Machine learning-based models for advanced fibrosis in nonalcoholic steatohepatitis patients:A cohort study

BACKGROUND The global prevalence of non-alcoholic steatohepatitis(NASH)and its associated risk of adverse outcomes,particularly in patients with advanced liver fibrosis,underscores the importance of early and accurate *** To develop a machine learning-based diagnostic model for advanced liver fibrosis in NASH *** A total of 749 patients who underwent liver biopsy at Beijing Ditan Hospital,Capital Medical University,between January 2010 and January 2020 were *** were randomly divided into training(n=522)and validation(n=224)*** machine learning models were applied to predict advanced liver fibrosis,with feature selection based on Shapley Additive Explanations(SHAP).The diagnostic performance of these models was compared to traditional scores such as the aspartate aminotransferase to platelet ratio index(APRI)and fibrosis index based on the 4 factors(FIB-4),using metrics including the area under the receiver operating characteristic curve(AUROC),decision curve analysis(DCA),and calibration *** The Extreme Gradient Boosting(XGBoost)model outperformed all other machine learning models,achieving an AUROC of 0.934(95%CI:0.914-0.955)in the training cohort and 0.917(95%CI:0.880-0.953)in the validation cohort(P<0.001).Incorporating liver stiffness measurement into the model further improved its performance,with an AUROC of 0.977(95%CI:0.966-0.980)in the training cohort and 0.970(95%CI:0.950-0.990)in the validation cohort,significantly surpassing APRI and FIB-4 scores(P<0.001).The XGBoost model also demonstrated superior clinical utility,as evidenced by DCA and calibration curve analysis in both *** The XGBoost model provides a highly accurate,non-invasive diagnosis of advanced liver fibrosis in NASH patients,outperforming traditional *** online tool based on this model has been developed to assist clinicians in evaluating the risk of advanced liver fibrosis.

Poly（ethylene oxide）-based polymer all-solid-state lithium metal batteries（ASSLBs） have received widespread attention due to their low cost,good process ability,and high energy ***,the growth of Li dendrites within polymer solid-state electrolytes damages the reversibility of Li anodes and still impedes their widespread *** efficient strategy is to construct a superior solid electrolyte ***,a stable interface enriched with Li3N and LiF is in-situ formed between Li anode and a terna ry salt *** terna ry salt electrolyte is innovatively designed by introducing lithium bis（trifluoromethane sulfonyl）imide（LiTFSI）,lithium bis（fluorosulfonyl）imide（LiFSI）,and LiNO3to poly（ethylene oxide） *** characterization indicates that LiNO3and LiFSI contribute to forming a Li3N-LiF-enriched interface and meanwhile LiTFSI ensures excellent ***,among various Li compound components,Li3N has high ionic conductivity,which is beneficial for reducing overpotential,while LiF has high interfacial energy which can enhance nucleation energy and suppress the formation of Li *** experimental results show that ASSLBs coupled with LiFePO4cathode display extremely excellent cycle stability approximately 2200 cycles at 2 C,with a final and corresponding discharge specific capacity of 96.7 mA h g-1. Additionally,a schematic illustration of the working mechanism for the Li3N-LiF interface is proposed.

The isospin asymmetry and quadrupole deformation value of drip-line nuclei are investigated using the Weizsäcker-Skyrme nuclear mass *** observe that for heavy nuclei at the neutron drip line,the Coulomb energy heightened by an aug-mented charge could not be mitigated completely by symmetry energy because of isospin asymmetry saturation but is resisted complementally by strong nuclear *** positions of saltation for the difference in proton numbers between two neighboring nuclei at the neutron drip line,and the isospin asymmetry of the neutron drip-line nucleus as a function of the neutron number distinctly correspond to the known magic numbers,which can serve as a reference to verify the undeter-mined neutron magic *** fitting of the binding energy difference between mirror nuclei(BEDbMN),a set of Coulomb energy coefficients with greater accuracy is obtained.A high-precision description of the BEDbMN is useful for accurately determining the experimentally unknown mass of the nucleus close to the proton drip line if the mass of its mirror nucleus is measured experimentally.

