Reperfusion after hypoxia-ischemia exacerbates brain injury with compensatory activation of the antiferroptosis system:based on a novel rat model

Hypoxic-ischemic encephalopathy,which predisposes to neonatal death and neurological sequelae,has a high morbidity,but there is still a lack of effective prevention and treatment in clinical practice.To better understand the pathophysiological mechanism underlying hypoxic-ischemic encephalopathy,in this study we compared hypoxic-ischemic reperfusion brain injury and simple hypoxic-ischemic brain injury in neonatal rats.First,based on the conventional RiceVannucci model of hypoxic-ischemic encephalopathy,we established a rat model of hypoxic-ischemic reperfusion brain injury by creating a common carotid artery muscle bridge.Then we performed tandem mass tag-based proteomic analysis to identify differentially expressed proteins between the hypoxic-ischemic reperfusion brain injury model and the conventional Rice-Vannucci model and found that the majority were mitochondrial proteins.We also performed transmission electron microscopy and found typical characteristics of ferroptosis,including mitochondrial shrinkage,ruptured mitochondrial membranes,and reduced or absent mitochondrial cristae.Further,both rat models showed high levels of glial fibrillary acidic protein and low levels of myelin basic protein,which are biological indicators of hypoxic-ischemic brain injury and indicate similar degrees of damage.Finally,we found that ferroptosis-related Ferritin(Fth1)and glutathione peroxidase 4 were expressed at higher levels in the brain tissue of rats with hypoxic-ischemic reperfusion brain injury than in rats with simple hypoxic-ischemic brain injury.Based on these results,it appears that the rat model of hypoxic-ischemic reperfusion brain injury is more closely related to the pathophysiology of clinical reperfusion.Reperfusion not only aggravates hypoxic-ischemic brain injury but also activates the anti-ferroptosis system.

CgVeA, a light signaling responsive regulator, is involved in regulation of chaetoglobosin A biosynthesis and conidia development in Chaetomium globosum

Cytochalasans,with diverse structures and pharmacological activities,are a class of compounds containing isoindolinone moieties fused to the tricyclic or tetracyclic ring system.Chaetoglobosin A(cheA),mainly produced by Chaetomium globosum,is the most abundant cytochalasan.However,limited understanding of transcriptional regulation of morphological development and cheA biosynthesis in C.globosum has hindered cheA application in agriculture and biomedical field.This study examined the regulatory role of CgVeA gene in C.globosum.CgVeA had significant effect on secondary metabolites production in C.globosum,similar to that reported in other filamentous fungi.Inactivation of CgVeA caused an obvious decrease in cheA production from 51.32 to 19.76 mg/L under dark conditions.In contrast,CgVeA overexpression resulted in a dramatic increase in cheA production,reaching 206.59 mg/L under light conditions,which was higher than that noted under dark condition.The RT-qPCR results confirmed that CgVeA,as a light responsive regulator,positively regulated cheA biosynthesis by controlling the expression of core genes of the cheA biosynthetic gene cluster and other relevant regulators.Electrophoretic mobility shift assays proved that CgVeA directly regulated LaeA,cheR,and p450,and indirectly regulated PKS.Moreover,CgVeA had a significant effect on the regulation of asexual spores production.When compared with wild-type C.globosum,CgVeA-silenced and CgVeA overexpression mutants presented remarkable differences in sporulation,irrespective of light or dark condition.Besides,CgVeA expression was speculated to negatively regulate spore formation.These findings illustrated the regulatory mechanism of a hypothetical global regulator,CgVeA,in C.globosum,suggesting its potential application in industrial-scale cheA biosynthesis.

Regulating Ni site in NiV LDH for efficient electrocatalytic production of formate and hydrogen by glycerol electrolysis

Energy-saving glycerol electrolysis with lower potential than water spitting endows a promising way for the concurrent production of value-added formate and high-purity hydrogen. However, there is still lack of efficient electrocatalysts at both anode and cathode for glycerol electrolysis. Herein, we report the activation of Ni site in NiV layered double hydroxide(LDH) by electrochemical and N_(2)/H_(2) plasma regulations for boosting the activity of glycerol oxidation reaction(GOR) and hydrogen evolution reaction(HER), respectively. Specifically, boosted GOR performance with a low overpotential(1.23 V at 10 mA·cm^(-2)) and a high Faradic efficiency(94%) is demonstrated by electrochemically regulated NiV LDH(ENiV LDH) with elevated valence state of Ni site. In situ Raman spectrum reveals the generation of Ni(Ⅲ) species by electrochemical regulation, and the highly active Ni(Ⅲ)can be regenerated with the process of electrochemical oxidation. Additionally, the possible reaction pathway is speculated based on the in situ Fourier transform infrared spectroscopy(FTIR) and high-performance liquid chromatography results. The plasma-regulated NiV LDH(PNiV LDH) with lower valence state of Ni site exhibits outstanding HER activity, displaying a low overpotential of 45 m V to deliver 10 mA·cm^(-2).When employing E-NiV LDH and P-NiV LDH as anode and cathode electrocatalyst, respectively, the assembled electrolyzer merely needs 1.25 V to achieve 10 m A·cm^(-2) for simultaneous production of formate and hydrogen, demonstrating remarkable 320 mV of lower potential than water electrolysis.

