＜正＞The development of dendrites involves several discrete morphological steps,which depends on the regulation of the *** RhoGTPases,including RhoA,Rac1 and Cdc4.,function as critical regulators of the actin cytoskeleton to play important roles in dendritic and spine morphology ***-activation of RhoA/Rock signaling causes a defect in dendritic outgrowth and branching,while down-regulation of RhoA/Rock signaling shows a more complex dendritic tree compared with ***,regulation of RhoA activity,especially down regulation of it,is critical for dendritic morphology during neuronal *** the present study,we have screened expression of 19 RhoA relative GTPase-activating proteins（RhoAGAPs）,negative regulators of RhoA,in the cortical *** are MPRIP,QLC1,DLC2,DLC3,P1190A,P190B,GRAF,GRAF2,Myosin9a,Myosin9b, RICS,ARAP1,ARAP2,P115 RhoGAP,srGAP1,GMIP,PARG1 and P50 *** loss-of-function analysis, we identify two of them,which highly expressed in neurons,regulate dendritic morphology *** of either of them decreases dendritic length and number both in cultured rat cortical neurons in vitro and developing cortical neurons in *** of RhoA/ROCK signaling restores the defects of dendritic morphology induced by knockdown of ***,these data suggest that some but not all of RhoAGAPs could modulate dendritic morphogenesis of rat cortical neurons via repression of RhoA/Rock signaling,providing novel insight into the physiological functions of RhoAGAPs in the dendritic development.

Red light at intensities above 10lx alters sleep–wake behavior in mice

Sleep is regulated by two mechanisms:the homeostatic process and the circadian *** affects sleep and alertness by entraining the circadian clock,and acutely inducing sleep/alertness,in a manner mediated by intrinsically photosensitive retinal ganglion *** intrinsically photosensitive retinal ganglion cells are believed to be minimally sensitive to red light,which is widely used for illumination to reduce the photic disturbance to nocturnal animals during the dark ***,the appropriate intensity of the red light is *** the present study,we recorded electroencephalograms and electromyograms of freely moving mice to investigate the effects of red light emitted by light-emitting diodes at different intensities and for different durations on the sleep–wake behavior of *** light was used as a ***,red light exerted potent sleep-inducing effects and changed the sleep architecture in terms of the duration and number of sleep episodes,the stage transition,and the EEG power density when the intensity was 4.0 ***,we lowered the light intensity and demonstrated that red light at or below 10 lx did not affect sleep–wake *** light markedly induced sleep and dis{4.pted sleep architecture even at an intensity as low as 10 *** findings highlight the importance of limiting the intensity of red light(⩽10 lx)to avoid optical influence in nocturnal behavioral experiments,particularly in the field of sleep and circadian research.

The global trend toward aging populations has resulted in an increase in the occurrence of Alzheimer's disease(AD)and associated socioeconomic *** metabolism of amyloid-β(Aβ)has been proposed as a significant pathomechanism in AD,supported by results of recent clinical trials using anti-Aβ***,the cognitive benefits of the current treatments are *** etiology of AD is multifactorial,encompassing Aβand tau accumulation,neuroinflammation,demyelination,vascular dysfunction,and comorbidities,which collectively lead to widespread neurodegeneration in the brain and cognitive ***,solely removing Aβfrom the brain may be insufficient to combat neurodegeneration and preserve *** attain effective treatment for AD,it is necessary to(1)conduct extensive {4. on various mechanisms that cause neurodegeneration,including advances in neuroimaging techniques for earlier detection and a more precise characterization of molecular events at scales ranging from cellular to the full system level;(2)identify neuroprotective intervention targets against different neurodegeneration mechanisms;and(3)discover novel and optimal combinations of neuroprotective intervention strategies to maintain cognitive function in AD *** Alzheimer's Disease Neuroprotection {4. Initiative's objective is to facilitate coordinated,multidisciplinary efforts to develop systemic neuroprotective strategies to combat *** aim is to achieve mitigation of the full spectrum of pathological processes underlying AD,with the goal of halting or even reversing cognitive decline.
