPL/TL） is a natural drug with novel anticancer effects. Preclinical studies indicated that TPL inhibits cell proliferation, induces cell apoptosis, inhibits tumor metastasis and enhances the effect of other therapeutic methods in various cancer cell lines. Multiple molecules and signaling pathways, such as caspases, heat-shock proteins, NF-κB, and deoxyribonucleic acid（DNA） repair-associated factors, are associated with the anti-cancer effect. TPL also improves chemoradiosensitivity in cancer therapy. Phase I trials indicate the potential clinical value of TPL use. However, further trials with larger sample sizes are needed to confirm these results.

背景:在肾发育过程中,成纤维细胞生长因子受体1,2的时空表达仍是一个有争议的问题,且其与肾脏发育的关系尚不清楚。目的:观察成纤维细胞生长因子受体1,2在小鼠肾发育过程中的动态表达,探求它们与肾发生发育的关系。方法:培育不同发育阶段的胎鼠(E12,14,16,18 d)和仔鼠(N1,3,7,14,24,40 d),应用免疫组织化学技术观察成纤维细胞生长因子受体1,2在不同肾组织中的时空表达,结合体视学和Western blot对它们的表达进行定量分析。结果与结论:①免疫组化显示:在E12 d时,成纤维细胞生长因子受体1主要定位在输尿管芽尖端的生后肾组织,随后表达在各期未成熟的肾小体、部分远曲小管和毛细血管袢,反应部位主要集中在生肾区;而成纤维细胞生长因子受体2开始即在输尿管芽和生后肾组织中均有表达,随着后肾发育,定位于未发育成熟的各期肾小体、远端小管、集合管和髓袢细段,成熟肾小体表达微弱;②体视学和Western blot检测显示:成纤维细胞生长因子受体1在生前表达较高,出生后逐渐下降,N7 d后表达很低;成纤维细胞生长因子受体2在肾脏的表达随着胚(日)龄的增加而升高,N7 d后趋于稳定;③结果表明:在肾的发育过程中,成纤维细胞生长因子受体1,2呈一定的时空性表达,推测二者可能参与了肾单位的发育与成熟,而在输尿管芽分支和形态的形成过程中,成纤维细胞生长因子受体2的作用更为显著。

目的:探讨辛夷散基于瞬时受体电位香草酸亚型1(TRPV1)通路治疗变应性鼻炎(AR)大鼠的疗效和作用机制。方法:将70只SD大鼠随机分为空白组,模型组,辛夷散低、中、高剂量组,激动剂组,阳性药组,每组10只。除空白组外,其余各组大鼠均采用卵清蛋白(OVA)致敏方法制作AR大鼠模型。造模成功后,除空白组外,其余各组采用相应药物治疗,且均OVA滴鼻维持致敏。治疗前后观察行为学评分,HE染色观察鼻黏膜组织形态学变化;ELISA检测血清白细胞介素(IL)-4、IL-5、IL-13、免疫球蛋白E(IgE)水平;ELISA检测鼻黏膜组织中TRPV1、P物质(SP)、降钙素基因相关肽(CGRP)、神经生长因子(NGF)水平。结果:与空白组比较,模型组外周血IgE、IL-4、IL-5、IL-13表达显著增多(P<0.01),鼻黏膜TRPV1、SP、CGRP、NGF表达显著增多(P<0.01);与模型组比较,辛夷散各剂量组外周血IgE、IL-4、IL-5、IL-13表达显著降低(P<0.05,P<0.01),鼻黏膜TRPV1、SP、CGRP、NGF表达显著降低(P<0.01);与阳性药组和激动剂组比较,辛夷散中剂量组外周血IgE、IL-4、IL-5、IL-13表达显著升高(P<0.01),鼻黏膜TRPV1、SP、CGRP、NGF表达显著升高(P<0.01)。结论:辛夷散可能是通过调控TRPV1通路减少SP、CGRP、IL-4释放从而抑制Th2细胞过度活化,降低痒觉神经敏化,缓解鼻痒,从而治疗AR。

基于脑胶质瘤发病过程中的信号通路寻找治疗靶点成为探索脑胶质瘤新型治疗方式的重点领域。铁死亡是一种由脂质过氧化驱动的铁依赖性的细胞死亡方式,已被证实其参与多种肿瘤(包括脑胶质瘤)的发生及进展进程。全文回顾铁死亡的经典代谢途径,总结已有研究中常用的铁死亡诱导剂、抑制剂以及脂质过氧化的评估方法,梳理铁死亡与脑胶质瘤相关研究中的最新发现,包括铁死亡关键调节因子在脑胶质瘤中的表达情况、脑胶质瘤生理环境对铁死亡的影响及脑胶质瘤治疗药物与铁死亡的关系等,为脑胶质瘤的靶向治疗药物研发提供参考。

Dear Editor,Soybean(Glycine max)is a major source of plant-based protein for people worldwide,providing a healthy,affordable,and environmentally friendly alternative to animal-based protein(Montgomery,2003).It has been estimated that there are currently over 12000 food products that contain soy protein(Montgomery,2003).Different food applications of soy protein require distinct properties that affect its function and sensory features.For instance,the sensory quality of soymilk is significantly affected by protein emulsibility,whereas tofu quality is determined by protein-gelling ability(Kinsella,1979).