Mesenchymal stem cells have neuroprotective effects that limit damage to the retina and photoreceptors,and which may be mediated by extracellular vesicles(or exosomes)released by mesenchymal stem cells.To investigate the neuroprotective effect of extracellular vesicles derived from umbilical cord mesenchymal stem cells on glaucoma,we established rat models of chronic ocular hypertension by injecting conjunctival fibroblasts into the anterior chamber to mimic optic nerve injury caused by glaucoma.One week after injury,extracellular vesicles derived from umbilical cord-derived mesenchymal stem cells were injected into the vitreous cavity.We found that extracellular vesicles derived from mesenchymal stem cells substantially reduced retinal damage,increased the number of retinal ganglion cells,and inhibited the activation of caspase-3.These findings suggest that mesenchymal stem cell-derived extracellular vesicles can help alleviate optic nerve injury caused by chronic ocular hypertension,and this effect is achieved by inhibiting cell apoptosis.

Superior low cycle fatigue property from cell structures in additively manufactured 316L stainless steel

We have investigated the low cycle fatigue(LCF)properties and the extent of strengthening in a dense additively manufactured stainless steel containing different volume fractions of cell structures but having all other microstructure characteristics the same.The samples were produced by laser powder bed fusion(L-PBF),and the concentration of cell structures was varied systematically by varying the annealing treatments.Load-controlled fatigue experiments performed on samples with a high fraction of cell structures reveal an up to 23 times increase in fatigue life compared to an essentially cell-free sample of the same grain configuration.Multiscale electron microscopy characterizations reveal that the cell structures serve as the soft barriers to the dislocation propagation and the partials are the main carrier for cyclic loading.The cell structures,stabilized by the segregated atoms and misorientation between the adjacent cells,are retained during the entire plastic deformation,hence,can continuously interact with dislocations,promote the formation of nanotwins,and provide massive 3D network obstacles to the dislocation motion.The compositional micro-segregation caused by the cellular solidification features serves as another nonnegligible strengthening mechanism to dislocation motion.Specifically,the cell structures with a high density of dislocation debris also appear to act as dislocation nucleation sites,very much like coherent twin boundaries.This work indicates the potential of additive manufacturing to design energy absorbent alloys with high performance by tailoring the microstructure through the printing process.

DEAR EDITOR,The Tibetan macaque(Macaca thibetana)is an endemic species in China belonging to the sinica group in the genus Macaca.Within the genus,it is the largest in body size and has a wide geographic distribution,with a population size second only to rhesus macaques(M.mulatta).Here,using PacBio long-read sequencing and Hi-C technology,we assembled the first chromosome-level genome of the Tibetan macaque,with a size of 2.82 Gb,contig N50 of 48.75 Mb,and scaffold N50 of 150.62 Mb,anchored to 22 chromosomes.

Real-time polymerase chain reaction for the diagnosis of necrotizing herpes stromal keratitis

AIM： To design, optimize and validate a rapid,internally controlled real-time polymerase chain reaction（RT-PCR） test for herpes simplex virus（HSV） in the diagnosis of necrotizing herpes stromal keratitis.· METHODS： Tears alone or together with corneal epithelium scrapings from 30 patients（30 eyes）suspected of necrotizing herpes stromal keratitis were tested for HSV DNA by RT-PCR. The samples were collected during the first visit and then on the subsequent 7, 14, 28, 42, and 56 d. The symptoms of the patients were scored before treatment to determine the correlation between HSV concentration in the corneal epithelium scrapings and clinical scores.·RESULTS： The positive rate（46.4%） in the corneal epithelium group before the therapy was significantly higher than that（13.3%） in the tears group（P =0.006）.There were 13 positive HSV patients before the therapy,the concentration of HSV DNA in corneal epithelium scrapings group was significantly higher than that in the tears group（paired t-test, P =0.0397）. Multilevel mixedeffects model analysis showed that the difference between the corneal epithelium scrapings group and the tears group was statistically significant（P =0.0049）. The Spearman rank correlation analysis indicated a positive correlation between the HSV concentration in the corneal epithelium scrapings and clinical scores before the treatment（r =0.844, P〈 0.0001）.· CONCLUSION： RT-PCR appears to be a powerful molecular tool for the diagnosis of necrotizing herpes stromal keratitis.