All-perovskite tandem solar cells have the potential to surpass the theoretical efficiency limit of single junction solar cells by reducing thermalization ***,the challenges encompass the oxidation of Sn^(2+)to Sn^(4+)and uncontrolled crystallization kinetics in Sn-Pb perovskites,leading to nonradiative recombination and compositional heterogeneity to decrease photovoltaic efficiency and operational ***,we introduced an ionic liquid additive,1-ethyl-3-methylimidazolium iodide (EMIMI) into Sn-Pb perovskite precursor to form low-dimensional Sn-rich/pure-Sn perovskites at grain boundaries,which mitigates oxidation of Sn^(2+)to Sn^(4+)and regulates the film-forming dynamics of Sn/Pb-based perovskite *** optimized single-junction Sn-Pb perovskite devices incorporating EMIMI achieved a high efficiency of 22.87%.Furthermore,combined with wide-bandgap perovskite sub-cells in tandem device,we demonstrate 2-terminal all-perovskite tandem solar cells with a power conversion efficiency of 28.34%,achieving improved operational stability.

VOCs(Volatile organic compounds)exert a vital role in ozone and secondary organic aerosol production,necessitating investigations into their concentration,chemical characteristics,and source apportionment for the effective implementation of measures aimed at preventing and controlling atmospheric *** to October 2020,onlinemonitoringwas conducted in the main urban area of Shijiazhuang to collect data on VOCs and analyze their concentrations and ***,the PMF(positive matrix factorization)method was utilized to identify the VOCs *** indicated that the TVOCs(total VOCs)concentration was(96.7±63.4μg/m^3),with alkanes exhibiting the highest concentration of(36.1±26.4μg/m^3),followed by OVOCs(16.4±14.4μg/m^3).The key active components were alkenes and aromatics,among which xylene,propylene,toluene,propionaldehyde,acetaldehyde,ethylene,and styrene played crucial roles as reactive *** sources derived from PMF analysis encompassed vehicle emissions,solvent and coating sources,combustion sources,industrial emissions sources,as well as plant sources,the contribution of which were 37.80%,27.93%,16.57%,15.24%,and 2.46%,***,reducing vehicular exhaust emissions and encouraging neighboring industries to adopt low-volatile organic solvents and coatings should be prioritized to mitigate VOCs levels.

A Mn-porphyrinic metal–organic framework immobilizing glucose oxidase for combined photodynamic/chemodynamic/starvation therapy

The combination of photodynamic therapy（PDT） and chemodynamic therapy（CDT） in tumor treatment has attracted considerable ***,tumor hypoxia and glutathione（GSH） overproduction in the tumor tissue restricted the progress of their ***,a Mn-porphyrinic metal-organic framework（Mn-TCPP） was constructed by the one-pot method and further used for immobilizing glucose oxidase（GOx） to obtain GOx@*** would rapidly exhaust endogenous glucose into hydrogen peroxide（H2O2） and gluconic acid,thus shutting off the energy supply of tumor cells for starvation ***-TCPP catalyzed H2O2to produce oxygen,regulating the hypoxic tumor microenvironment and in turn improving1O2generation under laser ***,Mn-TCPP can reduce reactive oxygen species consumption owing to the redox reaction between Mn3+and GSH,thus greatly enhancing ***,benefiting from GOx-mediated starvation therapy,Mn2+produced by Mn3+reduction can react with sufficient intracellular H2O2to generate ·OH with high cytotoxicity through a Fenton-like *** treatment by GOx@Mn-TCPP under laser irradiation in vitro,the cell viability of 4T1 and A549 tumor cells reached to 20%,reflecting GOx@Mn-TCPP could give full play to the advantages of PDT/CDT/starvation *** results in vivo demonstrated that GOx@Mn-TCPP mediated synergistic cascade therapy could significantly inhibit tumor growth and improve the therapeutic effect.

With the advancement of artificial intelligence,optic in-sensing reservoir computing based on emerging semiconductor devices is high desirable for real-time analog signal ***,we disclose a flexible optomemristor based on C_(27)H_(30)O_(15)/FeOx heterostructure that presents a highly sensitive to the light stimuli and artificial optic synaptic features such as short-and long-term plasticity(STP and LTP),enabling the developed optomemristor to implement complex analogy signal processing through building a real-physical dynamic-based in-sensing reservoir computing algorithm and yielding an accuracy of 94.88%for speech *** charge trapping and detrapping mediated by the optic active layer of C_(27)H_(30)O_(15) that is extracted from the lotus flower is response for the positive photoconductance memory in the prepared *** work provides a feasible organic−inorganic heterostructure as well as an optic in-sensing vision computing for an advanced optic computing system in future complex signal processing.

