Genome editing of PAR2 through targeted delivery of CRISPR-Cas9 system for alleviating acute lung inflammation via ERK/NLRP3/IL-1β and NO/iNOS signalling

Excessive and uncontrollable inflammatory responses in alveoli can dramatically exacerbate pulmonary disease progressions through vigorous cytokine releases,immune cell infiltration and protease-driven tissue *** is an urgent need to explore potential drug strategies for mitigating lung ***-activated receptor 2(PAR2)as a vital molecular target principally participates in various inflammatory diseases via intracellular signal ***,it has been rarely reported about the role of PAR2 in lung *** study applied CRISPR-Cas9 system encoding Cas9 and sg RNA(p Cas9-PAR2)for PAR2 knockout and fabricated an anionic human serum albuminbased nanoparticles to deliver p Cas9-PAR2 with superior inflammation-targeting efficiency and stability(TAP/p Cas9-PAR2).TAP/p Cas9-PAR2 robustly facilitated p Cas9-PAR2 to enter and transfect inflammatory cells,eliciting precise gene editing of PAR2 in vitro and in ***,PAR2 deficiency by TAP/p Cas9-PAR2 effectively and safely promoted macrophage polarization,suppressed proinflammatory cytokine releases and alleviated acute lung inflammation,uncovering a novel value of *** also revealed that PAR2-mediated pulmonary inflammation prevented by TAP/p Cas9-PAR2was mainly dependent on ERK-mediated NLRP3/IL-1β and NO/i NOS ***,this work indicated PAR2 as a novel target for lung inflammation and provided a potential nanodrug strategy for PAR2 deficiency in treating inflammatory diseases.

Spatio-temporal Evaluation of Multi-scale Cultivated Land System Resilience in Black Soil Region from 2000 to 2019:A Case Study of Liaoning Province,Northeast China

It is of great significance to systematically analyze the cultivated land system resilience(CLSR) for the black soil protection and national food *** CLSR is impacted by planting structure adjustment and cultivated land quality decline,posing major hidden dangers to food *** is urgent to evaluate the CLSR at multiple spatio-temporal *** study took Liaoning Province in the black soil region of Northeast China as an *** on the resilience theory,this study constructed the CLSR evaluation system from the input-feedback perspective at the provincial-scale and the city-scale,and used the rank-sum ratio comprehensive evaluation method(RSR) to analyze the key influencing factors of CLSR in Liaoning Province and its 14 cities from 2000 to *** results showed that:1) the time series changes of CLSR at the provincial-scale and the city-scale in Liaoning Province were similar,both showing an increasing trend.2) The CLSR in Liaoning Province presented a spatial pattern of ‘high in the west and low in the east’ at the city-scale.3) There were seven and six main influencing factors of CLSR at the provincial-scale and the city-scale,*** addition to the net income per capita of rural households,other influencing factors of CLSR were different at the provincial-scale and the *** feedback factors were dominant at the provincial-scale,and the input factors and feedback factors were dominant at the *** results could provide a reference for the utilization of black soil and draw on the experience of regional agricultural planning and adjustment.

The transient heat transfer behaviours in a packed bed have been studied using coupled computational fluid dynamics(CFD)and discrete element method(DEM).Intra-particle thermal diffusion,which is a crucial but rarely explored issue,is considered by introducing a one-dimensional diffusion equation in the DEM *** achieve a sufficiently long physical time of simulations,we further reduce the computational cost by applying periodic boundary condition to a small segment of the packed *** study demonstrates a shifting peak heat flow rate from the bottom to the top of the packed bed during the initial warming *** affirm that the influence of thermal diffusion within a particle is noteworthy only for particles exhibiting a high Biot number(e.g.,Bi=1.2).This insight enriches our understanding of heat transfer mechanisms in packed beds and their practical engineering applications.

Clinical Characteristics and Risk Factors of Left Ventricular Thrombus after Acute Myocardial Infarction： A Matched Case-control Study

Background： Left ventricular thrombus （LVT） is reported to be a common complication in acute myocardial infarction （AMI） patients. And it has the potential to cause systemic embolism. This retrospective study was to present the current situation of LVT in clinical practice, as well as to evaluate the clinical characteristics and the risk factors of LVT alter AMI. Methods： LVT cases （n = 96） were identified from 13,732 AMI （non-ST elevation myocardial infarction was excluded） patients in Fuwai Hospital＇s electronic medical records system from January 2003 to January 2013. The controls （n = 192） were gender- and age-matched AMI patients without LVT during this period. A conditional logistic regression （fitted by the Cox model） was performed to identity the independent risk factors. Results： The incidence of LVT after AMI was 0.7%. Univariate analysis indicated that the anterior myocardial infarction （especially extensive anterior myocardial infarction）, lower left ventricular ejection traction （LVEF）, LVEF ≤40%, severe regional wall motion abnonnalities （RWMA）, pericardial effusion, and left ventricular aneurysm were all related to LVT after AMI. The independent risk factors obtained from the conditional logistic regression analysis were lower LVEF （odds ratio （OR） = 0.891, 95% confidence interval （CI）： 0.828-0.960）, extensive anterior myocardial infarction （OR = 6.403, 95% CI： 1,769~3.169）, severe RWMA （OR = 7.348, 95% （7：1.32340.819）. and left ventricular aneurysm （OR = 6.955.95% CI： 1.673-28.921 ）. Conclusions： This study indicated that lower LVEF. extensive anterior myocardial infarction, severe RWMA, and left ventricular aneu,ysm were independent risk factors of LVT after AMI. It also suggested that further efforts are needed for the LVT diagnosis after AMI in clinical practice.