Effects of Bushen Yiqi Huoxue Decoction in Treatment of Patients with Diminished Ovarian Reserve:A Randomized Controlled Trial

Objective:To explore the therapeutic effect of Bushen Yiqi Huoxue Decoction BYHD)in patients with diminished ovarian reserve(DOR).Methods:A total of 180 patients with DOR diagnosed from December2013 to December 2014 were equally assigned into progynova and duphaston(E+D)group,Zuogui Pill group and BYHD group with 60 cases in each by computerized randomization.Patients received E+D,Zuogui Pill or BYHD for 12 months,respectively.Follicle stimulating hormone(FSH),luteinizing hormone(LH),estradiol(E_(2)),anti-Mullerian hormone(AMH),antral follicle count(AFC),ovarian volume,endometrial thickness,and the resistance indices(Rls)of ovarian arteries and uterine arteries were observed before and after treatment.Results:Nine women(4 from the E+D group,3 from the Zuogui Pill group,and 2 from the BYHD group)withdrew from the study.After 6 months,Zuogui Pill and BYHD significantly decreased FSH and LH and increased endometrial thickness and AMH(all P<0.01),BYHD also resulted in E2 elevation(P<0.05),ovary enlargement(P<0.05),AFC increase(P<0.01),and RI of ovarian arteries decrease(P<0.05).After 12 months,further improvements were observed in the Zuogui Pill and BYHD groups(all P<0.01),but BYHD showed better outcomes,with lower FSH,larger ovaries and a thicker endometrium compared with the Zuogui Pill group(all P<0.01).However,E+D only significantly increased endometrial thickness(P<0.01)and no significant improvements were observed in the RI of uterine arteries in the three groups.Conclusions:BYHD had a favorable therapeutic effect in patients with DOR by rebalancing hormone levels,promoting ovulation,and repairing the thin endometrium.The combination of tonifying Shen(Kidney),benefiting qi and activating blood circulation may be a promising therapeutic strategy for DOR.

Original article——Surgical treatment of IIIb-T4 lung cancer invading left atrium and great vessels

Background IIIb-T4 non-small cell lung cancer （NSCLC） is commonly considered a contraindication to surgery, although chemo-radiotherapy also achieves a poor survival rate. We reviewed our experience with T4 NSCLC patients who underwent surgery to explore the indications and prognostic factors of surgical treatment of lung cancer invading the left atrium and great vessels. Methods We investigated a cohort of 105 patients, 79 men and 26 women, who underwent surgery from May 1996 to July 2008. Their pathological staging was T4No-2M0. The median age was 59 years, ranging from 36 to 75 years. Patients were grouped based on invading sites： tumors invading the left atrium （LA group）, tumors invading the superior vena cava （SVC group）, and tumors invading the intrapericardial pulmonary artery （PA group）. Patients were further characterized based upon the type of operation, complete resection and incomplete resection groups, and on the lymph node pathological status, No, N1 and N2 groups. We calculated the overall five-year survival rate. Results All patients received resection of primary lesions, with partial resection of the left atrium in the LA group （n=-25）, angioplasty of superior vena cava in the SVC group （n=-23） and intrapericardial ligation of the pulmonary artery in the PA group （n=57）. Complete resection was possible in 77 patients （73.3%）. The overall survival rate of the 105 patients was 41.0% at 5 years; 36.0% for the LA group, 34.8% for the SVC group and 45.6% for the PA group. Pathological N status significantly influenced the overall 5-year survival rate; 61.5% for No, 51.1% for the N1 and 11.8% for the N2 groups （N2 group versus No group, P 〈0.0001, N2 versus N1 group, P〈0.0001）. Surgical resection also influenced survival; 49.4% for the complete resection group and 17.9% for the incomplete resection group （P 〈0.0001）. Evaluation of the learning curve of laser peripheral iridectomy:the 20^(th)case reaches the turning point

AIM:To explore the learning curve for ophthalmologists at the start of laser peripheral iridectomy(LPI)training.METHODS:The learning curve of 4 doctor groups without previous LPI experience was studied.Three main parameters of LPI were reviewed:total energy,argon energy and neodymium-doped yttrium aluminum garnet(Nd:YAG)energy.Procedures were evaluated in cohorts of 20 cases to identify the turning points of the three variables.RESULTS:There was no significant difference in terms of age or eye among the 4 doctor groups.There were stable trends on the learning curve for the Doctor A and C groups regarding total energy and argon energy.In addition,the turning points on the learning curve were determined after the 20th procedure for the Doctor B and D groups regarding total energy and argon energy.Moreover,the Nd:YAG energy was relatively stable since the first procedure.CONCLUSION:It requires approximately 20 procedures for a beginner to reach a turning point on the learning curve regarding LPI.It can serve as a point of reference or guideline for training beginners to perform LPI.