健康孕妇和子痫前期孕妇黄斑中心凹下不同脉络膜分层厚度变化的对比分析

目的观察正常女性、健康孕妇和子痫前期孕妇的黄斑中心凹下不同脉络膜分层厚度的变化和差异。方法经我院妇产科检查确诊的子痫前期患者22例22眼(子痫前期孕妇组)、孕20周以上的健康孕妇22例22眼(健康孕妇组)以及正常女性22例22眼(正常女性组)纳入研究。所有患者均进行最佳矫正视力(best corrected visual acuity,BCVA)、眼压、裂隙灯检查及光学相干断层扫描(optical coherence tomography,OCT)检查。采用OCT测量受检者黄斑中心凹下脉络膜厚度(subfoveal choroidal thickness,SFCT)、脉络膜大血管层厚度(large choroidal vessel thickness,LCVT)、脉络膜毛细血管层/中血管层厚度(medium choroidal vessel thickness,MCVT),并比较三组之间的差异。结果三组间年龄、等效球镜度数比较差异均无统计学意义(均为P>0.05);健康孕妇组与子痫前期孕妇组的孕周比较差异亦无统计学意义(P>0.05)。正常女性组、健康孕妇组、子痫前期孕妇组的SFCT分别为(263.45±69.66)μm、(310.55±55.63)μm、(350.41±54.32)μm;LCVT分别为(188.25±60.88)μm、(213.27±65.31)μm、(264.77±56.21)μm;MCVT分别为(75.20±33.65)μm、(97.27±22.14)μm、(85.64±20.93)μm。三组受检眼SFCT、LCVT、MCVT比较差异均有统计学意义(均为P<0.05)。健康孕妇组SFCT、MCVT较正常女性组厚,差异均统计学意义(均为P<0.05),而两组LCVT比较,差异无统计学意义(P=0.199)。子痫前期孕妇组SFCT、LCVT较健康孕妇组厚,差异均有统计学意义(均为P<0.05),而两组MCVT比较,差异无统计学意义(P=0.070)。结论子痫前期孕妇较健康孕妇SFCT、LCVT厚度增加,MCVT无明显差异。

Tanreqing Injection Inhibits Activation of NLRP3 Inflammasome in Macrophages Infected with Influenza A Virus by Promoting Mitophagy

Objective: To investigate the inhibitory effect of Tanreqing Injection(TRQ) on the activation of nucleotide-binding oligomerization domain-like receptor pyrin domain containing 3(NLRP3) inflammasome in macrophages infected with influenza A virus and the underlying mechanism based on mitophagy pathway. Methods:The inflammatory model of murine macrophage J774A.1 induced by influenza A virus [strain A/Puerto Rico/8/1934(H1N1), PR8] was constructed and treated by TRQ, while the mitochondria-targeted antioxidant Mito-TEMPO and autophagy specific inhibitor 3-methyladenine(3-MA) were used as controls to intensively study the anti-inflammatory mechanism of TRQ based on mitophagy-mitochondrial reactive oxygen species(mt ROS)-NLRP3 inflammasome pathway. The levels of NLRP3, Caspase-1 p20, microtubule-associated protein 1 light chain 3 Ⅱ(LC3Ⅱ) and P62 proteins were measured by Western blot. The release of interleukin-1β(IL-1β) was tested by enzyme linked immunosorbent assay, the mt ROS level was detected by flow cytometry, and the immunofluorescence and co-localization of LC3 and mitochondria were observed under confocal laser scanning microscopy. Results: Similar to the effect of Mito-TEMPO and contrary to the results of 3-MA treatment, TRQ could significantly reduce the expressions of NLRP3, Caspase-1 p20, and autophagy adaptor P62, promote the expression of autophagy marker LC3Ⅱ, enhance the mitochondrial fluorescence intensity, and inhibit the release of mt ROS and IL-1β(all P<0.01).Moreover, LC3 was co-localized with mitochondria, confirming the type of mitophagy. Conclusion: TRQ could reduce the level of mt ROS by promoting mitophagy in macrophages infected with influenza A virus, thus inhibiting the activation of NLRP3 inflammasome and the release of IL-1β, and attenuating the inflammatory response.