The Social Internet of Things(SIoT)integrates the Internet of Things(IoT)and social networks,taking into account the social attributes of objects and diversifying the relationship between humans and objects,which overcomes the limitations of the IoT’s focus on associations between *** Intelligence(AI)technology is rapidly *** is critical to build trustworthy and transparent systems,especially with system security issues coming to the *** paper emphasizes the social attributes of objects and uses hypergraphs to model the diverse entities and relationships in SIoT,aiming to build an SIoT hypergraph generation model to explore the complex interactions between entities in the context of intelligent *** hypergraph generation models impose too many constraints and fail to capture more details of real *** contrast,this paper proposes a hypergraph generation model that evolves dynamically over time,where only the number of nodes is *** combines node wandering with a forest fire model and uses two different methods to control the size of the *** new nodes are added,the model can promptly reflect changes in entities and relationships within *** results exhibit that our model can effectively replicate the topological structure of real-world *** also evaluate the vulnerability of the hypergraph under different attack strategies,which provides theoretical support for building a more robust intelligent SIoT hypergraph model and lays the foundation for building safer and more reliable systems in the future.

Molecular compositions and sources of organic aerosols at a rural site on the Guanzhong Plain,Northwest China:The importance of biomass burning

The concentration of PM_(2.5)has considerably reduced in recent years,but remains relatively high in *** particular,the increasing contribution of organic compounds to PM_(2.5)generates popular pressure for further reductions,resulting in an urgent need to study organic aerosol(OA).To investigate the molecular composition and source contribution of OA in the rural area of the Guanzhong Plain,Northwest China,PM_(2.5)samples were collected during 3–23 August 2016 and 5–20 January 2017 and studied for more than 100 organic tracer *** mean concentration of total measured organic compounds is 662±296 ng/m^(3)in summer and 3258±1925 ng/m^(3)in *** is the most abundant single compound found throughout the sampling period,which is a crucial tracer for biomass burning emissions,preliminary suggesting that biomass burning is an essential source of *** summer,organic compounds such as lipid compounds,sugar compounds,and polycyclic aromatic hydrocarbons(PAHs),more come from higher plants,wood burning,vehicle exhausts,plastic waste,and other direct emission *** PAHs(OPAHs),nitrophenols,and phthalic acids more come from the atmosphere through the oxidation reaction of aromatic precursors,especially photochemical ***,in winter,most of the increases in concentrations of organic compounds are attributed to biomass *** analysis of a haze event(14–19 January 2017)during the winter sampling period shows that the increases in the concentration of organic compounds are unaccompanied by strong secondary formation under lower relative humidity(49.1%±13.5%).The main reason for the growth of OA in this haze event is the accumulation of primary OA(POA).The source apportionment by the positive matrix factorization(PMF)model shows that biomass burning(37.1%)is the primary source of OA in the rural regions of the Guanzhong Plain,especially in winter(40.6%).The contribution of secondary formation decreases from 26.0%in summer t

Amplifying the intrinsic wettability of substrate material by changing the solid/liquid contact area is considered to be the main mechanism for controlling the wettability of rough or structured *** theoretical analysis and experimental exploration,we have found that in addition to this wettability structure amplification effect,the surface structure also simultaneously controls surface wettability by regulating the wetting state via changing the threshold Young angles of the Cassie-Baxter and Wenzel wetting *** wetting state regulation effect provides us with an alternative strategy to overcome the inherent limitation in surface chemistry by tailoring surface *** wetting state regulation effect created by multi-scale hierarchical structures is quite significant and plays is a crucial role in promoting the superhydrophobicity,superhydrophilicity and the transition between these two extreme wetting properties,as well as stabilizing the Cassie-Baxter superhydrophobic state on the fabricated lotus-like hierarchically structured Cu surface and the natural lotus leaf.

Background The aim of this study was to review current delivery room(DR)resuscitation intensity in Chinese tertiary neonatal intensive care units and to investigate the association between DR resuscitation intensity and short-term outcomes in preterm infants born at 24+0_31+6 weeks gestation age(GA).Methods This was a retrospective cross-sectional *** source population was infants born at 24+0_31+6 weeks'GA who were enrolled in the Chinese Neonatal Network 2019 *** infants were categorized into five groups:(1)regular care;(2)oxygen supplementation and/or continuous positive airway pressure(O2/CPAP);(3)mask ventilation;(4)endotracheal intubation;and(5)cardiopulmonary resuscitation(CPR).The association between DR resuscitation and shortterm outcomes was evaluated by inverse propensity score-weighted logistic *** Of 7939 infants included in this cohort,2419(30.5%)received regular care,1994(25.1%)received O,/CPAP,1436(18.1%)received mask ventilation,1769(22.3%)received endotracheal intubation,and 321(4.0%)received CPR in the *** maternal age and maternal hypertension correlated with a higher need for resuscitation,and antenatal steroid use tended to be associated with a lower need for resuscitation(P<0.001).Severe brain impairment increased significantly with increasing amounts of resuscitation in DR after adjusting for perinatal *** strategies vary widely between centers,with over 50%of preterm infants in eight centers requiring higher intensity *** Increased intensity of DR interventions was associated with increased mortality and morbidities in very preterm infants in *** is wide variation in resuscitative approaches across delivery centers,and ongoing quality improvement to standardize resuscitation practices is needed.
